New pollen and radiocarbon data from an 8.6-m coastal section, Cape Shpindler (69°43' N; 62°48' E), Yugorski Peninsula, document the latest Pleistocene and Holocene environmental history of this low Arctic region. Twelve AMS 14C dates indicate that the deposits accumulated since about 13,000 until 2000 radiocarbon years BP. A thermokarst lake formed ca. 13,000-12,800 years BP, when scarce arctic tundra vegetation dominated the area. By 12,500 years BP, a shallow lake existed at the site, and Arctic tundra with Poaceae, Cyperaceae, Salix, Saxifraga, and Artemisia dominated nearby vegetation. Climate was colder than today. Betula nana became dominant during the Early Preboreal period about 9500 years BP, responding to a warm event, which was one of the warmest during the Holocene. Decline in B. nana and Salix after 9500 years BP reflects a brief event of Preboreal cooling. A subsequent increase in Betula and Alnus fruticosa pollen percentages reflects amelioration of environmental conditions at the end of Preboreal period (ca. 9300 years BP). A decline in arboreal taxa later, with a dramatic increase in herb taxa, reflects a short cold event at about 9200 years BP. The pollen data reflect a northward movement of tree birch, peaking at the middle Boreal period, around 8500 years BP. Open Betula forest existed on the Kara Sea coast of the Yugorski Peninsula during the Atlantic period (8000-4500 years BP), indicating that climate was significantly warmer than today. Deteriorating climate around the Atlantic-Subboreal boundary (ca. 4500 years BP) is recorded by a decline in Betula percentages. Sedimentation slowed at the site, and processes of denudation and/or soil formation started at the beginning of the Subatlantic period, when vegetation cover on Yugorski Peninsula shifted to near-modern assemblages.

The chemical composition of surface associated metabolites of two Fucus species (Fucus vesiculosus and Fucus serratus) was analysed by means of gas chromatography-mass spectrometry (GC-MS) to describe temporal patterns in chemical surface composition. Method: The two perennial brown macroalgae F. vesiculosus and F. serratus were sampled monthly at Bülk, outer Kiel Fjord, Germany (54°27'21 N / 10°11'57 E) over an entire year (August 2012 - July 2013). Per month and species six non-fertile Fucus individuals were collected from mixed stands at a depth of 0.5 m under mid water level. For surface extraction approx. 50 g of the upper 5-10 cm apical thalli tips were cut off per species. The surface extraction of Fucus was performed according to the protocol of de Nys and co-workers (1998) with minor modifications (see Rickert et al. 2015). GC/EI-MS measurements were performed with a Waters GCT premier (Waters, Manchester, UK) coupled to an Agilent 6890N GC equipped with a DB-5 ms 30 m column (0.25 mm internal diameter, 0.25 mM film thickness, Agilent, USA). The inlet temperature was maintained at 250°C and samples were injected in split 10 mode. He carrier gas flow was adjusted to 1 ml min-1. Alkanes were used for referencing of retention times. Pollen analysis of Wisconsinan sediments from eleven localities in northern and central Illinois, combined with the results of older studies, allows a first general survey of the vegetational changes in Illinois during the last glaciation. In the late Altonian (after 40,000 B.P.), pine was already the most prevalent tree type in northern Illinois. Probably because of the influence of the last Altonian ice advance to northern Illinois, pine migrated to the south and reached south-central Illinois, which was at that time a region of prairie, with oak and hickory trees in favorable sites. Likewise in the late Altonian, spruce appeared in northern Illinois. Spruce also expanded its area to the south during the Wisconsinan, reaching south-central Illinois only after 21,000 B.P., in the early Woodfordian. Deciduous trees (predominantly oak) were present in south-central Illinois throughout the Wisconsinan. Their prevalence decreased to the north. The vegetation during the different subdivisions of the last glacial period in Illinois was approximately as follows: Late Altonian: Pine/spruce forest with some deciduous trees in northern and central Illinois; prairie and oak/hickory stands in south-central Illinois; immigration of pine. Farmdalian: Pine/spruce forest in central Illinois; deciduous trees and pine in south-central Illinois, with areas of open vegetation, perhaps similar to the present-day transition of prairie to forest in the northern Great Plains. Woodfordian: Northern and central Illinois ice covered; in south central Illinois, spruce and oak as dominant tree types, but also pine and grassland. During the Woodfordian, pine and spruce disappeared again from south-central Illinois, and oak/hickory forest and prairie again prevailed. The ice-free areas of northern Illinois become populated temporarily with spruce, but later there is proof of deciduous forest in this region. Pollen investigations in south-central Illinois have shown convincingly that deciduous trees could survive relatively close (less than 60 km) to the ice margin. Therefore the frequently presented view that arctic climatic conditions prevailed in North America during the last glaciation far south of the ice margin can be refuted for the Illinois area, confirming the opinion of other authors resulting from investigations of fossil mollusks and frost-soil features. The small number of localities investigated still permits no complete reconstruction of the vegetation zones and their possible movements in Illinois. During the Altonian and Farmdalian in Illinois, a vegetational zonation probably existed similar to that of today in North America. As the ice pushed southward as far as 39° 20' N. lat in the early Woodfordian, this zonation was apparently broken up under the influence of a relatively moderate climate. In any case, the Vandalia area, which was only about 60 km south of the ice, was at that time neither in a tundra zone nor in a zone of boreal coniferous forest.

