Both tissue plasminogen activator (t-PA) and plasminogen activator inhibitor 2 (PAI-2) are important proteolysis factors present in inflamed human periodontal tissues. The aim of the present study was to investigate the effect of lipopolysaccharide (LPS) on the synthesis of t-PA and PAI-2 by human gingival fibroblasts (HGF). LPS from different periodontal pathogens including Actinobacillus actinomycetemcomitans, Porphyromonas gingivalis and Fusobacterium nucleatum were extracted by the hot phenol water method. The levels of t-PA and PAI-2 secreted into the cell culture media were measured by enzyme-linked immunosorbent assays (ELISA). The mRNA for t-PA and PAI-2 were measured by RT-PCR. The results showed t-PA synthesis was increased in response to all types of LPS studied and PAI-2 level was increased by LPS from A. actinomycetemcomitans and F. nucleatum, but not P. gingivalis. When comparing the effects of LPS from non-periodontal bacteria (Escherichia coli and Salmonella enteritidis) with the LPS from periodontal pathogens, we found that the ratio of t-PA to PAI-2 was greater following exposure of the cells to LPS from periodontal pathogens. The highest ratio of t-PA to PAI-2 was found in those cells exposed to LPS from P. gingivalis. These results indicate that LPS derived from periodontal pathogens may cause unbalanced regulation of plasminogen activator and plasminogen activator inhibitor by HGF and such an effect may, in part, contribute to the destruction of periodontal connective tissue through dysregulated pericellular proteolysis.

Proton-bound dimers consisting of two glycerophospholipids with different headgroups were prepared using negative ion electrospray ionization and dissociated in a triple quadrupole mass spectrometer. Analysis of the tandem mass spectra of the dimers using the kinetic method provides, for the first time, an order of acidity for the phospholipid classes in the gas phase of PE < PA << PG < PS < PI. Hybrid density functional calculations on model phospholipids were used to predict the absolute deprotonation enthalpies of the phospholipid classes from isodesmic proton transfer reactions with phosphoric acid. The computational data largely support the experimental acidity trend, with the exception of the relative acidity ranking of the two most acidic phospholipid species. Possible causes of the discrepancy between experiment and theory are discussed and the experimental trend is recommended. The sequence of gas phase acidities for the phospholipid headgroups is found to (1) have little correlation with the relative ionization efficiencies of the phospholipid classes observed in the negative ion electrospray process, and (2) correlate well with fragmentation trends observed upon collisional activation of phospholipid \[M - H](-) anions. The colony of Queensland, established on 10 December 1859, passed its first Education Act in 1860. This enabled the government to provide for the education of all children from six to twelve years of age. The Board of General Education appointed its first Inspector of Schools in 1863, and he was followed by over 400 men — and later women — to inspect schools and teachers to guarantee conformity and efficiency. In 1875, education became a ministerial responsibility and the Department of Public Instruction was established. Teachers never seemed to enjoy a friendly working relationship with inspectors, and with the establishment of the Queensland Teachers Union, hostility increased. Eventually, in 1989, the Inspector of Schools position was abolished.

