The aim of this paper was to evaluate the effect of retention and detention reservoirs along with the regulation in channel flow upgrade on flood for an urban watershed located at Rio Claro, SP. For this purpose, modeling and simulation techniques were applied for runoff determination and its propagation in channel. The Soil Conservation Service – SCS hydrologic model as well as Pulz and non-linear Muskingum-Cunge model were used. The software IPHS1 was applied on simulations. The results pointed out that the combination of retention increasing and detention reservoir implementation (120,000 m3, corresponding to 1.5% of the watershed area) with the streamflow upgrade (n decreasing from 0,04 to 0,02) can minimize the flood on the investigated Servidão watershed. Further, after the proposed intervention, the flood was eliminated for the investigated times of recurrence: 5, 20, 50 and 100 years. The prognostic indicated that the available area occupation had a minor effect on flow increasing due to the observed high urbanization.

Watersheds are considered important study units when it comes to environmental planning, with regard to the optimal use of water resources. Water scarcity is predicted and feared by many societies, and proves to be an increasingly tangible problem nowadays. Still from the perspective of extreme events, this dissertation considers the study of flood waves in the sub-basin of the stream Claro, which belongs to the Corumbataí watershed. - SP, since thay can also have devastating effects for the population, A Decision Support System for Flood Routing Analysis in Complex Basins, ABC 6 software was applied in order to obtain hydrographs and peak flows in the sub-basin of the stream Claro, for return periods of 10 and 100 years, aiming to comprise events of different magnitudes. The model Soil Conservation Service (SCS) and the triangular SCS hydrograph were adopted for the simulations. Simultaneously, the Kokei Uehara method was applied for the obtainment of peak flow values under the same conditions, seeking to compare results. Data collection was performed using geoprocessing tools. For data entry in ABC 6, the fragmentation of sub-basin of the stream Claro was necessary, which generated 7 small watersheds, in order to fulfill a software demand, as the maximum drainage area it accepts is 50km² for each watershed analyzed. For RT = 10 and 100 years, respectively, the results of peak flow with use of ABC 6 were 46.10 and 95.45 m³/s, while for Kokei Uehara method, the results were 47.17 and 65.26 m³/s. The adoption of a single value of discretization time for all watersheds was indicated as limitation of ABC 6, which interfered in the final results. The transport of anthropogenic and natural contaminants to public-supply wells was evaluated in a part of the High Plains aquifer near York, Nebraska, as part of the U.S. Geological Survey National Water-Quality Assessment Program. The aquifer in the Eastern High Plains regional study area is composed of Quaternary alluvial deposits typical of the High Plains aquifer in eastern Nebraska and Kansas, is an important water source for agricultural irrigation and public water supply, and is susceptible and vulnerable to contamination. A six-layer, steady-state ground-water flow model of the High Plains aquifer near York, Nebraska, was constructed and calibrated to average conditions for the time period from 1997 to 2001. The calibrated model and advective particle-tracking simulations were used to compute areas contributing recharge and travel times from recharge areas to selected public-supply wells. Model results indicate recharge from agricultural irrigation return flow and precipitation (about 89 percent of inflow) provides most of the ground-water inflow, whereas the majority of ground-water discharge is to pumping wells (about 78 percent of outflow). Particle-tracking results indicate areas contributing recharge to public-supply wells extend northwest because of the natural ground-water gradient from the northwest to the southeast across the study area. Particle-tracking simulations indicate most ground-water travel times from areas contributing recharge range from 20 to more than 100 years but that some ground water, especially that in the lower confined unit, originates at the upgradient model boundary instead of at the water table in the study area and has travel times of thousands of years.

