In order to succeed in biological control programs, not only is it crucial to understand the number of natural enemies to be released but also on how many sites per area this releasing must be performed. These variables might differ deeply among egg parasitoid species and crops worked. Therefore, these trials were carried out to evaluate the parasitism (%) in eggs of Anticarsia gemmatalis and Pseudoplusia includens after the release of different densities of the egg parasitoid Trichogramma pretiosum. Field dispersal was also studied, in order to determine appropriate recommendations for the release of this parasitoid in soybean fields. The regression analysis between parasitism (%) and densities of the parasitoid indicated a quadratic effect for both A. gemmatalis and P. includens. The maximum parasitism within 24 h after the release was reached with densities of 25.6 and 51.2 parasitoids per host egg, respectively, for the two pests. Parasitism of T. pretiosum in eggs of P. includens decreased linearly as the distance of the pest eggs from the parasitoid release sites increased. For P. includens, the mean radius of T. pretiosum action and the area of parasitoid dispersal in the soybean crop were 8.01 m and 85.18 m(2), respectively. We conclude that for a successful biological control program of lepidopteran pests using T. pretiosum in soybean fields, a density of 25.6 parasitoids per host egg, divided into 117 sites per hectare, should be used.

A major issue for mass rearing of insects concerns sanitary conditions and disease. Microsporidian infection (Nosema sp.) in laboratory colonies of Diatraea saccharalis (Fabr.) (Lepidoptera: Crambidae), used in producing the parasitoid. Cotesia flavipes Cameron (Hymenoptera: Braconidae), is representative of the problems faced by growers and industry. Although C. flavipes has been produced for several years in Brazil for biological control of D. saccharalis, we have only recently observed that the parasitoid becomes infected when developing inside hosts infected with Nosema sp. We assessed the effects of Nosema sp. on C. flavipes, including the ability to locate and select hosts, and evaluated pathogen transmission. Third instar larvae of D. saccharalis were inoculated with Nosema sp. spores at different concentrations and were parasitized when larvae reached fifth instar. Heavily infected D. saccharalis larvae did not support parasitism. Parasitoids that developed in infected D. saccharalis larvae exhibited increased duration of larval and pupal stages, decreased adult longevity and number of offspring, and reduced tibia size compared to parasitoids developing in uninfected D. saccharalis larvae. Infection by Nosema sp. reduced the ability of the C. flavipes parasitoid to distinguish between volatiles released by the sugarcane infested by healthy larvae and pure air. Uninfected parasitoids preferred plants infested with uninfected hosts. But infected C. flavipes did not differentiate between uninfected hosts and those infected with Nosema sp. The pathogen is transmitted from host to parasitoids and parasitoids to hosts. Pathogenic effects of the microsporidium in C. flavipes are sufficiently severe to justify disease management efforts, particularly considering the importance of C. flavipes as a biological control agent in sugarcane. Plant extracts represent a great source of molecules, with insecticidal activity, which are used for pest control in several crop production systems. This work aimed to evaluate the toxicity of an aqueous extract of leaves of castor bean against larvae of Spodoptera frugiperda (Smith) (Lepidoptera: Noctuidae) in search for different classes of molecules with insecticidal activities by using in vitro assays. The effects of the castor bean leaf extract on the food utilization, development, and survival of S. frugiperda larvae was evaluated by feeding the larvae an artificial diet supplemented with different concentrations of the extract (0%, 1%, 2.5%, 5%, and 10% w/v). The effects observed were dose-dependent, and the highest concentration evaluated (10% w/v) was the one the most affected food utilization by altering the nutritional indices, as well as larval weight gain, development time, and survivorship. In vitro assays to detect saponins, lectins, and trypsin inhibitors in the castor bean leaf extract were performed, but only trypsin inhibitors were detected. No preference for the diet source was detected in S. frugiperda by feeding the larvae in choice experiments with diets containing different concentrations of the castor bean extract tested. The data obtained indicate the existence of a potential molecule in the tested extract of castor bean to be used as an alternative insecticide to be integrated in the management of S. frugiperda.

