The problem of desiccation cracks in soils has received increasing attention in the last few years, in both experimental investigations and modeling. Experimental research has been mainly focused on the behavior of slurries subjected to drying in plates of different shapes, sizes and thickness. The main objectives of these studies were to learn about the process of crack formation under controlled environmental conditions, and also to better understand the effect of different factors (e.g. soil type, boundary conditions, soil thickness) on the morphology of the crack network. As for the numerical modeling, different approaches have been suggested lately to describe the behavior of drying cracks in soils. One aspect that it is still difficult to describe properly is the crack pattern observed in desiccated soils. This work presents a novel technique to model the behavior of drying soils. The crack patter observed in desiccation tests on circular plates are simulated with the main objective of predicting the effect of soil thickness on crack pattern. Good agreement between experimental results and model prediction are observed.

Investigating tree's spatial patterns according to their size classes and according to their more abundant species can provide evidences about the structure of the vegetal community, since the spatial pattern is a key question for forestry ecology studies. The tree spatial organization patterns on the environment depend on several ecological processes and on the specific characteristics of each environment, so that the best understanding of this frame provides important elements for the knowledge on forestry formation. This paper aimed to study tree spatial patterns, according to the diameter classes and from four most abundant species in different forests, in order to provide evidences regarding to the ecology of each vegetal community. The spatial pattern description in each forestry formation was developed using Ripley's K function. The studied forestry formations were: Ombrophilous Forest, Cerradao, Seasonal Forest and Restinga Forest. In this work, a 10.24 ha plot was installed in each forestry formation, and every tree, with a circumference at breast height (CBH) larger than 15 cm were measured, georeferenced and identified. The obtained data highlights the aggregated character in tropical forests, as observed in every studied forest. The 'Cerraddo' and 'Restinga' forest trees showed close aggregate patterns. In the Ombrophilous forest, for all distance scales, the aggregate pattern was meaningful. In the seasonal forest, a random tendency was observed, although a meaningful aggregation was observed in all short distances. The spatial pattern by diameter classes was generally aggregated for trees smaller than 10 cm of diameter and between 10 and 20 cm and random for the others, proving the existence of a tendency which in young trees is more aggregated than in old ones. The spatial pattern of the dominant species is always strongly similar to the general pattern of each forestry formation. The differences between the spatial patterns of two or three coincident species, among the forestry formations, indicate that its pattern is influenced by each different environment. This stands out the importance of its self-ecology and of its ecological processes, intrinsic of each community that can explain the observed patterns.

In response to pathogen recognition by Toll-like receptors (TLRs) on their cell surface, macrophages release lipid mediators and cytokines that are widely distributed throughout the body and play essential roles in host responses. Granulocyte macrophage colony-stimulating factor (GM-CSF) is important for the immune response during infections to improve the clearance of microorganisms. In this study, we examined the release of mediators in response to TLR2 ligands by bone marrow-derived macrophages (BMDMs) primed with GM-CSF. We demonstrated that when stimulated with TLR2 ligands, non-primed BMDMs preferentially produced PGE(2) in greater amounts than LTB4. However, GM-CSF priming shifted the release of lipid mediators by BMDMs, resulting in a significant decrease of PGE(2) production in response to the same stimuli. The decrease of PGE(2) production from primed BMDMs was accompanied by a decrease in PGE-synthase mRNA expression and an increase in TNF-alpha and nitric oxide (NO) production. Moreover, some GM-CSF effects were potentiated by the addition of IFN-gamma. Using a variety of TLR2 ligands, we established that PGE(2) release by GM-CSF-primed BMDMs was dependent on TLR2 co-receptors (TLR1, TLR6), CD14, MyD88 and the nuclear translocation of NF kappa B but was not dependent on peroxisome proliferator-activated receptor-gamma (PPAR-gamma) activation. Indeed, GM-CSF priming enhanced TLR2, TLR4 and MyD88 mRNA expression and phospho-I kappa B alpha formation. These findings demonstrate that GM-CSF drives BMDMs to present a profile relevant to the host during infections.

