BACKGROUND: Epigenetic alterations have been implicated in the pathogenesis of solid tumors, however, proto-oncogenes activated by promoter demethylation have been sporadically reported. We used an integrative method to analyze expression in primary head and neck squamous cell carcinoma (HNSCC) and pharmacologically demethylated cell lines to identify aberrantly demethylated and expressed candidate proto-oncogenes and cancer testes antigens in HNSCC. METHODOLOGY/PRINCIPAL FINDINGS: We noted coordinated promoter demethylation and simultaneous transcriptional upregulation of proto-oncogene candidates with promoter homology, and phylogenetic footprinting of these promoters demonstrated potential recognition sites for the transcription factor BORIS. Aberrant BORIS expression correlated with upregulation of candidate proto-oncogenes in multiple human malignancies including primary non-small cell lung cancers and HNSCC, induced coordinated proto-oncogene specific promoter demethylation and expression in non-tumorigenic cells, and transformed NIH3T3 cells. CONCLUSIONS/SIGNIFICANCE: Coordinated, epigenetic unmasking of multiple genes with growth promoting activity occurs in aerodigestive cancers, and BORIS is implicated in the coordinated promoter demethylation and reactivation of epigenetically silenced genes in human cancers.

This deliverable outlines the design blueprints for the RAGE application scenario games and forms the rest of the scope for WP4’s tasks. The game designs have been developed in collaboration with application scenario partners in WP5, and informed by WP1, 2 & 3. Additionally peer-feedback has been provided by game developers across WP4. The designs outline the integration of the RAGE assets developed in WP2 and WP3. Each section provides in detail the game play descriptions, game dynamics and mechanics, pedagogies and technical implementation of the RAGE assets into the game applications as described in detailed in WP5’s application documents. The full description of the application objectives and associated learning outcomes has been provided in the project’s MS2 Application Scenario Outlines document.

Modern scientific world-view has undermined traditional myths, the functional survival of which seems to depend today in the West on a positivist justification. This would place them in the field of real History, through their study and revitalization by pseudoscientific disciplines such as the Atlantis and the ancient astronaut hypotheses. These have inspired new epic poems in (regular) verse that combine classic and/or biblical myths with a (pseudo)scientific modern world-view. For example, the critical rewriting of Noah’s myth by using the ancient astronaut hypothesis as a fictional device to produce a contemporary kind of plausibility allowed Abel Montagut to renew epic poetry, updating it also by adopting science fiction chronotopes in order to structure his fictional construction and to generate a high ethical sense for our time. Thus, his Poemo de Utnoa (1993) / La gesta d’Utnoa (1996), which has become a major classic of the literature in Esperanto thanks to its original version in this language, is a landmark of both science fiction and neo-biblical epics. This poem is written from a secular and purely literary perspective.

Este artículo analiza la imagen de la Ciudad de México configurada en la crónica mexicana a partir de los años setenta en especial en los textos de José Joaquín Blanco. Se detiene en el modo en que el género afronta la realidad de una megalópolis que ya solo puede ser asumida de modo fragmentario y múltiple. Entre la utopía y una visión apocalíptica de la ciudad, la crónica reconfigura el espacio y el tiempo urbanos, y formula una nueva identidad crítica gracias a la habilitación de espacios de expresión posibles dentro de las nuevas ciudades modificadas por el capital.

Programmed cell death (PCD) is executed by proteases, which cleave diverse proteins thus modulating their biochemical and cellular functions. Proteases of the caspase family and hundreds of caspase substrates constitute a major part of the PCD degradome in animals(1,2). Plants lack close homologues of caspases, but instead possess an ancestral family of cysteine proteases, metacaspases(3,4). Although metacaspases are essential for PCD(5-7), their natural substrates remain unknown(4,8). Here we show that metacaspase mcII-Pa cleaves a phylogenetically conserved protein, TSN (Tudor staphylococcal nuclease), during both developmental and stress-induced PCD. TSN knockdown leads to activation of ectopic cell death during reproduction, impairing plant fertility. Surprisingly, human TSN (also known as p100 or SND1), a multifunctional regulator of gene expression(9-15), is cleaved by caspase-3 during apoptosis. This cleavage impairs the ability of TSN to activate mRNA splicing, inhibits its ribonuclease activity and is important for the execution of apoptosis. Our results establish TSN as the first biological substrate of metacaspase and demonstrate that despite the divergence of plants and animals from a common ancestor about one billion years ago and their use of distinct PCD pathways, both have retained a common mechanism to compromise cell viability through the cleavage of the same substrate, TSN.