A new interglacial pollen sequence from the Döttinger dry maar in the Eifel region of the Rheinish Schield is presented. Palynology is used to correlated to several classical north German Holsteinian sites. The lake sediments reveal the complete interglacial and also 60 m of laminated sediments from the glacial preceding the Holsteinian. The interglacial section indicates limnic conditions in its lower part and telmatic conditions in its upper part with an intermediate episode of peat formation. Ash layers document internsive volcansim during the interglacial in the Eifel region. Some of the north German Holsteinian sites reval spikes of high abundance of Pinus, Beutal and Poaceae and/or setbacks of more demanding taxa during the interglacial, often interpreted as cold events. The Döttingen profile shows similar pattern, but with little response from the thermophilous pollen taxa. In the Döttingen sequence these vegetation 'anomalies' are preceded, or accompanied by phases of active volcanism. The role/interaction of climate and/or volcanism as a likely cause for these vegetation 'anomalies' ist still to be quantified.

Sporomorphs and dinoflagellate cysts from site GIK16867 in the northern Angola Basin record the vegetation history of the West African forest during the last 700 ka in relation to changes in salinity and productivity of the eastern Gulf of Guinea. During most cool and cold periods, the Afromontane forest, rather than the open grass-rich dry forest, expanded to lower altitudes partly replacing the lowland rain forest of the borderlands east of the Gulf of Guinea. Except in Stage 3, when oceanic productivity was high during a period of decreased atmospheric circulation, high oceanic productivity is correlated to strong winds. The response of marine productivity in the course of a climatic cycle, however, is earlier than that of wind vigour and makes wind-stress-induced oceanic upwelling in the area less likely. Monsoon variation is well illustrated by the pollen record of increased lowland rain forest that is paired to the dinoflagellate cyst record of decreased salinity forced by increased precipitation and run-off.

Fluxes of airborne freshwater diatoms (FD), phytoliths (PH), and pollen grains (PO) collected with sediment traps off Cape Blanc, northwest Africa, from 1988 till 1991 are presented. Both continental rainfall variations and wind mean strength and direction play a key role in the temporal fluctuations of the fluxes of eolian traces in the pelagic realm. Drier conditions in Northern Africa in 1987 could have preceded the high lithogenic input and moderate FD flux in 1988. The PH peak in summer 1988 was probably caused by increased wind velocity. Wetter rainy seasons of 1988/89 might have promoted a significant pollen production in summer 1989, and FD in late 1989 and early 1990, as well as contributed to the reduction of the lithogenic flux in 1989/90. Decreased fluxes of FD, PH and PO, and higher contribution of the 6-11 µm lithogenic fraction in 1991 would mainly reflect minor intensity and decreased amount of continental trade winds. Air-mass backward trajectories confirm that the Saharan Air Layer is predominantly involved in the spring/summer transport. Trade winds play a decisive role in the fall/winter months, but also contribute to the transport during late spring/summer. Origin of wind trajectories does not support a direct relationship between transporting wind-layers and material source areas in Northern Africa. High winter fluxes of eolian tracers and high amount of trade winds with continental origin in summer warn against a simplistic interpretation of the seasonal eolian signal preserved in the sediments off Cape Blanc, and the wind layer involved in its transport.

