Os elementos potencialmente tóxicos (EPTs) estão presentes nos solos em concentrações dependentes do material de origem e das ações antrópicas. A adição de EPTs ao solo pelas atividades antrópicas pode ocasionar risco à saúde humana, já que estes elementos podem ser acumulados no organismo por meio do contato dérmico com o solo, da inalação de partículas em suspensão, de ingestão de solo e de alimentos contaminados. A contaminação dos alimentos ocorre pelo cultivo em áreas com alta biodisponibilidade de EPTs, e nessa condição ocorre absorção e translocação para a parte aérea, com possível acúmulo dos metais nas porções comestíveis, como raízes, frutos e grãos. A biodisponibilidade dos EPTs é regulada pelas características químicas dos elementos e por atributos do solo, como a CTC, o pH e a matéria orgânica (MO). Sintomas de toxicidade e alterações morfológicas e fisiológicas podem aparecer dependendo da absorção e da movimentação dos EPTs nas plantas. Objetivou-se neste trabalho avaliar o efeito da adição de bário (Ba), de cádmio (Cd), de cobre (Cu), de níquel (Ni) e de zinco (Zn) em amostras de um Neossolo Quartzarênico e um Latossolo Vermelho distrófico, sob duas condições de saturação por bases (30% e 50 ou 70%, dependendo da cultura), no cultivo de arroz (Oryza sativa), alface (Lactuca sativa), girassol (Helianthus annuus) e tomate (Solanum lycopersicum). Os EPTs nos solos foram extraídos com EPA 3051a, Água Régia, DTPA, Mehlich 1, Mehlich 3, HNO3 (0,43 mol L-1) e CaCl2 (0,01 mol L-1), e seus teores correlacionados com os presentes nas raízes, na parte aérea, nos frutos e com a quantidade acumulada pelas plantas. Os fatores de bioconcentração (FBC) e de transferência (FT) foram calculados para as culturas. O índice SPAD (Soil Plant Analysis Development - Chlorophyll Meter) foi determinado na fase vegetativa da alface, do arroz e do girassol, enquanto a atividade fotossintética foi determinada pelo IRGA (Infrared gas analyzer). Os maiores teores de EPTs foram observados nas plantas cultivadas no Neossolo. As quantidades de Cu, Ni e Zn acumuladas nas plantas apresentaram correlação positiva com os teores extraídos pelo EPA 3051a e pela Água Régia. Os teores extraídos com HNO3 (0,43 mol L-1) apresentaram elevada correlação positiva com os teores reativos extraídos com DTPA e com Mehlich 3, e também com as quantidades de EPTs acumuladas pelas plantas. Os FBCs foram mais altos nos solos com baixa CTC, baixos teores de MO e baixos valores de pH. O arroz apresentou a menor translocação de Cd do sistema radicular para os grãos. O Cu, o Ni e o Zn causaram alterações no desenvolvimento da alface e do girassol, e diminuíram a transpiração e a condutância estomática da alface. O arroz apresentou a menor absorção de EPTs e a maior tolerância ao Ba, ao Cd, ao Ni e ao Zn, no entanto, as plantas apresentaram maiores condutividade estomática e transpiração.

La comparaison des résultats de l’analyse dendrochronologique et dendroclimatologique du Pin d’Alep de la forêt domaniale de Tlemcen a été réalisée en conditions stationnelles particulières. L’analyse de la croissance des cernes annuels et des rapports des écarts relatifs des cernes successifs montrent une nette tendance régressive chez les arbres jeunes. La sensibilité moyenne (SM) et les coefficients d’inter datation (SR) respectifs aux jeunes arbres et aux plus âgés confirment la dépendance assez forte des premiers aux facteurs climatiques particulièrement à la pluviométrie. Les résultats de ce travail ont permis d’établir une relation pluviométrie / accroissement radial en fonction de l’âge de leur formation. Ainsi, il est établi qu’à partir de ces résultats particulièrement ceux des valeurs des sensibilités moyennes (SM) que les conséquences probables des variations climatiques influentes sensiblement sur les jeunes sujets. Aussi pour éviter les changements d’ordre physio biologiques liés au vieillissement des arbres, il est préférable de comparer les cernes sur une période de 40 à 50 ans comme c’est le cas des six échantillons choisis dans la zone d’étude.