Objective: To describe the healing of marginal defects below or above 1 mm of dimension around submerged implants in a dog model. Material and methods: In 12 Labrador dogs, all mandibular premolars and first molars were extracted bilaterally. After 3 months of healing, full-thickness flaps were elevated in the edentulous region of the right side of the mandible. Two recipient sites were prepared and the marginal 5mm were widened to such an extent to obtain, after implant installation, a marginal gap of 0.5mm at the mesial site (small defect) and of 1.25mm at the distal site (large defect). Titanium healing caps were affixed to the implants and the flaps were sutured allowing a fully submerged healing. The experimental procedures were subsequently performed in the left side of the mandible. The timing of the experiments and sacrifices were planned in such a way to obtain biopsies representing the healing after 5, 10, 20 and 30 days. Ground sections were prepared and histomorphometrically analyzed. Results: The filling of the defect with newly formed bone was incomplete after 1 month of healing in all specimens. Bone formation occurred from the base and the lateral walls of the defects. A larger volume of new bone was formed in the large compared with the small defects. Most of the new bone at the large defect was formed between the 10- and the 20-day period of healing. After 1 month of healing, the outline of the newly formed bone was, however, located at a similar distance from the implant surface (about 0.4mm) at both defect types. Only minor newly formed bone in contact with the implant, starting from the base of the defects, was seen at the large defects (about 0.8mm) while a larger amount was detected at the small defects (about 2.2 mm). Conclusion: Marginal defects around titanium implants appeared to regenerate in 20-30 days by means of a distance osteogenesis. The bone fill of the defects was, however, incomplete after 1 month.

The Dipteran a native Brazilian insect that has become a valuable model system for developmental biology research because it provides an interesting opportunity to study a different type of insect oogenesis. Sequences from a cDNA library that was constructed with poly A + RNA from the ovaries of larvae at different ages were analyzed. Molecular characterization confirmed interesting findings, such as the presence of . The gene encodes a conserved RNA-binding protein that is required during early development for the maintenance and division of the primordial germ cells of Diptera. plays an important role in specifying the posterior regions of insect embryos and is important for abdomen formation. In the present work, we showed the spatial and temporal expression profiles of this important gene, which is involved in oogenesis and early development. Data mining techniques were used to obtain the complete sequence of . Bioinformatic tools were used to determine the following: (1) the secondary structure of the 3'-untranslated region of the mRNA, (2) the encoded protein of the isolated gene, (3) the conserved zinc-finger domains of the Nanos protein, and (4) phylogenetic analyses. Furthermore, RNA in situ hybridization and immunolocalization were used to determine mRNA and protein expression in the tissues that were studied and to define as a germ cell molecular marker.

The weakening mechanisms involved in the collapse of complex impact craters are controversial. The Araguainha impact crater, in Brazil, exposes a complex structure of 40 km in diameter, and is an excellent object to address this issue. Its core is dominated by granite. In addition to microstructural observations, magnetic studies reveal its internal fabric acquired during the collapse phase. All granite samples exhibit impact-related planar deformation features (PDFs) and planar fractures (PFs), which were overprinted by cataclasis. Cataclastic deformation has evolved from incipient brittle fracturing to the development of discrete shear bands in the center of the structure. Fracture planes are systematically decorated by tiny grains (<10 mu m) of magnetite and hematite, and the orientation of magnetic lineation and magnetic foliation obtained by the anisotropies of magnetic susceptibility (AMS) and anhysteretic remanence (AAR) are perfectly coaxial in all studied sites. Therefore, we could track the orientation of deformation features which are decorated by iron oxides using the AMS and AAR. The magnetic fabrics show a regular pattern at the borders of the central peak, with orientations consistent with the fabric of sediments at the crater's inner collar and complex in the center of the structure. Both the cataclastic flow revealed from microstructural observations and the structural pattern of the magnetic anisotropy match the predictions from numerical models of complex impact structures. The widespread occurrence of cataclasis in the central peak, and its orientations revealed by magnetic studies indicate that acoustic fluidization likely operates at all scales, including the mineral scales. The cataclastic flow made possible by acoustic fluidization results in an apparent plastic deformation at the macroscopic scale in the core. (C) 2012 Elsevier B.V. All rights reserved.

This study investigates two lanthanide compounds (La(3+) and Sm(3+)) obtained in water/ethyl alcohol solutions employing the anionic surfactant diphenyl-4-amine sulfonate (DAS) as ligand. Both sulfonates were characterized through IR, TG/DTG (O(2) and N(2)). The thermal treatment of both compounds at 1273 K under air leaves residues containing variable percentages of lanthanide oxysulfide/oxysulfate phases shown by synchrotron high-resolution XRD pattern including the Rietveld analysis. The phase distributions found in the residues evidence the differences in the relative stability of the precursors.

