I moderni sistemi embedded sono equipaggiati con risorse hardware che consentono l’esecuzione di applicazioni molto complesse come il decoding audio e video. La progettazione di simili sistemi deve soddisfare due esigenze opposte. Da un lato è necessario fornire un elevato potenziale computazionale, dall’altro bisogna rispettare dei vincoli stringenti riguardo il consumo di energia. Uno dei trend più diffusi per rispondere a queste esigenze opposte è quello di integrare su uno stesso chip un numero elevato di processori caratterizzati da un design semplificato e da bassi consumi. Tuttavia, per sfruttare effettivamente il potenziale computazionale offerto da una batteria di processoriè necessario rivisitare pesantemente le metodologie di sviluppo delle applicazioni. Con l’avvento dei sistemi multi-processore su singolo chip (MPSoC) il parallel programming si è diffuso largamente anche in ambito embedded. Tuttavia, i progressi nel campo della programmazione parallela non hanno mantenuto il passo con la capacità di integrare hardware parallelo su un singolo chip. Oltre all’introduzione di multipli processori, la necessità di ridurre i consumi degli MPSoC comporta altre soluzioni architetturali che hanno l’effetto diretto di complicare lo sviluppo delle applicazioni. Il design del sottosistema di memoria, in particolare, è un problema critico. Integrare sul chip dei banchi di memoria consente dei tempi d’accesso molto brevi e dei consumi molto contenuti. Sfortunatamente, la quantità di memoria on-chip che può essere integrata in un MPSoC è molto limitata. Per questo motivo è necessario aggiungere dei banchi di memoria off-chip, che hanno una capacità molto maggiore, come maggiori sono i consumi e i tempi d’accesso. La maggior parte degli MPSoC attualmente in commercio destina una parte del budget di area all’implementazione di memorie cache e/o scratchpad. Le scratchpad (SPM) sono spesso preferite alle cache nei sistemi MPSoC embedded, per motivi di maggiore predicibilità, minore occupazione d’area e – soprattutto – minori consumi. Per contro, mentre l’uso delle cache è completamente trasparente al programmatore, le SPM devono essere esplicitamente gestite dall’applicazione. Esporre l’organizzazione della gerarchia di memoria ll’applicazione consente di sfruttarne in maniera efficiente i vantaggi (ridotti tempi d’accesso e consumi). Per contro, per ottenere questi benefici è necessario scrivere le applicazioni in maniera tale che i dati vengano partizionati e allocati sulle varie memorie in maniera opportuna. L’onere di questo compito complesso ricade ovviamente sul programmatore. Questo scenario descrive bene l’esigenza di modelli di programmazione e strumenti di supporto che semplifichino lo sviluppo di applicazioni parallele. In questa tesi viene presentato un framework per lo sviluppo di software per MPSoC embedded basato su OpenMP. OpenMP è uno standard di fatto per la programmazione di multiprocessori con memoria shared, caratterizzato da un semplice approccio alla parallelizzazione tramite annotazioni (direttive per il compilatore). La sua interfaccia di programmazione consente di esprimere in maniera naturale e molto efficiente il parallelismo a livello di loop, molto diffuso tra le applicazioni embedded di tipo signal processing e multimedia. OpenMP costituisce un ottimo punto di partenza per la definizione di un modello di programmazione per MPSoC, soprattutto per la sua semplicità d’uso. D’altra parte, per sfruttare in maniera efficiente il potenziale computazionale di un MPSoC è necessario rivisitare profondamente l’implementazione del supporto OpenMP sia nel compilatore che nell’ambiente di supporto a runtime. Tutti i costrutti per gestire il parallelismo, la suddivisione del lavoro e la sincronizzazione inter-processore comportano un costo in termini di overhead che deve essere minimizzato per non comprometterre i vantaggi della parallelizzazione. Questo può essere ottenuto soltanto tramite una accurata analisi delle caratteristiche hardware e l’individuazione dei potenziali colli di bottiglia nell’architettura. Una implementazione del task management, della sincronizzazione a barriera e della condivisione dei dati che sfrutti efficientemente le risorse hardware consente di ottenere elevate performance e scalabilità. La condivisione dei dati, nel modello OpenMP, merita particolare attenzione. In un modello a memoria condivisa le strutture dati (array, matrici) accedute dal programma sono fisicamente allocate su una unica risorsa di memoria raggiungibile da tutti i processori. Al crescere del numero di processori in un sistema, l’accesso concorrente ad una singola risorsa di memoria costituisce un evidente collo di bottiglia. Per alleviare la pressione sulle memorie e sul sistema di connessione vengono da noi studiate e proposte delle tecniche