Changes in bioavailability of phosphorus (P) during pedogenesis and ecosystem development have been shown for geogenic calcium phosphate (Ca-P). However, very little is known about long-term changes of biogenic Ca-P in soil. Long-term transformation characteristics of biogenic Ca-P were examined using anthropogenic soils along a chronosequence from centennial to millennial time scales. Phosphorus fractionation of Anthrosols resulted in overall consistency with the Walker and Syers model of geogenic Ca-P transformation during pedogenesis. The biogenic Ca-P (e.g., animal and fish bones) disappeared to 3% of total P within the first ca. 2,000 years of soil development. This change concurred with increases in P adsorbed on metal-oxides surfaces, organic P, and occluded P at different pedogenic time. Phosphorus K-edge X-ray absorption near-edge structure (XANES) spectroscopy revealed that the crystalline and therefore thermodynamically most stable biogenic Ca-P was transformed into more soluble forms of Ca-P over time. While crystalline hydroxyapatite (34% of total P) dominated Ca-P species after about 600-1,000 years, beta-tricalcium phosphate increased to 16% of total P after 900-1,100 years, after which both Ca-P species disappeared. Iron-associated P was observable concurrently with Ca-P disappearance. Soluble P and organic P determined by XANES maintained relatively constant (58-65%) across the time scale studied. Disappearance of crystalline biogenic Ca-P on a time scale of a few thousand years appears to be ten times faster than that of geogenic Ca-P.

A revista, que teria completado 100 anos em outubro de 2005, caracterizou-se por uma postura didático-pedagógica e pela disseminação de produtos quadrinhísticos genuinamente nacionais, dando oportunidade para a atuação de dezenas de desenhistas brasileiros e propiciando o aparecimento de diversos personagens de histórias em quadrinhos. Na década de 1960, desgastada pelo tempo e sofrendo a concorrência da televisão, a revista encerrou suas atividades, ficando na lembrança de seus leitores por sua abnegação a um modelo de educação para a infância que privilegiava o civismo, os bons costumes e a religiosidade

Scientific collections are important sources of material for many areas of ornithological research. Although firearms (particularly shotguns) have been the standard for avian scientific collecting for more than 100 years, their use is restricted in many areas of the world. We describe a cheap, relatively silent, and effective air shotgun for collecting birds weighing up to 50 g at distances up to 4 m. This air shotgun is capable of shooting birdshot, uses hollow metal rivets connected to plastic straws as shot shells, and represents a simple adaptation of any 0.177 or 0.22 cal single-shot, break-barrel air rifle with at least 25 joules of muzzle energy. This air shotgun will be especially useful for focused sampling of birds (and other small vertebrates) in situations where firearm use or transport is restricted.

Nitroglycerin (GIN) has been clinically used to treat angina pectoris and acute heart episodes for over 100 years. The effects of GTN have long been recognized and active research has contributed to the unraveling of numerous metabolic routes capable of converting GIN to the potent vasoactive messenger nitric oxide. Recently, the mechanism by which minute doses of GIN elicit robust pharmacological responses was revisited and eNOS activation was implicated as an important route mediating vasodilation induced by low GTN doses (1-50 nM). Here, we demonstrate that at such concentrations the pharmacologic effects of nitroglycerin are largely dependent on the phosphatidylinositol 3-kinase, Akt/PKB, and phosphatase and tensin homolog deleted on chromosome 10 (PTEN) signal transduction axis. Furthermore, we demonstrate that nitroglycerin-dependent accumulation of 3,4,5-InsP(3), probably because of inhibition of PTEN, is important for eNOS activation, conferring a mechanistic basis for GIN pharmacological action at pharmacologically relevant doses. During the last century, great improvements have been made in rectal cancer management regarding preoperative staging, pathologic assessment, surgical technique, and multimodal therapies. Surgically, there was a move from a strategy characterized by simple perineal excision to complex procedures performed by means of a laparoscopic approach, and more recently with the aid of robotic systems. Perhaps the most important advance is that rectal cancer is no longer a fatal disease as it was at the beginning of the 20th century. This achievement is definitely due in part to Ernest Mile's contribution regarding lymphatic spread of tumor cells, which helped clarify the natural history of the disease and the proper treatment alternatives. He advocated a combined approach with the rationale to clear "the zone of upward spread." The aim of the present paper is to present a brief review concerning the evolution of rectal cancer surgery, focusing attention on Miles' abdominoperineal excision of the rectum (APR) and its controversies and refinements over time. Although APR has currently been restricted to a small proportion of patients with low rectal cancer, recent propositions to excise the rectum performing a wider perineal and a proper pelvic floor resection have renewed interest on this procedure, confirming that Ernest Miles' original ideas still influence rectal cancer management after more than 100 years.

