Vetores de expressão são ferramentas moleculares úteis para investigar a função de genes tanto em sistemas procarióticos quanto eucarióticos. O fungo Metarhizium anisopliae, utilizado no controle biológico de artrópodes, é bem caracterizado em nível molecular. Genes candidatos a participar do processo de infecção de hospedeiros tem sido isolados utilizando estratégias em que o gene candidato é predefinido (enzimas hidrolíticas, por exemplo) ou estratégias mais globais como projetos de ESTs e a análise de bibliotecas de subtração. A superexpressão tem sido o método adotado para verificar a participação de genes isolados no processo de infecção. Esta estratégia tem sido baseada no promoter heterólogo PgpdA de Aspergillus nidulans. Neste trabalho, o gene que codifica o fator de alongamento de tradução tef-1 α de Metarhizium anisopliae foi clonado e a sua região promotora foi localizada e utilizada na construção de um vetor de expressão. Somente uma cópia do gene tef-1α está presente no genoma de M. anisopliae e o seu perfil de expressão foi analisado. Uma árvore filogenética foi construída baseada nos ortógos de tef-1 α e mostrou uma alta correlação com o fungo Cordyceps taii. A região de 639 pb à montante do codon de iniciação (ATG) foi utilizada com sucesso para a expressão do gene repórter sGFP e do gene bar, que confere resistência ao glifosinato de amônio, em M. anisopliae. Os transformantes construídos não apresentaram alteração na sua virulência em bioensaios com carrapatos, em relação a linhagem receptora. Além disso, demonstramos que o nível de expressão permite a detecção óptica da fluorescência de sGFP durante a infecção dos carrapatos. Desta forma, o vetor desenvolvido será uma ferramenta útil para a superexpressão em Metarhizium.

Esta tese de mestrado descreve o desenvolvimento, implementação e teste de um sistema de medição de energia concebido para um ambiente doméstico, baseado no circuito integrado medidor de energia Teridian 71M6515H. O sistema desenvolvido envia periodicamente os valores monitorizados para uma base de dados, através de uma rede sem fios com base no protocolo IEEE 802.11 (Wi-Fi). Os dados podem ser acedidos remotamente em tempo real, através de uma página na internet, onde é possível consultar a quantidade de energia consumida acumulada. Na primeira fase deste trabalho realizou-se o enquadramento do tema no contexto atual, realizando-se o estudo de alguns sistemas domésticos de monitorização do consumo de energia elétrica existentes no mercado. Numa segunda fase foi realizado o estudo dos componentes a serem utilizados, o desenho da placa de circuito impresso e o desenvolvimento do firmware, para que todo o processamento e manipulação dos dados fossem realizados pela unidade de aquisição de dados. Em seguida procedeu-se à construção da unidade de aquisição e à programação para o registo dos valores numa base de dados. A última fase consistiu nos testes de funcionamento da unidade de aquisição em conjunto com a aplicação de registo de dados. Com a implementação do sistema desenvolvido o consumidor doméstico poderá ter conhecimento, em tempo real, do custo de funcionamento dos equipamentos que possui e assim tomar decisões para os utilizar de forma mais racional, o que se pode traduzir numa economia dos recursos energéticos.

Nesta dissertação de mestrado é desenvolvido um sistema de replicação de circuitos integrados digitais (combinatórios e sequenciais), por observação do seu normal funcionamento. O sistema desenvolvido carateriza-se pela capacidade de extrair e descrever na linguagem VHDL o comportamento de um circuito integrado digital em funcionamento, utilizando técnicas não invasivas e automatizadas, suportado por um vasto conjunto de algoritmos de aquisição e análise de dados. O sistema desenvolvido assenta em dois módulos principais: um módulo de software que consiste numa plataforma de algoritmos de análise, controlo e gestão do sistema (alojada num computador) e um módulo de aquisição de dados (hardware) que consiste num circuito capaz de realizar as medições necessárias para o funcionamento do sistema, comandado pelo módulo de software. A comunicação entre os dois módulos é efetuada via porta série. Os algoritmos desenvolvidos realizam uma análise da correspondência entre entradas e saídas procurando aplicar uma aproximação a um circuito combinatório se possível, caso contrário são utilizados métodos heurísticos para efetuar uma aproximação a um circuito sequencial através de uma máquina de estados. Entradas ou saídas constantes são previamente identificados e excluídos do processo de análise, para efeitos de simplificação. Os resultados obtidos demonstram que é possível replicar o comportamento observado em circuitos digitais (combinatórios e sequenciais) desde que o número de amostras recolhidas seja adequado. Verifica-se ainda que o método desenvolvido replica a funcionalidade do circuito integrado nas condições onde o circuito está inserido.
Sistema de detecção e isolamento de falhas em sistemas dinâmicos baseado em identificação paramétrica