The present star formation rate (SFR) in the inner Galaxy is puzzling for the chemical evolution models (CEM). No static CEM is able to reproduce the peak of the SFR in the 4 kpc ring. The main reason is probably a shortage of gas, which could be due to the dynamical effects produced by the galactic bar, not considered by these models. We developed a CEM that includes radial gas flows in order to mimic the effects of the galactic bar in the first 5 kpc of the galactic disk. In this model, the star formation (SF) is a two-step process: first, the diffuse gas forms molecular clouds. Then, stars form from cloud-cloud collisions or by the interaction between massive stars and the molecular gas. The former is called spontaneous and the latter induced SF. The mass in the different phases of each region changes by the processes associated with the stellar formation and death by: the SF due to spontaneous fragmentation of gas in the halo; formation of gas clouds in the disk from the diffuse gas; induced SF in the disk due to the interaction between massive stars and gas clouds; and finally, the restitution of the diffuse gas associated to these process of cloud and star formation. In the halo, the star formation rate for the diffuse gas follows a Schmidt law with a power n = 1.5. In the disk, the stars form in two steps: first, molecular clouds are formed from the diffuse gas also following a Schmidt law with n=1.5 and a proportionality factor. Including a specific pattern of radial gas flows, the CEM is able to reproduce with success the peak in the SFR at 4 kpc (fig. 1).

The barred spiral galaxy M83 (NGC5236) has been observed in the 12CO J=1–0 and J=2–1 millimetre lines with the Swedish-ESO Submillimetre Telescope (SEST). The sizes of the CO maps are 100×100, and they cover the entire optical disk. The CO emission is strongly peaked toward the nucleus. The molecular spiral arms are clearly resolved and can be traced for about 360º. The total molecular gas mass is comparable to the total Hi mass, but H2 dominates in the optical disk. Iso-velocity maps show the signature of an inclined, rotating disk, but also the effects of streaming motions along the spiral arms. The dynamical mass is determined and compared to the gas mass. The pattern speed is determined from the residual velocity pattern, and the locations of various resonances are discussed. The molecular gas velocity dispersion is determined, and a trend of decreasing dispersion with increasing galactocentric radius is found. A total gas (H2+Hi+He) mass surface density map is presented, and compared to the critical density for star formation of an isothermal gaseous disk. The star formation rate (SFR) in the disk is estimated using data from various star formation tracers. The different SFR estimates agree well when corrections for extinctions, based on the total gas mass map, are made. The radial SFR distribution shows features that can be associated with kinematic resonances. We also find an increased star formation efficiency in the spiral arms. Different Schmidt laws are fitted to the data. The star formation properties of the nuclear region, based on high angular resolution HST data, are also discussed.

Investigations on formation and specification of neural precursor cells in the central nervous system of the Drosophila melanogaster embryoSpecification of a unique cell fate during development of a multicellular organism often is a function of its position. The Drosophila central nervous system (CNS) provides an ideal system to dissect signalling events during development that lead to cell specific patterns. Different cell types in the CNS are formed from a relatively few precursor cells, the neuroblasts (NBs), which delaminate from the neurogenic region of the ectoderm. The delamination occurs in five waves, S1-S5, finally leading to a subepidermal layer consisting of about 30 NBs, each with a unique identity, arranged in a stereotyped spatial pattern in each hemisegment. This information depends on several factors such as the concentrations of various morphogens, cell-cell interactions and long range signals present at the position and time of its birth. The early NBs, delaminating during S1 and S2, form an orthogonal array of four rows (2/3,4,5,6/7) and three columns (medial, intermediate, and lateral) . However, the three column and four row-arrangement pattern is only transitory during early stages of neurogenesis which is obscured by late emerging (S3-S5) neuroblasts (Doe and Goodman, 1985; Goodman and Doe, 1993). Therefore the aim of my study has been to identify novel genes which play a role in the formation or specification of late delaminating NBs.In this study the gene anterior open or yan was picked up in a genetic screen to identity novel and yet unidentified genes in the process of late neuroblast formation and specification. I have shown that the gene yan is responsible for maintaining the cells of the neuroectoderm in an undifferentiated state by interfering with the Notch signalling mechanism. Secondly, I have studied the function and interactions of segment polarity genes within a certain neuroectodermal region, namely the engrailed (en) expressing domain, with regard to the fate specification of a set of late neuroblasts, namely NB 6-4 and NB 7-3. I have dissected the regulatory interaction of the segment polarity genes wingless (wg), hedgehog (hh) and engrailed (en) as they maintain each other’s expression to show that En is a prerequisite for neurogenesis and show that the interplay of the segmentation genes naked (nkd) and gooseberry (gsb), both of which are targets of wingless (wg) activity, leads to differential commitment of NB 7-3 and NB 6-4 cell fate. I have shown that in the absence of either nkd or gsb one NB fate is replaced by the other. However, the temporal sequence of delamination is maintained, suggesting that formation and specification of these two NBs are under independent control.

