OBJECTIVE: To assess the frequency and severity of the anomalous origin of the left coronary artery (ALCA) from the pulmonary artery (PA). DESIGN OF THE STUDY: Prospective study of case series between March 1991 and December 1994. SETTING: Referral-based Paediatric Cardiology Department of a Tertiary Care Center. PATIENTS AND METHODS: Five consecutive patients (pts) with anomalous origin of the LCA from the PA; there were three infants aged 4 months and two children one 8 year and one 9 year old. There were three girls and two boys. All pts had clinical and 2D-echo and Doppler investigation prior to cardiac catheterization (CC). Indication for CC was based in the association of symptoms and signs of myocarditis or dilated cardiomyopathy of acute or subacute onset and electrocardiographic (ECG) signs of ischemia in infants. In older patients (pts) diagnosis was suspected mainly from ECG. During CC in all pts, aortograms and when necessary selective coronary angiograms were performed. Surgical correction was performed in all children. In two pts stress exercise ECG and stress Thallium studies before and after surgery were performed. RESULTS: two pts had "adult" an three had "infantile" type of ALCA from the PA. CC was performed and diagnosis was confirmed at surgery in all cases. In one child, correct diagnosis was made by ECO prior to CC and in one case LCA to PA fistula was suspected on Colour-Doppler study. No complications were attributed to CC. Several types of surgery were performed: reimplantation of the ALCA from the PA to the aorta (three pts); tunnel connection of the aorta to the ALCA via the PA (one pt) and left internal mammary to LCA anastomosis (one pt). Two infants died intraoperatively due to extensive myocardial infarction and poor left ventricular function. All the three survivors are asymptomatic after a mean follow up of 34 months. Two oldest pts are currently in New York Heart Association functional class I with normal ECG and improved myocardial perfusion on Thallium scan despite almost total occlusion of LCA at the site of implantation in the aorta as diagnosed on coronary angiogram. CONCLUSIONS: ALCA from PA is associated with major morbidity and mortality. Diagnosis should be suspected in pts with unexplained myocardial ischemia on ECG and even more if it is associated to clinical signs of dilated cardiomyopathy or myocarditis. Careful assessment on ECO and pulsed Doppler and colour flow mapping should make the diagnosis in most cases. Although surgery can be performed based only on ECO diagnosis, we strongly advise for angiography in all cases as in our experience there are false negative diagnosis by ECO. Preoperative Thallium studies can be useful for the selection of the type of surgery as pts with very little viable myocardium will not survive the establishment of a direct systemic to coronary blood flow and may be candidates for heart transplantation.

Com a crescente preocupação em dinamizar as exportações e potenciar os seus efeitos na economia, muitos trabalhos têm tentado encontrar fatores potenciadores do sucesso das empresas no mercado internacional (dimensão, produtividade pré exportadora, idade, fase do ciclo de produção, relacionamento prévio com o exterior, etc.). Temas como a seleção natural do mercado e a aprendizagem pela exportação, são transversais e incontornáveis nos trabalhos empíricos que abordam o estudo das exportações ao nível das empresas. No entanto, não nos devemos esquecer que uma das principais motivações das empresas, é a maximização do lucro. Com efeito, uma nova onda de trabalhos tem-se voltado para a o impacto que as exportações têm sobre a rentabilidade das empresas. Utilizando um modelo de efeitos fixos com dados em painel, aplicado a uma base de dados de empresas portuguesas, com espetro temporal entre 2008 e 2012, este trabalho encontra evidências e que as exportações são um fraco potenciador da rentabilidade das empresas. Do ponto de vista da organização do presente trabalho, no primeiro capítulo será apresentada uma breve revisão de literatura enquadradora do tema; no segundo capítulo será apresentada a base de dados, tratamento e a abordagem econométrica; por último será apresentada uma conclusão, com os resultados principais do trabalho e com algumas questões que poderão ser abordadas no futuro.