di partizionamento delle strutture dati. Queste tecniche richiedono che una singola entità di tipo array venga trattata nel programma come l’insieme di tanti sotto-array, ciascuno dei quali può essere fisicamente allocato su una risorsa di memoria differente. Dal punto di vista del programma, indirizzare un array partizionato richiede che ad ogni accesso vengano eseguite delle istruzioni per ri-calcolare l’indirizzo fisico di destinazione. Questo è chiaramente un compito lungo, complesso e soggetto ad errori. Per questo motivo, le nostre tecniche di partizionamento sono state integrate nella l’interfaccia di programmazione di OpenMP, che è stata significativamente estesa. Specificamente, delle nuove direttive e clausole consentono al programmatore di annotare i dati di tipo array che si vuole partizionare e allocare in maniera distribuita sulla gerarchia di memoria. Sono stati inoltre sviluppati degli strumenti di supporto che consentono di raccogliere informazioni di profiling sul pattern di accesso agli array. Queste informazioni vengono sfruttate dal nostro compilatore per allocare le partizioni sulle varie risorse di memoria rispettando una relazione di affinità tra il task e i dati. Più precisamente, i passi di allocazione nel nostro compilatore assegnano una determinata partizione alla memoria scratchpad locale al processore che ospita il task che effettua il numero maggiore di accessi alla stessa.

Graphene and graphenic derivatives have rapidly emerged as an extremely promising system for electronic, optical, thermal, and electromechanical applications. Several approaches have been developed to produce these materials (i.e. scotch tape, CVD, chemical and solvent exfoliation). In this work we report a chemical approach to produce graphene by reducing graphene oxide (GO) via thermal or electrical methods. A morphological and electrical characterization of these systems has been performed using different techniques such as SPM, SEM, TEM, Raman and XPS. Moreover, we studied the interaction between graphene derivates and organic molecules focusing on the following aspects: - improvement of optical contrast of graphene on different substrates for rapid monolayer identification1 - supramolecular interaction with organic molecules (i.e. thiophene, pyrene etc.)4 - covalent functionalization with optically active molecules2 - preparation and characterization of organic/graphene Field Effect Transistors3-5 Graphene chemistry can potentially allow seamless integration of graphene technology in organic electronics devices to improve device performance and develop new applications for graphene-based materials. The development and characterization of biomolecule sensor formats based on the optical technique Surface Plasmon Resonance (SPR) Spectroscopy and electrochemical methods were investigated. The study can be divided into two parts of different scope. In the first part new novel detection schemes for labeled targets were developed on the basis of the investigations in Surface-plamon Field Enhanced Spectroscopy (SPFS). The first one is SPR fluorescence imaging formats, Surface-plamon Field Enhanced Fluorescence Microscopy (SPFM). Patterned self assembled monolayers (SAMs) were prepared and used to direct the spatial distribution of biomolecules immobilized on surfaces. Here the patterned monolayers would serve as molecular templates to secure different biomolecules to known locations on a surface. The binding processed of labeled target biomolecules from solution to sensor surface were visually and kinetically recorded by the fluorescence microscope, in which fluorescence was excited by the evanescent field of propagating plasmon surface polaritons. The second format which also originates from SPFS technique, Surface-plamon Field Enhanced Fluorescence Spectrometry (SPFSm), concerns the coupling of a fluorometry to normal SPR setup. A spectrograph mounted in place of photomultiplier or microscope can provide the information of fluorescence spectrum as well as fluorescence intensity. This study also firstly demonstrated the analytical combination of surface plasmon enhanced fluorescence detection with analyte tagged by semiconducting nano- crystals (QDs). Electrochemically addressable fabrication of DNA biosensor arrays in aqueous environment was also developed. An electrochemical method was introduced for the directed in-situ assembly of various specific oligonucleotide catcher probes onto different sensing elements of a multi-electrode array in the aqueous environment of a flow cell. Surface plasmon microscopy (SPM) is utilized for the on-line recording of the various functionalization steps. Hybridization reactions between targets from solution to the different surface-bound complementary probes are monitored by surface-plasmon field-enhanced fluorescence