Die vorliegende Arbeit behandelt die Entwicklung des 570 Ma alten, neoproterozoischen Agardagh - Tes-Chem Ophioliths (ATCO) in Zentralasien. Dieser Ophiolith liegt südwestlich des Baikalsees (50.5° N, 95° E) und wurde im frühen Stadium der Akkretion des Zentralasiatischen Mobilgürtels auf den nordwestlichen Rand des Tuvinisch-Mongolischen Mikrokontinentes aufgeschoben. Bei dem Zentralasiatische Mobilgürtel handelt es sich um einen riesigen Akkretions-Subduktionskomplex, der heute das größte zusammenhängende Orogen der Erde darstellt. Im Rahmen dieser Arbeit wurden eine Reihe plutonischer und vulkanischer Gesteine, sowie verschiedene Mantelgesteine des ATCO mittels mikroanalytischer und geochemischer Verfahren untersucht (Elektronenstrahlmikrosonde, Ionenstrahlmikrosonde, Spurenelement- und Isotopengeochemie). Die Auswertung dieser Daten ermöglichte die Entwicklung eines geodynamisch-petrologischen Modells zur Entstehung des ATCO. Die vulkanischen Gesteine lassen sich aufgrund ihrer Spurenelement- und Isotopenzusammensetzung in inselbogenbezogene und back-arc Becken bezogene Gesteine (IA-Gesteine und BAB-Gesteine) unterscheiden. Darüber hinaus gibt es eine weitere, nicht eindeutig zuzuordnende Gruppe, die hauptsächlich mafische Gänge umfasst. Der grösste Teil der untersuchen Vulkanite gehört zur Gruppe der IA-Gesteine. Es handelt sich um Al-reiche Basalte und basaltische Andesite, welche aus einem evolvierten Stammmagma mit Mg# 0.60, Cr ~ 180 µg/g und Ni ~ 95 µg/g hauptsächlich durch Klinopyroxenfraktionierung entstanden sind. Das Stammmagma selbst entstand durch Fraktionierung von ca. 12 % Olivin und geringen Anteilen von Cr-Spinell aus einer primären, aus dem Mantel abgeleiteten Schmelze. Die IA-Gesteine haben hohe Konzentrationen an inkompatiblen Spurenelementen (leichte-(L)- Seltenerdelement-(SEE)-Konzentrationen etwa 100-fach chondritisch, chondrit-normierte (La/Yb)c von 14.6 - 5.1), negative Nb-Anomalien (Nb/La = 0.37 - 0.62) und niedrige Zr/Nb Verhältnisse (7 - 14) relativ zu den BAB-Gesteinen. Initiale eNd Werte liegen bei etwa +5.5, initiale Bleiisotopenverhältnisse sind: 206Pb/204Pb = 17.39 - 18.45, 207Pb/204Pb = 15.49 - 15.61, 208Pb/204Pb = 37.06 - 38.05. Die Anreicherung lithophiler inkompatibler Spurenelemente (LILE) in dieser Gruppe ist signifikant (Ba/La = 11 - 130) und zeigt den Einfluss subduzierter Komponenten an. Die BAB-Gesteine repräsentieren Schmelzen, die sehr wahrscheinlich aus der gleichen Mantelquelle wie die IA-Gesteine stammen, aber durch höhere Aufschmelzgrade (8 - 15 %) und ohne den Einfluss subduzierter Komponenten entstanden sind. Sie haben niedrigere Konzentrationen an inkompatiblen Spurenelementen, flache SEE-Muster ((La/Yb)c = 0.6 - 2.4) und höhere initiale eNd Werte zwischen +7.8 und +8.5. Nb Anomalien existieren nicht und Zr/Nb Verhältnisse sind hoch (21 - 48). Um die geochemische Entwicklung der vulkanischen Gesteine des ATCO zu erklären, sind mindestens drei Komponenten erforderlich: (1) eine angereicherte, ozeaninselbasalt-ähnliche Komponente mit hoher Nb Konzentration über ~ 30 µg/g, einem niedrigen Zr/Nb Verhältnis (ca. 6.5), einem niedrigen initialen eNd Wert (um 0), aber mit radiogenen 206Pb/204Pb-, 207Pb/204Pb- und 208Pb/204Pb-Verhältnissen; (2) eine N-MORB ähnliche back-arc Becken Komponente mit flachem SEE-Muster und einem hohen initialen eNd Wert von mindestens +8.5, und (3) eine Inselbogen-Komponente aus einer verarmten