The present research aims at contributing to the area of detection and diagnosis of failure through the proposal of a new system architecture of detection and isolation of failures (FDI, Fault Detection and Isolation). The proposed architecture presents innovations related to the way the physical values monitored are linked to the FDI system and, as a consequence, the way the failures are detected, isolated and classified. A search for mathematical tools able to satisfy the objectives of the proposed architecture has pointed at the use of the Kalman Filter and its derivatives EKF (Extended Kalman Filter) and UKF (Unscented Kalman Filter). The use of the first one is efficient when the monitored process presents a linear relation among its physical values to be monitored and its out-put. The other two are proficient in case this dynamics is no-linear. After that, a short comparative of features and abilities in the context of failure detection concludes that the UFK system is a better alternative than the EKF one to compose the architecture of the FDI system proposed in case of processes of no-linear dynamics. The results shown in the end of the research refer to the linear and no-linear industrial processes. The efficiency of the proposed architecture may be observed since it has been applied to simulated and real processes. To conclude, the contributions of this thesis are found in the end of the text

This work deals with the development of a prototype of a helicopter quadrotor for monitoring applications in oil facilities. Anomaly detection problems can be resolved through monitoringmissions performed by a suitably instrumented quadrotor, i.e. infrared thermosensors should be embedded. The proposed monitoring system aims to reduce accidents as well as to make possible the use of non-destructive techniques for detection and location of leaks caused by corrosion. To this end, the implementation of a prototype, its stabilization and a navigation strategy have been proposed. The control strategy is based on dividing the problem into two control hierarchical levels: the lower level stabilizes the angles and the altitude of the vehicle at the desired values, while the higher one provide appropriate references signals to the lower level in order the quadrotor performs the desired movements. The navigation strategy for helicopter quadrotor is made using information provided by a acquisition image system (monocular camera) embedded onto the helicopter. Considering that the low-level control has been solved, the proposed vision-based navigation technique treats the problem as high level control strategies, such as, relative position control, trajectory generation and trajectory tracking. For the position control we use a control technique for visual servoing based on image features. The trajectory generation is done in a offline step, which is a visual trajectory composed of a sequence of images. For the trajectory tracking problem is proposed a control strategy by continuous servovision, thus enabling a navigation strategy without metric maps. Simulation and experimental results are presented to validate the proposal

The sharing of knowledge and integration of data is one of the biggest challenges in health and essential contribution to improve the quality of health care. Since the same person receives care in various health facilities throughout his/her live, that information is distributed in different information systems which run on platforms of heterogeneous hardware and software. This paper proposes a System of Health Information Based on Ontologies (SISOnt) for knowledge sharing and integration of data on health, which allows to infer new information from the heterogeneous databases and knowledge base. For this purpose it was created three ontologies represented by the patterns and concepts proposed by the Semantic Web. The first ontology provides a representation of the concepts of diseases Secretariat of Health Surveillance (SVS) and the others are related to the representation of the concepts of databases of Health Information Systems (SIS), specifically the Information System of Notification of Diseases (SINAN) and the Information System on Mortality (SIM)

Nowadays, where the market competition requires products with better quality and a constant search for cost savings and a better use of raw materials, the research for more efficient control strategies becomes vital. In Natural Gas Processin Units (NGPUs), as in the most chemical processes, the quality control is accomplished through their products composition. However, the chemical composition analysis has a long measurement time, even when performed by instruments such as gas chromatographs. This fact hinders the development of control strategies to provide a better process yield. The natural gas processing is one of the most important activities in the petroleum industry. The main economic product of a NGPU is the liquefied petroleum gas (LPG). The LPG is ideally composed by propane and butane, however, in practice, its composition has some contaminants, such as ethane and pentane. In this work is proposed an inferential system using neural networks to estimate the ethane and pentane mole fractions in LPG and the propane mole fraction in residual gas. The goal is to provide the values of these estimated variables in every minute using a single multilayer neural network, making it possibly to apply inferential control techniques in order to monitor the LPG quality and to reduce the propane loss in the process. To develop this work a NGPU was simulated in HYSYS R software, composed by two distillation collumns: deethanizer and debutanizer. The inference is performed through the process variables of the PID controllers present in the instrumentation of these columns. To reduce the complexity of the inferential neural network is used the statistical technique of principal component analysis to decrease the number of network inputs, thus forming a hybrid inferential system. It is also proposed in this work a simple strategy to correct the inferential system in real-time, based on measurements of the chromatographs which may exist in process under study