microscopy (SPFM) using targets that are either labeled with organic dyes or with semiconducting quantum dots for color-multiplexing. This study provides a new approach for the fabrication of (small) DNA arrays and the recording and quantitative evaluation of parallel hybridization reactions. In the second part of this work, the ideas of combining the SP optical and electrochemical characterization were extended to tethered bilayer lipid membrane (tBLM) format. Tethered bilayer lipid membranes provide a versatile model platform for the study of many membrane related processes. The thiolipids were firstly self-assembled on ultraflat gold substrates. Fusion of the monolayers with small unilamellar vesicles (SUVs) formed the distal layer and the membranes thus obtained have the sealing properties comparable to those of natural membranes. The fusion could be monitored optically by SPR as an increase in reflectivity (thickness) upon formation of the outer leaflet of the bilayer. With EIS, a drop in capacitance and a steady increase in resistance could be observed leading to a tightly sealing membrane with low leakage currents. The assembly of tBLMs and the subsequent incorporation of membrane proteins were investigated with respect to their potential use as a biosensing system. In the case of valinomycin the potassium transport mediated by the ion carrier could be shown by a decrease in resistance upon increasing potassium concentration. Potential mediation of membrane pores could be shown for the ion channel forming peptide alamethicin (Alm). It was shown that at high positive dc bias (cis negative) Alm channels stay at relatively low conductance levels and show higher permeability to potassium than to tetramethylammonium. The addition of inhibitor amiloride can partially block the Alm channels and results in increase of membrane resistance. tBLMs are robust and versatile model membrane architectures that can mimic certain properties of biological membranes. tBLMs with incorporated lipopolysaccharide (LPS) and lipid A mimicking bacteria membranes were used to probe the interactions of antibodies against LPS and to investigate the binding and incorporation of the small antimicrobial peptide V4. The influence of membrane composition and charge on the behavior of V4 was also probed. This study displays the possibility of using tBLM platform to record and valuate the efficiency or potency of numerous synthesized antimicrobial peptides as potential drug candidates.

In der vorliegenden Arbeit wurden experimentelle Untersuchungen zu gepfropften Polymerfilmen durchgeführt. Dabei wurden endgepfropfte poly-methyl-methacrylate (PMMA) Bürsten hergestellt durch „grafting from“ Methoden und polystyrol (PS)/ poly-vinyl-methyl-ether (PVME) Polymerfilme gepfropft auf UV sensitiven Oberflächen untersucht. Zur Strukturuntersuchung wurden die hergestellten Systeme wurden mit Rasterkraftmikroskopie (engl.: Surface Probe Microscopy, SPM), Röntgen - und Neutronenreflektivitätsmessungen, sowie mit Röntgenstreuung unter streifenden Einfall (engl.: Grazing Incidence Small Angle X-Ray Scattering, GISAXS) untersucht. rnEs wurde gezeigt, dass ein aus der Transmissionsstreuung bekanntes Model auch für auch für die GISAXS Analyse polydisperser Polymerdomänen und Kolloidsysteme verwendet werden kann. Der maximale Fehler durch die gemachten Näherungen wurde auf < 20% abgeschätzt.rnErgebnisse aus der Strukturanalyse wurden mit mechanischen Filmeigenschaften verknüpft. Dazu wurden mechanische Spannungsexperimente durchgeführt. Hierzu wurden die zu untersuchenden Filme selektiv auf einzelne Mikro-Federbalken-Sensoren (engl.: Micro Cantilever Sensor, MCS) der MCS Arrays aufgebracht. Dies wurde durch Maskierungstechniken und Mikro-Kontaktdrucken bewerkstelligt. rnPhasenübergansexperimente der gepfropften PS/PVME Filme haben gezeigt, dass die Möglichkeit einer Polymer/Polymer Phasenseparation stark von Propfpunktdichte der gebundenen Polymerketten mit der Oberfläche abhängt. PS/PVME Filmsysteme mit hohen Pfropfpunktdichten zeigten keinen Phasenübergang. Bei niedrig gepfropften Filmsystemen waren hingegen Polymer/Polymer Phasenseparationen zu beobachten. Es wurde geschlussfolgert, dass die gepfropften Polymersysteme einen hinreichenden Grad an entropischen Freiheitsgraden benötigen um eine Phasenseparation zu zeigen. Mechanische Spannungsexperimente haben dabei das Verstehen der Phasenseparationsmechanismen möglich gemacht.rnAus Quellexperimenten dichtgepfropfter PMMA Bürsten, wurden Lösungsmittel-Polymer Wechselwirkungsparameter (-Parameter) bestimmt. Dabei wurde festgestellt, dass sich die erhaltenen Parameter aufgrund von Filmbenetzung und entropischen Effekten maßgeblich von den errechneten Bulkwerten unterscheiden. Weiterhin wurden nicht reversible Kettenverschlaufungseffekt beobachtet.