Plant mines are structures with the form of a cavity caused by consumption of host plant tissue by the insect's miner larvae. Plant mines are more common in leaves, but in Cipocereus minensis, a species in which the leaves are modified spines, the miner activity is restricted to the stem. The aim of this paper was to document the morphological and anatomical differences in the infected and uninfected stems of C. minensis due to the feeding habit of the mining agent. Fresh tissue samples of non-mined and mined young stem of C minensis were collected and examined in transverse sections. We hypothesize that the infection begins following mating when the females scratch the surface of the stem or while they feed on fruits and lay eggs, which subsequently develop into larvae, invading the cactus stem. The insect's miner larvae had mostly consumed the parenchyma tissue towards the center of the stem, and periderm formed along the entire path of the insect. This meristematic tissue or "wound periderm" is a common response for compartmentalization to isolate the damaged tissue, in this case the incubating chamber, in which the eggs will be placed. There were no signs of consumption of vascular tissue in the infested samples, further suggesting a compartmentalized infestation. The nest chamber was found in the stem pith region, with periderm surrounding an insect's miner pupa inside identified as a member of the Cerambycidae. The mining insect depends on a host plant to complete the life cycle; however, the nature of this partnership and the long-term effects of the insect on the plant tissue are unknown. The complex mechanisms by which herbivorous insects control the morphogenesis of the plant host are discussed. We propose that C. minensis has a recognition system to identify insect attack and evaluate the effectiveness of early response triggering compartmentalized defense mechanisms by protecting the injured area with a new layer of periderm.

We have characterized in vitro and in vivo effects of trypsin inhibitors from Theobroma seeds on the activity of trypsin- and chymotrypsin-like proteins from Lepidopteran pest insects. The action of semipurified trypsin inhibitors from Theobroma was evaluated by the inhibition of bovine trypsin and chymotrypsin activities determined by the hydrolysis of N-Benzoyl-DL-Arginine-p-Nitroanilide (BAPA) and N-Succinyl-Ala-Ala-Pho-Phe p-Nitroanilide (S-(Ala)2ProPhe-pNA). Proteinase inhibitor activities from Theobroma cacao and T. obovatum seeds were the most effective in inhibiting trypsin-like proteins, whereas those from T. obovatum and T. sylvestre were the most efficient against chymotrypsin-like proteins. All larvae midgut extracts showed trypsin-like proteolytic activities, and the putative trypsin inhibitors from Theobroma seeds significantly inhibited purified bovine trypsin. With respect to the influence of Theobroma trypsin inhibitors on intact insects, the inclusion of T. cacao extracts in artificial diets of velvet bean caterpillars (Anticarsia gemmatalis) and sugarcane borer (Diatraea saccharalis) produced a significant increase in the percentage of adult deformation, which is directly related to both the survival rate of the insects and oviposition.

Aflatoxin (AFL) contamination of corn is a serious economic and food security issue. Although a variety of technical solutions for reducing AFL contamination of corn have been proposed, only a few have produced satisfactory results. A successful approach is a biocontrol strategy consisting of using non-flatoxigenic strains of Aspergillus flavus to replace indigenous AFL-producing isolates. The main objective of the present thesis was to investigate the dynamic and contamination of AFL/A. flavus in corn in Northern Italy. The study also investigated the role of the key-pest of corn, the European Corn Borer (ECB), on AFL contamination and dispersal of A. flavus propagules in corn. Finally, the study evaluated the feasibility of bioplastic-based granules entrapping a non-aflatoxigenic A. flavus strain for the biocontrol of this fungus in corn. The 2-year field study demonstrated the efficacy of the bioplastic formulation to reduce AFL contamination in corn. More precisely, although AFL contamination varied among the two years, application of 15 and 30 kg ha-1 of granules reduced AFL contamination to up 60 and 85% in 2009 and 2010 respectively. Microbiological analysis showed that the relative abundance of non-aflatoxigenic soil isolates significantly increased after 1 month from granules application (mid-May) and throughout the corn-growing season. These findings were consistent with data obtained using a bioplastic-based bait specifically developed to selectively isolate Aspergilli from soil and other environmental samples. In addition, field and laboratory evaluations showed that the level of damages produced by ECB larvae were not significantly correlated to A. flavus infestation and AFL contamination. Taking together, these findings demonstrated that AFL contamination of corn in Northern Italy was variable, but above the EU limit for human consumption. First proposed in the USA, this study showed the practical possibility of this formulation to be use for reducing AFL contamination in corn in the EU.