The pantropical family Eriocaulaceae includes ten genera and c. 1,400 species, with diversity concentrated in the New World. The last complete revision of the family was published more than 100 years ago, and until recently the generic and infrageneric relationships were poorly resolved. However, a multi-disciplinary approach over the last 30 years, using morphological and anatomical characters, has been supplemented with additional data from palynology, chemistry, embryology, population genetics, cytology and, more recently, molecular phylogenetic studies. This led to a reassessment of phylogenetic relationships within the family. In this paper we present new data for the ITS and trnL-F regions, analysed separately and in combination, using maximum parsimony and Bayesian inference. The data confirm previous results, and show that many characters traditionally used for differentiating and circumscribing the genera within the family are homoplasious. A new generic key with characters from various sources and reflecting the current taxonomic changes is presented.

Age-related physiological changes in the gastrointestinal tract, as well as modification in lifestyle, nutritional behaviour, and functionality of the host immune system, inevitably affect the gut microbiota. The study presented here is focused on the application and comparison of two different microarray approaches for the characterization of the human gut microbiota, the HITChip and the HTF-Microb.Array, with particular attention to the effects of the aging process on the composition of this ecosystem. By using the Human Intestinal Tract Chip (HITChip), recently developed at the Wageningen University, The Netherland, we explored the age-related changes of gut microbiota during the whole adult lifespan, from young adults, through elderly to centenarians. We observed that the microbial composition and diversity of the gut ecosystem of young adults and seventy-years old people is highly similar but differs significantly from that of the centenarians. After 100 years of symbiotic association with the human host, the microbiota is characterized by a rearrangement in the Firmicutes population and an enrichment of facultative anaerobes. The presence of such a compromised microbiota in the centenarians is associated with an increased inflammation status, also known as inflamm-aging, as determined by a range of peripheral blood inflammatory markers. In parallel, we overtook the development of our own phylogenetic microarray with a lower number of targets, aiming the description of the human gut microbiota structure at high taxonomic level. The resulting chip was called High Taxonomic level Fingerprinting Microbiota Array (HTF-Microb.Array), and was based on the Ligase Detection Reaction (LDR) technology, which allowed us to develop a fast and sensitive tool for the fingerprint of the human gut microbiota in terms of presence/absence of the principal groups. The validation on artificial DNA mixes, as well as the pilot study involving eight healthy young adults, demonstrated that the HTF-Microb.Array can be used to successfully characterize the human gut microbiota, allowing us to obtain results which are in approximate accordance with the most recent characterizations. Conversely, the evaluation of the relative abundance of the target groups on the bases of the relative fluorescence intensity probes response still has some hindrances, as demonstrated by comparing the HTF.Microb.Array and HITChip high taxonomic level fingerprints of the same centenarians.

