Haematopoiesis and blood cells` functions can be influenced by dietary concentration of nutrients. This paper studied the effects of dietary protein:energy ratio on the growth and haematology of pacu, Piaractus mesopotamicus. Fingerling pacu (15.5 +/- 0.4 g) were fed twice a day for 10 weeks until apparent saciety with diets containing 220, 260, 300, 340 or 380 g kg(-1) crude protein (CP) and 10.88, 11.72, 12.55, 13.39, 14.22 MJ kg(-1) digestible energy (DE) in a totally randomized experimental design, 5 x 5 factorial scheme (n=3). Weight gain and specific growth rate were affected (P < 0.05) by protein level only. Protein efficiency ratio decreased (P < 0.05) with increasing dietary protein at all levels of dietary energy. Daily feed intake decreased (P < 0.05) with increasing dietary energy. Mean corpuscular haemoglobin concentration was affected (P < 0.05) by DE and interaction between dietary CP and DE. Total plasma protein increased (P < 0.05) with dietary protein and energy levels. Plasma glucose decreased (P < 0.05) with increasing dietary protein. The CP requirement and optimum protein:energy ratio for weight gain of pacu fingerlings, determined using broken-line model, were 271 g kg(-1) and 22.18 g CP MJ(-1) DE respectively. All dietary CP and DE levels studied did not pose damages to fish health.

The objective of this experiment was to evaluate the effects of replacing coastcross hay NDF by soybean hull (SH) NDF on the lactation performance and eating behavior of ewes and also on the performance of their lambs. Fifty-six Santa Ines lactating ewes (56.1 +/- 6.8 kg of initial BW; mean +/- SD) were penned individually and used in a randomized complete block design with 14 blocks and 4 treatments. Diets were formulated to provide similar concentrations of NDF (56%) and CP (16%). The SH NDF replaced 33 (SH33), 67 (SH67), or 100% (SH100) of the NDF contributed by coastcross hay in a 70% forage-based diet (SH0), resulting in SH inclusion rates of 0, 25, 54, and 85% of the dietary DM. Once a week, from the second to the eighth week of lactation (weaning time), ewes were separated from their lambs, stimulated by a 6-IU i.v. oxytocin injection, and hand milked to empty the udder. After 3 h, milk production was obtained after the same procedure. Quadratic effect for milk production (142.4, 179.8, 212.6, and 202.9 g/3 h) and cubic effect for DMI (2.27, 2.69, 3.25, and 3.00 kg/d) were observed as SH inclusion increased from 0 to 85% of the dietary DM. Milk fat (7.59, 7.86, 7.59, and 7.74%), protein (4.53, 4.43, 4.40, and 4.55%), and total solids (18.24, 18.54, 18.39, and 18.64%) did not differ among the 70% forage-based diet and diets with SH NDF replacing 33, 67, or 100% of the NDF. A linear increase in lactose concentration was observed with SH inclusion. Ewe BW gain during the trial showed a cubic response (0.37, 0.03, 4.80, and 2.80 kg) with SH inclusion. The preweaning ADG of lambs increased linearly, and ADG of lambs after weaning decreased linearly with SH inclusion. Final BW of lambs (2 wk after weaning) did not differ among treatments. Eating behavior observations were conducted with 44 ewes. The same facilities, experimental design, dietary treatments, and feeding management were used. Observations were visually recorded every 5 min for a 24-h period when ewes were 46 +/- 6.8 d in milk. Eating time (min/d, min/g of DMI, and min/g of NDF intake) and time expended in rumination and chewing activities (min/g of DMI and min/g of NDF intake) decreased linearly with the addition of SH in the diets. The inclusion of SH improved DMI and milk production, also reflecting on the BW of lambs at weaning. Milk performance was not affected when SH NDF replaced 100% of hay NDF.

Nitrogen fertilization in common bean crops under no-tillage and conventional systems. Nitrogen fertilizer is necessary for high yields in common bean crops and N responses under conditions of no-tillage and conventional systems are still basic needs. Thus, the objective of this research was to evaluate the effect of N application and common bean yield in no-tillage and conventional systems. The experimental design was a randomized block in a factorial scheme (2x8+1) with four replications. The treatments were constituted by the combination of two N doses (40 and 80 kg ha(-1)) applied at side dressing at eight distinct stadia during vegetative development of the common bean (V(4-3), V(4-4), V(4-5), V(4-6), V(4-7), V(4-8), V(4-9) and V(4-10)), in addition to a control plot without N in side dressing. The experiment was conducted over two years (2002 and 2003) in no-tillage on millet crop residues and conventional plow system. It was concluded that N fertilizer at the V(4) stadium of common bean promotes similar seed yields in no-tillage and conventional systems. Yield differences between no-tillage and conventional systems are inconsistent in the same agricultural area.