Huntington’s disease (HD) is an autosomal neurodegenerative disorder affecting approximately 5-10 persons per 100,000 worldwide. The pathophysiology of HD is not fully understood but the age of onset is known to be highly dependent on the number of CAG triplet repeats in the huntingtin gene. Using 1H NMR spectroscopy this study biochemically profiled 39 brain metabolites in post-mortem striatum (n=14) and frontal lobe (n=14) from HD sufferers and controls (n=28). Striatum metabolites were more perturbed with 15 significantly affected in HD cases, compared with only 4 in frontal lobe (P<0.05; q<0.3). The metabolite which changed most overall was urea which decreased 3.25-fold in striatum (P<0.01). Four metabolites were consistently affected in both brain regions. These included the neurotransmitter precursors tyrosine and L-phenylalanine which were significantly depleted by 1.55-1.58-fold and 1.48-1.54-fold in striatum and frontal lobe, respectively (P=0.02-0.03). They also included L-leucine which was reduced 1.54-1.69-fold (P=0.04-0.09) and myo-inositol which was increased 1.26-1.37-fold (P<0.01). Logistic regression analyses performed with MetaboAnalyst demonstrated that data obtained from striatum produced models which were profoundly more sensitive and specific than those produced from frontal lobe. The brain metabolite changes uncovered in this first 1H NMR investigation of human HD offer new insights into the disease pathophysiology. Further investigations of striatal metabolite disturbances are clearly warranted.

The Dot/Icm type IV secretion system (T4SS) of Legionella pneumophila is crucial for the pathogen to survive in protozoa and cause human disease. Although more than 275 effector proteins are delivered into the host cell by the T4SS, the function of the majority is unknown. Here we have characterized the Dot/Icm effector LtpD. During infection, LtpD localized to the cytoplasmic face of the membrane of the Legionella-containing vacuole (LCV). In A549 lung epithelial cells, ectopically expressed LtpD localized to large vesicular structures that contained markers of endosomal compartments. Systematic analysis of LtpD fragments identified an internal 17-kDa fragment, LtpD471-626, which was essential for targeting ectopically expressed LtpD to vesicular structures and for the association of translocated LtpD with the LCV. LtpD471-626 bound directly to phosphatidylinositol 3-phosphate [PtdIns(3)P] in vitro and colocalized with the PtdIns(3)P markers FYVE and SetA in cotransfected cells. LtpD was also found to bind the host cell enzyme inositol (myo)-1 (or 4)-monophosphatase 1, an important phosphatase involved in phosphoinositide production. Analysis of the role of LtpD in infection showed that LtpD is involved in bacterial replication in THP-1 macrophages, the larvae of Galleria mellonella, and mouse lungs. Together, these data suggest that LtpD is a novel phosphoinositide- binding L. pneumophila effector that has a role in intracellular bacterial replication. © 2013, American Society for Microbiology.