Palynologie und Sedimentologie der Interglazialprofile Döttingen, Bonstorf, Munster und Bilshausen Zusammenfassung In der vorliegenden Dissertation wurden vier dem Holstein-Interglazial zugehörige Bohrkerne sowie ein rhumezeitlicher Bohrkern palynologisch und sedimentologisch bearbeitet. Die holsteinzeitlichen Bohrkerne stammen aus Kieselgurlagerstätten der Lüneburger Heide (Bonstorf und Munster) und aus einem Trockenmaar (Döttingen) in der Eifel. Der rhumezeitliche Kern stammt aus der Typlokalität Bilshausen im Harzvorland. Neben Prozentwertdiagrammen werden Pollendichte- und wenn möglich Polleninfluxwerte vorgestellt, die insbesondere für die Lokalitäten Hetendorf/Bonstorf und Munster/Breloh bisher nicht verfügbar waren. Mit dem Profil Döttingen konnte erstmals eine sowohl vollständige als auch nicht innerhalb des klassischen Aufkommens holsteinzeitlicher Fundstellen im norddeutschen Tiefland positionierte Holsteinsequenz aus dem deutschen Mittelgebirge dokumentiert werden. Das erhaltene Pollendiagramm bestätigt die aus den norddeutschen Profilen bekannte holsteintypische Vegetationsabfolge, durch die das Holstein gegenüber anderen Interglazialen wie Holozän, Eem oder Rhume palynologisch definiert ist. Neben der grundsätzlichen Übereinstimmung der Pollensequenz unterscheidet sich das Profil Döttingen aber deutlich im prozentualen Aufkommen der beteiligten Taxa von den norddeutschen Profilen. So wird eine hohe Alnus-Präsenz als Merkmal deutscher Holsteinprofile bestätigt, jedoch ist die, in den norddeutschen Lokalitäten durchhaltend hohe oder dominante Beteiligung von Pinus im deutschen Mittelgebirge nicht vorhanden und muss daher auf die Standortbedingungen Norddeutschlands zurückgeführt werden. Abies dagegen ist im Holstein der Mittelgebirge wesentlich präsenter als im norddeutschen Flachland. Im Profil Döttingen wurden insgesamt 10 Tephralagen gefunden. Auf eine dieser Tephren folgt ein „Birken-Kiefern-Gräser Vorstoß“, der palynostratigraphisch dem älteren „Birken-Kiefern Vorstoß“ in Munster/Breloh entspricht. Als eine Typologie des Holstein kann das in den Profilen Döttingen und Munster bestätigte intraholsteinzeitliche Carpinus-Minimum verstanden werden. An Hand sedimentologischer und palynologischer Befunde aus dem Bohrkern MU 2 muss die Existenz zweier, in der Literatur postulierter, postholsteinzeitlicher, „Nachschwankungen“ in Munster/Breloh in Frage gestellt, wenn nicht abgelehnt werden. In Kern MU 2 fallen palynostratigraphische Grenzen häufig mit Sandeinschaltungen zusammen. Eine dieser Sandeinschaltungen, nämlich unmittelbar vor dem älteren „Birken-Kiefern-Vorstoß“, korreliert in ihrer stratigraphischen Position mit der den „Birken-Kiefern-Gräser-Vorstoß“ im Profil Döttingen einleitenden Tephralage. Es gelang die Dauer des intraholsteinzeitlichen Carpinus Minimums auf etwa 1500±100 Jahre zu bestimmen und eine interne Zweigliederung zu dokumentieren. Im rhumezeitlichen Kern von Bilshausen (BI 1) konnten zahlreiche Störungen nachgewiesen werden. Insbesondere im Teufenbereich des Bilshausener „Birken-Kiefern-Vorstoßes“ deuten diese auf eine möglicherweise verfälschte Überlieferung. Der palynologisch markante „Lindenfall“ von Bilshausen liegt im Bereich einer isoklinalen Schichtenverfaltung. Die in der Literatur im Horizont des „Lindenfalls“ beschriebene „Bilshausentephra“ wurde nicht gefunden. Warvenzählungen an den Kernen MU 1, MU 2 und BI 1 ermöglichten Pollenzonendauern in Holstein- und Rhume-Interglazial zu bestimmen. Dabei wurde mittels den Warvenzählungen, unter zu Hilfenahme von Literaturdaten und von Schätzwerten eine Dauer für das Holstein-Interglazial sensu stricto (Pollenzonen I-XIV) von 15400-17800 Jahren und für das Rhume-Interglazial von wahrscheinlich 22000 Jahren bis maximal 26000 Jahren ermittelt.

