O metilmercúrio (MeHg) é a forma mais tóxica do mercúrio. A exposição ao MeHg gera estresse oxidativo, podendo afetar a retina, pois esta possui alta vulnerabilidade em função do seu elevado conteúdo de ácidos graxos poliinsaturados e consumo de oxigênio. Nesse contexto, a administração de antioxidantes exógenos obtidos pela dieta, como os presentes na Euterpe oleracea (açaí), poderia ser uma forma de prevenir esse desequilíbrio e suas consequências. Portanto, o objetivo deste trabalho foi avaliar o possível efeito protetor da Euterpe oleracea nas alterações eletrofisiológicas causadas pelo MeHg na retina. Para tal, foi realizada gavagem com MeHgCl (5 mg/Kg) ou solução salina (NaCl 0,9%) durante 7 dias e pré-tratamento com ração enriquecida com polpa de açaí (10%) por 28 dias. Foram utilizados ratos Wistar divididos em 4 grupos: Grupo MeHg (recebeu ração padrão e MeHgCl); MeHg+Açaí (ração enriquecida com açaí e MeHgCl); Açaí (ração enriquecida com açaí e NaCl); Veículo (ração padrão e NaCl). Um dia após a última gavagem os animais foram submetidos ao eletrorretinograma de campo total (ffERG) para obtenção da resposta escotópica (de bastonetes, mista 1 e mista 2) e fotópica (de cones e de flicker em 12; 18; 24 e 30Hz). No dia seguinte ao ffERG foi aplicado o teste campo aberto para avaliar a atividade locomotora dos animais. Posteriormente, foi feita medição de peroxidação lipídica no tecido retiniano pelo método TBARS. A análise estatística foi feita pelo teste ANOVA de uma via com pós-teste de Tukey, considerando significativo p<0,05. Os resultados do campo aberto e da massa corporal não apresentaram diferença entre os grupos. O MeHg reduziu a amplitude das seguintes respostas: onda-b da resposta de bastonetes (Veículo: 114,6±23,6 μV e MeHg: 41,2±9,6 μV); onda-a (Veículo: 8,4±1,4 μV e MeHg: 3,4±0,3 μV) e onda-b (Veículo: 176,7±17,8 μV e MeHg: 69,5±12,0 μV) na resposta mista 1; onda-a (Veículo: 103,1 ±23,3 μV e MeHg: 40,2±9,6 μV) e onda-b (Veículo: 281±,38,3 μV e MeHg: 138,6±14 μV) da resposta mista 2; onda-a (Veículo: 27,2 ±3,6 μV e MeHg: 7,5±1,8 μV) e onda-b (Veículo: 139,3±16,1 μV e MeHg: 54,4±10 μV) da resposta de cones; onda-b nas frequências 12 Hz (Veículo: 67,7±10μV e MeHg: 28,6±6,9 μV), 18 Hz (Veículo: 31,3±3,4 μV e MeHg: 14,2± 2,3 μV) e 24 Hz (Veículo: 21,0±1,8μV e MeHg: 11,0± 1,1μV) e 30 Hz (Veículo: 10,9±0,6μV e MeHg: 6,0± 1,1μV). O tempo implícito das ondas não foi alterado em nem uma das respostas. O pré-tratamento com Euterpe oleracea evitou a redução de amplitude de ambas as ondas nas respostas mista 1 (onda-a: 8,3±0,6 μV; onda b: 144,1±7,1 μV) e mista 2 (onda-a: 106,4±13,6μV; onda b: 275,2±27,6 μV), assim como da onda-b da resposta de cones (104,5±5,9 μV) e fotópica de flicker em 12 Hz (67,2±9,1 μV), 18 Hz (29,5±4,8 μV) e 24 Hz (21,9±2,4 μV). A peroxidação lipídica no tecido retiniano do grupo MeHg (294,9±205,8%) foi maior que a do Veículo (100±25,1%) e o açaí protegeu contra esse dano oxidativo (MeHg+Açaí: 111,2±26,1%). Nossos resultados demonstraram alteração difusa na resposta eletrofisiológica e aumento na peroxidação lipídica da retina induzidos pelo MeHg e proteção exercida pelo açaí nesses dois parâmetros. Assim, a Euterpe oleracea poderia ser utilizada como importante alternativa para amenizar as alterações causadas pelo MeHg na retina.