In recent decades, Organic Thin Film Transistors (OTFTs) have attracted lots of interest due to their low cost, large area and flexible properties which have brought them to be considered the building blocks of the future organic electronics. Experimentally, devices based on the same organic material deposited in different ways, i.e. by varying the deposition rate of the molecules, show different electrical performance. As predicted theoretically, this is due to the speed and rate by which charge carriers can be transported by hopping in organic thin films, transport that depends on the molecular arrangement of the molecules. This strongly suggests a correlation between the morphology of the organic semiconductor and the performance of the OTFT and hence motivated us to carry out an in-situ real time SPM study of organic semiconductor growth as an almost unprecedent experiment with the aim to fully describe the morphological evolution of the ultra-thin film and find the relevant morphological parameters affecting the OTFT electrical response. For the case of 6T on silicon oxide, we have shown that the growth mechanism is 2D+3D, with a roughening transition at the third layer and a rapid roughening. Relevant morphological parameters have been extracted by the AFM images. We also developed an original mathematical model to estimate theoretically and more accurately than before, the capacitance of an EFM tip in front of a metallic substrate. Finally, we obtained Ultra High Vacuum (UHV) AFM images of 6T at lying molecules layer both on silicon oxide and on top of 6T islands. Moreover, we performed ex-situ AFM imaging on a bilayer film composed of pentacene (a p-type semiconductor) and C60 (an n-type semiconductor).

Intense research is being done in the field of organic photovoltaics in order to synthesize low band-gap organic molecules. These molecules are electron donors which feature in combination with acceptor molecules, typically fullerene derivarntives, forming an active blend. This active blend has phase separated bicontinuous morphology on a nanometer scale. The highest recorded power conversionrnefficiencies for such cells have been 10.6%. Organic semiconductors differ from inorganic ones due to the presence of tightly bonded excitons (electron-hole pairs)resulting from their low dielectric constant (εr ≈2-4). An additional driving force is required to separate such Frenkel excitons since their binding energy (0.3-1 eV) is too large to be dissociated by an electric field alone. This additional driving force arises from the energy difference between the lowest unoccupied molecular orbital (LUMO) of the donor and the acceptor materials. Moreover, the efficiency of the cells also depends on the difference between the highest occupied molecular orbital (HOMO) of the donor and LUMO of the acceptor. Therefore, a precise control and estimation of these energy levels are required. Furthermore any external influences that change the energy levels will cause a degradation of the power conversion efficiency of organic solar cell materials. In particular, the role of photo-induced degradation on the morphology and electrical performance is a major contribution to degradation and needs to be understood on a nanometer scale. Scanning Probe Microscopy (SPM) offers the resolution to image the nanometer scale bicontinuous morphology. In addition SPM can be operated to measure the local contact potential difference (CPD) of materials from which energy levels in the materials can be derived. Thus SPM is an unique method for the characterization of surface morphology, potential changes and conductivity changes under operating conditions. In the present work, I describe investigations of organic photovoltaic materials upon photo-oxidation which is one of the major causes of degradation of these solar cell materials. SPM, Nuclear Magnetic Resonance (NMR) and UV-Vis spectroscopy studies allowed me to identify the chemical reactions occurring inside the active layer upon photo-oxidation. From the measured data, it was possible to deduce the energy levels and explain the various shifts which gave a better understanding of