Flugfähige Insekten sind äußerst leistungsfähige Tiere. Ihre Flugmuskulatur ist das Gewebe mit der höchsten ATP-Umsatzrate im Tierreich. Der hohe Energieumsatz ist möglich durch einen vollständig aeroben Stoffwechsel der Flugmuskulatur, der durch die effiziente Sauerstoffversorgung über das Tracheensystem gewährleistet wird. Andererseits haben Insekten einen offenen Blutkreislauf, d.h. ihre Gewebe werden nicht über Kapillaren mit Substraten versorgt, sondern von der Hämolymphe umspült, die daher eine hohe Konzentration an energieliefernden Substraten haben muss. Als schnell verfügbares Substrat nutzen Wanderheuschrecken bei Beginn eines Fluges als Hauptsubstrat Trehalose, die in hoher Konzentration als Hämolymphzucker vorliegt (20 bis 40mal höhere Konzentration als Glucose). Trehalose ist, anders als Glucose, ein nicht-reduzierender Zucker und daher nicht toxisch. Allerdings muss das Disaccharid Trehalose zu Glucose hydrolysiert werden, bevor sie im Zellstoffwechsel verwertet werden kann. Diese Funktion erfüllt die Trehalase (EC 3.2.1.28), ein Enzym, das membrangebunden ist und nach Zellfraktionierung in der Mikrosomenfraktion erscheint. Es ist schon lange offensichtlich, dass die Aktivität der Trehalase regulierbar sein muss und zwar reversibel (eine Eigenschaft, die für Hydrolasen ungewöhnlich ist), der Mechanismus ist allerdings bislang nicht klar, da alle üblichen Typen von Aktivitätsregulation nicht verwirklicht zu sein scheinen. Die meisten Autoren vermuten, dass die Regulation über den Transport des Substrats erfolgt. Ein Trehalosetransporter konnte allerdings bisher in der Flugmuskulatur von Locusta nicht nachgewiesen werden. In dieser Arbeit stelle ich Experimente vor, die dafür sprechen, dass Trehalase als Ektoenzym aktiv ist (overte Form), während eine inaktive Form (latente Form) in Vesikeln im Cytoplasma vorliegt und per Exocytose reversibel in die Plasmamembran transloziert werden kann. Für die Testung dieser Arbeitshypothese nutzte ich Trehazolin, einen sehr spezifischen Inhibitor der Trehalase, der äußerst fest und dauerhaft im aktiven Zentrum des Enzyms bindet. Dazu war es nötig, die Flugmuskulatur zu fraktionieren, um die Effekte von Trehazolin auf die verschiedenen Formen der Trehalase (gebunden, löslich, overt, latent) zu analysieren. Mit der Arbeitshypothese vereinbar sind die folgenden Befunde: (1) In die Hämolymphe injiziertes Trehazolin hemmt bevorzugt die overte Trehalase und erst bei höheren Dosen und nach längerer Zeit die latente Form. (2) Trehazolin wirkt in hoher Dosis (50µg pro Tier) auch nach Verfütterung, allerdings stark abgeschwächt, da nach 24 Stunden ein signifikanter Effekt nur auf die overte, aber nicht auf die latente Form sichtbar war. (3) In einem Langzeitversuch über 30 Tage führte die einmalige Injektion von 20µg Trehazolin zu einer schnellen Hemmung der overten Trehalase, der eine verzögerte Hemmung der latenten Aktivität folgte. Der Zeitverlauf von Hemmung und Erholung spricht für eine Vorläufer-Produkt-Beziehung zwischen latenter und overter Form. (4) Flugaktivität der Tiere führt zu einer starken Verminderung der latenten Aktivität, falls Trehazolin in der Hämolymphe der Tiere vorhanden war. (5) Neuropeptide könnten die Translokation fördern. Insulin hat einen entsprechenden Effekt, der aber unabhängig ist von der Flugaktivität. (6) Der PI3-Kinasehemmstoff Wortmannin stabilisiert die latente Form der Trehalase. Auch andere Organe als die Flugmuskulatur besitzen Trehalase, aber mit deutlich geringerer Aktivität. In der Sprungmuskulatur könnte auch eine latente Form vorhanden sein, für Darm und Gehirn ist das nicht wahrscheinlich.