O presente relatório visa documentar a aplicação das competências adquiridas academicamente em ambiente empresarial, sob a forma de estágio curricular que decorreu na Steelgreen, S.A., que surgiu de uma parceria entre o Grupo Domingos Da Silva Teixeira (DST) e Ferlito – Ferros do Litoral, S.A.. O tema predominante deste documento será o aço, uma vez que a empresa se dedica ao corte e moldagem de armaduras. Inicialmente, será efetuada uma breve apresentação da empresa, descrevendo a sua metodologia de trabalho, pormenorizando-se todos os procedimentos executados desde a receção da matéria-prima até à sua expedição. O estágio curricular decorreu principalmente no âmbito do Departamento de Produção. Assim, neste documento enquadrar-se-á a principal atividade a cargo deste: a produção industrial. Serão explicados todos os procedimentos executados durante o processo de conceção das encomendas. A comercialização do aço requer que uma série de requisitos legais sejam cumpridos. Um capítulo deste documento será dedicado ao enquadramento desta matéria-prima na legislação em vigor. Para finalizar, serão descritos todos os trabalhos solicitados no decorrer do estágio curricular, expondo todos os procedimentos executados para atingir o objetivo proposto.

Candida albicans is an opportunistic human pathogen that is capable of causing superficial and systemic infections in immunocompromised patients. Extracts of Sapindus saponaria have been used as antimicrobial agents against various organisms. In the present study, we used a combination of two-dimensional polyacrylamide gel electrophoresis (2D-PAGE) and matrix-assisted laser desorption ionization-time of flight mass spectrometry (MALDI-TOF MS) to identify the changes in protein abundance of C. albicans after exposure to the minimal inhibitory concentration (MIC) and sub-minimal inhibitory concentration (sub-MIC) of the butanolic extract (BUTE) of S. saponaria and also to fluconazole. A total of six different proteins with greater than 1.5 fold induction or repression relative to the untreated control cells were identified among the three treatments. In general, proteins/enzymes involved with the glycolysis (GPM1, ENO1, FBA1), amino acid metabolism (ILV5, PDC11) and protein synthesis (ASC1) pathways were detected. In conclusion, our findings reveal antifungal-induced changes in protein abundance of C. albicans. By using the previously identified components of the BUTE of S. saponaria(e.g., saponins and sesquiterpene oligoglycosides), it will be possible to compare the behavior of compounds with unknown mechanisms of action, and this knowledge will help to focus the subsequent biochemical work aimed at defining the effects of these compounds.

RESUMO - O fenómeno stress relacionado com o trabalho é um tema atual, bastante debatido e alvo de diversas investigações. No entanto, os estudos portugueses sobre o stress relacionado com o trabalho em profissionais de saúde não são muito frequentes, apesar de desempenharem uma atividade considerada de alto risco em termos de stress. O síndrome de burnout é a consequência de prolongados níveis de stress relacionado com o trabalho e compreende três dimensões: exaustão emocional (EE), despersonalização (DSP) e realização pessoal (RP). O presente estudo demonstra-se como não experimental, descritivo e transversal que pretende contribuir para uma melhor compreensão do stress relacionado com o trabalho e burnout em técnicos de radiologia. Estudaram-se 73 técnicos de radiologia do Centro Hospitalar de Lisboa Central. 76,4% da população em estudo é do sexo feminino, 58,9% tem idades compreendidas entre os 41 e 60 anos, 53,4% dos profissionais são casados e 56,1% tem entre 1 e 20 anos de serviço como técnico de radiologia nos locais em estudo. Foram utilizados três questionários, um sócio-demográfico, o questionário Perceived Stress Scale (versão portuguesa do Instituto de Prevenção do Stress e Saúde Ocupacional) e o questionário Maslach Burnout Inventory (versão portuguesa traduzida pela Drª Alexandra Pinto. Verifica-se que 50,7% dos profissionais em estudo percecionam o seu nível de stress relacionado com o trabalho como elevado. Na análise da perceção do stress relacionado com o trabalho, foram os homens que surgiram com um nível mais elevado, assim como os profissionais dos 20 aos 40 anos, os solteiros e aqueles com um tempo de serviço entre 1 e 20 anos. Em relação ao burnout verificamos que 66,2% da população em estudo apresenta nível médio a elevado de EE, 57,9% de DSP e 66,2% de RP. Podemos verificar que são as mulheres que surgem com valores mais elevados de EE e RP, enquanto os homens apresentam valores mais elevados na dimensão DSP. Os mais novos (20 a 40 anos) e os solteiros apresentam nível médio a elevado de EE e DSP, enquanto os mais velhos (41 aos 60 anos) e os casados demonstram níveis mais elevados de RP. Estes dados demonstram um nível médio a elevado de burnout vivenciado pelos técnicos de radiologia dos hospitais em estudo.

