The tonoplast calcium Ca2+/H+ antiport system of maize (Zea mays L. cv LG 11) roots was characterized using the ''pH jump'' technique in order to avoid interference from the tonoplast proton and Ca2+ pumps. Ca2+ uptake was recorded in the presence of different inhibitors and divalent ions. Chemical modification of amino acid residues of the antiport was used to elucidate the amino acid residues participating in the Ca2+ transport activity. The Ca2+/H+ antiport activity was found to be strongly inhibited by ruthenium red and verapamil, whereas diethylstilbestrol was less effective. Vanadate, erythrosin B, cyclopiazonic acid, bafilomycin, thapsigargin, N,N'-dicyclohexylcarbodiimide (DCCD) and 4,4'-diisothiocyanatostilbene-2,2'-disulfonate (DIDS) were without effect. Lanthanum and divalent ions were strongly inhibitory (Cd2+ > Mn2+ > Sr2+ > Ba2+). While reagents modifying sulfhydryl groups (N-ethylmaleimide and 5,5'-dithio-bis(2-nitrobenzoate)) did not affect the antiport activity, modification of trytophan residues (N-bromosuccinimide) was strongly inhibitory. We conclude that ruthenium red, verapamil, lanthanum and divalent cations directly inhibit Ca2+ uptake independent of the function of the proton and Ca2+ pumps. Moreover, the results of chemically modified amino acid residues suggest that sulfhydryl groups are not involved in Ca2+ transport, while tryptophan residues seem important for this translocation.

The subcellular localization, distribution and the steady state level of calmodulin from maize roots (Zea mays L., cv. LG 11) were studied. To analyze the subcellular localization, 2-day old root membranes were fractionated by sucrose density gradient centrifugation and immunoblotting was done with an antibody raised against a vertebrate calmodulin (SWant) which recognized the plant calmodulin. Calmodulin was principally associated with high density fractions and particularly plasmalemma. For studying the distribution of calmodulin in various zones of Zea mays roots, a micro method of membrane preparation was developed. Most of the calmodulin was present in microsomes isolated from the root apex corresponding to the first 4 mm of a 15 +/- 2 mm root. An identical distribution was found by studying the steady state level of the protein by Northern blotting using a cDNA clone of Zea mays calmodulin.

AIM: This study aims to investigate the clinical and demographic factors influencing gentamicin pharmacokinetics in a large cohort of unselected premature and term newborns and to evaluate optimal regimens in this population. METHODS: All gentamicin concentration data, along with clinical and demographic characteristics, were retrieved from medical charts in a Neonatal Intensive Care Unit over 5 years within the frame of a routine therapeutic drug monitoring programme. Data were described using non-linear mixed-effects regression analysis ( nonmem®). RESULTS: A total of 3039 gentamicin concentrations collected in 994 preterm and 455 term newborns were included in the analysis. A two compartment model best characterized gentamicin disposition. The average parameter estimates, for a median body weight of 2170 g, were clearance (CL) 0.089 l h(-1) (CV 28%), central volume of distribution (Vc ) 0.908 l (CV 18%), intercompartmental clearance (Q) 0.157 l h(-1) and peripheral volume of distribution (Vp ) 0.560 l. Body weight, gestational age and post-natal age positively influenced CL. Dopamine co-administration had a significant negative effect on CL, whereas the influence of indomethacin and furosemide was not significant. Both body weight and gestational age significantly influenced Vc . Model-based simulations confirmed that, compared with term neonates, preterm infants need higher doses, superior to 4 mg kg(-1) , at extended intervals to achieve adequate concentrations. CONCLUSIONS: This observational study conducted in a large cohort of newborns confirms the importance of body weight and gestational age for dosage adjustment. The model will serve to set up dosing recommendations and elaborate a Bayesian tool for dosage individualization based on concentration monitoring.

Quantitative analyses of abscisic acid in the elongating zone of a single maize root (Zea mays L. cv LG 11) were performed by gas chromatography-mass spectrometry using negative chemical ion ionization. Data showed that the more abscisic acid, the slower the growth, but a large dispersion of individual values was observed. We assume that abscisic acid is perhaps not correlated only to the growth rate.