Execution of programmed cell death (PCD) in nonmetazoan organisms is morphologically different from apoptotic PCD in animals and lacks a number of key molecular components of apoptotic machinery, including caspases. Yet protozoan, fungal, and plant cells exhibit caspase-like proteolytic activities, which increase in a PCD-dependent manner. This poses a question whether nonmetazoan organisms contain structurally dissimilar proteases that functionally substitute for caspases. Putative ancestors of caspases, metacaspases, are candidates for this role; however, their distinct substrate specificity raises doubts. The identification of a common biological target of caspases and metacaspases and previously unknown functions unrelated to cell death of metacaspases provide new food for thought.

Approaches exploiting trait distribution extremes may be used to identify loci associated with common traits, but it is unknown whether these loci are generalizable to the broader population. In a genome-wide search for loci associated with the upper versus the lower 5th percentiles of body mass index, height and waist-to-hip ratio, as well as clinical classes of obesity, including up to 263,407 individuals of European ancestry, we identified 4 new loci (IGFBP4, H6PD, RSRC1 and PPP2R2A) influencing height detected in the distribution tails and 7 new loci (HNF4G, RPTOR, GNAT2, MRPS33P4, ADCY9, HS6ST3 and ZZZ3) for clinical classes of obesity. Further, we find a large overlap in genetic structure and the distribution of variants between traits based on extremes and the general population and little etiological heterogeneity between obesity subgroups.

The androgen receptor (AR) is a key regulator of prostate growth and the principal drug target for the treatment of prostate cancer. Previous studies have mapped AR targets and identified some candidates which may contribute to cancer progression, but did not characterize AR biology in an integrated manner. In this study, we took an interdisciplinary approach, integrating detailed genomic studies with metabolomic profiling and identify an anabolic transcriptional network involving AR as the core regulator. Restricting flux through anabolic pathways is an attractive approach to deprive tumours of the building blocks needed to sustain tumour growth. Therefore, we searched for targets of the AR that may contribute to these anabolic processes and could be amenable to therapeutic intervention by virtue of differential expression in prostate tumours. This highlighted calcium/calmodulin-dependent protein kinase kinase 2, which we show is overexpressed in prostate cancer and regulates cancer cell growth via its unexpected role as a hormone-dependent modulator of anabolic metabolism. In conclusion, it is possible to progress from transcriptional studies to a promising therapeutic target by taking an unbiased interdisciplinary approach.

The androgen receptor (AR) initiates important developmental and oncogenic transcriptional pathways. The AR is known to bind as a homodimer to 15-base pair bipartite palindromic androgen-response elements; however, few direct AR gene targets are known. To identify AR promoter targets, we used chromatin immunoprecipitation with on-chip detection of genomic fragments. We identified 1,532 potential AR-binding sites, including previously known AR gene targets. Many of the new AR target genes show altered expression in prostate cancer. Analysis of sequences underlying AR-binding sites showed that more than 50% of AR-binding sites did not contain the established 15 bp AR-binding element. Unbiased sequence analysis showed 6-bp motifs, which were significantly enriched and were bound directly by the AR in vitro. Binding sequences for the avian erythroblastosis virus E26 homologue (ETS) transcription factor family were also highly enriched, and we uncovered an interaction between the AR and ETS1 at a subset of AR promoter targets.

HDL has long been known for its role in reverse cholesterol transport, thought in part to explain the well-recognized links between low levels of HDL-C and cardiovascular disease. The past decade has seen increasing evidence from epidemiological, basic science and early human intervention studies that HDL biology is more complex and may influence the onset and progression of type 2 diabetes. Research has identified multiple potential pathways by which higher HDL particle concentrations or functional improvements may ameliorate the development and progression of the disease. These include promotion of insulin secretion and pancreatic islet beta-cell survival, promotion of peripheral glucose uptake, and suppression of inflammation. The relationships between HDL-C levels, commonly used in clinical practice, and HDL particle number, size and various HDL functions is complex, and is intimately linked with triglyceride metabolism. The complexity of these relationships is amplified in diabetes, which negatively impacts multiple aspects of lipoprotein biology. This article reviews the rationale for, and potential of, HDL-based anti-diabetic pharmacotherapy, with an emphasis on the particular challenges posed by diabetes-related HDL dysfunction, and on the difficulties of selecting appropriate targets and HDL-related biomarkers for research and for clinical practice. We discuss aspects of HDL metabolism that are known to be altered in type 2 diabetes, potentially useful measures of HDL-targeted therapy in diabetes, and review early intervention studies in humans. These areas provide a firm foundation for further research and knowledge expansion in this intriguing area of human health and disease.