This study aimed to evaluate the average behavior, the genotype x environment (GxE), adaptability and stability of seven soybean cultivars at three sowing dates in Uberlandia-MG. The tests were conducted at Capim Branco Farm, belonging to the Federal University of Uberlandia. Sowing was held on october 29 (1st season), november 24 (2nd season) and december 17 (3rd season) 2007. The experimental design was a randomized, seven genotypes (UFUS Xavante, UFUS Riqueza, UFUS Guarani, UFUS Milionaria, Msoy 8001, Msoy 8411 and Msoy 8914) with three replications in each of three sowing dates. Means were compared by Tukey test at 5% probability. Analysis of adaptability and phenotypic stability of genotypes was performed using the Eberhart and Russell (1966), Lin and Binns (1988) modified by Carneiro (1998) and centroid (NASCIMENTO et al., 2009). For grain yield, the cultivar UFUS Xavante was classified as specific adaptability to environment and high stability. The other cultivars were classified as being of general adaptability. For oil content, the cultivars Msoy 8914 and UFUS Xavante behaved as high stability and was classified as having high adaptability. For the character content of protein, all cultivars behaved as wide adaptability and low stability.

Yield is closely linked to the plant yield components and depend directly of the genotype interaction with the environment. Essays were installed in the experimental field of the Bahia Foundation in the Maria Gabriela farm in the county of Sao Desiderio - BA in the year 2006-2007. The aim of this work was to evaluate yield components of five soybean cultivars with different maturation cycles indicated for the Western Region of Bahia in different sowing periods. The experimental design was in random blocks in 4 x 5 factorial scheme (four sowing periods: Ep1 first - 11/29/2006, Ep2 second - 12/14/2006, Ep3 third - 12/28/2007, Ep4 fourth - 01/12/2007 and five cultivars: M-SOY 8411, BRS Corisco, BRS 263 [Diferente], BRS Barreiras e M-SOY 9350) with four repetitions. The following characteristics were evaluated: total number of pods per plant, total number of beans per plant, mass of 1000 beans and yield. Plant yield components, total number of pods per plant, total number of beans per plant, mass of 1000 beans, reduced with sowing delay and showed compensation effect between cultivars and sowing periods. Late sowing Ep3 (28/12/2006) and Ep4 (12/01/2007) were not favourable to raise yield of soybean in the Western Region of Bahia.

This work aimed at evaluating the total carotenoids production by a newly isolated Sporidiobolus pararoseus. Bioproduction was carried out in an orbital shaker, using 10% (w/v) of inoculum (25 A degrees C, 180 rpm for 35 h), incubated for 120 h in a dark room. Liquid N(2) and dimethylsulphoxide (DMSO) were used for cell rupture, and carotenoids were extracted with a solution of acetone/methanol (7:3, v/v). Optimization of carotenoids bioproduction was achieved by experimental design technique. Initially, a Plackett-Burman design was used for the screening of the most important factors, after the statistical analysis, a complete second-order design was carried out to optimize the concentration of total carotenoids in a conventional medium. Maximum concentration of 856 mu g/L of total carotenoids was obtained in a medium containing 60 g/L of glucose, 15 g/L of peptone, and 15 g/L of malt extract, 25 A degrees C, initial pH 4.0 and 180 rpm. Fermentation kinetics showed that the maximum concentration of total carotenoids was reached after 102 h of fermentation and that carotenoids bioproduction was associated with cell growth.

Nutrition in bean plants and anthracnose intensity in function of silicon and copper application. The objective of this work was to evaluate the effect of calcium silicate and copper sulfate on anthracnose intensity and nutrition of bean plants. The experiment was conducted using an experimental design in randomized blocks following a 4 x 4 factorial arrangement , (four levels of calcium silicate and four levels of copper sulfate) and two additional treatments (plants without inoculation and plants sprinkled with Benomyl). Four evaluations of the incidence and severity of anthracnose were done, in addition to measuring, total leaf area. At the end of the evaluations, incidence: and data were integrated over time, obtaining the area under disease progress curve (AUDPC). Contents of N, P, K, Ca, Mg, B, Cu, Fe, Mn, Zn, Si and lignin were determined in the aerial Part. A linear decrease of the intensity AUDPC was observed with the increase of the doses of calcium silicate. The severity AUDPC was influenced by the doses of copper, obtaining a reduction of 35% on the higher dosage. The supply of silicon and copper altered the content of the K, mg, S, Zn, Ca and Si in the aerial part of the bean plants.