A dissertação “Cozinha a Baixa Temperatura: Aplicação ao caso da Confeção de Carne de Novilho” inicia-se pelo enquadramento histórico e respetiva conceptualização. São enumerados e explicados os intervenientes e procedimentos na Cozinha a Baixa Temperatura-Cozinha em Vácuo. Enquanto estudo, a cozinha a baixa temperatura foi alvo de críticas positivas no que confere aos benefícios nutricionais, à maximização das potencialidades do produto (conservação, textura, sabor, entre outros…), ao incremento das vantagens organizacionais e económicas (gestão de cozinhas/restaurantes); alertando para o fato de que um incorreto embalamento comprometerá as vantagens da técnica em estudo, ou seja são enumerados ainda todos os requisitos necessários a uma correta e eficaz cozinha em vácuo. Os diferentes tipos de cozinha a vácuo são igualmente abordados (indireta e dupla cozedura), bem como a influência da relação existente entre o tempo e a temperatura e as consequentes modificações químicas e fisiológicas. No intuito de introduzir uma vertente prática e enquanto projeto pessoal e profissional foi desenvolvida uma máquina para cozinhar a baixa temperatura (roner). Neste âmbito foram enumerados os materiais utilizados e explicado o seu funcionamento. Porque conceber um roner implica testá-lo, foi então decidido utilizar carne de vitela (capitação 100gr) durante 1 hora com temperaturas entre os 54 ºC e os 70 ºC. Foram realizados testes de textura em todas as amostras de carne recorrendo ao texturómetro TA XT Plus e respetivo software para o tratamento dos resultados. Em suma, analisando os resultados obtidos, a relação tempo e temperatura origina modificações químicas e fisiológicas na carne, ou seja uma carne a 54 ºC aporta uma menor gelatinização do colagénio originando uma carne menos macia, de tom rosa avermelhado, onde se dá uma menor contração das fibras musculares e que consequentemente origina uma menor quantidade de sucos expelidos da carne; já o contrário acontece numa confeção a 60 ºC, em que se verifica um tom rosa uniforme onde acontece uma maior contração das fibras musculares originando uma maior quantidade de sucos expelidos pela carne. Os resultados dos ensaios instrumentais de textura mostraram que a carne confecionada a 60 ºC se apresentou menos firme e menos dura, e portanto mais macia.