Hoje em dia, as ameaças são cada vez mais frequentes e sofisticadas, do que alguma vez registado. Todo o tipo de empresas/organizações e informação estão sujeitas a estas ameaças. Estes ataques são cada vez mais recorrentes, deixando para trás um rasto de várias quebras de segurança. Existem uma serie de ciberataques que já deixaram a sua marca na historia. Uma das mais notórias, foi o caso da Estónia em 2007, por um grupo pro-kremlin de Transnístria em que vários servidores governamentais, fornecedores de serviço, servidores da banca, entre outros foram alvo de uma serie de ataques, na sua maioria de DDoS (Distributed Denial of Service1),e botnets2. O seu método era tao complicado que o governo da Estónia achava que estavam a ser apoiados pelo governo russo. Isto resultou na paragem de um país ate que o problema fosse normalizado. Considerado um ato de hacktivismo3 pelo que representava algo muito importante para a população russa, um ícone, “the Bronze Soldier of Tallinn”, um elaborado cemitério da altura soviética que o governo da Estónia queria recolocar. Hoje em dia, não só enfrentamos adversários mais sofisticados, como a informação que valorizam é cada vez mais alargada. Estes grupos conseguem fazer coisas inimagináveis com os bits4 mais aparentemente inócuos de informações recolhidas. Como tal, é preciso tomar medidas para garantir a segurança dos cidadãos quando navegam no ciberespaço, no qual as fronteiras são desconhecidas, onde a regulação é insuficiente e a segurança é ainda muito precoce. No plano nacional pode-se afirmar que Portugal possui as capacidades necessárias à proteção do seu ciberespaço. Com a criação do Centro Nacional de Cibersegurança (CNCS), Portugal atingiu um dos objetivos principais da sua estratégia nacional de cibersegurança, em assegurar um ciberespaço livre e seguro e em implementar as medidas e instrumentos necessários à antecipação, deteção, reação e recuperação de situações que, face à iminência ou ocorrência de incidentes ou ciberataques, ponham em causa o funcionamento dos organismos do estado, das infraestruturas críticas e dos interesses nacionais. Partindo de uma analise à estrutura organizacional da cibersegurança em Portugal este trabalho pretende dar um contributo para o que se considera ser uma necessidade, o desenvolvimento de um quadro situacional para a cibersegurança com o objetivo de melhorar o nível de awareness nacional contribuindo assim para o desenvolvimento do modelo de maturidade do CNCS relativamente á prevenção e deteção de incidentes no ciberespaço nacional. Neste sentido foram formulados um conjunto de estudos com o objetivo de dar a entender ao leitor toda a estrutura de um centro de cibersegurança na qual se destaca a proposta de desenvolvimento de um quadro situacional para a cibersegurança em Portugal.

Relación entre anchoveta y ambiente a diferentes escalas temporales. Bol Inst Mar Perú 25(1-2):13-21.- Se estudió la relación del ambiente y la anchoveta peruana (Engraulis ringens) en el Ecosistema de Afloramiento Peruano (EAP). Se hipotetiza que en la zona de afloramiento comprendida entre 5°S - 13°S se presenta una relación negativa entre la temperatura, concentración de oxígeno y desembarques de anchoveta en escalas de tiempo decadales. En series de tiempo mensuales observadas entre 1950 – 2008, se analizó la temperatura superficial, subsuperficial, oxígeno, clorofila-a y desembarques de anchoveta y sardina, aplicando métodos estadísticos y espectrales para obtener modos temporales decadales y caracterizar la modulación a baja frecuencia de sus ciclos estacionales. También se usaron datos de reanálisis para caracterizar los cambios estacionales a partir de los forzamientos (remoto y local) ambientales del EAP. Se encontró una fuerte asociación a escala decadal entre las series ambientales costeras y los desembarques de anchoveta y se sugiere que la covariabilidad ambiental en escalas temporales interanuales, intraestacionales, decadales, seculares frente al EAP se incrementará durante la próxima década.