Molasses-based liquid supplements fed ad libitum are widely used to provide additional metabolisable energy, non-protein N (NPN) and other nutrients to grazing cattle, but it is often difficult to achieve target intakes of supplementary nutrients. Experiments examined the effects of increasing concentrations of phosphoric acid, urea and ammonium sulfate on the voluntary intake (VI) of molasses-based supplements offered ad libitum to heifers grazing tropical pastures. In Experiment 1, the VI of a supplement containing 78 g urea/kg and 26 g phosphoric acid/kg as-fed (M80U+PA) was 3.61 g DM/kg liveweight (LW) per day, and provided 181 mg NPN and 32.4 mg phosphorus (P)/kg LW per day. Increasing the urea content of the supplement to 137 g/kg (M140U+PA) or 195 g/kg (M200U+PA) reduced VI of supplement DM, NPN and P by up to 76%, 44% and 80%, respectively. VI of supplement containing ammonium sulfate (M140+AS+PA) was lower (P < 0.05) than that of M140U+PA supplement, and tended (P > 0.05) to be lower than that of M200U+PA supplement. In experiment 2, the VI by heifers of a supplement containing 200 g urea/kg (M200U) was 1.53 g supplement DM/kg LW per day, which provided 186 mg NPN/kg LW per day. Inclusion of 49 g phosphoric acid/kg as-fed in this supplement (M190U+50PA) reduced (P < 0.05) VI of supplement DM and NPN by 33% and 36%, respectively, while inclusion of 97 g phosphoric acid/kg (M180U+100PA) reduced (P < 0.05) VI of supplement DM and NPN by 43% and 48%, respectively. The M190U+50PA and M180U+100PA supplements provided 16 and 26 mg P/kg LW per day, respectively. Heifers not fed supplements gained 0.07 kg/day, and the M200U supplement increased (P < 0.05) LW gain to 0.18 kg/day. LW gain was further increased (P < 0.05) by the M190U+50PA to 0.28 kg/day, indicating a growth response to supplementary P. No adverse effects of the supplements on animal health were observed in any of the experiments. In conclusion, addition of urea and/or phosphoric acid to molasses supplements effectively reduced VI of supplementary DM, NPN and P, and in the circumstances of Experiment 2, both molasses-urea and P supplements increased heifer LW.

The density-matrix renormalization group (DMRG) method is used for a comparative study of low-lying excitations in trans-polyacetylene (t-PA) and transversely substituted t-PA (TS-t-PA). We have employed the Pariser-Parr-Pople model Hamiltonian which incorporates long-range electronic correlations to model these systems. We find some fundamental differences in the excited states of the t-PA and TS-t-PA. We find that the lowest two-photon allowed excited state in TS-t-PA is not made up of two triplet excitons and the gap to this state is nonzero even for undimerized chains in the thermodynamic limit. Contrary to earlier results for the Hubbard model, we find that the lowest two-photon state is always below the first optically allowed state in all the systems studied here making TS-t-PA systems only weakly fluorescent materials. Nonresonant tumbling averaged linear and third harmonic generation optic coefficients of TS-t-PA systems are also much smaller than that of t-PA.

We present a systematic study to explore the effect of important process variables on the composition and structure of niobium nitride thin films synthesized by Reactive Pulsed Laser Deposition (RPLD) technique through ablation of high purity niobium target in the presence of low pressure nitrogen gas. Secondary Ion Mass Spectrometry has been used in a unique way to study and fix gas pressure, substrate temperature and laser fluence, in order to obtain optimized conditions for one variable in single experimental run. The x-ray diffraction and electron microscopic characterization have been complemented by proton elastic backscattering spectroscopy and x-ray photoelectron spectroscopy to understand the incorporation of oxygen and associated non-stoichiometry in the metal to nitrogen ratio. The present study demonstrates that RPLD can be used for obtaining thin film architectures using non-equilibrium processing. Finally the optimized NbN thin films were characterized for their hardness using nano-indentation technique and found to be similar to 30 GPa at the deposition pressure of 8 Pa. A protease inhibitor from the seeds of Butea monosperma (BmPI) was purified, characterized and studied for its influence on developmental physiology of Helicover-pa armigera. BmPI on two-dimensional separations indicated the presence of a 14 kDa protein with an isoelectric point in the acidic region (pl 5.6). Multiple Sequence Analysis data suggested that the BmPI contains a sequence motif which is conserved in various trypsin and chymotrypsin inhibitors of Kunitz-type. The inhibitor exhibited trypsin inhibitory activity in a broad range of pH (4-10) and temperature (10-80 degrees C). The enzyme kinetic studies revealed BmPI as a competitive inhibitor with a K-i value of 1.2 x 10(-9) M. In vitro studies with BmPI indicated measurable inhibitory activity on total gut proteolytic enzymes of H. armigera (IC(50)2.0 mu g/ml) and bovine trypsin. BmPI supplemented artificial diet caused dose dependent mortality and reduction in growth and weight. The fertility and fecundity of H. armigera, declined whereas the larval-pupal duration of the insect life cycle extended. These detrimental effects on H. armigera suggest the usefulness of BmPl in insect pest management of food crops. (C) 2014 Elsevier Ltd. All rights reserved.