Klinische Studien haben gezeigt, dass die allergenspezifische Immuntherapie (SIT) eine effektive Therapieoption für allergische Erkrankungen ist. Obwohl dieses Therapieverfahren seit über 100 Jahren existiert, sind die zugrunde liegenden Suppressionsmechanismen jedoch nicht vollständig verstanden. Bisher wird angenommen, dass der Behandlungserfolg der SIT auf einer Blockade durch allergenspezifische Antikörper, einer Verschiebung des Th1-Th2-Gleichgewichtes und/oder auf einer Suppression durch regulatorische T-Zellen (Tregs) basiert. Um die Effekte der SIT in einer chronischen Erkrankung in vivo untersuchen zu können, wurde in dieser Doktorarbeit ein Mausmodell für chronisches Asthma entwickelt, das die Situation im Menschen nach einer SIT nachahmt. rnDurch eine SIT war es möglich, allergeninduzierte Asthmasymptome wie Atemwegshyperreagibilität (AHR), Eosinophilie in der Lunge, IgE-Produktion und Atemwegsentzündung im Modell zu unterdrücken. Bemerkenswert ist, dass durch OVA-spezifische Immuntherapie (OVA-IT) ebenfalls eine Verringerung der strukturellen Veränderungen im Lungengewebe im chronischen Krankheitsverlauf erreicht wurde.rnDes Weiteren wurde in diesem Modell nach den Prozessen gesucht, die für die toleranzinduzierende Wirkung der SIT verantwortlich sein können. Dabei wurde im Vergleich zur Placebo-behandelten Gruppe eine erhöhte Antwort spezifischer IgG1-Antikörper, eine verstärkte Th1-Antwort, sowie eine erhöhte Frequenz von FoxP3+ Tregs und von IL-10-produzierenden T-Zellen (Tr1-Zellen) nach OVA-IT festge-stellt. Zur weiteren Untersuchung der von SIT-induzierten T-Zellantworten wurden Mausmodelle des allergischen Asthmas mit einem akuten Verlauf gewählt.rnDie Bedeutung der Th1-Zellen für die SIT wurde in T-bet-/- Mäusen untersucht, welche aufgrund des Fehlens des Transkriptionsfaktors T-bet keine stabile Th1-Antwort induzieren können. Durch SIT war es möglich, allergeninduzierte Asthmasymptome wie AHR, eosinophile Granulozyten in der Lunge, IgE-Produktion und Atemwegsentzündung in den T-bet-/- Tieren im gleichen Maße wie in den Wildtyptieren zu unterdrücken. Diese Untersuchung zeigte, dass die SIT auch ohne funktionelle Th1-Zellen die allergische Entzündung unterdrücken kann. rnDie Rolle der Tregs für die SIT wurde in DO11.10 Mäusen und DO11.10 RAG-/- Mäusen untersucht. In beiden Stämmen konnte nach SIT eine Induktion OVA-spezifischer Tregs nachgewiesen werden. In DO11.10 RAG-/- Mäusen können durch den Knockout im rag2-Gen keine natürlichen, d.h. im Thymus gereiften, Tregs entstehen. Im Blut von DO11.10 RAG-/- Mäusen war direkt nach Durchführung der OVA-IT eine FoxP3+ Treg-Population detektierbar. Demnach wird durch die OVA-IT eine de-novo-Induktion von FoxP3+ Tregs in Gang gesetzt. In Abwesenheit der natürlichen Tregs zeigte sich weiterhin, dass diese Zellen zur Produktion von IL-10 in T-Zellen und somit zum Erfolg der SIT beitragen.rnDie Rolle der FoxP3+ Tregs bei der SIT wurde in DEREG Mäusen untersucht. Eine Depletion der FoxP3+ Tregs in DEREG Mäusen während der Durchführung der OVA-IT hob die protektiven Effekte der Therapie jedoch nur teilweise auf. rnUm die Rolle des regulatorischen Zytokins IL-10 bei der SIT zu untersuchen, wurde ein blockierender Antikörper gegen den IL-10-Rezeptor (anti-IL-10R) im chronischen Modell des allergischen Asthmas mit SIT angewendet. Anti-IL-10R hob die protektive Wirkung der SIT auf die AHR, die Atemwegsentzündung und die strukturellen Veränderungen im Lungengewebe auf. Somit ist die protektive Wirkung der SIT abhängig vom IL-10-Signalweg.rnZusammenfassend stellt diese Arbeit die Bedeutung der SIT für allergische Erkrankungen heraus. SIT kann durch die positive Beeinflussung der allergiebedingten, strukturellen Veränderungen in der Lunge auch für Asthmapatienten große Vorteile bringen. Die aus Studien bekannten Mechanismen konnten im Modell bestätigt werden und wurden im weiteren Verlauf untersucht. Die Arbeit stellt im Besonderen die Bedeutung der IL-10-produzierenden und FoxP3+ Tregs für die Effektivität der SIT in den Vordergrund. Zudem ist durch die Etablierung eines neuen Mausmodells der SIT für chronisches allergisches Asthma ein Mittel zur weiteren Erforschung der zugrunde liegenden Prozesse dieser erfolgreichen Therapie geschaffen worden. rn
