O objectivo principal deste artigo é exemplificar a utilização de uma metodologia de especificação de sistemas digitais, baseada em Redes de Petri orientadas por objectos, para obter de uma forma rápida e simplificada um protótipo em VHDL do sistema pretendido. É considerado como exemplo um sistema digital, para o qual se efectua a especificação no modelo RdP-shobi e a partir da qual se pode gerar automaticamente código VHDL. Este exemplo permite demonstrar acerca da capacidade desta metodologia no projecto de sistemas digitais, suportado por princípios de orientação por objectos e por uma ferramenta de EDA concebida para o efeito.

The new complex, [Zr(pda)2]n (1, pda2- = N,N'-bis(neo-pentyl)-ortho-phenylenediamide, n = 1 or 2), prepared by the reaction of 2 equiv of pdaLi2 with ZrCl4, reacts rapidly with halogen oxidants to afford the new product ZrX2(disq)2 (3, X = Cl, Br, I; disq- = N,N'-bis(neo-pentyl)-ortho-diiminosemiquinonate) in which each redox-active ligand has been oxidized by one electron. The oxidation products 3a-c have been structurally characterized and display an unusual parallel stacked arrangement of the disq- ligands in the solid state, with a separation of approximately 3 A. Density functional calculations show a bonding-type interaction between the SOMOs of the disq- ligands to form a unique HOMO while the antibonding linear combination forms a unique LUMO. This orbital configuration leads to a closed-shell-singlet ground-state electron configuration (S = 0). Temperature-dependent magnetism measurements indicate a low-lying triplet excited state at approximately 750 cm-1. In solution, 3a-c show strong disq--based absorption bands that are invariant across the halide series. Taken together these spectroscopic measurements provide experimental values for the one- and two-electron energies that characterize the pi-stacked bonding interaction between the two disq- ligands.

Electricity price forecasting has become an important area of research in the aftermath of the worldwide deregulation of the power industry that launched competitive electricity markets now embracing all market participants including generation and retail companies, transmission network providers, and market managers. Based on the needs of the market, a variety of approaches forecasting day-ahead electricity prices have been proposed over the last decades. However, most of the existing approaches are reasonably effective for normal range prices but disregard price spike events, which are caused by a number of complex factors and occur during periods of market stress. In the early research, price spikes were truncated before application of the forecasting model to reduce the influence of such observations on the estimation of the model parameters; otherwise, a very large forecast error would be generated on price spike occasions. Electricity price spikes, however, are significant for energy market participants to stay competitive in a market. Accurate price spike forecasting is important for generation companies to strategically bid into the market and to optimally manage their assets; for retailer companies, since they cannot pass the spikes onto final customers, and finally, for market managers to provide better management and planning for the energy market. This doctoral thesis aims at deriving a methodology able to accurately predict not only the day-ahead electricity prices within the normal range but also the price spikes. The Finnish day-ahead energy market of Nord Pool Spot is selected as the case market, and its structure is studied in detail. It is almost universally agreed in the forecasting literature that no single method is best in every situation. Since the real-world problems are often complex in nature, no single model is able to capture different patterns equally well. Therefore, a hybrid methodology that enhances the modeling capabilities appears to be a possibly productive strategy for practical use when electricity prices are predicted. The price forecasting methodology is proposed through a hybrid model applied to the price forecasting in the Finnish day-ahead energy market. The iterative search procedure employed within the methodology is developed to tune the model parameters and select the optimal input set of the explanatory variables. The numerical studies show that the proposed methodology has more accurate behavior than all other examined methods most recently applied to case studies of energy markets in different countries. The obtained results can be considered as providing extensive and useful information for participants of the day-ahead energy market, who have limited and uncertain information for price prediction to set up an optimal short-term operation portfolio. Although the focus of this work is primarily on the Finnish price area of Nord Pool Spot, given the result of this work, it is very likely that the same methodology will give good results when forecasting the prices on energy markets of other countries.