Este estudo investigou a toxicidade aguda oral, o efeito antinociceptivo em modelos de nocicepção química e térmica, bem como a atividade anti-inflamatória em modelos de carragenina e óleo de cróton do extrato hidroetanólico de partes aéreas de Portulaca pilosa (EHEPp). Identificou também alguns possíveis mecanismos envolvidos na antinocicepção do extrato, além dos seus efeitos sobre o sistema nervoso central de ratos. No teste de toxicidade aguda oral, o tratamento com EHEPp (2000 mg/kg) não causou óbitos. No teste de contorções abdominais induzidas por ácido acético, o EHEPp (100, 200, 400 e 600 mg/kg), por via oral (v.o.), reduziu significantemente o número de contorções em 18.18, 33.25, 47.27, 65.81 e 73.94%, respectivamente. No teste da placa quente, o tratamento com EHEPp (200, 400 e 600 mg/kg, v.o.) não alterou a latência ao estímulo térmico de 50 ± 0,5 ºC. No teste da formalina, o tratamento com EHEPp (200,400 e 600mg/kg, v.o.) reduziu de maneira significativa o tempo de lambida nas fases neurogênica (1ª fase) em 38.79, 60.61 e 75.18 %, e inflamatória (2ª fase) em 49.23, 53.03 e 87.53 %, respectivamente. A administração prévia de naloxona reverteu, significativamente, o efeito do EHEPp (600 mg/kg, v.o.) em ambas as fases do teste da formalina. O pré-tratamento com o L-NAME e azul de metileno reverteu o efeito do EHEPp (600 mg/kg, v.o.) de maneira significante em ambas as fases do teste da formalina. O pré-tratamento com o fármaco glibenclamida também reverteu de maneira significativa o efeito do EHEPp (600 mg/kg, v.o.) em ambas as fases do teste da formalina. O EHEPp, na dose 600 mg/kg, v.o., não afetou a atividade locomotora dos ratos submetidos ao teste do campo aberto. No teste de edema de pata induzido por carragenina e edema de orelha induzido pelo óleo cróton, o EHEPp (400 e 600 mg/kg, v.o.) não inibiu a formação de edema de maneira significante em ambos os testes. Os resultados deste estudo mostraram que o HEEPp, oralmente, apresentou baixa toxicidade e sua atuação antinociceptiva observada na fase neurogênica pode envolver interações periféricas com receptores opióides e ativação da via NO/GCs/GMPc/ KATP. Já a atividade antinociceptiva observada na fase inflamatória parece não depender de inibição da via bioquímica fosfolipase A2/ciclo-oxigenases, mas de interações periféricas com receptores opióides e com a via NO/GCs/GMPc/KATP.

Recently was observed that pyrethroids decrease motor coordination and that calcium channels can be important targets for this effect. To contribute with this observation, this work studied the motor coordination and exploration (using hole-board apparatus), and locomotion (using open-field apparatus) of rats exposed to following treatments: permethrin (PM), PM plus calcium gluconate (CG) and PM plus amlodipine (AML). The results obtained show that CG or AML alone not changed the motor coordination while PM decreases it. CG kept the effect of permethrin; AML, however, decreased the values of permethrin to the control. Locomotor activity and exploration, which could confound results of motor coordination, were not modified by treatments. The concentration of PM in brain tissue was increased by the CG and AML. The neurosomatic index (weight brain/body weight) was increased by the PM and PM + CG. In conclusion, the combined results here obtained indicates that the calcium ion and the channels in which it is involved can be important targets for the toxic effect of pyrethroid insecticide permethrin on motor nerve activity of rats. Cocaine and anabolic-androgenic steroids are substances commonly co-abused. The use of anabolic steroids and cocaine has increased among adolescents. However, few studies investigated the consequences of the interaction between anabolic-androgenic steroids in animals' model of adolescence. We examined the effects of acute and repeated testosterone administration on cocaine-induced locomotor activity in adult and adolescent rats. Rats received ten once-daily subcutaneous (s.c.) injections of testosterone (10mg/kg) or vehicle. Three days after the last testosterone or vehicle injections rats received an intraperitoneal (i.p.) challenge injection of either saline or cocaine (10mg/kg). A different subset of rats was treated with a single injection of testosterone (10mg/kg) or vehicle and three days later was challenged with cocaine (10mg/kg, i.p.) or saline. Immediately after cocaine or saline injections the locomotor activity was recorded during forty minutes. Our results demonstrated that repeated testosterone induced locomotor sensitization to cocaine in adolescent but not adult rats.