A maize (Zea mays L. cv LG 11) root homogenate was prepared and centrifuged to sediment the mitochondria. The pellet (6 KP) and the supernatant (6 KS) were collected and fractionated on linear sucrose density gradients. Marker enzymes were used to study the distribution of the different cell membranes in the gradients. The distribution of the ATP- and pyrophosphate-dependent proton pumping activities was similar after 3 hours of centrifugation of the 6 KS or the 6 KP fraction. The pumps were clearly separated from the mitochondrial marker cytochrome c oxidase and the plasmalemma marker UDP-glucose-sterolglucosyl-transferase. The pyrophosphate-dependent proton pump might be associated with the tonoplast, as the ATP-dependent pump, despite the lack of a specific marker for this membrane. However, under all the conditions tested, the two pumps overlapped the Golgi markers latent UDPase and glucan synthase I and the ER marker NADH-cytochrome c reductase. It is therefore not possible to exclude the presence of proton pumping activities on the Golgi or the ER of maize root cells. The two pumps (but especially the pyrophosphate-dependent one) were more active (or more abundant) in the tip than in the basal part of maize roots, indicating that these activities might be important in growth processes.

Tonoplast-enriched membranes were prepared from maize (Zea mays L. cv LG 11) primary roots, using sucrose nonlinear gradients. The functional molecular size of the tonoplast ATP-and PPi-dependent proton pumps were analyzed by radiation inactivation. Glucose-6-phosphate dehydrogenase (G6PDH) was added as an internal standard. Frozen samples (-196 degrees C) of the membranes were irradiated with (60)Co for different periods of time. After thawing the samples, the activities of G6PDH, ATPase, and PPase were tested. By applying target theory, the functional sizes of the ATPase and PPase in situ were found to be around 540 and 160 kilodaltons, respectively. The two activities were solubilized and separated by gel filtration chromatography. The different polypeptides copurifying with the two pumps were analyzed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis. Two bands (around 59 and 65 kilodaltons) were associated with the ATPase activity, whereas a double band (around 40 kilodaltons) was recovered with the PPase activity.

O uso de leguminosas constitui boa alternativa para suprir N e aumentar o rendimento das culturas comerciais. A aceitação desta prática por agricultores e extensionistas, entretanto, depende da demonstração de sua vantagem econômica em relação ao sistema tradicional. Neste estudo, avaliou-se o efeito do cultivo intercalar de quatro leguminosas estivais de cobertura do solo [feijão de porco (Canavalia ensiformis L.), guandu (Cajanus cajan L.) cv. roxo anão, mucuna cinza (Stizolobium niveum Kuntze) e soja preta (Glycine sp)] sobre a receita líquida da cultura do milho, em três doses de N (0, 60 e 120 kg ha-1), comparativamente ao sistema tradicional de milho exclusivamente (testemunha). Dois experimentos foram realizados, durante quatro e cinco anos, respectivamente, em sistemas de preparo convencional (PC) e preparo reduzido (PR), num Latossolo Vermelho distroférrico, em Chapecó (SC), sendo os resultados apresentados na média das safras. A cultura do milho, sem leguminosa e sem adubação nitrogenada, apresentou uma receita líquida de 45 R$ ha-1 no PC e de 72 R$ ha-1 no PR. O cultivo intercalar das leguminosas promoveu incrementos na receita líquida da cultura do milho que variaram de 64 a 312 R$ ha-1 (142 a 693%), no PC, e de 57 a 147 R$ ha-1 (79 a 204%), no PR, na ausência de adubação nitrogenada. Mesmo com aplicação de N, as leguminosas promoveram aumento na receita líquida da cultura do milho, porém de menor magnitude. O cultivo intercalar de leguminosas estivais de cobertura do solo demonstrou-se uma alternativa viável para aumentar a receita líquida da cultura do milho. Os benefícios econômicos obtidos com esta prática provavelmente compensam o aumento em mão-de-obra.

Cultivos em vasos têm sido utilizados para avaliar a capacidade dos solos em suprir potássio para as plantas. Com este objetivo, foi realizado um estudo com amostras de dois Planossolos, um Chernossolo e um Gleissolo do estado do Rio Grande do Sul, nos quais a cultura do arroz irrigado por alagamento não tem respondido à adubação potássica. Os solos foram cultivados durante 50 dias, em casa de vegetação, com arroz (Oriza sativa.l cv. BR-IRGA 409). Foram quantificados o potássio acumulado na parte aérea das plantas e os teores de K trocável e K não-trocável, extraídos dos solos por NH4OAc 1 mol L-1 pH 7 e por HNO3 1 mol L-1 fervente, respectivamente, antes e depois do cultivo. O K acumulado pelas plantas de arroz diferiu entre os solos, decrescendo na seqüência: Gleissolo > Chernossolo > Planossolo 1 > Planossolo 2. O K trocável não foi a única forma do elemento no solo que supriu o nutriente às plantas de arroz, ocorrendo contribuição de formas de K não-trocável.