Työssä tutkittiin Salossa sijaitsevan Korvenmäen kaatopaikan kaasunkeräysjärjestelmää. Tavoitteena oli selvittää kaatopaikkakaasun talteenoton merkittävimmät haasteet, löytää ratkaisuja ongelmiin ja hajuhaittojen vähentämiseen sekä arvioida keräysjärjestelmän parannusten ja kaatopaikalle tulevan pintarakenteen vaikutus talteenottoon ja hajuhaittoihin. Korvenmäen kaatopaikalla on käytössä vaakakeräysjärjestelmä, jonka merkittävimpiä ongelmia ovat olleet kaatopaikkakaasun matala talteenottoaste, happipitoisuuden nousu, järjestelmän hankala säädettävyys sekä kaatopaikalla ja ympäristössä havaitut hajuhaitat. Muodostuvan ja karkaavan kaatopaikkakaasun määrää arvioitiin Jouko Petäjän metaanilaskentamallin avulla. Tulosten perusteella kaasua muodostuu runsaasti, mutta suurin osa siitä pääsee karkaamaan ympäristöön, sillä arvioitu kaatopaikkakaasun talteenottoaste on tällä hetkellä vain 11 %. Kaasujen käsittelykäytäntöjä suomalaisilla kaatopaikoilla selvitettiin kyselytutkimuksella. Tutkimuksen mukaan kaasunkeräyksen ongelmat ja hajuhaitat ovat yleisiä, mutta erilaisilla toimenpiteillä niitä voidaan huomattavasti vähentää. Korvenmäelle suositeltavia kehitystoimia ovat kaasulinjoihin yhdistettyjen suotoveden keräys- ja tarkkailukaivojen tiivistäminen, vanhojen vaakalinjojen kunnon tarkistus ja korjaus sekä pystykaivojen asentaminen kaasunkeräyksen tehostamiseksi. Pintarakenne tulee valmistuttuaan vähentämään karkaavan kaasun ja kaasulinjoihin imeytyvän ilman määrää. Hajuhaittojen hallinnassa tärkeintä on toimiva kaasunkeräys, mutta lisäksi suositellaan rikkivetyä hapettavien päivittäispeittomateriaalien levittämistä kaatopaikan pinnalle. Kehitystoimenpiteiden myötä kaatopaikkakaasun talteenottoaste tulee paranemaan ja hajuhaitat vähenevät. Kerättävän kaasumäärän kasvaessa mikroturbiinilaitoksen kapasiteettia voidaan lisätä, jotta kaasu saadaan mahdollisimman tehokkaasti hyödynnettyä energiana.

A ground source heat pump assisted by an array of photovoltaic (PV)-thermal modules was studied in this work. Extracting heat from an array of PV modules should improve the performance of both the PV cells and the heat pump. A series of computer simulations compare the performance of a ground source heat pump with a short ground circuit, used to provide space heating and domestic hot water at a house in southern England. The results indicate that extracting heat from an array of PV-thermal modules would improve the performance of a ground source heat pump with an undersized ground loop. Nevertheless, open air thermal collectors could be more effective, especially during winter. In one model more electricity was saved in ohmic heating than was generated by cooling the PV cells. Cooling the PV modules was found to increase their electrical output up to 4%, but much of the extra electricity was consumed by the cooling pumps.

5-HT(1A) receptor plays an important role in the delayed onset of antidepressant action of a class of selective serotonin reuptake inhibitors. Moreover, 5-HT(1A) receptor levels have been shown to be altered in patients suffering from major depression. In this work, hologram quantitative structure-activity relationship (HQSAR) studies were performed on a series of arylpiperazine compounds presenting affinity to the 5-HT(1A) receptor. The models were constructed with a training set of 70 compounds. The most significant HQSAR model (q(2) = 0.81, r(2) = 0.96) was generated using atoms, bonds, connections, chirality, and donor and acceptor as fragment distinction, with fragment size of 6-9. Predictions for an external test set containing 20 compounds are in good agreement with experimental results showing the robustness of the model. Additionally, useful information can be obtained from the 2D contribution maps.

This research was commenced with the idea to promote an strategic evaluation of the sustentability of the free Internet access, analysing its process of value creation, its sources of revenue generation, its business models development as well as its capacity to generate adequated returns to the investors. It was concluded that the free Internet access is not dead. Indeed, what has occoured was a migration from a model completely based on delivering a single service, generally dependent of publicity sells revenues, to a hibrid based business model with different types of services and with revenues been generated from multiple sources. Nevertheless, not all players were capable to evolute their business models and survive, what occasioned a huge number of fails, mergers and acquisitions, resulting in a concentration all over the segment.