The variation in the Ca:Mg ratio in amendments used to neutralize soil acidity is one way of altering the availability of those nutrients to the plants in acid soils. The objective of the work was to evaluate the effect of different proportions of calcium and magnesium in the form of CaCO(3) and MgCO(3) Oil the nutrient uptake, and initial production of dry matter by corn plants. The study was carried out in greenhouse conditions, in Lages, SC, with a completely randomized experimental design, with three replications. The treatments were the application of equivalent to 21.0 t ha(-1) of lime, using mixtures of CaCO(3) and MgCO(3) in several proportions to obtain different Ca:Mg ratios (1: 1, 2:1, 4:1, 8:1, 16:1 and 32:1), on a Humic Alic Cambisol, with 310 g kg(-1) of clay. The application of treatments caused the following Ca:Mg ratios in the CEC: 1. 1: 1, 2.1:1, 4.0:1, 8.1:1, 16.4:1 and 31.8:1. The high concentrations of exchangeable Ca in soil caused by addition of lime with high Ca content inhibited the uptake of Mg and K by the corn plants. The increase in the soil Ca:Mg ratio reduced the dry matter production and height of plants in the initial stage of development.

Due to the lack of information about the effects of soybean fertilization anticipation and the use of Eleusine coracana (L.) Gaerm. (ANSB Pe-de-galinha 5352) as cover culture, and the need for new techniques for the management of the agro-ecosystem in a more conservationist, sustainable and functional manner the goal of this research was to study the effects of anticipated fertilization on the production of dry matter in E. coracana. The experiments were carried out in an Oxisol, during the growing seasons of 200112002, 200212003 and 200312004, in Piracicaba, SP Brazil (22`50`25""S and 48`01`65""W). The experimental design was of totally randomized blocks and twelve treatments (lev- els of anticipated fertilization) were used with three repetitions. The base fertilization of soybean culture was partially anticipated, in the sowing for the finger-millet crop. Approximately 70 days after sowing, samples of the finger millet plants were collected in order to evaluate the dry matter production and to desiccate them thereafter It is concluded that the anticipation of phosphorus and potassium fertilization of soybean increases the dry matter production of E. coracana; in addition, E. coracana holds a great potential for the production of plant residues and it can be used in culture rotation or in no-tillage production systems.

An adequate supply of nitrogen is essential for high yield of common bean seeds; however, the responses to this nutrient in no-tillage systems can vary in function of the species and the amount of straw present. The aim of this research was to evaluate response of the common bean to N in no-tillage systems over maize, millet and Brachiaria brizantha crop residues. Three experiments were conducted, one for each gramineous species. The experimental design was a randomized block in a 5x2x2 factorial scheme, with five N doses (0, 30, 60, 90, 120 kg ha(-1)), in two distinct stadia during the vegetative development (3(rd) and 6(th) trifoliate leaf) and two common bean cultivars (IPR Juriti and Perola), with four replications. The IPR Juriti produced a greater number of pods per plant and showed lower mass per 100 seeds than Perola. It did not show variation in bean yield when the sidedressing N application was carried out in the 3(rd) or 6(th) trifoliate leaf stadia. Based on the obtained results, one concludes that the seed productivity of common beans increases linearly with N doses in a no-tillage system over maize residues.

This work aimed to evaluate the effect of different concentrations of IBA (indolbutyric acid) in the rooting and growth of Eucalyptus urophylla cuttings. The experimental design used was the randomized blocks, in factorial with an arrangement of split plots, with three concentrations of IBA (2.000; 5.000 and 8.000 mg L(-1)), two ways of the application of plant gowth regulators (paste and powder), and three period of evaluations (30, 45 and 60 days). The experiment was carried out in the Farm Buriti de Prata, an entreprise property of Souza Cruz, in the city of Prata - MG in 2003. After the preparation of the cuttings, patterned in 10 cm of length and 0.8 cm of diameter, those were immersed in mixtures of IBA for 10 seconds, in the forms of dry powder and paste and than planted in plastic tubes contening Plantmax substrate with vermiculite. The cuttings were transported to greenhouse with controled humidity, where they remained for 60 days. The variables studied were: height of plant in the 30(th), 45(th) and 60(th) days after seeding; fresh mass of the aerial part and roots. The IBA applied in powder form as well as in paste form, resulted in an greater seedling growth. To the 60 days, the seedlings presented greater growth, being significantly superior to the heights measured in other times of evaluation. The application of 2.000 mg L(-1) and 5,000 mg L(-1) resulted in significant increases on weight of fresh mass of the aerial part and root system.

