The methanol extract of the flowers of Paepalanthus geniculatus Kunth. showed radical-scavenging activity in the TEAC assay. An analytical approach based on HPLC:ESIMSn was applied to obtain the metabolite profile of this extract and led to the rapid identification of 19 polyphenolic compounds comprising flavonoids and naphthopyranones. The new naphthopyranone (10, 16), quercetagetin (1, 5, 7, 13), and galetine derivatives (9, 11, 17, 19), and a flavonol glucoside cyclodimer in the truxillate form (12), were identified. Compounds 2, 6, and 7 showed the highest antioxidant capacity and ability to affect the levels of intracellular ROS in human prostate cancer cells (PC3).

Este trabalho apresenta os resultados de atividade alelopática, antimicrobiana e antioxidante dos extratos orgânicos (hexano, acetato de etila e metanol) das folhas de Pyrostegia venusta (Ker Gawl.) Miers (Bignoniaceae). Para alelopatia, foi estudado o desenvolvimento de Cucumis sativus (pepino), sendo avaliados o comprimento da raiz principal, o número de raízes secundárias e o comprimento do hipocótilo. Os dois primeiros parâmetros foram afetados por todos os três extratos testados enquanto o comprimento do hipocótilo só não foi afetado pelo extrato acetato de etila. Quanto à atividade antimicrobiana, avaliada pelo ensaio de CIM, o extrato hexânico apresentou inibição moderada frente ao Staphylococcus aureus (0,9 mg mL-1) e forte ao Enterococcus hirae (0,5 mg mL-1). O extrato acetato de etila apresentou forte atividade frente Candida albicans (0,3 mg mL-1) enquanto o extrato metanólico não mostrou-se ativo para os microrganismos testados. Por outro lado, o extrato metanólico apresentou a maior capacidade de seqüestrar radicais livres (Concentração Efetiva 50%-CE50 =102,0 ± 56,9 mg mL-1, com TCE50 = 30 min) no ensaio com DPPH (2,2-difenil-1-picril-hidrazila) e o maior teor de compostos fenólicos (116,2 ± 83,0 mg ácido gálico g amostra-1), avaliado pelo ensaio de Folin-Ciocalteau.

Phenylpropanoid glycosides, 1 '-O-benzyl-alpha-(L)-rhamnopyranosyl-(1 ''-> 6 ')-beta-(D)-glucopyranoside (1) and alpha-(L)-Xylopyranosyl(4 '', 2 ')-(3-O-beta-(D)-glucopyranosyl)-1 '-O-E-caffeoyl-beta-(D)-glucopyranoside (2), together with the known derivatives, 1,6-di-O-caffeoyl- beta-(D)-glucopyrano side (3), 1-O-(E)-caffeoyl-beta-(D)-glucopyranoside (4) and 1-O-(E)-feruloyl-beta-(D)-glucopyranoside (5), were isolated from leaves of Coussarea hydrangeifolia. Their structures were determined by IR, HRESIMS, and I D and 2D NMR experiments, and their antioxidant activities, evaluated by assaying the free radical scavenging capacity using the DPPH (1,1-diphenyl-2-picrylhydrazyl) radical as substrate. The antioxidant activities of 3 and 4 (IC50 values of 15.0 and 19.2 mu M, respectively) were comparable to that of the standard positive control caffeic acid, whilst 2 and 5 were only weakly active and 1 was inactive. A study using two classification methods (SDA and SIMCA) was carried out in this work with the aim of investigating the relationship between the structure of flavonoid compounds and their free-radical-scavenging ability. In this work, we report the use of chemometric methods (SDA and SIMCA) able to select the most relevant variables (steric, electronic, and topological) responsible for this ability. The results obtained with the SDA and SIMCA methods agree perfectly with our previous model, in which we used other chemometric methods (PCA, HCA and KNN) and are also corroborated with experimental results from the literature. This is a strong indication of how reliable the selection of variables is.