L’utilisation de méthodes d’investigation cérébrale avancées a permis de mettre en évidence la présence d’altérations à court et à long terme à la suite d’une commotion cérébrale. Plus spécifiquement, des altérations affectant l’intégrité de la matière blanche et le métabolisme cellulaire ont récemment été révélées par l’utilisation de l’imagerie du tenseur de diffusion (DTI) et la spectroscopie par résonance magnétique (SRM), respectivement. Ces atteintes cérébrales ont été observées chez des athlètes masculins quelques jours après la blessure à la tête et demeuraient détectables lorsque les athlètes étaient à nouveau évalués six mois post-commotion. En revanche, aucune étude n’a évalué les effets neurométaboliques et microstructuraux dans la phase aigüe et chronique d’une commotion cérébrale chez les athlètes féminines, malgré le fait qu’elles présentent une susceptibilité accrue de subir ce type de blessure, ainsi qu’un nombre plus élevé de symptômes post-commotionnels et un temps de réhabilitation plus long. Ainsi, les études composant le présent ouvrage visent globalement à établir le profil d’atteintes microstructurales et neurométaboliques chez des athlètes féminines par l’utilisation du DTI et de la SRM. La première étude visait à évaluer les changements neurométaboliques au sein du corps calleux chez des joueurs et joueuses de hockey au cours d’une saison universitaire. Les athlètes ayant subi une commotion cérébrale pendant la saison ont été évalués 72 heures, 2 semaines et 2 mois après la blessure à la tête en plus des évaluations pré et post-saison. Les résultats démontrent une absence de différences entre les athlètes ayant subi une commotion cérébrale et les athlètes qui n’en ont pas subie. De plus, aucune différence entre les données pré et post-saison a été observée chez les athlètes masculins alors qu’une diminution du taux de N-acetyl aspartate (NAA) n’a été mise en évidence chez les athlètes féminines, suggérant ainsi un impact des coups d’intensité sous-clinique à la tête. La deuxième étude, qui utilisait le DTI et la SRM, a révélé des atteintes chez des athlètes féminines commotionnées asymptomatiques en moyenne 18 mois post-commotion. Plus spécifiquement, la SRM a révélé une diminution du taux de myo-inositol (mI) au sein de l’hippocampe et du cortex moteur primaire (M1) alors que le DTI a mis en évidence une augmentation de la diffusivité moyenne (DM) dans plusieurs faisceaux de matière blanche. De iii plus, une approche par région d’intérêt a mis en évidence une diminution de la fraction d’anisotropie (FA) dans la partie du corps calleux projetant vers l’aire motrice primaire. Le troisième article évaluait des athlètes ayant subi une commotion cérébrale dans les jours suivant la blessure à la tête (7-10 jours) ainsi que six mois post-commotion avec la SRM. Dans la phase aigüe, des altérations neuropsychologiques combinées à un nombre significativement plus élevé de symptômes post-commotionnels et dépressifs ont été trouvés chez les athlètes féminines commotionnées, qui se résorbaient en phase chronique. En revanche, aucune différence sur le plan neurométabolique n’a été mise en évidence entre les deux groupes dans la phase aigüe. Dans la phase chronique, les athlètes commotionnées démontraient des altérations neurométaboliques au sein du cortex préfrontal dorsolatéral (CPDL) et M1, marquées par une augmentation du taux de glutamate/glutamine (Glx). De plus, une diminution du taux de NAA entre les deux temps de mesure était présente chez les athlètes contrôles. Finalement, le quatrième article documentait les atteintes microstructurales au sein de la voie corticospinale et du corps calleux six mois suivant une commotion cérébrale. Les analyses n’ont démontré aucune différence au sein de la voie corticospinale alors que des différences ont été relevées par segmentation du corps calleux selon les projections des fibres calleuses. En effet, les athlètes commotionnées présentaient une diminution de la DM et de la diffusivité radiale (DR) au sein de la région projetant vers le cortex préfrontal, un volume moindre des fibres de matière blanche dans la région projetant vers l’aire prémotrice et l’aire motrice supplémentaire, ainsi qu’une diminution de la diffusivité axiale (DA) dans la région projetant vers l’aire pariétale et temporale. En somme, les études incluses dans le présent ouvrage ont permis d’approfondir les connaissances sur les effets métaboliques et microstructuraux des commotions cérébrales et démontrent des effets délétères persistants chez des athlètes féminines. Ces données vont de pair avec la littérature scientifique qui suggère que les commotions cérébrales n’entraînent pas seulement des symptômes temporaires.