We demonstrate a non-contact technique to apply calibrated and localized forces in the micro-Newton to milli-Newton range using an air microjet. An electromagnetically actuated diaphragm controlled by a signal generator is used to generate the air microjet. With a nozzle diameter of 150 mu m, the microjet diameter was maintained to a maximum of 1 mm at a distance of 5 mm from the nozzle. The force generated by the microjet was measured using a commercial force sensor to determine the velocity profile of the jet. Axial flow velocities of up to 25 m s(-1) were obtained at distances as long as 6 mm. The microjet exerted a force up to 1 mu N on a poly dimethyl siloxane (PDMS) micropillar (50 mu m in diameter, 157 mu m in height) and 415 mu N on a PDMS membrane (3 mm in diameter, 28 mu m thick). We also demonstrate that from a distance of 6 mm our microjet can exert a peak pressure of 187 Pa with a total force of about 84 mu N on a flat surface with 8 V operating voltage. Out of the cleanroom fabrication and robust design make this system cost effective and durable.

ZnO thin films were deposited on the substrates of (100) gamma-LiAlO2 at 400, 550 and 700 degrees C using pulsed laser deposition (PLD) with the fixed oxygen pressure of 20 Pa, respectively. When the substrate temperature is 400 degrees C, the grain size of the film is less than 1 mu m observed by Leitz microscope and measured by X-ray diffraction (XRD). As the substrate temperature increases to 550 degrees C, highly-preferred c-orientation and high-quality ZnO film can be attained. While the substrate temperature rises to 700 degrees C, more defects appears on the surface of film and the ZnO films become polycrystalline again possibly because more Li of the substrate diffused into the ZnO film at high substrate temperature. The photoluminescence (PL) spectra of ZnO films at room temperature show the blue emission peaks centered at 430 nm. We suggest that the blue emission corresponds to the electron transition from the level of interstitial Zn to the valence band. Meanwhile, the films grown on gamma-LiAlO2 (LAO) exhibit green emission centered at 540 nm, which seemed to be ascribed to excess zinc and/or oxygen vacancy in the ZnO films caused by diffusion of Li. from the substrates into the films during the deposition.

O objetivo central desta pesquisa consiste em verificar a existência de semelhanças entre o planejamento estratégico situacional e o orçamento participativo implementado em Belém durante o período de 1997 a 2000; assim como verificar a existência de semelhanças entre a teoria da escolha pública e o orçamento participativo em Belém no mesmo período. Para alcançar tal objetivo, serão abordadas resumidamente, a teoria da escolha pública e o planejamento estratégico situacional, como base teórica para desenvolver o estudo. Supõe-se que essas teorias fundamentam o orçamento participativo, instrumento mais conhecido de participação popular no processo da gestão pública. Para alcançar esse fim, utilizou-se o método de estudo de caso, por ser o mais indicado para esse tipo de pesquisa. Os resultados sugerem que o orçamento participativo, do modo como foi implementado na Capital do Estado do Pará, no período de 1997 a 2000 possui características da teoria e do método aqui levantados. No que diz respeito à teoria da escolha pública, características em comum com o orçamento participativo foram observadas, como reconhecer que os agentes políticos são motivados por interesses pessoais, além de admitir que a escolha política, no processo político, é resultado das preferências dos agentes envolvidos nas escolhas que permitem passar de preferências diversas de cada indivíduo para uma única escolha coletiva. Quanto ao planejamento estratégico situacional, também foram observados conceitos e características em comum com o orçamento participativo. Ambos reconhecem a existência de vários atores dentro do processo de planejamento, que passa a ser discutido diretamente com a população, inserindo todos os setores do município no processo de gestão. Através de reivindicações, que são entendidas como problemas, o planejamento passa a ser dividido com a população, e esta passa a ser atuante no planejamento do espaço em que vive. Outra característica em comum é a subjetividade, que diz respeito ao entendimento que cada ator tem de seu próprio problema, e do problema de outros atores. Pôde-se observar que a participação popular na gestão pública é instrumento de elevada importância para impulsionar o desenvolvimento social, sendo também incentivada nas formas previstas em Lei. No entanto, tal incentivo acarreta fatores diversos que por vezes fogem ao controle dos gestores, além de significar uma divisão de poderes.