Diethylpropion (DEP) is a stimulant drug widely used for weight control in Brazil and other American countries. However, its effects on behavior and addiction potential are not yet well known. Data suggest that sensitization resulting from pre-exposure to psychostimulants could be a possible risk factor in subsequent drug addiction. The purpose of this investigation was to verify whether pre-exposure to DEP would sensitize rats to the motor activating effect and to the rewarding value of DEP. Two experiments were conducted. In both experiments rats were pre-exposed to DEP (20 mg/kg) or vehicle for 7 consecutive days. The acute effect of DEP (0.0, 1.0, 2.5 or 5.0 mg/kg) on motor activity (Experiment 1) and induction of Conditioned Place Preference-CPP (Experiment 2) were then measured. Results from Experiment 1 showed that 2.5 and 5.0 mg/kg DEP increased motor activity. Sensitization of this motor effect was observed. In Experiment 2, the doses of 2.5 and 5.0 mg/kg DEP induced CPP, indicating their rewarding value. However, no sensitization effect was observed. The results suggest that DEP at low doses has psychostimulant and rewarding properties. It is recommended that more effort should be dedicated to elucidating DEP abuse Potential. Because GABA(A) receptors containing alpha 2 subunits are highly represented in areas of the brain, such as nucleus accumbens (NAcc), frontal cortex, and amygdala, regions intimately involved in signaling motivation and reward, we hypothesized that manipulations of this receptor subtype would influence processing of rewards. Voltage-clamp recordings from NAcc medium spiny neurons of mice with alpha 2 gene deletion showed reduced synaptic GABA(A) receptor-mediated responses. Behaviorally, the deletion abolished cocaine`s ability to potentiate behaviors conditioned to rewards (conditioned reinforcement), and to support behavioral sensitization. In mice with a point mutation in the benzodiazepine binding pocket of alpha 2-GABA(A) receptors (alpha 2H101R), GABAergic neurotransmission in medium spiny neurons was identical to that of WT (i.e., the mutation was silent), but importantly, receptor function was now facilitated by the atypical benzodiazepine Ro 15-4513 (ethyl 8-amido-5,6-dihydro-5-methyl-6-oxo-4H-imidazo [1,5-a] [1,4] benzodiazepine-3-carboxylate). In alpha 2H101R, but not WT mice, Ro 15-4513 administered directly into the NAcc-stimulated locomotor activity, and when given systemically and repeatedly, induced behavioral sensitization. These data indicate that activation of alpha 2-GABA(A) receptors (most likely in NAcc) is both necessary and sufficient for behavioral sensitization. Consistent with a role of these receptors in addiction, we found specific markers and haplotypes of the GABRA2 gene to be associated with human cocaine addiction.