Ankle sprains are the most common injuries in sports, usually causing damage to the lateral ligaments. Recurrence has as usual result permanent instability, and thus loss of proprioception. This fact, together with residual symptoms, is what is known as chronic ankle instability, CAI, or FAI, if it is functional. This problem tries to be solved by improving musculoskeletal stability and proprioception by the application of bandages and performing exercises. The aim of this study has been to review articles (meta-analisis, systematic reviews and revisions) published in 2009-2015 in PubMed, Medline, ENFISPO and BUCea, using keywords such as “sprain instability”, “sprain proprioception”, “chronic ankle instability”. Evidence affirms that there does exist decreased proprioception in patients who suffer from CAI. Rehabilitation exercise regimen is indicated as a treatment because it generates a subjective improvement reported by the patient, and the application of bandages works like a sprain prevention method limiting the range of motion, reducing joint instability and increasing confidence during exercise. As podiatrists we should recommend proprioception exercises to all athletes in a preventive way, and those with CAI or FAI, as a rehabilitation programme, together with the application of bandages. However, further studies should be generated focusing on ways of improving proprioception, and on the exercise patterns that provide the maximum benefit.

Työn tavoite on selvittää erilaisia käyttöpääoman ennustusmalleja, jotka pohjautuvat kysyntäennusteeseen. Tavoitteena on esittää malleja niin pitkän kuin lyhyenkin aikavälin ennustamiseen. Pitkällä aikavälillä mallien on tarkoitus ennustaa yrityksissä sitoutuvan käyttöpääoman kokonaistarvetta, kun taas lyhyen aikavälin malleilla pureudutaan käyttöpääomaerien (lopputuotevarasto ja myyntisaamiset) ennustamiseen tuotekohtaisesti. Työssä pyritään myös vastaamaan, miksi käyttöpääomaa kannattaa ennustaa ja miten sen hallintaa voidaan tehostaa. Työ on toteutettu kirjallisuuskatsauksena, mitä on elävöitetty laskuesimerkein. Laskuesimerkkien tarkoitus on antaa lukijalle kuva käyttöpääoman ennustamisen todellisista mahdollisuuksista, jotta työ ei jäisi pelkäksi sanahelinäksi. Työssä esitetyt ennustusmallit on valittu matemaattisen yksinkertaisuutensa ehdolla, jotta niiden käyttäminen yritysmaailmassa olisi mahdollista. Käyttöpääomatarpeen ennustamisen suurimpana haasteena voidaan pitää kysyntäennusteen luotettavuutta. Vuositasolla käyttöpääomatarpeen keskimääräinen osuus suhteessa liikevaihtoon on helpohko ennustaa liiketoiminnan pysyessä melko vakiona. Tarve voi kuitenkin vaihdella hyvin paljon kysynnän vaihdellessa kuukausi-, viikko- ja päivätasolla. Jos kysyntäennustetta saadaan tarkennettua, lopputuotevaraston kokoa ja siihen sitoutuvaa käyttöpääomaa on mahdollista pienentää, koska varmuusvaraston määrittelyssä käytettävä hajonta pienenee. Myyntisaamisten osalta lähitulevaisuudessa sitoutuvaan käyttöpääomaan vaikuttaa eniten asiakaskohtainen maksukäyttäytyminen, koska myyntisaamisiin vaikuttava kysyntä on suurimmalta osaltaan jo toteutunut.

