Biomechanical gait parameters—ground reaction forces (GRFs) and plantar pressures—during load carriage of young adults were compared at a low gait cadence and a high gait cadence. Differences between load carriage and normal walking during both gait cadences were also assessed. A force plate and an in-shoe plantar pressure system were used to assess 60 adults while they were walking either normally (unloaded condition) or wearing a backpack (loaded condition) at low (70 steps per minute) and high gait cadences (120 steps per minute). GRF and plantar pressure peaks were scaled to body weight (or body weight plus backpack weight). With medium to high effect sizes we found greater anterior-posterior and vertical GRFs and greater plantar pressure peaks in the rearfoot, forefoot and hallux when the participants walked carrying a backpack at high gait cadences compared to walking at low gait cadences. Differences between loaded and unloaded conditions in both gait cadences were also observed.

A generic optical biosensing strategy was developed that relies on the absorbance enhancement phenomenon occurring in a multiple scattering matrix. Experimentally, inserts made of glass fiber membrane were placed into microplate wells in order to significantly lengthen the trajectory of the incident light through the sample and therefore increase the corresponding absorbance. Enhancement factor was calculated by comparing the absorbance values measured for a given amount of dye with and without the absorbance-enhancing inserts in the wells. Moreover, the dilution of dye in solutions with different refractive indices (RI) clearly revealed that the enhancement factor increased with the ΔRI between the membrane and the surrounding medium, reaching a maximum value (EF>25) when the membranes were dried. On this basis, two H2O2-biosensing systems were developed based on the biofunctionalization of the glass fiber inserts either with cytochrome c or horseradish peroxidase (HRP) and the analytical performances were systematically compared with the corresponding bioassay in solution. The efficiency of the absorbance-enhancement approach was particularly clear in the case of the cytochrome c-based biosensor with a sensitivity gain of 40 folds and wider dynamic range. Therefore, the developed strategy represents a promising way to convert standard colorimetric bioassays into optical biosensors with improved sensitivity.

The regenerating urodele limb is a useful model system in which to study, in vivo, the controls of cell proliferation and differentiation. Techniques are available which enable one to experimentally manipulate mitogenic influences upon the blastema, as well the morphogenesis of the regenerating 11mb. Although classical regeneration studies have generated a wealth of knowledge concerning tissue interactions, little 1s known about the process at the level of gene expression. The aim of this project was to clone potentially developmentally regulated genes from a newt genomic library for use in future studies of gene expression during limb regeneration. We decided to clone the cytoskeletal actin gene for the following reasons: 1. its expression reflects the proliferative and differentiatlve states of cells in other systems 2. the high copy number of cytoplasmic actin pseudogenes in other vertebrates and the high degree of evolutionary sequence conservation among actin genes increased the chance of cloning one of the newt cytoplasmic actin genes. 3. Preliminary experiments indicated that a newt actin could probably be identified using an available chick ~-actln gene for a molecular probe. Two independent recombinant phage clones, containing actin homologous inserts, were isolated from a newt genomic library by hybridization with the chick actin probe. Restriction mapping identified actin homologous sequences within the newt DNA inserts which were subcloned into the plasmid pTZ19R. The recombinant plasmids were transformed into the Escherichia coli strain, DHsa. Detailed restriction maps were produced of the 5.7Kb and 3.1Kb newt DNA inserts in the plasmids, designated pTNAl and pTNA2. The short «1.3 Kb) length of the actin homologous sequence in pTNA2 indicated that it was possibly a reverse transcript pseudogene. Problems associated with molecular cloning of DNA sequences from N. viridescens are discussed with respect to the large genome size and abundant highly repetitive DNA sequences.