PURPOSE: To introduce a new k-space traversal strategy for segmented three-dimensional echo planar imaging (3D EPI) that encodes two partitions per radiofrequency excitation, effectively reducing the number excitations used to acquire a 3D EPI dataset by half. METHODS: The strategy was evaluated in the context of functional MRI applications for: image quality compared with segmented 3D EPI, temporal signal-to-noise ratio (tSNR) (the ability to detect resting state networks compared with multislice two-dimensional (2D) EPI and segmented 3D EPI, and temporal resolution (the ability to separate cardiac- and respiration-related fluctuations from the desired blood oxygen level-dependent signal of interest). RESULTS: Whole brain images with a nominal voxel size of 2 mm isotropic could be acquired with a temporal resolution under half a second using traditional parallel imaging acceleration up to 4× in the partition-encode direction and using novel data acquisition speed-up of 2× with a 32-channel coil. With 8× data acquisition speed-up in the partition-encode direction, 3D reduced excitations (RE)-EPI produced acceptable image quality without introduction of noticeable additional artifacts. Due to increased tSNR and better characterization of physiological fluctuations, the new strategy allowed detection of more resting state networks compared with multislice 2D-EPI and segmented 3D EPI. CONCLUSION: 3D RE-EPI resulted in significant increases in temporal resolution for whole brain acquisitions and in improved physiological noise characterization compared with 2D-EPI and segmented 3D EPI. The tightest open conformations of a few prime knots (3(1), 4(1), 5(1), 5(2), and 6(3)) found with the use of the SONO algorithm are presented and discussed. The conformations are obtained from the tightest closed conformations of the knots by cutting and opening them at different locations. The length of the rope engaged in each of the open conformations is calculated. The curvature and torsion profiles of the unique tightest conformations of the 3(1) and 4(1) open knots are presented and discussed. In particular, symmetry properties of the knots reflected within their curvature and torsion profiles are analysed. Connections with the physics of polymers are discussed.

Continuation elleptique du Tristan en prose, qui s'inscrit dans l'interstice séparant la naissance de Tristan du remariage de Méliadus avec la fille du roi Hoël, le Roman de Meliadus (1235-1240) est une oeuvre fondamentalement ouverte, de par son inachèvement et de par le dialogue constant qu'il instaure avec les autres romans arthuriens. S'il revendique sa filiation et assume son statut de récit puîné, les réminiscences qu'il exhibe masquent aussi les gauchissements, les infléchissements qui lui permettent de faire du neuf avec du vieux. C'est ce jeu - aux deux sens du terme - que cette étude se propose de mettre en lumière et de voir fonctionner, non seulement dans le Roman de Meliadus proprement dit, mais également dans trois de ses relectures, qui actualisent et renouvellent la signification du roman en profondeur. La première est une continuation qui date de la toute fin du XIIIe ou du début du XIVe siècle et qui est aujourd'hui conservée par le seul manuscrit Ferrell 5. La deuxième actualisation retenue est celle qu'offre Meliadus de Leonnoys, l'imprimé publié en 1528 par Galliot du Pré, puis en 1532 par Denis Janot, fruit d'un minutieux travail de découpage et de remontage. La dernière enfin est l'extrait paru en 1776 dans la Bibliothèque Universelle des Romans sous le titre Méliadus de Léonnois. An ellipitic continuation of the Prose Tristan, which inscribes itself in the space separating the birth of Tristan from Meliadus' new marriage with king Hoël's daughter, the Meliadus' romance (1235-1240) is essentially an open text on account of its incompleteness and the dialogue it establishes with other arthurian romances. Even asserting filiation status, the reminiscences also show the reshaping and the inflection that allow the text to transform old into new. Analyzing this game is the central purpose of this work; to observe the operation in the Meliadus' romance, as well as in three of its recuperations that profoundly renew the significance of the novel; beginning with a continuation from the end of the 13th century or the early 14th century, preserved nowadays in only one manuscript (Ferrel 5); followed by the meticulous work of cutting and reassembling offered by the Meliadus of Leonnoys (printed by Galliot du Pré in 1528 first and again by Denis Janot in 1532) and finally an excerpt published in 1776 in the Bibliothèque Universelle des Romans with the title Méliadus of Leonnois.