Tutkimuksen tarkoituksena ja tavoitteena on ymmärtää osuuskunnan ja erityisesti osuuskaupan edustajiston tehtäviä ja roolia. Tutkimuksessa halutaan tunnistaa osuuskunnan edustajiston tehtävät ja rooli siten, että ne parhaalla mahdollisella tavalla lisäisivät edustajiston jäsenten vaikuttavuutta ja vaikuttavuuden tunnetta. Tarkoituksena on kehittää toimintaa sellaiseksi, että edustajiston jäsenet kokevat osuuskunnan omakseen ja roolinsa siellä tärkeäksi, ja tätä kautta sitoutuvat tehtäviinsä ja ottavat vastuuta. Tutkimuksessa selvitetään, kuinka edustajisto voi tulevaisuudessa toteuttaa edustuksellista rooliaan parhaalla mahdollisella tavalla huomioiden jäsenten osuuskuntatoimintaa koskevat arvot, tavoitteet sekä odotukset. Tutkimus suoritettiin laadullisena haastattelututkimuksena, johon kuului kuusi puolistrukturoitua haastattelua huhtikuussa 2016. Esimerkkiorganisaationa tässä tutkimuksessa käytettiin Helsingin Osuuskauppa Elannon edustajistoa. Tutkimuksen tulokset osoittavat, että edustajiston jäsenet tuntevat roolinsa ja tehtävänsä vaikuttavaksi, mutta ongelma näyttäisi olevan, että tätä mielikuvaa ei ole pystytty siirtämään eteenpäin jäsenistölle. Jotta mielikuva ja tieto välittyisi ulospäin pitäisi tutkimusta tehdä vielä lisää sekä lisätä mainontaa, koulutusta ja valistusta edustajiston tehtävien ja roolin tiimoilta. Edustajat kokevat tulosten mukaan roolinsa tärkeäksi, mutta parannettavaa löytyi osuuskunnan omaksi tuntemisessa sekä sitä kautta sitoutumisessa. Omistuksen tunteen kehittyminen auttaisi sitoutumisessa työhön. Kollektiivisena tällainen omistamisen tunne palvelisi myös osuuskunnan etuja edustajien paremman työpanoksen ja yhteisten päämäärien kautta. Tällainen vahvemman siteen syntyminen edesauttaisi myös mielikuvien ja ennakkoluulojen muuttamista, joita osuuskunnan jäsenillä näyttäisi olevan edustajistosta ei kollektiivisena yhteisönä.

The screening for genes in metagenomic libraries from soil creates opportunities to explore the enormous genetic and metabolic diversity of microorganisms. Rivers are ecosystems with high biological diversity, but few were examined using the metagenomic approach. With this objective, a metagenomic library was constructed from DNA soil samples collected at three different points along the Jundiaí-river (Rio Grande do Norte-Brazil). The points sampled are from open area, rough terrain and with the direct incidence of sunlight. This library was analyzed functionally and based in sequence. For functional analysis Luria-Bertani solid medium (LB) with NaCl concentration varied from 0.17M to 0.85M was used for functional analysis. Positives clones resistant to hypersaline medium were obtained. The recombinant DNAs were extracted and transformed into Escherichia coli strain DH10B and survival curves were obtained for quantification of abiotic stress resistance. The sequences of clones were obtained and submitted to the BLASTX tool. Some clones were found to hypothetical proteins of microorganisms from both Archaea and Bacteria division. One of the clones showed a complete ORF with high similarity to glucose-6-phosphate isomerase which participates in the synthesis of glycerol pathway and serves as a compatible solute to balance the osmotic pressure inside and outside of cells. Subsequently, in order to identify genes encoding osmolytes or enzymes related halotolerance, environmental DNA samples from the river soil, from the water column of the estuary and ocean were collected and pyrosequenced. Sequences of osmolytes and enzymes of different microorganisms were obtained from the UniProt and used as RefSeqs for homology identification (TBLASTN) in metagenomic databases. The sequences were submitted to HMMER for the functional domains identification. Some enzymes were identified: alpha-trehalose-phosphate synthase, L-ectoina synthase (EctC), transaminase L-2 ,4-diaminobutyric acid (EctB), L-2 ,4-diaminobutyric acetyltransferase (EctA), L-threonine 3 dehydrogenase (sorbitol pathway), glycerol-3-phosphate dehydrogenase, inositol 3-phosphate dehydrogenase, chaperones, L-proline, glycine betaine binding ABC transporter, myo-inositol-1-phosphate synthase protein of proline simportadora / PutP sodium-and trehalose-6-phosphate phosphatase These proteins are commonly related to saline environments, however the identification of them in river environment is justified by the high salt concentration in the soil during prolonged dry seasons this river. Regarding the richness of the microbiota the river substrate has an abundance of halobacteria similar to the sea and more than the estuary. These data confirm the existence of a specialized response against salt stress by microorganisms in the environment of the Jundiaí river