Mantelquelle, welche durch die abtauchende Platte geochemisch modifiziert wurde. Die geochemische Entstehung der ATCO Vulkanite lässt sich dann am besten durch eine Kombination aus Quellenkontamination, fraktionierte Kristallisation und Magmenmischung erklären. Geodynamisch gesehen entstand der ATCO sehr wahrscheinlich in einem intraozeanischen Inselbogen - back-arc System. Bei den untersuchten Plutoniten handelt es sich um ultramafische Kumulate (Wehrlite und Pyroxenite) sowie um gabbroische Plutonite (Olivin-Gabbros bis Diorite). Die geochemischen Charakteristika der mafischen Plutonite sind deutlich unterschiedlich zu denen der vulkanischen Gesteine, weshalb sie sehr wahrscheinlich ein späteres Entwicklungsstadium des ATCO repräsentieren. Die Spurenelement-Konzentrationen in den Klinopyroxenen der ultramafischen Kumulate sind extrem niedrig, mit etwa 0.1- bis 1-fach chondritischen SEE-Konzentrationen und mit deutlich LSEE-verarmten Mustern ((La/Yb)c = 0.27 - 0.52). Berechnete Gleichgewichtsschmelzen der ultramafischen Kumulate zeigen grosse Ähnlichkeit zu primären boninitischen Schmelzen. Die primären Magmen waren daher boninitischer Zusammensetzung und entstanden in dem durch vorausgegangene Schmelzprozesse stark verarmten Mantelkeil über einer Subduktionszone. Niedrige Spurenelement-Konzentrationen zeigen einen geringen Einfluss der abtauchenden Platte an. Die Spurenelement-Konzentrationen der Gabbros sind ebenfalls niedrig, mit etwa 0.5 - 10-fach chondritischen SEE-Konzentrationen und mit variablen SEE-Mustern ((La/Yb)c = 0.25 - 2.6). Analog zu den Vulkaniten der IA-Gruppe haben alle Gabbros eine negative Nb-Anomalie mit Nb/La = 0.01 - 0.31. Die initialen eNd Werte der Gabbros variieren zwischen +4.8 und +7.1, mit einem Mittelwert von +5.9, und sind damit identisch mit denen der IA-Vulkanite. Bei den untersuchten Mantelgesteinen handelt es sich um teilweise serpentinisierte Dunite und Harzburgite, die alle durch hohe Mg/Si- und niedrige Al/Si-Verhältnisse gekennzeichnet sind. Dies zeigt einen refraktären Charakter an und steht in guter Übereinstimmung mit den hohen Cr-Zahlen (Cr#) der Spinelle (bis zu Cr# = 0.83), auf deren Basis der Aufschmelzgrad der residuellen Mantelgesteine berechnet wurde. Dieser beträgt etwa 25 %. Die geochemische Zusammensetzung und die petrologischen Daten der Ultramafite und Gabbros lassen sich am besten erklären, wenn man für die Entstehung dieser Gesteine einen zweistufigen Prozess annimmt. In einer ersten Stufe entstanden die ultramafischen Kumulate unter hohem Druck in einer Magmenkammer an der Krustenbasis, hauptsächlich durch Klinopyroxen-Fraktionierung. Bei dieser Magmenkammer handelte es sich um ein offenes System, dem von unten laufend neue Schmelze zugeführt wurde, und aus dem im oberen Bereich evolviertere Schmelzen geringerer Dichte entwichen. Diese evolvierten Schmelzen stiegen in flachere krustale Bereiche auf und bildeten dort meist isolierte Intrusionskörper. Diese Intrusionskörper erstarrten ohne Magmen-Nachschub, weshalb petrographisch sehr unterschiedliche Gesteine entstehen konnten. Eine geochemische Modifikation der abkühlenden Schmelzen erfolgte allerdings durch die Assimilation von Nebengestein. Da innerhalb der Gabbros keine signifikante Variation der initalen eNd Werte existiert, handelte es sich bei dem assimilierten Material hauptsächlich um vulkanische Gesteine des ATCO und nicht um ältere, möglicherweise kontinentale Kruste.