Pig meat production was valued at €290 (£198) million at farm gate in Republic of Ireland (ROI) in 2007. In Northern Ireland (NI) in 2006, pig meat was estimated to account for almost seven percent of gross turnover in the food and drinks processing sector at £190 (€280) million. Whilst researching for this report it emerged that comparable figures for the value of the pig meat industry on ROI and NI are not available. This report showed that pig production on the IOI has changed from a small-scale enterprise carried out by a large number of mixed farmers to a modern industry comprised of a small number of specialist producers operating large-scale units. Most products for retailers are prepared and packed in specialised cutting and processing units which may or may not be integrated in the slaughter plant. For some pork products, various additives such as salt, herbs and flavour enhancers are added. Pork products are then stored and transported, frozen or chilled to wholesale, retail and catering facilities for ultimate sale to consumers.

La fibrosi pulmonar idiopàtica (FPI) és una malaltia intersticial letal, d'etiologia desconeguda i per la que no es disposa de cap tractament efectiu. Creiem que l’elasticitat normal del pulmó té propietats anti-fibròtiques, de manera que la FPI només progressarà si es produeix un enduriment sostingut del teixit. Per això hem dissenyat dos assaigs que ens permetran examinar els efectes pro-fibròtics de l'enduriment extracel•lular sobre fibroblasts primaris de pulmó. Els dos assaigs d'enduriment es basen en gels de poliacrilamida 2D i en gels de col•lagen 3D. Els fibroblasts s'han cultivat en el model d'enduriment 3D en presència o absència de la citoquina pro-fibròtica TGF-β1 i s'ha analitzat com la combinació de l’enduriment extracelular i la TGF-b1 modifiquen el fenotip de les cèl.lules. Els resultats preliminars mostren canvis en l'expressió d'alguns gens en resposta a l'enduriment i al TGF-b1, així com diferències entre cel•lules normals i fibròtiques. A més, suggereixen que l'expressió de COL1A1 i MMP-1 és mecanosensible, i que la seva desregulació podria estar associada a l’enduriment anòmal característic dels pulmons amb FPI.

Six individuals of the palm A. phalerata, in Poconé floodplains of Mato Grosso, were sprayed with a synthetic pyrethroid (0.25% concentration) in order to study the biomass, diversity, and richness of the canopy arthropods. A total of 17,188 (238.7±80.6 ind./m²) arthropods belonging to 22 Orders, was collected in a 72 m² funnel area. Two hours after spraying, 58.9% of the total number fell into the funnels, 37.6% was obtained by shaking the trees, and finally, 3.5% after cutting and washing all the palm leaves. The Coleoptera (27.4%), Hymenoptera-Formicidae (19.0%), Collembola (13.6%), Psocoptera (10.7%), Diptera (9.0%) and Araneae (6.4%) were the predominant. The total biomass was 15.1 g dry weight (0.4mg/m²; 0.13+0.04/tree). A total of 4,715 beetles representing 48 families and 326 morphospecies were obtained. Tenebrionidae (22.9%), Curculionidae (22.0%), Carabidae (10.9%) and Staphylinidae (7.9%) were the most abundant, while Curculionidae (44 spp.), Staphylinidae (40 spp.) and Chrysomelidae (34 spp.) presented the largest number of morphospecies. Herbivores (37.5%) were the dominant in the trophic guilds of adult Coleoptera, followed by predators (35.4%), fungivores (14.6%), and saprophages (12.5%). Although most arthropod Orders were represented in all the palms sampled, analysis of variance showed no significant differences in their composition, however there was a significant difference in their frequency of occurrence.