The triterpenes, 22beta-hydroxypristimerin and cognatine, were isolated together with the known compounds pristimerin, maytenin, 20alpha-hydroxymaytenin, 22beta-hydroxymaytenin, netzahualcoyol, netzahualcoyondiol and netzahualcoyone from root bark of Chei-Ioclinium cognatum. The structures of the isolated compounds were elucidated by interpretation of their spectral data, including gHMQC and gHMBC experiments. The isolates were investigated for their radical scavenging abilities through a spectrophotometric assay involving reduction of 2,2-diphenyl-picryl hydrazyl (DPPH). (C) 2004 Elsevier Ltd. The bioassay directed fractionation of the EtOH extract from leaves of Cryptocarya ashersoniana seedlings led to the isolation of two flavonol glucosides: iso-quercitrin and hyperin, which exhibited free radical scavenging activity towards DPPH (IC50 34.4 muM and 32.7 muM, respectively) and were compared to standard compounds rutin (IC50 27.0 muM) and catechin (IC50 41.4 muM). Investigation of extracts from the seedlings roots and stems afforded one antifungal styrylpyrone: goniothalamine, and two dihydropyrones: 6-propyl-5,6-dihydro-2-pyrone and the new 6-[(4'-ethyl-9'-oxabicyclo[3.3.1]non-6'-en-3'-yl)methyl]-5,6-dihydro-2H-pyran-2-one, which had its structure determined by detailed analysis of MS and NMR data, including 2D experiments.

A new quinonemethide triterpene named as salacin, has been isolated from the root bark of Salacia campestris in addition to the known pristimerin, maytenin, 20 alpha-hydroxymaytenin, and netzahualcoyene. Salacin was identified on the basis of NMR-spectral and mass spectrometric analysis. The free-radical scavenging activities of the quinonemethide triterpenes salacin (1), pristimerin (2), maytenin (3), 20a-hydroxymaytenin (4), and netzahualcoyene (5) towards DPPH have been evaluated and showed absorbance variation (AA) of 19, 20, 39, 28, 55, and 10%, respectively, having rutin (74% at 50 pm) and BHT (7% at 50 mu M) as standard compounds.

Type-1 diabetic patients experience hyperketonemia caused by an increase in fatty acid metabolism. Thus, the aim of this study was to measure the effect of ketone bodies as suppressors of oxidizing species produced by stimulated neutrophils. Both acetoacetate and 3-hydroxybutyrate have suppressive effect on the respiratory burst measured by luminol-enhanced chemiluminescence. Through measurements of hypochlorous acid production, using neutrophils or the myeloperoxidase/H2O2/Cl- system, it was found that acetoacetate but not 3-hydroxybutyrate is able to inhibit the generation of this antimicrobial oxidant. The superoxide anion scavenging properties were confirmed by ferricytochrome C reduction and lucigenin-enhanced chemiluminescence assays. However, ketone bodies did not alter the rate of oxygen uptake by stimulated neutrophils, measured with an oxygen electrode. A strong inhibition of the expression of the cytokine IL-8 by cultured neutrophils was also observed; this is discussed with reference to the antioxidant-like property of acetoacetate. © 2004 Pharmaceutical Society of Japan.

The EtOH crude extract from the leaves of Nectandra grandiflora collected in the Atlantic Forest, Brazil, showed antioxidant activity towards β-carotene in a TLC assay. The bioassay-guided fractionation led to the isolation of protocatechuic acid and two flavonol glycosides: afzelin and quercetrin, which showed free radical scavenging activity towards DPPH (ΔA 32, 16 and 73% at 50 μmol L-1) and were compared to commercial antioxidants rutin (81% at 50 μmol L-1) and BHT (9% at 50 μmol L-1), used as standard compounds. Additionally, three inactive neolignans, including the new bicycle[3.2.1]octane neolignan 2′-oxo-piperol B were obtained and characterized by spectrometric methods. ©2005 Sociedade Brasileira de Química.
