The psbA2 gene of a unicellular cyanobacterium, Microcystis aeruginosa K-81, encodes a D1 protein homolog in the reaction center of photosynthetic Photosystem II. The expression of the psbA2 transcript has been shown to be light-dependent as assessed under light and dark (12/12 h) cycling conditions. We aligned the 5′-untranslated leader regions (UTRs) of psbAs from different photosynthetic organisms and identified a conserved sequence, UAAAUAAA or the ‘AU-box’, just upstream of the SD sequences. To clarify the role of 5′-upstream cis-elements containing the AU-box for light-dependent expression of psbA2, a series of deletion and point mutations in the region were introduced into the genome of heterologous cyanobacterium Synechococcus sp. strain PCC 7942, and psbA2 expression was examined. A clear pattern of light-dependent expression was observed in recombinant cyanobacteria carrying the K-81 psbA2 –38/+36 region (which includes the minimal promoter element and a light-dependent cis-element with the AU-box), +1 indicating the transcription start site. A constitutive pattern of expression, in which the transcripts remained almost stable under dark conditions, was obtained in cells harboring the –38/+14 region (the minimal element), indicating that the +14/+36 region with the AU-box is important for the observed light-dependent expression. Point mutations analyses within the AU-box also revealed that changes in number, direction and identity (as assayed by adenine/uridine nucleotide substitutions) influenced the light-dependent pattern of expression. The level of psbA2 transcripts increased markedly in CG- or deletion-box mutants in the dark, strongly indicating that the AU- (AT-) box acts as a negative cis-element. Furthermore, characterization of transcript accumulation in cells treated with rifampicin suggests that psbA2 5′-mRNA is unstable in the dark, supporting the view that the light-dependent expression is controlled at the post-transcriptional level. We discuss various mechanisms that may lead to altered mRNA stability such as the binding of factor(s) or ribosomes to the 5′-UTR and possible roles of the AU-box motif and the SD sequence.

O entendimento da comunidade fitoplanctônica em sistemas instáveis, como por exemplo reservatórios, necessita conhecimento de escalas de variabilidade. Com base nisso, um estudo sobre a heterogeneidade espacial e variabilidade temporal de dois reservatórios com diferentes graus de trofia, no Estado de São Paulo foi realizado em 20 estações no reservatório de Salto Grande e em 19 no reservatório do Lobo, em 3 dias consecutivos, em quatro períodos: outubro de 1999, janeiro, abril e junho e julho de 2000. Para tanto foram determinadas as concentrações de nutrientes totais e dissolvidos, material em suspensão, carbono inorgânico, clorofila a, biomassa, densidade, composição e produtividade primária da comunidade fitoplanctônica e os perfis de oxigênio dissolvido, temperatura, pH e condutividade. Os dois reservatórios tiveram estruturas espaciais semelhantes com a formação de três zonas distintas. A zona de rio, misturada, com menor penetração de luz e maior concentração de nutrientes, a zona de transição, e a zona lacustre, mais estratificada, com maior penetração de luz e menor concentração de nutrientes. Apesar dessa compartimentalização a heterogeneidade espacial no reservatório de Salto Grande foi maior que no reservatório do Lobo, sobretudo em função do gradiente longitudinal de nutrientes e luz. A variabilidade diária (3 dias) nos dois reservatórios não foi significativa na determinação da comunidade fitoplanctônica. A escala de variabilidade sazonal, nos dois reservatórios, foi determinada, principalmente pela variação nos padrões de estratificação e mistura sendo, assim, determinante na composição da comunidade fitoplanctônica. Essa influência foi mais evidente no reservatório do Lobo. A variação temporal e heterogeneidade espacial das mais abundantes espécies e grupos taxonômicos da comunidade fitoplanctônica, (Microcystis aeruginosa, Anabaena crassa e Anabaena circinalis em Salto Grande e Aphanocapsa delicatissima, Coelastrum reticulatum e Aulacoseira granulata no Lobo) nos dois reservatórios foram determinados pelos complexos processos de estratificação e mistura e da disponibilidade de luz. Os resultados obtidos são importantes para o entendimento da variabilidade ambiental de reservatórios tropicais e no planejamento de amostragens que visem o gerenciamento desses sistemas.

As represas construídas em cascata no rio Tietê (Barra Bonita, Bariri, Ibitinga, Promissão, Nova Avanhandava e Três Irmãos) constituem em sistemas artificiais com grande importância ecológica, econômica e social. Com base nos dados físico-químicos e biológicos das seis represas, foram avaliadas as condições limnológicas, abundância da comunidade fitoplanctônica e zooplanctônica e o estado de trofia dos reservatórios através da aplicação do índice do estado trófico, desenvolvido por Carlson (1977). A utilização da concentração de fósforo para determinação do índice é o que deu melhor resultado, mostrando uma redução gradual de sua concentração nas represas seqüenciais, principalmente no período do verão. Os reservatórios que se mostraram eutróficos como o de Barra Bonita, Bariri e Ibitinga apresentaram uma maior abundância e freqüência de florescimento das algas pertencentes a classe Cyanophyceae principalmente das espécies Microcystis aeruginosa. Algumas espécies de Rotifera, como Asplanchna sieboldi, Brachionus caliciflorus e Kellicottia bostoniensis, e de Copepoda Calanoida Notodiaptomus iheringi, também serviram de indicativos do estado trófico dos sistemas, já que esses estão associados a ambientes eutróficos.