Cohabitation for 14 days with Ehrlich tumor-bearing mice was shown to increase locomotor activity, to decrease hypothalamic noradrenaline (NA) levels, to increase NA turnover and to decrease innate immune responses and decrease the animals' resistance to tumor growth. Cage mates of a B16F10 melanoma-bearer mice were also reported to show neuroimmune changes. Chemosignals released by Ehrlich tumor-bearing mice have been reported to be relevant for the neutrophil activity changes induced by cohabitation. The present experiment was designed to further analyze the effects of odor cues on neuroimmune changes induced by cohabitation with a sick cage mate. Specifically, the relevance of chemosignals released by an Ehrlich tumor-bearing mouse was assessed on the following: behavior (open-field and plus maze); hypothalamic NA levels and turnover; adrenaline (A) and NA plasmatic levels; and host resistance induced by tumor growth. To comply with such objectives, devices specifically constructed to analyze the influence of chemosignals released from tumor-bearing mice were employed. The results show that deprivation of odor cues released by Ehrlich tumor-bearing mice reversed the behavioral, neurochemical and immune changes induced by cohabitation. Mice use scents for intraspecies communication in many social contexts. Tumors produce volatile organic compounds released into the atmosphere through breath, sweat, and urine. Our results strongly suggest that volatile compounds released by Ehrlich tumor-injected mice are perceived by their conspecifics, inducing the neuroimmune changes reported for cohabitation with a sick companion. This study investigated whether perinatal exposure to picrotoxin, a GABA(A) antagonist, modifies the effect of muscimol, a GABA(A) agonist, on the sexual behavior of adult male rats. Two hours after birth and then once daily during the next 9 days of lactation, dams received picrotoxin (0.75 mg/kg subcutaneously) or saline (1 ml/kg subcutaneously). The adult male offspring from the picrotoxin and saline groups received saline (1 ml/kg intraperitoneally) or muscimol (1 mg/kg intraperitoneally), and 15 min later, their sexual behavior was assessed. Muscimol treatment in the saline-exposed group increased the mount and intromission latencies. However, these effects were absent in the picrotoxin-exposed groups. The latencies to first ejaculation, postejaculatory mount, and intromission were decreased in both picrotoxin-exposed groups relative to the saline-exposed groups. The picrotoxin + muscimol-treated rats required more intromissions to ejaculate and the picrotoxin-exposed groups made more ejaculations than the saline-exposed groups. Thus, muscimol treatment did not increase the mount and intromission latencies following picrotoxin exposure, but increased the ejaculation frequency, which did not differ between the picrotoxin + muscimol and the picrotoxin + saline groups. Subterranean organisms are excellent models for chronobiological studies, yet relatively few taxa have been investigated with this focus. Former results were interpreted as a pattern of regression of circadian locomotor activity rhythms in troglobitic (exclusively subterranean) species. In this paper we report results of experiments with cave fishes showing variable degrees of troglomorphism (reduction of eyes, melanic pigmentation and other specializations related to the hypogean life) submitted to light-dark cycles, preceded and followed by several days in constant darkness. Samples from seven species have been monitored in our laboratory for the detection of significant circadian rhythms in locomotor activity: S. typhlops, an extremely troglomophic species, presented the lowest number of significant components in the circadian range (only one individual out of eight in DD1 and three other fish in LD), all weak (low values of spectral power). Higher incidence of circadian components was observed for P. kronei - only one among six studied catfish without significant circadian rhythms under DD1 and DD2; spectral powers were generally high. Intermediate situations were observed for the remaining species, however all of them presented relatively strong significant rhythms under LD. Residual oscillations (circadian rhythms in DD2) were detected in at least part of the studied individuals of all species but S. typhlops, without a correlation with spectral powers of LD rhythms, i.e., individuals exhibiting residual oscillations were not necessarily those with the strongest LD rhythms. In conclusion, the accumulated evidence for troglobitic fishes strongly supports the hypothesis of external, environmental selection for circadian locomotor rhythms.

Daily rhythmic processes are coordinated by circadian clocks, which are present in numerous central and peripheral tissues. In mammals, two circadian clocks, the food-entrainable oscillator (FEO) and methamphetamine-sensitive circadian oscillator (MASCO), are "black box" mysteries because their anatomical loci are unknown and their outputs are not expressed under normal physiological conditions. In the current study, the investigation of the timekeeping mechanisms of the FEO and MASCO in mice with disruption of all three paralogs of the canonical clock gene, Period, revealed unique and convergent findings. We found that both the MASCO and FEO in Per1(-/-)/Per2(-/-)/Per3(-/-) mice are circadian oscillators with unusually short (similar to 21 h) periods. These data demonstrate that the canonical Period genes are involved in period determination in the FEO and MASCO, and computational modeling supports the hypothesis that the FEO and MASCO use the same timekeeping mechanism or are the same circadian oscillator. Finally, these studies identify Per1(-/-)/Per2(-/-)/Per3(-/-) mice as a unique tool critical to the search for the elusive anatomical location(s) of the FEO and MASCO.