Background: Optimal valganciclovir (VGC) dosage and duration for cytomegalovirus (CMV) prophylaxis in kidney transplant recipients remains controversial. This study aimed to determine GCV blood levels and efficacy/safety observed under low-dose oral VGC in kidney transplant recipients. Secondly, to quantify the variability of GCV blood levels, and its potential clinical impact. Methods: In this prospective study, each patient at risk for CMV undergoing kidney transplantation received low-dose VGC (450 mg qd) prophylaxis for 3 months, unless GFR was below 40 mL/min, in which case the dose was adapted to 450 mg every other day. GCV levels, at trough (Ctrough) and at peak (C3h) were measured monthly and CMV viremia was assessed during and after prophylaxis using real time quantitative Polymerase Chain Reaction. Adverse effects were recorded on each GCV sampling. Patients were followed up to one year after transplantation. Results: 38 kidney recipients (19 D+/R+, 11 D+/R-, 8 D-/R+) received 3-month VGC prophylaxis. Most patients (mean GFR of 59 mL/min) received 450 mg qd but the dose was reduced to 450 mg every other day in 6 patients with mean GFR of 22 mL/min. Average GCV C3h and Ctrough (regressed at 24h or 48h) were 3.9 mg/L (CV 33%, range: 1.3-8.2) and 0.4 mg/L (CV 111%, range 0.1-3.3). Population pharmacokinetic analysis showed a fair dispersion of the parameters mainly influenced by renal function. Despite this variability, patients remained aviremic during VGC prophylaxis. Neutropenia and thrombocytopenia (grade 2-4) were reported in 4% and 3% of patients respectively. During follow-up, asymptomatic CMV viremia was reported in 25% patients. One year after transplantation, 12% patients (all D+/R-) had developed a CMV disease, which was treated with a therapeutic 6-week course of oral VGC. Conclusion: Average GCV blood levels after oral administration of low-dose VGC in kidney transplant recipients were comparable to those previously reported with oral GCV prophylaxis, efficacious and well tolerated. Thus, a 3-month course of low-dose VGC is appropriate for the renal function of most kidney transplant recipients.

Estudar o efeito da calagem e do método de secagem na produtividade do amendoim (Arachis hypogea L., cv. Botutatu) foi o objetivo deste trabalho, conduzido num solo Latossolo Vermelho-Escuro, textura média, em São Manuel, São Paulo. Os tratamentos consistiram de ausência ou presença de calagem (2,05 Mg ha-1) e secagem à sombra, ao sol e duas formas combinadas desta última com estufa. A calagem eliminou a fitotoxicidade de manganês, melhorando a nodulação e a nutrição nitrogenada, que, conseqüentemente, levaram ao aumento do número de ramificações, de vagens por planta e da produtividade. Com a calagem, observou-se também redução nas perdas durante a colheita. Das formas de secagem, a realizada à sombra e a combinada campo-estufa foram as que proporcionaram maiores produtividades, por permitirem melhor maturação dos frutos e menores perdas na colheita.

The objective of this paper was to describe the radiation and energy balance, during the lettuce (Lactuca sativa, L. cv. Verônica) crop cycle inside a polyethylene greenhouse. The radiation and energy balance was made inside a tunnel greenhouse with polyethylene cover (100 mum) and in an external area, both areas with 35 m². Global, reflected and net radiation, soil heat flux and air temperature (dry and humid) were measured during the crop cycle. A Datalogger, which operated at 1 Hz frequency, storing 5 minutes averages was utilized. The global (Kl>¯) and reflected (Kl>) radiations showed that the average transmission of global radiation (Kl>¯in / Kl>¯ex) was almost constant, near to 79.59%, while the average ratio of reflected radiation (Kl>in / Kl>ex) was 69.21% with 8.47% standard-deviation. The normalized curves of short-wave net radiation, in relation to the global radiation (K*/ Kl>¯), found for both environments, were almost constant at the beginning of cycle; this relation decreased in the final stage of culture. The normalized relation (Rn/ Kl>¯) was bigger in the external area, about 12%, when the green culture covered the soil surface. The long-wave radiation balance average (L*) was bigger outside, about 50%. The energy balance, estimated in terms of vertical fluxes, showed that, for the external area, in average, 83.07% of total net radiation was converted in latent heat evaporation (LE), and 18% in soil heat flux (G), and 9.96% in sensible heat (H), while inside of the greenhouse, 58.71% of total net radiation was converted in LE, 42.68% in H, and 28.79% in G.
A rotação de cultura reduz a matocompetição e aumenta o teor de clorofila e a produtividade do arroz