A preocupação acrescida com a condição e aparência física, refletem-se em cuidados com o estado de saúde e bem-estar e são, nos últimos anos, responsáveis pela expansão dos ginásios e health clubs.Mas, atividade física e a alimentação estão intimamente ligadas, tanto no que respeita à estética como também, com maiores preocupações, no que respeita à saúde. Daí que as duas vertentes estejam ligadas à imagem de pessoas saudáveis e vigorosas. Na verdade, a capacidade de rendimento do organismo melhora com a nutrição adequada. A avaliação da composição corporal é um importante aspeto na determinação da condição física e permite-nos observar as alterações fisiológicas produzidas pelos programas de atividade física e/ou alimentares, oferecendo informações quanto a sua eficiência ou possíveis correções a serem efetuadas. Pretendeu-se com o presente estudo caracterizar os hábitos alimentares e a composição corporal dos indivíduos praticantes de exercício físico num ginásio do Concelho de Coimbra. A amostra foi constituída por 50 indivíduos, 22 do sexo masculino e 28 do sexo feminino, com idades compreendidas entre os 18 e os 57 anos. Estes indivíduos têm em comum o facto de praticarem exercício físico num mesmo ginásio, de uma forma sistemática há pelo menos 6 meses. Para a caracterização dos hábitos de AF dos participantes no estudo foi utilizada a versão curta do International Physical Activity Questionnaire (IPAQ). Para avaliação nutricional recorreu-se ao Questionário Semiquantitativo de Frequência Alimentar (QSFA), elaborado pelo Serviço de Higiene e Epidemiologia da Faculdade de Medicina da Universidade do Porto. A conversão dos alimentos em nutrientes foi realizada recorrendo ao programa informático Food Processor Plus ® versão 7.0. Foram ainda avaliados os seguintes parâmetros antropométricos: peso, altura, perímetro abdominal, IMC, massa gorda e massa magra. As análises estatísticas foram efetuadas com recurso ao Programa Estatístico SPSS (Statistical Package for the Sciences) para o Windows, versão 20.0. O nível de significância estatística foi mantida em 5% (p<0,05). A análise dos dados permitiu retirar as seguintes conclusões: Os frequentadores de ginásio são na sua maioria indivíduos do sexo feminino (56%), jovens com uma idade média de 27±7 anos. A maioria dos participantes do estudo praticava EF (exercício físico) há 39 meses, no entanto no grupo existe uma grande variabilidade, existindo indivíduos que praticam EF há 6 meses, e outros que o fazem há 180 meses. Praticam EF com uma frequência média de 3,5±1,3 sessões/semana a que corresponde uma prática de EF 6,40±3,2 h/semana. A maioria apresentava um Perímetro Abdominal (PA) de 85,2±6,3 cm, Índice de Massa Corporal (IMC) normal (23,1± 2,6 kg/m2), valores de Massa Gorda (MG) (23,9%) e de Massa Muscular (MM) de 34,6±7,9%. Observou-se uma diminuição da MG com o tempo de prática de exercício de 25,3% (praticantes entre 6-12 meses) vs 20,9% (praticantes> 48 meses). Esta influência temporal, embora sem significado estatístico, verifica-se também relativamente ao perímetro abdominal e IMC, embora neste último numa razão inversa, caracterizada pelo seu aumento. O tipo de exercício praticado influencia a composição corporal: praticantes de exercício aeróbio apresentam um perímetro abdominal inferior (82,3±6,0 cm) aos praticantes de outro tipo de exercícios porém acompanhado de uma percentagem de massa gorda superior (29,4±6,0%).Os praticantes de exercício de resistência apresentam valores de MG de 16,0±7,4%. No estudo por nós realizado, constatou-se inadequação na quantidade energética consumida, face às necessidades efetivas da amostra. Embora a ingestão de hidratos de carbono, de fibra e de lípidos se apresente dentro dos limites recomendados a ingestão de proteína ultrapassa os valores recomendados.Os resultados obtidos mostram que o tipo de exercício bem como o tempo de prática de exercício influenciam a composição corporal. Verificou-se ainda que a alimentação dos participantes no estudo não é adequada do ponto de vista nutricional e para além disso os hábitos nutricionais não parecem influir na composição corporal da amostra. Mais estudos serão necessários para que os hábitos alimentares dos praticantes de exercício em ginásios e health clubs sejam bem conhecidos e os profissionais da área tenham mais informações e possam orientar melhor os praticantes de exercício físico regular em ginásios e health clubs de modo a otimizar os resultados pretendidos através de um aconselhamento nutricional individualizado.

High salinity is a severe constraint on tomato growth and productivity in many regions and situations. To obtain an ideal gene donor for improving the salt tolerance of tomato cultivars, the potential of tolerance response to salinity were evaluated for 14 tomato accessions including wild and cultivated species. By investigation of seed germination and seedling survival, a common cultivar, Solanum lycopersicum 'moneymaker', is evidenced significantly salt-tolerant among them and correspondingly, a wild accession, Solanum cheesmanniae 'LA0317', is most vulnerable to salinity. The performance of Moneymaker and LA0317 upon salinity was then compared in detail for their growth inhibition and some physiological changes. Complete dominance of Moneymaker and its high gene identity in tomato species lead us to use it in microarray experiment and apply it as gene donor for salt tolerance. The results indicated some mechanism differences between Moneymaker and LA0317 in salt response, proposed the potentially high salt tolerance of cultivated tomato and implied that Moneymaker is a valuable gene donor in this field, potentially minimizing the growth inhibition and yield reduction in transgenic plants.