The central point of this work is the investigation of neurogenesis in chelicerates and myriapods. By comparing decisive mechanisms in neurogenesis in the four arthropod groups (Chelicerata, Crustacea, Insecta, Myriapoda) I was able to show which of these mechanisms are conserved and which developmental modules have diverged. Thereby two processes of embryonic development of the central nervous system were brought into focus. On the one hand I studied early neurogenesis in the ventral nerve cord of the spiders Cupiennius salei and Achaearanea tepidariorum and the millipede Glomeris marginata and on the other hand the development of the brain in Cupiennius salei.rnWhile the nervous system of insects and crustaceans is formed by the progeny of single neural stem cells (neuroblasts), in chelicerates and myriapods whole groups of cells adopt the neural cell fate and give rise to the ventral nerve cord after their invagination. The detailed comparison of the positions and the number of the neural precursor groups within the neuromeres in chelicerates and myriapods showed that the pattern is almost identical which suggests that the neural precursors groups in these arthropod groups are homologous. This pattern is also very similar to the neuroblast pattern in insects. This raises the question if the mechanisms that confer regional identity to the neural precursors is conserved in arthropods although the mode of neural precursor formation is different. The analysis of the functions and expression patterns of genes which are known to be involved in this mechanism in Drosophila melanogaster showed that neural patterning is highly conserved in arthropods. But I also discovered differences in early neurogenesis which reflect modifications and adaptations in the development of the nervous systems in the different arthropod groups.rnThe embryonic development of the brain in chelicerates which was investigated for the first time in this work shows similarities but also some modifications to insects. In vertebrates and arthropods the adult brain is composed of distinct centres with different functions. Investigating how these centres, which are organised in smaller compartments, develop during embryogenesis was part of this work. By tracing the morphogenetic movements and analysing marker gene expressions I could show the formation of the visual brain centres from the single-layered precheliceral neuroectoderm. The optic ganglia, the mushroom bodies and the arcuate body (central body) are formed by large invaginations in the peripheral precheliceral neuroectoderm. This epithelium itself contains neural precursor groups which are assigned to the respective centres and thereby build the three-dimensional optical centres. The single neural precursor groups are distinguishable during this process leading to the assumption that they carry positional information which might subdivide the individual brain centres into smaller functional compartments.rn

Eine wichtige Voraussetzung für das Verständnis der Spezifizierungsmechanismen unterschiedlicher Zelltypen im embryonalen Gehirn ist die detaillierte Kenntnis des neuroektodermalen Ursprungs seiner neuralen Stammzellen (Neuroblasten, NB), sowie der Morphologie und zellulären Komposition der daraus hervorgehenden Zellstammbäume (ZSBe). In der vorliegenden Arbeit wurde die Entstehung und Topologie von 21 embryonalen ZSBen im anteriorsten Gehirnteil, dem Protocerebrum, charakterisiert, mit besonderem Fokus auf solche ZSBe, die den Pilzkörper konstituieren. Pilzkörper sind prominente, paarige Neuropilzentren, die eine wichtige Rolle bei der Verarbeitung olfaktorischer Informationen, beim Lernen und bei der Gedächtnisbildung spielen. In dieser Arbeit konnte erstmalig die Embryonalentwicklung der Pilzkörper ab dem Zeitpunkt der Entstehung ihrer NBen im procephalen Neuroektoderm (pNE), bis hin zum funktionellen Gehirnzentrum in der frühen Larve auf Ebene individueller ZSBe bzw. einzelner Neurone beschrieben werden. Mittels der klonalen Di-Markierungstechnik konnte ich zeigen, dass die vier NBen der Pilzkörper (PKNBen) jeder Gehirnhemisphäre innerhalb des NE aus dem ventralen Bereich der mitotischen Domäne B (δB) hervorgehen. Ein in diesem Bereich liegendes proneurales Feld beherbergt etwa 10-12 Zellen, die alle das Potential haben sich zu PKNBen zu entwickeln. Des Weiteren zeigen diese Untersuchungen, dass die PKNBen (und weitere NBen der δB) aus benachbarten NE-Zellen hervorgehen. Dieser Befund impliziert, dass der Mechanismus der lateralen Inhibition in diesem Bereich des NE keine Rolle spielt. Weiterhin stellte sich heraus, dass jeder PKNB eine ihm eigene Position im sich entwickelnden Pilzkörperkortex besetzt und eine spezifische Kombination