Background: Considering that an increasing number of patients are victims of mutilator surgical resections, these studies are important for treatment success of rehabilitation of patients presenting oronasal communication.Purpose: The aim of this study was to assess the stress distribution through photoelasticity in palatal obturator prostheses with different attachment systems for implants.Methods: Two photoelastic models were obtained from an experimental maxillary model presenting an oronasal communication. One model was fabricated without implant, and the other with 2 implants 10 mm in length inserted in the left crest. Four colorless palatal obturator prostheses were fabricated. One prosthesis presented no attachment system, whereas the remaining prostheses were adapted to 3 attachment systems. The assembly was positioned in a circular polariscope for application of axial load.Results: The results were based on photographic records of stress in the photoelastic model submitted to loading. The records revealed higher stress concentration on the bar-clip system followed by the O'ring/bar-clip and O'ring systems, respectively. A homogeneous stress distribution was observed on the photoelastic model with the mucous-supported prosthesis.Conclusions: The attachment systems generated different characteristics of stress distribution that was concentrated surrounding the implants. The bar-clip system exhibited the highest stress concentration on the alveolar crest.

É possível que organismos apresentem uma capacidade relacional básica. Sendo assim, relações de equivalência seriam geradas por contingências de reforçamento. Resultados inconsistentes obtidos em testes de formação de classes de equivalência seriam devido ao desenvolvimento de procedimentos experimentais insuficientes no sentido de gerar o desempenho planejado. Tomando como base a hipótese de que não apenas os estímulos antecedentes, mas também as respostas e estímulos reforçadores, se específicos para cada classe, tornam-se membros das mesmas, o presente trabalho avaliou o desempenho de um macaco-prego (Cebus apella), com história pré-experimental de discriminações condicionais de identidade e arbitrárias e uso de comparações vazias (máscaras), em testes de simetria BA após treino de discriminações condicionais com reforçamento específico, pelotas de alimento de sabor diferente para cada discriminação. Durante o treino foi usado o procedimento de máscara para garantir relações de controle coerentes com as planejadas. Após o treino, foi realizado o teste de simetria com reforçamento específico, cujos resultados foram inconsistentes com a formação de classes. É possível que o sujeito necessitasse de mais sessões de treino com reforçadores específicos e/ou que as similaridades entre os estímulos reforçadores de cada classe tenha gerado uma generalização entre eles, o que os tornaria não reforçadores específicos, mas sim, reforçadores comuns.

In der vorliegenden Arbeit wurden die bioinformatischen Methoden der Homologie-Modellierung und Molekularen Modellierung dazu benutzt, die dreidimensionalen Strukturen verschiedenster Proteine vorherzusagen und zu analysieren. Experimentelle Befunde aus Laborversuchen wurden dazu benutzt, die Genauigkeit der Homologie-Modelle zu erhöhen. Die Ergebnisse aus den Modellierungen wurden wiederum dazu benutzt, um neue experimentelle Versuche vorzuschlagen. Anhand der erstellten Modelle und bekannten Kristallstrukturen aus der Protein-Datenbank PDB wurde die Struktur-Funktionsbeziehung verschiedener Tyrosinasen untersucht. Dazu gehörten sowohl die Tyrosinase des Bakteriums Streptomyces als auch die Tyrosinase der Hausmaus. Aus den vergleichenden Strukturanalysen der Tyrosinasen resultierten Mechanismen für die Monophenolhydroxylase-Aktivität der Tyrosinasen sowie für den Import der Kupferionen ins aktive Zentrum. Es konnte der Beweis geführt werden, daß die Blockade des CuA-Zentrums tatsächlich der Grund für die unterschiedliche Aktivität von Tyrosinasen und Catecholoxidasen ist. Zum ersten Mal konnte mit der Maus-Tyrosinase ein vollständiges Strukturmodell einer Säugetier-Tyrosinase erstellt werden, das dazu in der Lage ist, die Mechanismen bekannter Albino-Mutationen auf molekularer Ebene zu erklären. Die auf der Basis des ermittelten 3D-Modells gewonnenen Erkenntnisse über die Wichtigkeit bestimmter Aminosäuren für die Funktion wurde durch gerichtete Mutagenese an der rekombinant hergestellten Maus-Tyrosinase getestet und bestätigt. Weiterhin wurde die Struktur der Tyrosinase des Krebses Palinurus elephas durch eine niedrigaufgelöste 3D-Rekonstruktion aus elektronenmikroskopischen Bildern aufgeklärt. Der zweite große Themenkomplex umfasst die Strukturanalyse der Lichtsammlerkomplexe LHCI-730 und LHCII. Im Falle des LHCII konnte der Oligomerisierungszustand der LHCMoleküle mit diskreten Konformationen des N-Terminus korreliert werden. Auch hier kam eine Kombination von Homologie-Modellierung und einer experimentellen Methode, der Elektronen-Spin-Resonanz-Messung, zum Einsatz. Die Änderung des Oligomerisierungszustands des LHCII kontrolliert den Energiezufluß zu den Photosystemen PS I und PS II. Des Weiteren wurde ein vollständiges Modell des LHCI-730 erstellt, um die Auswirkungen gerichteter Mutagenese auf das Dimerisierungsverhalten zu untersuchen. Auf Basis dieses Modells wurden die Wechselwirkungen zwischen den Monomeren Lhca1 und Lhca4 evaluiert und potentielle Bindungspartner identifiziert.