Trying to pass someone walking toward you in a narrow corridor is a familiar example of a two-person motor game that requires coordination. In this study, we investigate coordination in sensorimotor tasks that correspond to classic coordination games with multiple Nash equilibria, such as "choosing sides," "stag hunt," "chicken," and "battle of sexes". In these tasks, subjects made reaching movements reflecting their continuously evolving "decisions" while they received a continuous payoff in the form of a resistive force counteracting their movements. Successful coordination required two subjects to "choose" the same Nash equilibrium in this force-payoff landscape within a single reach. We found that on the majority of trials coordination was achieved. Compared to the proportion of trials in which miscoordination occurred, successful coordination was characterized by several distinct features: an increased mutual information between the players' movement endpoints, an increased joint entropy during the movements, and by differences in the timing of the players' responses. Moreover, we found that the probability of successful coordination depends on the players' initial distance from the Nash equilibria. Our results suggest that two-person coordination arises naturally in motor interactions and is facilitated by favorable initial positions, stereotypical motor pattern, and differences in response times.

The Monitor National Marine Sanctuary (MNMS) was the nation’s first sanctuary, originally established in 1975 to protect the famous civil war ironclad shipwreck, the USS Monitor. Since 2008, sanctuary sponsored archeological research has branched out to include historically significant U-boats and World War II shipwrecks within the larger Graveyard of the Atlantic off the coast of North Carolina. These shipwrecks are not only important for their cultural value, but also as habitat for a wide diversity of fishes, invertebrates and algal species. Additionally, due to their unique location within an important area for biological productivity, the sanctuary and other culturally valuable shipwrecks within the Graveyard of the Atlantic are potential sites for examining community change. For this reason, from June 8-30, 2010, biological and ecological investigations were conducted at four World War II shipwrecks (Keshena, City of Atlanta, Dixie Arrow, EM Clark), as part of the MNMS 2010 Battle of the Atlantic (BOTA) research project. At each shipwreck site, fish community surveys were conducted and benthic photo-quadrats were collected to characterize the mobile conspicuous fish, smaller prey fish, and sessile invertebrate and algal communities. In addition, temperature sensors were placed at all four shipwrecks previously mentioned, as well as an additional shipwreck, the Manuela. The data, which establishes a baseline condition to use in future assessments, suggest strong differences in both the fish and benthic communities among the surveyed shipwrecks based on the oceanographic zone (depth). In order to establish these shipwrecks as sites for detecting community change it is suggested that a subset of locations across the shelf be selected and repeatedly sampled over time. In order to reduce variability within sites for both the benthic and fish communities, a significant number of surveys should be conducted at each location. This sampling strategy will account for the natural differences in community structure that exist across the shelf due to the oceanographic regime, and allow robust statistical analyses of community differences over time.