the physics of the device. In addition, I was able to quantify the degradation by correlating the local changes in the CPD and conductivity to the device characteristics, i.e., open circuit voltage and short circuit current. Furthermore, time-resolved electrostatic force microscopy (tr-EFM) allowed us to probe dynamic processes like the charging rate of the individual donor and acceptor domains within the active blend. Upon photo-oxidation, it was observed, that the acceptor molecules got oxidized first preventing the donor polymer from degrading. Work functions of electrodes can be tailored by modifying the interface with monomolecular thin layers of molecules which are made by a chemical reaction in liquids. These modifications in the work function are particularly attractive for opto-electronic devices whose performance depends on the band alignment between the electrodes and the active material. In order to measure the shift in work function on a nanometer scale, I used KPFM in situ, which means in liquids, to follow changes in the work function of Au upon hexadecanethiol adsorption from decane. All the above investigations give us a better understanding of the photo-degradation processes of the active material at the nanoscale. Also, a method to compare various new materials used for organic solar cells for stability is proposed which eliminates the requirement to make fully functional devices saving time and additional engineering efforts.

Die vorliegende Arbeit behandelt die Anwendung der Rasterkraftmikroskopie auf die Untersuchung mesostrukturierter Materialien. Mesostrukturierte Materialien setzen sich aus einzelnen mesoskopen Bausteinen zusammen. Diese Untereinheiten bestimmen im Wesentlichen ihr charakteristisches Verhalten auf äußere mechanische oder elektrische Reize, weshalb diesen Materialien eine besondere Rolle in der Natur sowie im täglichen Leben zukommt. Ein genaues Verständnis der Selbstorganisation dieser Materialien und der Wechselwirkung der einzelnen Bausteine untereinander ist daher von essentieller Bedeutung zur Entwicklung neuer Synthesestrategien sowie zur Optimierung ihrer Materialeigenschaften. Die Charakterisierung dieser mesostrukturierten Materialien erfolgt üblicherweise mittels makroskopischer Analysemethoden wie der dielektrischen Breitbandspektroskopie, Thermogravimetrie sowie in Biegungsexperimenten. In dieser Arbeit wird gezeigt, wie sich diese Analysemethoden mit der Rasterkraftmikroskopie verbinden lassen, um mesostrukturierte Materialien zu untersuchen. Die Rasterkraftmikroskopie bietet die Möglichkeit, die Oberfläche eines Materials abzubilden und zusätzlich dazu seine quantitativen Eigenschaften, wie die mechanische Biegefestigkeit oder die dielektrische Relaxation, zu bestimmen. Die Übertragung makroskopischer Analyseverfahren auf den Nano- bzw. Mikrometermaßstab mittels der Rasterkraftmikroskopie erlaubt die Charakterisierung von räumlich sehr begrenzten Proben bzw. von Proben, die nur in einer sehr kleinen Menge (<10 mg) vorliegen. Darüberhinaus umfasst das Auflösungsvermögen eines Rasterkraftmikroskops, welche durch die Größe seines Federbalkens (50 µm) sowie seines Spitzenradius (5 nm) definiert ist, genau den Längenskalenbereich, der einzelne Atome mit der makroskopischen Welt verbindet, nämlich die Mesoskala. In dieser Arbeit werden Polymerfilme, kolloidale Nanofasern sowie Biomineralien ausführlicher untersucht.rnIm ersten Projekt werden mittels Rasterkraftmikroskopie dielektrische Spektren von mischbaren Polymerfilmen aufgenommen und mit ihrer lokalen Oberflächenstruktur korreliert. Im zweiten Projekt wird die Rasterkraftmikroskopie eingesetzt, um Biegeexperimente an kolloidalen Nanofasern durchzuführen und so ihre Brucheigenschaften genauer zu untersuchen. Im letzten Projekt findet diese Methode Anwendung bei der Charakterisierung der Biegeeigenschaften von Biomineralien. Des Weiteren erfolgt eine Analyse der organischen Zusammensetzung dieser Biomineralien. Alle diese Projekte demonstrieren die vielseitige Einsetzbarkeit der Rasterkraftmikroskopie zur Charakterisierung mesostrukturierter Materialien. Die Korrelation ihrer mechanischen und dielektrischen Eigenschaften mit ihrer topographischen Beschaffenheit erlaubt ein tieferes Verständnis der mesoskopischen Materialien und ihres Verhaltens auf die Einwirkung äußerer Stimuli.rn