This qualitative study examines the attributes or perceptions of service providers and overseers as to the effectiveness of intensive family preservation services provided by a social services agency in Tucson, Arizona. The services provided are patterned after the Homebuilders' model developed in 1974 in Tacoma, Washington. Data collection was generated from interviews and focus groups with the in-home service providers, the program supervisor, and investigators and case managers with Child Protective Services (CPS). Although placement prevention rates (PPR) are the dependent variable in most studies on this form of intervention, this study seeks to understand those characteristics of the model that contribute to successful outcomes with client families. Those appear to be the short-term intervention coupled with a non-judgmental approach to client families and the clinical supervision provided by the program supervisor.

The recent determination of the myosin head atomic structure has led to a new model of muscle contraction, according to which mechanical torque is generated in the catalytic domain and amplified by the lever arm made of the regulatory domain [Fisher, A. J., Smith, C. A., Thoden, J., Smith, R., Sutoh, K., Holden, H. M. & Rayment, I. (1995) Biochemistry 34, 8960–8972]. A crucial aspect of this model is the ability of the regulatory domain to move independently of the catalytic domain. Saturation transfer–EPR measurements of mobility of these two domains in myosin filaments give strong support for this notion. The catalytic domain of the myosin head was labeled at Cys-707 with indane dione spin label; the regulatory domain was labeled at the single cysteine residue of the essential light chain and exchanged into myosin. The mobility of the regulatory domain in myosin filaments was characterized by an effective rotational correlation time (τR) between 24 and 48 μs. In contrast, the mobility of the catalytic domain was found to be τR = 5–9 μs. This difference in mobility between the two domains existed only in the filament form of myosin. In the monomeric form, or when bound to actin, the mobility of the two domains in myosin was indistinguishable, with τR = 1–4 μs and >1,000 μs, respectively. Therefore, the observed difference in filaments cannot be ascribed to differences in local conformations of the spin-labeled sites. The most straightforward interpretation suggests a flexible hinge between the two domains, which would have to stiffen before force could be generated.

Progress toward understanding the biology of prostate cancer has been slow due to the few animal research models available to study the spectrum of this uniquely human disease. To develop an animal model for prostate cancer, several lines of transgenic mice were generated by using the prostate-specific rat probasin promoter to derive expression of the simian virus 40 large tumor antigen-coding region. Mice expressing high levels of the transgene display progressive forms of prostatic disease that histologically resemble human prostate cancer, ranging from mild intraepithelial hyperplasia to large multinodular malignant neoplasia. Prostate tumors have been detected specifically in the prostate as early as 10 weeks of age. Immunohistochemical analysis of tumor tissue has demonstrated that dorsolateral prostate-specific secretory proteins were confined to well-differentiated ductal epithelial cells adjacent to, or within, the poorly differentiated tumor mass. Prostate tumors in the mice also display elevated levels of nuclear p53 and a decreased heterogeneous pattern of androgen-receptor expression, as observed in advanced human prostate cancer. The establishment of breeding lines of transgenic mice that reproducibly develop prostate cancer provides an animal model system to study the molecular basis of transformation of normal prostatic cells and the factors influencing the progression to metastatic prostate cancer.