PURPOSE: Saccharopolyspora rectivirgula is the principal cause of farmer's lung disease (FLD). Serodiagnosis is based on immunoprecipitation techniques or enzyme immunoassays with homemade crude antigens and is not standardized. We aimed to produce specific recombinant antigens for the development of a standardized ELISA. EXPERIMENTAL DESIGN: We recruited 41 patients and 43 healthy exposed controls from five university hospital pneumology departments in France and Switzerland. S. rectivirgula proteins were extracted, separated by 2D electrophoresis, and subjected to Western blotting, with sera from FLD patients or controls. FLD-specific proteins were identified by MS and were produced as recombinant antigens. The diagnostic performance of ELISA tests using the recombinant antigens was assessed with all the sera from FLD patients and controls. RESULTS: We identified 25 FLD-specific proteins, some of which play important roles in transport, nutrition, or virulence. We produced 17 of these proteins as recombinant antigens and assessed their suitability for inclusion in the ELISA test. A combination of three of these proteins (SR1FA, SR17, and SR22) proved remarkably effective at discriminating between patients and controls, with a sensitivity of 83% and a specificity of 77%. CONCLUSIONS AND CLINICAL RELEVANCE: The recombinant antigens produced in this study constitute a major step toward the improvement of diagnostic performance and the standardization of FLD serodiagnosis.

The CD8(+)-T-cell response to Moloney murine leukemia virus (M-MuLV)-associated antigens in C57BL/6 mice is directed against an immunodominant gag-encoded epitope (CCLCLTVFL) presented in the context of H-2D(b) and is restricted primarily to cytotoxic T lymphocytes (CTL) expressing the Valpha3.2 and Vbeta5.2 gene segments. We decided to examine the M-MuLV response in congenic C57BL/6 Vbeta(a) mice which are unable to express the dominant Valpha3.2(+) Vbeta5.2(+) T-cell receptor (TCR) due to a large deletion at the TCR locus that includes the Vbeta5.2 gene segment. Interestingly, M-MuLV-immune C57BL/6 Vbeta(a) mice were still able to reject M-MuLV-infected tumor cells and direct ex vivo analysis of peripheral blood lymphocytes from these immune mice revealed a dramatic increase in CD8(+) cells utilizing the same Valpha3.2 gene segment in association with two different Vbeta segments (Vbeta3 and Vbeta17). Surprisingly, all these CTL recognized the same immunodominant M-MuLV gag epitope. Analysis of the TCR repertoire of individual M-MuLV-immune (C57BL/6 x C57BL/6 Vbeta(a))F(1) mice revealed a clear hierarchy in Vbeta utilization, with a preferential usage of the Vbeta17 gene segment, whereas Vbeta3 and especially Vbeta5.2 were used to much lesser extents. Sequencing of TCRalpha- and -beta-chain junctional regions of CTL clones specific for the M-MuLV gag epitope revealed a diverse repertoire of TCRbeta chains in Vbeta(a) mice and a highly restricted TCRbeta-chain repertoire in Vbeta(b) mice, whereas TCRalpha-chain sequences were highly conserved in both cases. Collectively, our data indicate that the H-2D(b)-restricted M-MuLV gag epitope can be recognized in a hierarchal fashion by different Vbeta domains and that the degree of beta-chain diversity varies according to Vbeta utilization.