Negli ultimi anni uno dei settori di ricerca più interessanti in ambito tecnologico è sicuramente quello dell'elettronica organica. Lo sviluppo è spinto dai vantaggi che portano i dispositivi basati su materiali organici: bassi costi di produzione, facilità di fabbricazione su grandi aree e flessibilità. In questa tesi andiamo ad esaminare un transistor organico a effetto di campo (OFET) dal punto di vista macroscopico e microscopico, cercando di mettere in relazione le sue caratteristiche morfologiche ed elettriche. Il dispositivo sottoposto ai test è un OFET realizzato con TIPS-pentacene come semiconduttore. Dai risultati ottenuti si evince che le prestazioni elettriche del transistor sono fortemente legate alla microstruttura assunta dal materiale organico durante la deposizione. I primi due capitoli illustrano i principi di funzionamento degli OFET e la tecnica SPM (scanning probe microscopy) utilizzata per l'indagine microscopica. Il terzo e quarto capitolo descrivono rispettivamente gli apparati sperimentali e i risultati ottenuti dall'indagine su due aree diverse del dispositivo.

Hereditary spastic paraplegia (HSP) associated with thin corpus callosum is a rare autosomal recessive neurodegenerative disorder characterized by an abnormally thin corpus callosum, normal motor development, slowly progressive spastic paraparesis and cognitive deterioration. To investigate and localize abnormalities in the brains of two Chinese patients with HSP-TCC, with mutations in the spatacsin gene. Diffusion tensor imaging (DTI) was used to determine the mean diffusion (MD) and fractional anisotropy (FA) in the brains of the patients in comparison to 20 healthy subjects. Voxel-based analysis (VBA) of both the diffusion and anisotropy values were performed using statistical parametric mapping (SPM). Significant changes with MD increase and FA reduction were found in the already known lesions including the corpus callosum, cerebellum and thalamus. In addition, changes were also found in regions that appear to be normal in conventional MRI, such as the brain stem, internal capsule, cingulum and subcortical white matter including superior longitudinal fascicle and inferior longitudinal fascicle. Neither increase in FA nor reduction in MD was detected in the brain. Our study provides clear in vivo MR imaging evidence of a more widespread brain involvement of HSP-TCC. MD is more sensitive than FA in detecting lesions in thalamus and subcortical white matter, suggesting that MD may be a better marker of the disease progression.

BACKGROUND Hypoglycin A, found in seeds of Acer negundo, appears to cause seasonal pasture myopathy (SPM) in North America and is implicated in atypical myopathy (AM) in Europe. Acer negundo is uncommon in Europe. Thus, the potential source of hypoglycin A in Europe is unknown. HYPOTHESIS AND OBJECTIVES We hypothesized that seeds of Acer pseudoplatanus were the source of hypoglycin A in Europe. Our objective was to determine the concentration of hypoglycin A in seeds of A. pseudoplatanus trees located in pastures where previous cases of AM had occurred. ANIMALS None. METHODS University of Berne records were searched to retrospectively identify 6 farms with 10 AM cases and 11 suspected AM deaths between 2007 and 2011. During October 2012, A. pseudoplatanus seeds were collected from 2 to 6 trees per pasture on 6 AM farms (7 pastures) from trees in or close to 2 pastures on 2 control farms where AM had not been previously reported. Hypoglycin A in seeds was analyzed by GC-MS. RESULTS Acer pseudoplatanus trees were identified on all AM pastures. Hypoglycin A was detected in all A. pseudoplatanus seeds in highly variable concentrations ranging from 0.04 to 2.81 μg/mg (mean 0.69) on AM farms and 0.10 to 9.12 μg/mg (mean 1.59) on control farms. CONCLUSION AND CLINICAL IMPORTANCE Preventing horses from grazing pastures containing A. pseudoplatanus seeds during late fall and early spring might be the best means to prevent AM.