Trimeresurus stejnegeri venom, which contains TSV-PA (a specific plasminogen activator sharing 60-70% sequence homology with venom fibrinogen-clotting enzymes), also possesses fibrinogen-clotting activity in vitro. A fibrinogen-clotting enzyme (stejnobin) has been purified to homogeneity by gel filtration and ion-exchange chromatography on a Mono-Q column. It is a single-chain glycoprotein with a mol. wt of 44,000. The NH2-terminal amino acid sequence of stejnobin shows great homology with venom fibrinogen-clotting enzymes and TSV-PA. Like TSV-PA, stejnobin was able to hydrolyse several chromogenic substrates. Comparative study of substrate specificities of stejnobin and other venom proteases purified in our laboratory was carried out on five chromogenic substrates. Stejnobin clotted human fibrinogen with a specific activity of 122 NIH thrombin-equivalent units/mg protein. However, stejnobin did not act on other blood coagulation factors, such as factor X, prothrombin and plasminogen. Diisopropyl fluorophosphate and phenylmethanesulfonyl fluoride inhibited its activity, whereas ethylenediamine tetracetic acid had no effect on it, indicating that it is a serine protease. Although stejnobin showed strong immunological cross-reaction with polyclonal antibodies raised against TSV-PA, it was interesting to observe that, unlike the case of TSV-PA, these antibodies did not inhibit the amidolytic and fibrinogen-clotting activities of stejnobin. Changes of plasminogen activators (PA) during different stages of development of the corpus luteum, and their possible physiological role in luteolysis were studied in rhesus monkeys. It was demonstrated for the first time that monkey corpus luteal cells not only produce PA, but that the function of the corpus luteum is also closely related to the activity of this enzyme system. Generally, the life span for a corpus luteum in monkey is approximately 14-16 days, its demise beginning thereafter. In the present study, we found that urokinase in the corpus luteum is higher on day 5 and day 10 after human chorionic gonadotrophin injection, while the tissue type (t) PA is mainly produced on day 13 when luteolysis may take place. Progesterone production remained high on day 5 and day 10 and decreased dramatically from day 13, indicating the important role of tPA but not urokinase (u) PA in suppressing luteal function. When purified tPA (but not uPA) monoclonal antibody was added to luteal cell culture to neutralize endogenously produced tPA activity, progesterone production in the cells was increased significantly. Interestingly, prolactin alone was capable of increasing PA production by luteal cells; prolactin together with luteinizing hormone, however, had a synergistic luteotrophic effect.

Random amplified polymorphic DNA (RAPD) molecular markers specific for one, two or three clones have been identified from five gynogenetic clones of silver crucian carp (Carassius auratus gibelio Bloch) using RAPD markers developed earlier. In this study, three RAPD markers (RA1-PA, RA2-EF and RA4-D) produced by Opj-1, and two RAPD DNA fragments (RA3-PAD and RA5-D) produced by Opj-7, were selected for molecular cloning and sequencing. Sequence data indicated that there were identical 801-bp nucleotide sequences in the shared marker RA1-PA cloned respectively from clones P and A, and the shared marker RA2-EF (which was cloned from clones E and F), were also of identical 958-by nucleotide sequences. The nucleotide sequences of the shared marker RA3-PAD fragments were also similar for 1181 by among clones P, A and D. The specific fragment RA4-D was composed of 628 bp, and the fragment RA5-D from clone D contained 385 nucleotides. According to the nucleotide sequences, we designed and synthesized five pairs of sequence characterized amplified regions (SCAR) primers to identify the specific fragments in these gynogenetic clones of silver crucian carp. Only individuals from clones P and A amplified a specific band using a pair of SCI-PA primers synthesized according to the marker RA1-PA sequences, whereas no products were detected in individuals from clones D, E and F. The PCR products amplified using SC2-EF and SC3-PAD primers were as expected. Furthermore, the pair of SC4-D primers amplified specific bands only in individuals from clone D, although weak bands could be produced in all individuals of the five clones when lower annealing temperatures were used. However, an additional pair of SC5-D primers designed from the RA5-D marker sequences could amplify a DNA band in individuals from clones P, A and D, and the same weak band was produced in clone E, whereas no products were detected in individuals from clone F. Searches in GenBank revealed that the 385-bp DNA fragment from RA5-D was homologous to the 5' end of gonadotropin I beta subunit 2 gene and growth hormone gene. No homologous sequences were found for other markers in GenBank. The SCAR markers identified in this study will offer a powerful, easy, and rapid method for discrimination of different clones and for genetic analyses that examine their origins and unique reproductive modes in crucian carp. Furthermore, they will likely benefit future selective breeding programs as reliable and reproducible molecular markers. (C) 2001 Elsevier Science B.V. All rights reserved.