Previous works have studied the characteristics and peculiarities of P2P networks, especially security information aspects. Most works, in some way, deal with the sharing of resources and, in particular, the storage of files. This work complements previous studies and adds new definitions relating to this kind of systems. A system for safe storage of files (SAS-P2P) was specified and built, based on P2P technology, using the JXTA platform. This system uses standard X.509 and PKCS # 12 digital certificates, issued and managed by a public key infrastructure, which was also specified and developed based on P2P technology (PKIX-P2P). The information is stored in a special file with XML format which is especially prepared, facilitating handling and interoperability among applications. The intention of developing the SAS-P2P system was to offer a complementary service for Giga Natal network users, through which the participants in this network can collaboratively build a shared storage area, with important security features such as availability, confidentiality, authenticity and fault tolerance. Besides the specification, development of prototypes and testing of the SAS-P2P system, tests of the PKIX-P2P Manager module were also performed, in order to determine its fault tolerance and the effective calculation of the reputation of the certifying authorities participating in the system

This work deals with an on-line control strategy based on Robust Model Predictive Control (RMPC) technique applied in a real coupled tanks system. This process consists of two coupled tanks and a pump to feed the liquid to the system. The control objective (regulator problem) is to keep the tanks levels in the considered operation point even in the presence of disturbance. The RMPC is a technique that allows explicit incorporation of the plant uncertainty in the problem formulation. The goal is to design, at each time step, a state-feedback control law that minimizes a 'worst-case' infinite horizon objective function, subject to constraint in the control. The existence of a feedback control law satisfying the input constraints is reduced to a convex optimization over linear matrix inequalities (LMIs) problem. It is shown in this work that for the plant uncertainty described by the polytope, the feasible receding horizon state feedback control design is robustly stabilizing. The software implementation of the RMPC is made using Scilab, and its communication with Coupled Tanks Systems is done through the OLE for Process Control (OPC) industrial protocol

In general, the designs of equipment takes into account the effects and processes of deterioration it will undergo and arrives at an approximate useful life. However, changes in operational processes and parameters, the action of external agents, the kind of maintenance conducted, the means of monitoring, and natural and accidental occurrences completely modify the desired performance of the equipment. The discontinuities that occur in anisotropic materials often and due to different factors evolve from being subcritical to critical acquiring the status of defect and compromising the physical integrity of the equipment. Increasingly sophisticated technological means of detection, monitoring and assessment of these discontinuities are required to respond ever more rapidly to the requirements of industry. This paper therefore presents a VPS (Virtual Pipe System) computational tool which uses the results of ultrasonic tests on equipment, plotting the discontinuities found in models created in the CAD and CAE systems, and then simulates the behavior of these defects in the structure to give an instantaneous view of the final behavior. This paper also presents an alternative method of conventional ultrasonic testing which correlates the integrity of an overlay (carbon steel and stainless steel attached by welding) and the reflection of ultrasonic waves coming from the interface between the two metals, thus making it possible to identify cracks in the casing and a shift of the overlay

The central question of the present study is to identify the epistemological knowledge that the teachers-trainees possess regarding the characteristics (properties) of the decimal numbering system; its purpose is to offer a contribution to the pedagogic practice of the teachers who work within the Basic Literacy Cycle, in terms of what concerns both the acquisition of contents and the development of the knowledge that helps them in the elaboration of adequate strategies to working with the Decimal Numbering System in the classroom. The study is based on the constructivist sociointeractionist approach to teaching Mathematics and it constitutes, in itself, a methodological intervention with the teachers-trainees engaged in the Professional Qualification Program in Basic Education of the Federal University of Rio Grande do Norte. The foundations of the study were found in investigations of researchers who had carried out studies on the construction of numerical writing, showing, for instance, that the construction process of ideas and procedures involved in groupings and changes to base 10 take a lot longer to be accomplished than one can imagine. A set of activities was then elaborated which could not only contribute to the acquisition of contents but that could also make the teachers-trainees reflect upon their teaching practices in the classroom so that in this way they will be able to elaborate more consistent didactic approaches, taking into consideration the previous knowledge of the students and also some obstacles that often appear along the way. Even when teachers have access to the most appropriate dicactic resources, the lack of knowledge of the content and of the real meaning of that content make the Decimal Numbering System, a subject of fundamental importance, be taught most times in a mechanical way. The analisys of the discussions and behaviours of the teachers-trainees during the activities reavealed that they made them reflect upon their current practices in the classroom and that, as a whole, the aims of each of the activities carried out with the teachers-trainers were reached