Relaxation of the upper age limits for solid organ transplantation coupled with improvements in post-transplant survival have resulted in greater numbers of elderly patients receiving immunosuppressant drugs such as tacrolimus. Tacrolimus is a potent agent with a narrow therapeutic window and large inter- and intraindividual pharmacokinetic variability. Numerous physiological changes occur with aging that could potentially affect the pharmacokinetics of tacrolimus and, hence, patient dosage requirements. Tacrolimus is primarily metabolised by cytochrome P450 (CYP) 3A enzymes in the gut wall and liver. It is also a substrate for P-glycoprotein, which counter-transports diffused tacrolimus out of intestinal cells and back into the gut lumen. Age-associated alterations in CYP3A and P-glycoprotein expression and/or activity, along with liver mass and body composition changes, would be expected to affect the pharmacokinetics of tacrolimus in the elderly. However, interindividual variation in these processes may mask any changes caused by aging. More investigation is needed into the impact aging has on CYP and P-glycoprotein activity and expression. No single-dose, intense blood-sampling study has specifically compared the pharmacokinetics of tacrolimus across different patient age groups. However, five population pharmacokinetic studies, one in kidney, one in bone marrow and three in liver transplant recipients, have investigated age as a co-variate. None found a significant influence for age on tacrolimus bioavailability, volume of distribution or clearance. The number of elderly patients included in each study, however, was not documented and may have been only small. It is likely that inter- and intraindividual pharmacokinetic variability associated with tacrolimus increase in elderly populations. In addition to pharmacokinetic differences, donor organ viability, multiple co-morbidity, polypharmacy and immunological changes need to be considered when using tacrolimus in the elderly. Aging is associated with decreased immunoresponsiveness, a slower body repair process and increased drug adverse effects. Elderly liver and kidney transplant recipients are more likely to develop new-onset diabetes mellitus than younger patients. Elderly transplant recipients exhibit higher mortality from infectious and cardiovascular causes than younger patients but may be less likely to develop acute rejection. Elderly kidney recipients have a higher potential for chronic allograft nephropathy, and a single rejection episode can be more devastating. There is a paucity of information on optimal tacrolimus dosage and target trough concentration in the elderly. The therapeutic window for tacrolimus concentrations may be narrower. Further integrated pharmacokinetic-pharmaco-dynamic studies of tacrolimus are required. It would appear reasonable, based on current knowledge, to commence tacrolimus at similar doses as those used in younger patients. Maintenance dose requirements over the longer term may be lower in the elderly, but the increased variability in kinetics and the variety of factors that impact on dosage suggest that patient care needs to be based around more frequent monitoring in this age group.

Maternal recognition of pregnancy in marsupials occurs in more subtle ways than it does in eutherians. For instance, unlike in eutherians, the plasma progesterone profiles of pregnant and non-pregnant animals are similar during the luteal phase. It is typically during the brief luteal phase that both gestation and parturition occur in marsupials. Yet histological and physiological changes have been documented between gravid and non-gravid uteri in certain monovular marsupials and between pregnant and non-pregnant animals in polyovular marsupials. Early pregnancy factor (EPF), a 10.8-kDa serum protein known to be homologous to chaperonin 10, is associated with maternal immunosuppression, embryonic development and pregnancy in eutherian mammals. It has been reported in two Australian marsupials: the dasyurid Sminthopsis macroura and the phalangerid Trichosurus vulpecula. This paper documents its occurrence in the New World didelphid Monodelphis domestica. EPF is detectable by rosette inhibition assay in the peripheral circulation of pregnant but not of non-pregnant or pseudopregnant animals. Our work focuses on the embryo–maternal signalling role of EPF during pregnancy. Because progesterone-driven changes are similar in pregnant and non-pregnant marsupials, these animals are an excellent laboratory model in which to investigate the role of EPF in orchestrating the physiological changes necessary to sustain pregnancy.