A rotação de culturas é uma prática importante em sistemas agrícolas sustentáveis pelos efeitos benéficos tanto na fertilidade do solo como na redução da matocompetição, entre outros. Este trabalho foi conduzido no campo experimental da UEMA, em São Luís, MA. Avaliou-se o efeito da rotação de culturas na incidência de plantas invasoras, nos teores de clorofila e na produtividade do arroz (Oryza sativa L. cv. Guarani). Cultivou-se o arroz com e sem adubação nitrogenada, bem como em rotação com caupi (Vigna unguiculata (L.) Walp.), crotalária (Crotalaria paulina Schrank) e mucuna (Mucuna aterrima Piper & Tracy). No primeiro ciclo a cobertura morta das leguminosas, deixada sobre o solo para a safra seguinte, foi máxima na crotalária, intermediária na mucuna e mínima no caupi. No segundo ciclo, cultivado em plantio direto sem uso de herbicidas, a rotação com mucuna e crotalária reduziu a biomassa, a cobertura e a densidade das invasoras. O teor de clorofila foi maior na rotação com crotalária do que na testemunha. A produtividade do arroz foi maior na rotação com crotalária e mucuna do que no arroz em cultivo sucessivo com ou sem N. Concluiu-se que a rotação do arroz com leguminosas aumenta a produtividade do arroz e reduz a matocompetição.

Com o objetivo de avaliar os efeitos de diferentes doses de cloreto de mepiquat aplicadas parceladamente no algodoeiro (Gossypium hirsutum L.) cv. CNPA-ITA 90, dois experimentos foram conduzidos durante os anos agrícolas de 1993/94 e 1994/95, na Fazenda Itamarati, Ponta Porã, MS. As doses estudadas foram: 1) 0,0; 2) 12,5+12,5+25,0 (50,0); 3) 25,0+25,0+25,0 (75,0); 4) 0,0+ 50,0+50,0 (100,0) e 5) 12,5+62,5+50,0 (125,0) g ha-1 de cloreto de mepiquat. O delineamento experimental utilizado foi o de blocos casualizados, com quatro repetições. As aplicações foram efetuadas aos 34, 47 e 62 dias após a emergência (DAE), em 1993/94, e aos 42, 60 e 73 DAE no ano de 1994/95, quando as plantas apresentavam altura média de 51, 84 e 93 cm e 61, 79 e 97 cm, respectivamente. Com o aumento da dose de cloreto de mepiquat, verificou-se redução da altura de plantas, da matéria seca foliar, do caule e do total da parte vegetativa, do número de nós, de ramos, do comprimento de ramos, do número de frutos danificados e do número de maçãs, e aumento do número de capulhos totalmente abertos.

The effect of intercropping on plant water status, gas exchange and productivity of maize (Zea mays L.) cv. Centralmex, and cowpea (Vigna unguiculata L. (Walp)) cv. Pitiuba were evaluated under semi-arid conditions at the Embrapa-Centro de Pesquisa Agropecuária do Trópico Semi-Árido (CPATSA) at Petrolina, PE, Brazil. The treatments were: maize and cowpea as sole crops, at a population of 40,000 plants ha-1, and intercropped at a population of 20,000 plants ha-1. The results obtained in this paper appear to be related to the degree of competition experienced by the components, mainly for water and light. Maize intercropped had higher values of leaf water potential, stomatal conductance, transpiration and photosynthesis than as sole crop. Intercropped cowpea had higher values of leaf water potential but lower stomatal conductance, transpiration and photosynthesis than sole cowpea. Maize productivity increased 18% in relation to sole crop whereas a 5% decrease was observed with cowpea. Despite these facts the Land Equivalent Ratio obtained was 1.13 indicating intercropping advantage over the sole system. The higher partial Land Equivalent Ratio observed for maize suggests that this specie was the main component influencing the final productivity of the intercropping system studied.

The productivity and the translocation of assimilates and nitrogen (N) were compared after inoculation of wheat (Triticum aestivum L., cv. BR-23) seeds with two strains of Azospirillum brasilense (strains 245 and JA 04) under field conditions. The inoculation of wheat seeds was done with a peat inoculant at sowing time. Plant material for evaluations were collected at anthesis and maturity. No differences in grain yield and in the translocation of assimilates resulting from inoculation were detected. Differences were observed in relation to N rates (0, 15, and 60 kg ha-1). N content in the grain increased significantly in the bacteria-inoculated treatments in which N was not added. This increase in N content in the grain with inoculation was probably due to higher N uptake after anthesis without any significant contribution on the grain yield. Such increment was of 8.4 kg ha-1 of N representing 66% more N than in no inoculated treatment. Regardless of the inoculation and the rate of N applied, it was observed that about 70% of the N accumulated at anthesis was translocated from vegetative parts to the grain.