Erythrocyte concentrates (ECs) are the major labile blood product being transfused worldwide, aiming at curing anemia of diverse origins. In Switzerland, ECs are stored at 4 °C up to 42 days in saline-adenine-glucose-mannitol (SAGM). Such storage induces cellular lesions, altering red blood cells (RBCs) metabolism, protein content and rheological properties. A hot debate exists regarding the impact of the storage lesions, thus the age of ECs on transfusion-related clinical adverse outcomes. Several studies tend to show that poorer outcomes occur in patients receiving older blood products. However, no clear association was demonstrated up to date. While metabolism and early rheological changes are reversible through transfusion of the blood units, oxidized proteins cannot be repaired, and it is likely such irreversible damages would affect the quality of the blood product and the efficiency of the transfusion. In vivo, RBCs are constantly exposed to oxygen fluxes, and are thus well equipped to deal with oxidative challenges. Moreover, functional 20S proteasome complexes allow for recognition and proteolysis of fairly oxidized protein, and some proteins can be eliminated from RBCs by the release of microvesicles. The present PhD thesis is involved in a global research project which goal is to characterize the effect of processing and storage on the quality of ECs. Assessing protein oxidative damages during RBC storage is of major importance to understand the mechanisms of aging of stored RBCs. To this purpose, redox proteomic-based investigations were conducted here. In a first part, cysteine oxidation and protein carbonylation were addressed via 2D-DIGE and derivatization-driven immunodetection approaches, respectively. Then, the oxidized sub- proteomes were characterized through LC-MS/MS identification of proteins in spots of interest (cysteine oxidation) or affinity-purified carbonylated proteins. Gene ontology annotation allowed classifying targets of oxidation according to their molecular functions. In a third part, the P20S activity was evaluated throughout the storage period of ECs, and its susceptibility to highly oxidized environment was investigated. The potential defensive role of microvesiculation was also addressed through the quantification of eliminated carbonylated proteins. We highlighted distinct protein groups differentially affected by cysteine oxidation, either reversibly or irreversibly. In addition, soluble extracts showed a decrease in carbonylation at the beginning of the storage and membrane extracts revealed increasing carbonylation after 4 weeks of storage. Engaged molecular functions revealed that antioxidant (AO) are rather reversibly oxidized at their cysteine residue(s), but are irreversibly oxidized through carbonylation. In the meantime, the 20S proteasome activity is decreased by around 40 % at the end of the storage period. Incubation of fresh RBCs extracts with exogenous oxidized proteins showed a dose-dependent and protein-dependent inhibitory effect. Finally, we proved that the release of microvesicles allows the elimination of increasing quantities of carbonylated proteins. Taken together, these results revealed an oxidative pathway model of RBCs storage, on which further investigation towards improved storage conditions will be based. -- Les concentrés érythrocytaires (CE) sont le produit sanguin le plus délivré au monde, permettant de traiter différentes formes d'anémies. En Suisse, les CE sont stocké à 4 °C pendant 42 jours dans une solution saline d'adénine, glucose et mannitol (SAGM). Une telle conservation induit des lésions de stockage qui altèrent le métabolisme, les protéines et les propriétés rhéologique du globule rouge (GR). Un débat important concerne l'impact du temps de stockage des CE sur les risques de réaction transfusionnelles, certaines études tentant de démontrer que des transfusions de sang vieux réduiraient l'espérance de vie des patients. Cependant, aucune association concrète n'a été prouvée à ce jour. Alors que les modifications du métabolisme et changement précoces des propriétés rhéologiques sont réversibles suite à la transfusion du CE, les protéines oxydées ne peuvent être réparées, et il est probable que de telles lésions affectent la qualité et l'efficacité des produits sanguins. In vivo, les GR sont constamment exposés à l'oxygène, et sont donc bien équipés pour résister aux lésions oxydatives. De plus, les complexes fonctionnels de proteasome 20S reconnaissent et dégradent les protéines modérément oxydées, et certaines protéines peuvent être éliminées par les microparticules. Cette thèse de doctorat est imbriquée dans un projet de recherche global ayant pour objectif la caractérisation des effets de la préparation et du stockage sur la qualité des GR. Evaluer les dommages oxydatifs du GR pendant le stockage est primordial pour comprendre les mécanismes de vieillissement des produits sanguin. Dans ce but, des recherches orientées redoxomique ont été conduites. Dans une première partie, l'oxydation des cystéines et la carbonylation des protéines sont évaluées par électrophorèse bidimensionnelle différentielle et par immunodétection de protéines dérivatisées. Ensuite, les protéines d'intérêt ainsi que les protéines carbonylées, purifiées par affinité, sont identifiées par spectrométrie de masse en tandem. Les protéines cibles de l'oxydation sont classées selon leur fonction moléculaire. Dans une troisième partie, l'activité protéolytique du protéasome 20S est suivie durant la période de stockage. L'impact du stress oxydant sur cette activité a été évalué en utilisant des protéines exogènes oxydées in vitro. Le potentiel rôle défensif de la microvesiculation a également été étudié par la quantification des protéines carbonylées éliminées. Dans ce travail, nous avons observé que différents groupes de protéines sont affectés par l'oxydation réversible ou irréversible de leurs cystéines. De plus, une diminution de la carbonylation en début de stockage dans les extraits solubles et une augmentation de la carbonylation après 4 semaines dans les extraits membranaires ont été montrées. L'incubation d'extraits de GR en début de stockage avec des protéines oxydées exogènes montre un effet inhibiteur « dose-dépendant » et « protéine-dépendant ». Enfin, les microvésicules s'avèrent éliminer des quantités croissantes de protéines carbonylées. La synthèse de ces résultats permet de modéliser une voie oxydative du stockage des GRs, à partir de laquelle de futures recherches seront menées avec pour but l'amélioration des conditions de stockage.