Many therapeutic agents are commercialized under their racemic form. The enantiomers can show differences in the pharmacokinetic and pharmacodynamic profile. The use of a pure enantiomer in pharmaceutical formulations may result in a better therapeutic index and fewer adverse effects. Atropine, an alkaloid of Atropa belladonna, is a racemic mixture of l-hyoscyamine and d-hyoscyamine. It is widely used to dilate the pupil. To quantify these enantiomers in ophthalmic solutions, an HPLC method was developed and validated using a Chiral AGP (R) column at 20 degrees C. The mobile phase consisted of a buffered phosphate solution (containing 10 mM 1-octanesulfonic acid sodium salt and 7.5 mM triethylamine, adjusted to pH 7.0 with orthophosphoric acid) and acetonitrile (99 + 1, v/v). The flow rate was 0.6 mL/min, with UV detection at 205 nm. In the concentration range of 14.0-26.0 mu g/mL, the method was found to be linear (r > 0.9999), accurate (with recovery of 100.1-100.5%), and precise (RSD system: <= 0.6%; RSD intraday: <= 1.1%; RSD interday: <= 0.9%). The method was specific, and the standard and sample solutions were stable for up to 72 h. The factorial design assures robustness with a variation of +/-10% in the mobile phase components and 2 degrees C of column temperature. The complete validation, including stress testing and factorial design, was studied and is presented in this research.

A 2(3-1) factorial experimental design was used to evaluate the performance of a perforated rotating disc contactor to extract alpha-toxin from the fermented broth of Clostridium perfringens Type A by aqueous two-phase system of polyethylene glycol-phosphate salts. The influence of three independent variables, specifically the dispersed phase flowrate, the continuous phase flowrate and the disc rotational speed, was investigated on the hold up, the mass transfer coefficient, the separation efficiency and the purification factor, taken as the response variables. The optimum dispersed phase flowrate was 3.0 mL/min for all these responses. Besides, maximum values of hold up (0.80), separation efficiency (0. 10) and purification factor (2.4) were obtained at this flowrate using the lowest disc rotational speed (35 rpm), while the optimum mass transfer coefficient (0. 165 h(-1)) was achieved at the highest agitation level (140 rpm). The results of this study demonstrated that the dispersed phase flowrate strongly influenced the performance of PRDC, in that both the mass transfer coefficient and hold up increased with this parameter. This study dealt with the influence of both the feeding time and light intensity on the fed-batch culture of the cyanobacterium Spirulina (Arthrospira) platensis using ammonium chloride as a nitrogen source. For this purpose, a 2 2 plus star central composite experimental design combined with response surface methodology was employed, and the maximum cell concentration (X-m), the cell productivity (P-X), and the yield of biomass on nitrogen (Y-X/N) were selected as the response variables. The optimum values of X-m (1,833 mgL(-1)) and Y-X/N (5.9 gg(-1)) estimated by the model at light intensity of 13 klux and feeding time of 17.2 days were very close to those obtained experimentally under these conditions (X-m = 1,771 +/- 41 mg L-1; Y-X/N = 5.7 +/- 0.17 gg(-1)). The cell productivity was a decreasing function of the ammonium chloride feeding time and a quadratic function of the light intensity. The protein and lipid contents of dry biomass collected at the end of cultivations were shown to decrease with increasing light intensity.

A partial pseudo-ternary phase diagram has been studied for the cethyltrimethylammonium bromide/isooctane:hexanol:butanol/potassium phosphate buffer system, where the two-phase diagram consisting of the reverse micelle phase (L-2) in equilibrium with the solvent is indicated. Based on these diagrams two-phase systems of reverse micelles were prepared with different compositions of the compounds and used for extraction and recovery of two enzymes, and the percentage of enzyme recovery yield monitored. The enzymes glucose-6-phosphate dehydrogenase (G6PD) and xylose redutase (XR) obtained from Candida guilliermondii yeast were used in the extraction procedures. The recovery yield data indicate that micelles having different composition give selective extraction of enzymes. The method can thus be used to optimize enzyme extraction processes.