In Australian freshwaters, Anabaena circinalis, Microcystis spp. and Cylindrospermopsis raciborskii are the dominant toxic cyanobacteria. Many of these Surface waters are used as drinking water resources. Therefore, the National Health and Medical Research Council of Australia set a guideline for MC-LR toxicity equivalents of 1.3 mug/l drinking, water. However, due to lack of adequate data, no guideline values for paralytic shellfish poisons (PSPs) (e.g. saxitoxins) or cylindrospermopsin (CYN) have been set. In this spot check. the concentration of microcystins (MCs), PSPs and CYN were determined by ADDA-ELISA, cPPA, HPLC-DAD and/or HPLC-MS/MS, respectively, in two water treatment plants in Queensland/Australia and compared to phytoplankton data collected by Queensland Health, Brisbane. Depending on the predominant cyanobacterial species in a bloom, concentrations of up to 8.0, 17.0 and 1.3 mug/l were found for MCs, PSPs and CYN, respectively. However, only traces (< 1.0 mug/l) of these toxins were detected in final water (final product of the drinking water treatment plant) and tap water (household sample). Despite the low concentrations of toxins detected in drinking water, a further reduction of cyanobacterial toxins is recommended to guarantee public safety. (C) 2004 Elsevier Ltd. Cyanobacteria ("blue-green algae") are known to produce a diverse repertoire of biologically active secondary metabolites. When associated with so-called "harmful algal blooms", particularly in freshwater systems, a number of these metabolites have been associated—as "toxins", or commonly "cyanotoxins"—with human and animal health concerns. In addition to the known water-soluble toxins from these genera (i.e. microcystins, cylindrospermopsin, and saxitoxins), our studies have shown that there are metabolites within the lipophilic extracts of these strains that inhibit vertebrate development in zebrafish embryos. Following these studies, the zebrafish embryo model was implemented in the bioassay-guided purification of four isolates of cyanobacterial harmful algal blooms, namely Aphanizomenon, two isolates of Cylindrospermopsis, and Microcystis, in order to identify and chemically characterize the bioactive lipophilic metabolites in these isolates. ^ We have recently isolated a group of polymethoxy-1-alkenes (PMAs), as potential toxins, based on the bioactivity observed in the zebrafish embryos. Although PMAs have been previously isolated from diverse cyanobacteria, they have not previously been associated with relevant toxicity. These compounds seem to be widespread across the different genera of cyanobacteria, and, according to our studies, suggested to be derived from the polyketide biosynthetic pathway which is a common synthetic route for cyanobacterial and other algal toxins. Thus, it can be argued that these metabolites are perhaps important contributors to the toxicity of cyanobacterial blooms. In addition to the PMAs, a set of bioactive glycosidic carotenoids were also isolated because of their inhibition of zebrafish embryonic development. These pigmented organic molecules are found in many photosynthetic organisms, including cyanobacteria, and they have been largely associated with the prevention of photooxidative damage. This is the first indication of these compounds as toxic metabolites and the hypothesized mode of action is via their biotransformation to retinoids, some of which are known to be teratogenic. Additional fractions within all four isolates have been shown to contain other uncharacterized lipophilic toxic metabolites. This apparent repertoire of lipophilic compounds may contribute to the toxicity of these cyanobacterial harmful algal blooms, which were previously attributed primarily to the presence of the known water-soluble toxins.^

The wide-spread impact of exotic fishes especially Oreochromis niloticus and Lates niloticus together with over fishing in the Victoria and Kyoga lake basins during the 1950s and 1960s, caused endemic species such as the previously most important Oreochromis esculentus to become virtually extinct in the two lakes by the 1970s. Based on reports of presence of this native species in some satellite lakes within the two lake basins, a set of satellite lakes in the Victoria basin (Nabugabo lakes: Kayanja and Kayugi), were sampled between 1997-2002 with an objective of assessing their value as conservation sites for O. esculentus. Other satellite lakes (Mburo and Kachera) also in the Victoria basin, and Lemwa, Kawi and Nabisojjo, in the Kyoga basin, were sampled for comparison. Among the Nabugabo lakes, O. esculentus was more abundant in Lake Kayanja (20.1 %) ofthe total fish catch by weight compared to Lake Kayugi (1.4 %). The largest fish examined (38.7 cm TL) was caught in Lake Kayugi, (also the largest in all satellite lakes sampled), while the smallest (6.6 cm TL) was from Lake Kayanja. Fish from Lake Kayugi had a higher condition factor K (1.89± 0.02) than that from Lake Kayanja (1.53±0.0I), which was the second highest (compared with other satellite lakes) to Lake Kawi (1.92±0.2). Diatoms, especially Aulacoseira, which were previously known to be the best food for O. esculentus in Lake Victoria were mostly encountered (93.2 %) in fish stomachs from Lake Kayugi. In Lake Kayanja the dominant food item was the blue green algae (Planktolyngbya) while Microcystis was the most abundant diet item in fish from other satellite lakes. There were more male than female fish (ratio 1:0.91 and 1: 0.79 in lakes Kayugi and Kayanja respectively). This is comparable to the situation in Lake Victoria before the species got depleted. The highest mean fecundity was (771±218 eggs) recorded in Lake Kayugi compared to Lake Kayanja (399±143). Based on the results from Lake Kayugi, where diatoms dominated the diet of O. esculentus and where the largest, most fecund and healthy fish were found, this lake would be a most valuable site for the conservation of O. esculentus and the best source of fish, for restocking and for captive-propagation. This lake is therefore recommended for protection from over exploitation and misuse.
