O tomateiro (Solanum lycopersicum L. = Lycopersicon esculentum Mill.) é uma das plantas oleráceas mais cultivadas no Brasil e o seu fruto, o tomate, é uma das olerícolas mais consumidas na mesa do brasileiro e muito usado pela agroindústria. Uma das doenças do tomateiro mais temidas pelos produtores é a requeima, causada pelo fungo Phytophtora infestans, que provoca grande destruição na cultura em pouco tempo. Este trabalho teve o objetivo de caracterizar isolados de P. infestans, coletados de tomateiros, quanto ao grupo de compatibilidade, à viruªncia e à resistência ao fungicida mefenoxan. Vinte e seis isolados foram caracterizados quanto ao grupo de compatibilidade; 24 foram caracterizados quanto à resistência ao mefenoxan; e determinou-se o espectro de viruªncia de 14 isolados. Todos os 26 isolados testados foram classificados como do grupo A1 de compatibilidade. Em relação à viruªncia, todos os isolados foram virulentos à cultivar de tomate 'IPA-5'. A maioria foi virulenta em plantas de tomate com os genes Ph1 (92,86%) ou Ph2 (78,57%) e uma pequena parte dos isolados foram virulentos em plantas com o gene Ph3 (21,43%). Quanto à resistência ao mefenoxan, a freqüência de isolados sensíveis, intermediários e resistentes foram de 16,67%, 16,67% e 66,66%, respectivamente. Há evidências da não-reprodução sexuada e formação de oósporos nos campos de tomate do país, retardando o aparecimento da doença. Devido a um maior número de isolados resistentes ao mefenoxan, o controle da requeima com este fungicida pode ser ineficiente em campo. Uma vez que o patógeno apresentou um amplo espectro de viruªncia, a utilização de cultivares com resistência vertical a requeima não é recomendada. O manejo integrado da requeima é a forma mais eficaz de se controlar a doença.

The main objective of the present work is the study of a profitable process not only in the extraction and selective separation of lycopene and β-carotene, two compounds present in tomato, but also in its potential application to food industry wastes. This is one of the industries that produce larger amounts of wastes, which are rich in high value biomolecules with great economic interest. However, the conventional methods used to extract this kind of compounds are expensive which limits their application at large scale. Lycopene and βcarotene are carotenoids with high commercial value, known for their antioxidant activity and benefits to human health. Their biggest source is tomato, one of the world’s most consumed fruits, reason for which large quantities of waste is produced. This work focuses on the study of diverse solvents with a high potential to extract carotenoids from tomato, as well as the search for more environmentally benign solvents than those currently used to extract lycopene and β-carotene from biomass. Additionally, special attention was paid to the creation of a continuous process that would allow the fractionation of the compounds for further purification. Thus, the present work started with the extraction of both carotenoids using a wide range of solvents, namely, organic solvents, conventional salts, ionic liquids, polymers and surfactants. In this stage, each solvent was evaluated in what regards their capacity of extraction as well as their penetration ability in biomass. The results collected showed that an adequate selection of the solvents may lead to the complete extraction of both carotenoids in one single step, particularly acetone and tetrahydrofuran were the most effective ones. However, the general low penetration capacity of salts, ionic liquids, polymers and surfactants makes these solvents ineffective in the solid-liquid extraction process. As the organic solvents showed the highest capacity to extract lycopene and βcarotene, in particular tetrahydrofuran and acetone, the latter solvent used in the development process of fractionation, using to this by strategic use of solvents. This step was only successfully developed through the manipulation of the solubility of each compound in ethanol and n-hexane. The results confirmed the possibility of fractionating the target compounds using the correct addition order of the solvents. Approximately, 39 % of the β-carotene was dissolved in ethanol and about 64 % of lycopene was dissolved in n-hexane, thus indicating their separation for two different solvents which shows the selective character of the developed process without any prior stage optimization. This study revealed that the use of organic solvents leads to selective extraction of lycopene and β-carotene, allowing diminishing the numerous stages involved in conventional methods. At the end, it was possible to idealize a sustainable and of high industrial relevance integrated process, nevertheless existing the need for additional optimization studies in the future.

Les peroxyrédoxines (PRXs) forment une famille de peroxydases communes à tous les organismes vivants et ubiquitaires dans la cellule. Leur particularité provient d’un ou deux résidus cystéines accomplissant un cycle d’oxydo-réduction à €™aide d’un donneur d’électron. Ces protéines thiols sensibles au potentiel redox sont impliquées dans le mécanisme de détoxification du H2O2, une mo©cule oxydante induite lors de situations de stress. Les PRXs pourraient être induites par le stress et régu©es par phosphorylation. En effet, des expérimentations in vitro ont démontré que la nuc©oside diphosphate kinase 1 (NDPK1) a la capacité de phosphoryler une PRX cytosolique de pomme de terre. Ce mémoire décrit les travaux expérimentaux effectués pour caractériser la fonction de la PRX. Pour cela, le clonage d’une isoforme a été effectué, suivi d’une caractérisation biochimique et d’une étude d’expression de la protéine. Les données de séquençage révèlent qu’il s’agit d’une PRX de type II phylogénétiquement liée aux PRXs cytosoliques. €™ADNc codant pour cette peroxyrédoxine (PRX1) a été cloné chez Solanum chacoense. Une protéine recombinante portant une étiquette (6xHis) en N-terminale a été produite. Des essais enzymatiques ont confirmé la fonction antioxydante de la protéine recombinante et un anticorps polyclonal a été généré chez le lapin puis utilisé en conjonction avec un anticorps anti-NDPK1 pour déterminer les patrons d’expression généraux de ces protéines chez Solanum lycopersicum et Solanum tuberosum lors de situations de stress. Les données démontrent que les deux protéines sont généralement co-exprimées mais pas co-régu©es et que la PRX1 est induite en certaines situations de stress.

The combined effects of shoot pruning (one or two stems) and inflorescence thinning (five or ten flowers per inflorescence) on greenhouse tomato yield and fruit quality were studied during the dry season (DS) and rainy season (RS) in Central Thailand. Poor fruit set, development of undersized (mostly parthenocarpic) fruits, as well as the physiological disorders blossom-end rot (BER) and fruit cracking (FC) turned out to be the prevailing causes deteriorating fruit yield and quality. The proportion of marketable fruits was less than 10% in the RS and around 65% in the DS. In both seasons, total yield was significantly increased when plants were cultivated with two stems, resulting in higher marketable yields only in the DS. While the fraction of undersized fruits was increased in both seasons when plants were grown with a secondary stem, the proportions of BER and FC were significantly reduced. Restricting the number of flowers per inflorescence invariably resulted in reduced total yield. However, in neither season did fruit load considerably affect quantity or proportion of the marketable yield fraction. Inflorescence thinning tended to promote BER and FC, an effect which was only significant for BER in the RS. In conclusion, for greenhouse tomato production under climate conditions as they are prevalent in Central Thailand, the cultivation with two stems appears to be highly recommendable whereas the measures to control fruit load tested in this study did not proof to be advisable.