Physiological changes that take place at cellular level are usually reflective of their level of gene expression. Different formulation excipients have an impact on physiological behavior of the exposed cells and in turn affect transporter genes, enterocyte-mediated metabolism and toxicity biomarkers. The aim of this study was to prepare solid dispersion of paracetamol and evaluate genetic changes that occur in Caco-2 cell lines during the permeability of paracetamol alone and paracetamol solid dispersion formulations. Paracetamol-PEG 8000 solid dispersion was prepared by melt fusion method and the formulation was characterised using differential scanning calorimetry (DSC), scanning electron microscopy (SEM) and Fourier transform infrared spectroscopy (FTIR). Formulation of solid dispersion resulted in the conversion of crystalline drug into an amorphous form. Permeability studies showed that paracetamol absorption was higher from the solid dispersion formulation. DNA microarrays analysis was carried out in order to investigate the involvement of any efflux/uptake transporters in paracetamol or its solid dispersion permeability. Neither transporter carriers nor efflux proteins were found to be involved in the absorption of paracetamol or its PEG solid dispersion. Gene expression analysis established that paracetamol toxicity was potentially reduced upon formulation into solid dispersion when ATP binding cassette (ABC) and solute carrier transporter (SLC) genes were analyzed.

The development of in vitro techniques to model the surface-associated mode of growth is a prerequisite to understanding more fully the physiological changes involved in such a growth strategy. Key factors believed to influence bacterial persistence in chronic infections are those of the biofilm mode of growth and slow growth rate. Methods for controlling Pseudomonas aeruginosa biofilm population growth rates were investigated in this project. This microorganism was incompatible with the in vitro 47mm diameter membrane filter-based biofilm technique developed for the study of Escherichia coli and Staphylococcus epidermidis by Gilbert et al (Appl. Environ. Microbiol. 1989, 55, 1308-1311). Two alternative methods were designed. The first comprised a 25mm diameter cellulose acetate membrane filter supported in an integral holder. This was found to be limited to the study of low microbial population densities with low flow rates. The second, based on a cylindrical cellulose fibre depth filter, permitted rapid flow rates to be studied and allowed growth rate control of biofilm and eluted cells. Model biofilms released cells to the perfusing medium as they grew and divided. The viability of released cells was reduced during, and shortly after, inclusion of ciprofloxacin in the perfusate. Outer membrane profiles of biofilm populations exhibited at least two bands not apparent in planktonic cells grown in batch and chemostat culture, and LPS profiles of biofilm populations showed variation with growth rate. Cell surface hydrophobicity of resuspended biofilm cells varied little with growth rate, whilst it decreased markedly for cells released from the biofilms as growth rate increased. Cells released from the biofilm were more hydrophilic than their sessile counterparts. Differing growth rates, LPS profiles and hydrophobicity are proposed to have a bearing on the release of cells from the adherent population.

The thesis investigates the ocular response to silicone-hydrogel (SiH) contact lens wear, a relatively new contact lens material that has a higher modulus of rigidity and different surface coating than used in conventional hydrogel materials. The properties of SiH materials differ significantly from conventional hydrogels and, using subjective and objective means of assessment, the thesis examines how these properties affect reflection and biometry, ocular physiology, tear film characteristics, symptomatology, adverse events and complications. A range of standard and newly designed investigative techniques were employed, and latter involving novel imaging techniques, for the objective assessment of physiological changes which occur with contact lens wear. The study is the first to combine these techniques with biochemical analyses of the tear film composition. Forty-seven subjects were fitted with SiH lenses and randomly allocated to one of the two materials currently on the market (Lotrafilcon A or Balafilcon A) on an either daily or continuous wear basis. An additional control group of 14 age-matched non-contact lens wearers were monitored over the same period. Measurements were taken before and 1, 3, 6, 12 and 18 months after initial fitting. The findings reported in this thesis will enable contact lens practitioners and manufacturers to understand further the optical, physiological and biochemical nature of the ocular response to SiH contact lenses and hence facilitate the development of this important generation of contact lens material.