der Transkriptionsfaktoren Dachshund, Eyeless und Retinal homeobox exprimiert. Dadurch konnte jeder der vier PKNBen in den betreffenden frühembryonalen NB-Karten einem der ca. 105 NBen pro Gehirnhemisphäre zugeordnet werden. Die PKNBen bringen individuelle ZSBe hervor, die Pilzkörper-intrinsische γ-Neurone beinhalten, aber auch jeweils charakteristische Sets an Interneuronen, die nicht am Aufbau des Pilzkörpers beteiligt sind. Diese verschiedenen Neuronentypen entstehen in einer zeitlichen Abfolge, die für jeden PKNBen spezifisch ist. Ihre embryonalen ZSBe sind aber nicht nur durch individuelle Sets an frühgeborenen ni-Neuronen charakterisiert, sondern auch durch spezifische Unterschiede in der Anzahl ihrer γ-Neurone, welche jedoch, wie ich zeigen konnte, nicht durch Apoptose reguliert wird. Weiterhin konnte ich zeigen, dass γ-Neurone, in einer PKNB Klon-abhängigen Weise, spezifische Unterschiede in der räumlich-zeitlichen Innervation des Pedunkels, der Calyx und der Loben aufweisen. Im Weiteren wurde die Expression verschiedener molekularer Marker in diesen ZSBen charakterisiert, u.a. die Expression verschiedener Gal4-Fliegenstämme, und solcher Transkriptionsfaktoren, die eine wichtige Rolle bei der temporären Spezifizierung im VNS spielen. So werden hb, Kr, pdm1 auch in Nachkommenzellen der PKNBen exprimiert und haben möglicherweise eine Funktion bei ihrer temporären Spezifizierung. Diese Arbeit gibt auch erstmalig Einblick in die vollständige spätembryonale/frühlarvale Morphologie anderer protocerebraler Gehirnzellstammbäume aus δB und δ1. Die Beschreibungen dieser ZSBe beinhalten Angaben zu deren Zellzahl, Zelltypen, der Lage der ZSBe im Gehirn, axonalen/dendritischen Projektionsmustern sowie dem Entstehungsort des NBen.

This study aims at a comprehensive understanding of the effects of aerosol-cloud interactions and their effects on cloud properties and climate using the chemistry-climate model EMAC. In this study, CCN activation is regarded as the dominant driver in aerosol-cloud feedback loops in warm clouds. The CCN activation is calculated prognostically using two different cloud droplet nucleation parameterizations, the STN and HYB CDN schemes. Both CDN schemes account for size and chemistry effects on the droplet formation based on the same aerosol properties. The calculation of the solute effect (hygroscopicity) is the main difference between the CDN schemes. The kappa-method is for the first time incorporated into Abdul-Razzak and Ghan activation scheme (ARG) to calculate hygroscopicity and critical supersaturation of aerosols (HYB), and the performance of the modied scheme is compared with the osmotic coefficient model (STN), which is the standard in the ARG scheme. Reference simulations (REF) with the prescribed cloud droplet number concentration have also been carried out in order to understand the effects of aerosol-cloud feedbacks. In addition, since the calculated cloud coverage is an important determinant of cloud radiative effects and is influencing the nucleation process two cloud cover parameterizations (i.e., a relative humidity threshold; RH-CLC and a statistical cloud cover scheme; ST-CLC) have been examined together with the CDN schemes, and their effects on the simulated cloud properties and relevant climate parameters have been investigated. The distinct cloud droplet spectra show strong sensitivity to aerosol composition effects on cloud droplet formation in all particle sizes, especially for the Aitken mode. As Aitken particles are the major component of the total aerosol number concentration and CCN, and are most sensitive to aerosol chemical composition effect (solute effect) on droplet formation, the activation of Aitken particles strongly contribute to total cloud droplet formation and thereby providing different cloud droplet spectra. These different spectra influence cloud structure, cloud properties, and climate, and show regionally varying sensitivity to meteorological and geographical condition as well as the spatiotemporal aerosol properties (i.e., particle size, number, and composition). The changes responding to different CDN schemes are more pronounced at lower altitudes than higher altitudes. Among regions, the subarctic regions show the strongest changes, as the lower surface temperature amplifies the effects of the activated aerosols; in contrast, the Sahara desert, where is an extremely dry area, is less influenced by changes in CCN number concentration. The aerosol-cloud coupling effects have been examined by comparing the prognostic CDN simulations (STN, HYB) with the reference simulation (REF). Most pronounced effects are found in the cloud droplet number concentration, cloud water distribution, and cloud radiative