Seit Jahrtausenden veränderten Menschen ihre Umwelt, direkt, etwa durch landwirtschaftliche Tätigkeiten, oder indirekt, durch Veränderungen der atmosphärischen Zusammensetzung und der biogeochemischen Zyklen. Der andauernde menschliche Einfluss führte in vielen Landökosystemen zur Entwicklung komplexer Verbindungen zwischen Mensch und Natur. Um die Rolle des Menschen und des Klimas auf Landökosysteme zu verstehen und zu entwirren, sind paläoökologische Untersuchungen an Seesedimenten und Mooren eine vielversprechende Methode. Sie setzen aber zeitlich und taxonomisch hochaufgelöste Multiproxy-Studien (z.B. Pollen-, Sporen-, Kieselalgen-, Zuckmücken- und Holzkohleanalysen) an geschichteten Sedimenten voraus. Die Zeit von der Spätantike bis ins Mittelalter ist besonders geeignet, um die Wechselwirkungen Mensch-Klima-Umwelt zu untersuchen. Einerseits wurde die Umwelt bereits zuvor durch viele tausend Jahre menschliche Landnutzung verändert, andererseits zeichnen sich lokal bis regional deutliche Landschaftsveränderungen ab: Nach einem starken Einbruch der landwirtschaftlichen Tätigkeit in den Wirren der Völkerwanderungszeit, in der die damaligen politischen und wirtschaftlichen Systeme in West-, Mittel- und Südeuropa kollabierten, erholte sich die Landnutzung im aufblühenden Frankenreich der Merowinger und noch deutlicher der Karolinger. Im 12./13. Jh. kam es zu einer Phase intensiver Städtegründungen (Kap. 4.2). Für diese Zeit stehen auch dendroklimatische Rekonstruktionen zur Verfügung, welche Niederschlag und Temperatur entkoppeln können (Büntgen et al. 2011). Am Beispiel von paläoökologischen Resultaten (Pollen, Sporen, Holzkohle) aus dem Lauerzersee diskutieren wir die Umweltveränderungen vom 5. bis ins 14. Jh. exemplarisch. Es ist zurzeit die beste vorhandene Sequenz aus dem Schweizer Mittelland und für diese Region repräsentativ.

Standard stereotaxic reference systems play a key role in human brain studies. Stereotaxic coordinate systems have also been developed for experimental animals including non-human primates, dogs, and rodents. However, they are lacking for other species being relevant in experimental neuroscience including sheep. Here, we present a spatial, unbiased ovine brain template with tissue probability maps (TPM) that offer a detailed stereotaxic reference frame for anatomical features and localization of brain areas, thereby enabling inter-individual and cross-study comparability. Three-dimensional data sets from healthy adult Merino sheep (Ovis orientalis aries, 12 ewes and 26 neutered rams) were acquired on a 1.5 T Philips MRI using a T1w sequence. Data were averaged by linear and non-linear registration algorithms. Moreover, animals were subjected to detailed brain volume analysis including examinations with respect to body weight (BW), age, and sex. The created T1w brain template provides an appropriate population-averaged ovine brain anatomy in a spatial standard coordinate system. Additionally, TPM for gray (GM) and white (WM) matter as well as cerebrospinal fluid (CSF) classification enabled automatic prior-based tissue segmentation using statistical parametric mapping (SPM). Overall, a positive correlation of GM volume and BW explained about 15% of the variance of GM while a positive correlation between WM and age was found. Absolute tissue volume differences were not detected, indeed ewes showed significantly more GM per bodyweight as compared to neutered rams. The created framework including spatial brain template and TPM represent a useful tool for unbiased automatic image preprocessing and morphological characterization in sheep. Therefore, the reported results may serve as a starting point for further experimental and/or translational research aiming at in vivo analysis in this species.