Loss of optic nerve head (ONH) axons in primary open angle glaucoma (POAG) has been attributed to both mechanical and vascular factors. Confocal scanning laser ophthalmoscopy (cSLO) provides a promising tool for the topographic follow-up of the ONH in glaucoma, while scanning laser Doppler flowmetry (SLDF) facilitates the rapid non-invasive assessment of retinal capillary blood flow. The purposes of these investigations were to optimise the techniques and explore their potential to classify and monitor disease. Preliminary investigations explored the reproducibility and validity of cSLO and SLDF and showed that: For cSLO: In a model eye, measurements are accurate over a range of axial lengths. For best reproducibility, seven images per visit are required, with a contour line located on Elschnig's scleral ring and transferred automatically between images. For SLDF: Three perfusion images are required for optimum reproducibility. Physiological changes induced by gas perturbation can be measured. Cross-sectional comparison of groups of normal subjects and early POAG patients showed that: cSLO parameters differentiate the early POAG group. Blood volume measured by SLDF showed group differences in superior nasal retina only. Longitudinal investigation of ONH topography, haemodynamic and visual field indices in normal subjects and POAG patients showed that: cSLO detects topographical change over time more frequently in the POAG group. Important parameters include: C:D area ratio, cup and rim area, mean depth in contour, volumes above and below reference and surface. Factor analysis identified "cup" and "rim" factors that can be used to detect change over time in individual patients. Blood flow changes were most apparent in the inferior nasal peripapillary retina of the POAG group. Perimetry is of clinical value for the identification of glaucoma but is less sensitive than cSLO for monitoring glaucomatous change.

Current knowledge of the long-term, low dose effects of carbamate (CB) anti-cholinesterases on skeletal muscle or on the metabolism and regulation of the molecular forms of acetylcholinesterase (AChE) is limited. This is largely due to the reversible nature of these inhibitors and the subtle effects they induce which has generally made their study difficult and preliminary investigations were conducted to determine suitable study methods. A sequential extraction technique was used to rapidly analyse AChE molecular form activity at the mouse neuromuscular junction and also in peripheral parts of muscle fibres. AChE in the synaptic cleft involved in the termination of cholinergic transmission was successfully assessed by the assay method and by an alternative method using a correlation equation which represented the relationship between synaptic AChE and the prolongation of extra-cellular miniature endplate potentials. It was found that inhibition after in vivo Carbamate (CB) dosing could not be maintained during tissue analysis because CB-inhibited enzyme complexes decarbamoylated vary rapidly and could not be prevented even when maintained on ice. The methods employed did not therefore give a measure of inhibition but presented a profile of metabolic responses to continual, low dose CB treatment. Repetitive and continual infusion with low doses of the CBs: pyridostigmine and physostigmine induced a variety of effects on mouse skeletal muscle. Both compounds induced a mild myopathy in the mouse diaphragm during continual infusion which was characterised by endplate deformation without necrosis; such deformation persisted on termination of treatment but had recovered slightly 14 days later. Endplate and non-endplate AChE molecular forms displayed selective responses to CB treatment. During treatment endplate AChE was reduced whereas non-endplate AChE was largely unaffected, and after treatment, endplate AChE recovered, whereas non-endplate AChE was up-regulated. The mechanisms by which these responses become manifest are unclear but may be due to CB-induced effects on nerve-mediated muscle activity, neurotrophic factors or morphological and physiological changes which arise at the neuromuscular junction. It was concluded that, as well as inhibiting AChE, CBs also influence the metabolism and regulation of the enzyme and induce persistent endplate deformation; possible detrimental effects of long-term, low-dose determination requires further investigation.

Hypoxia and ocean acidification are two consequences of anthropogenic activities. These global trends occur on top of natural variability. In environments such as estuarine areas, short-term acute pH and O2 fluctuations are occurring simultaneously. The present study tested the combined effects of short-term seawater acidification and hypoxia on the physiology and energy budget of the thick shell mussel Mytilus coruscus. Mussels were exposed for 72 h to six combined treatments with three pH levels (8.1, 7.7 and 7.3) and two dissolved oxygen (DO) levels (2 mg/L, 6 mg/L). Clearance rate (CR), food absorption efficiency (AE), respiration rate (RR), ammonium excretion rate (ER), O:N ratio and scope for growth (SFG) were significantly reduced, and faecal organic dry weight ratio (E) was significantly increased at low DO. Low pH did not lead to a reduced SFG. Interactive effects of pH and DO were observed for CR, E and RR. Principal component analysis (PCA) revealed positive relationships among most physiological indicators, especially between SFG and CR under normal DO conditions. These results demonstrate that Mytilus coruscus was sensitive to short-term (72 h) exposure to decreased O2 especially if combined with decreased pH levels. In conclusion, the short-term oxygen and pH variation significantly induced physiological changes of mussels with some interactive effects.