effect. The aerosol-cloud coupling generally increases cloud droplet number concentration; this decreases the efficiency of the formation of weak stratiform precipitation, and increases the cloud water loading. These large-scale changes lead to larger cloud cover and longer cloud lifetime, and contribute to high optical thickness and strong cloud cooling effects. This cools the Earth's surface, increases atmospheric stability, and reduces convective activity. These changes corresponding to aerosol-cloud feedbacks are also differently simulated depending on the cloud cover scheme. The ST-CLC scheme is more sensitive to aerosol-cloud coupling, since this scheme uses a tighter linkage of local dynamics and cloud water distributions in cloud formation process than the RH-CLC scheme. For the calculated total cloud cover, the RH-CLC scheme simulates relatively similar pattern to observations than the ST-CLC scheme does, but the overall properties (e.g., total cloud cover, cloud water content) in the RH simulations are overestimated, particularly over ocean. This is mainly originated from the difference in simulated skewness in each scheme: the RH simulations calculate negatively skewed distributions of cloud cover and relevant cloud water, which is similar to that of the observations, while the ST simulations yield positively skewed distributions resulting in lower mean values than the RH-CLC scheme does. The underestimation of total cloud cover over ocean, particularly over the intertropical convergence zone (ITCZ) relates to systematic defficiency of the prognostic calculation of skewness in the current set-ups of the ST-CLC scheme.rnOverall, the current EMAC model set-ups perform better over continents for all combinations of the cloud droplet nucleation and cloud cover schemes. To consider aerosol-cloud feedbacks, the HYB scheme is a better method for predicting cloud and climate parameters for both cloud cover schemes than the STN scheme. The RH-CLC scheme offers a better simulation of total cloud cover and the relevant parameters with the HYB scheme and single-moment microphysics (REF) than the ST-CLC does, but is not very sensitive to aerosol-cloud interactions.

Homeorhetic Assemblies indaga le potenzialità tettoniche ed architettoniche derivanti dallo studio dei sistemi biologici decentralizzati, dei loro comportamenti e delle relazioni dinamiche con la colonia in termini di processi adattativi e costruttivi continui nel tempo. La ragione di questo interesse è radicata nei principi dell’ecologia applicata al design ed alle tecnologie di fabbricazione contemporanee, che vanno al di là della mera imitazione formale: ci si è quindi chiesto come raggiungere una spazialità complessa ed articolata, omogeneità di prestazioni ed una struttura continua caratterizzata da molti elementi aventi le stesse caratteristiche di forma e materiale. L’ecologia è lo studio di un insieme di economie, ovvero rapporti di scambio, tra un organismo ed il suo ambiente e l’efficenza dei pattern distributivi che derivano da queste relazioni sono fondamentali al fine del successo evolutivo del sistema stesso. I sistemi su cui ci si è concentrati sono caratterizzati dalla capacità di creare strutture a buon mercato (con l’uso di istruzioni semplici ed un unico materiale) e ad elevato grado di complessità ed efficienza, armonizzando l’aspetto formale con l’organizzazione materica e fisiologica. Il modello di comportamento considerato riguarda le dinamiche alla base della creazione degli alveari naturali creati dalle api millifere. Queste caratteristiche sono state codificate nella programmazione di un sistema multi agente composto da agenti autonomi in grado di interagire in un ambiente eterogeneo e capaci di depositare selettivamente elementi in una struttura composta da springs e particles, periodicamente stabilizzata ed ottimizzata. In un tale sistema, a priori sono note solo le relazioni locali per i singoli agenti ed il comportamento strutturale generale, mentre gli oggetti e gli eventi emergono in maniera non predeterminata come risultato di queste interazioni nello spazio e nel tempo. I risultati appaiono estremamente complessi ed eterogenei nella loro organizzazione spaziale, pur emergendo un set di elementi identificabili nella loro specifica singolarità (come ad esempio superfici, colonne, capriate etc...) ma che generano strutture continue, e creano grande differenziazione di densità e di disposizione dei singoli elementi all’interno della struttura. La ridondanza strutturale ottenuta è una scelta deliberata e permessa dall’automatizzazione della fase di costruzione attraverso la programmazione di robot, tramite i quali si intende realizzare un prototipo fisico delle strutture ottenute.