Recombinant human adenovirus (Ad) vectors are being extensively explored for their use in gene therapy and recombinant vaccines. Ad vectors are attractive for many reasons, including the fact that (1) they are relatively safe, based on their use as live oral vaccines, (2) they can accept large transgene inserts, (3) they can infect dividing and postmitotic cells, and (4) they can be produced to high titers. However, there are also a number of major problems associated with Ad vectors, including transient foreign gene expression due to host cellular immune responses, problems with humoral immunity, and the creation of replication competent adenoviruses (RCA). Most Ad vectors contain deletions in the E1 region that allow for insertion of a transgene. However, the E1 gene products are required for replication and thus must be supplied in trans by a helper ceillille that will allow for the growth and packaging of the defective virus. For this purpose the 293 cell line (Graham et al., 1977) is used most often; however, homologous recombination between the vector and the cell line often results in the generation of RCA. The presence of RCA in batches of adenoviral vectors for clinical use is a safety risk because tlley . may result in the mobilization and spread of the replication-defective vector viruses, and in significant tissue damage and pathogenicity. The present research focused on the alteration of the 293 cell line such that RCA formation can be eliminated. The strategy to modify the 293 cells involved the removal of the first 380 bp of the adenovirus genome through the process of homologous recombination. The first step towards this goal involved identifying and cloning the left-end cellular-viral jUl1ction from 293 cells to assemble sequences required for homologous recombination. Polymerase chain reaction (PCR) was performed to clone the junction, and the clone was verified through sequencing. The plasn1id PAM2 was then constructed, which served as the targeting cassette used to modify the 293 cells. The cassette consisted of (1) the cellular-viral junction as the left-end region of homology, (2) the neo gene to use for positive selection upon tranfection into 293 cells, (3) the adenoviral genome from bp 380 to bp 3438 as the right-end region of homology, and (4) the HSV-tk gene to use for negative selection. The plasmid PAM2 was linearized to produce a double strand break outside the region of homology, and transfected into 293 cells using the calcium-phosphate technique. Cells were first selected for their resistance to the drug G418, and subsequently for their resistance to the drug Gancyclovir (GANC). From 17 transfections, 100 pools of G418f and GANCf cells were picked using cloning lings and expanded for screening. Genomic DNA was isolated from the pools and screened for the presence of the 380 bps using PCR. Ten of the most promising pools were diluted to single cells and expanded in order to isolate homogeneous cell lines. From these, an additional 100 G41Sf and GANef foci were screened. These preliminary screening results appear promising for the detection of the desired cell line. Future work would include further cloning and purification of the promising cell lines that have potentially undergone homologous recombination, in order to isolate a homogeneous cell line of interest.

The ease of production and manipulation has made plasmid DNA a prime target for its use in gene transfer technologies such as gene therapy and DNA vaccines. The major drawback of plasmid however is its stability within mammalian cells. Plasmid DNA is usually lost by cellular mechanisms or as a result of mitosis by simple dilution. This study set out to search for mammalian genomic DNA sequences that would enhance the stability of plasmid DNA in mammalian cells.Creating a plasmid based genomic DNA library, we were able to screen the human genome by transfecting the library into Human Embryonic Kidney (HEK 293) Cells. Cells that contained plasmid DNA were selected, using G418 for 14 days. The resulting population was then screened for the presence of biologically active plasmid DNA using the process of transformation as a detector.A commercially available plasmid DNA isolation kit was modified to extract plasmid DNA from mammalian cells. The standardized protocol had a detection limit of -0.6 plasmids per cell in one million cells. This allowed for the detection of 45 plasmids that were maintained for 32 days in the HEK 293 cells. Sequencing of selected inserts revealed a significantly higher thymine content in comparison to the human genome. Sequences with high A/T content have been associated with Scaffold/Matrix Attachment Region (S/MAR) sequences in mammalian cells. Therefore, association with the nuclear matrix might be required for the stability of plasmids in mammalian cells.

The description of the image reads "No. 115 - The Rapids, Niagara - From the Terrapin Tower". On the reverse it reads "The Rapids, Niagara, From the Terrapin Tower (taken instantaneously). From this commanding point of view the rapids are seen to great perfection - the Tower standing just on the edge of the Horse-shoe Fall. Some idea of the rapidity with which these waters hurry onward to the mighty abyss may be formed from the fact that the river descends nearly fifty-one feet in a distance of three-quarters of a mile. Gathering force as they approach nearer the edge of the Falls, they dash and foam amid the rocks which speck their surface, and with torturous writhes dash onward with a fury grand and beautiful in the extreme. Whole flocks of water-fowl have often been seen going to destruction among these rapids. Pleased with being carried by the stream, they have indulged in the pleasure till the rapidity of the current has rendered it impossible for them to rise and thus have they been carried down and washed over the Fall. At one time during the months of September and October, sufficient quantities of dead water-fowl have been found every morning below the Fall to afford ample subsistence for the garrison at the fort. Cette thèse étudie la représentation de la machine chez Robida. La partie centrale de notre recherche s’intéresse à révéler ses significations et interroge sa mise en scène littéraire et visuelle dans chacun des romans de la trilogie d’anticipation scientifique la plus connue de l’auteur-illustrateur. La quête se transforme en un voyage continu entre le lisible et le visible, le dit et le non-dit, la description littéraire et l’imagination, la réalité et la fiction. Nous nous intéressons à l’évolution de la vision de Robida : dans Le Vingtième siècle, l’image de la machine bienfaisante, facilitant la vie de l’homme, économisant du temps et de l’argent, et contribuant largement à son bonheur et à son divertissement, à part quelques accidents très limités, se traduit par une complémentarité avantageuse entre le texte d’une part et les vignettes, les tableaux et les hors-textes se trouvant dans le récit, d’autre part. Celle-ci se transforme, dans La Guerre au vingtième siècle, en une inquiétude vis-à-vis de l’instrumentalisation de la machine pour la guerre, qui s’exprime par une projection de la narration vers l’illustration in-texte, et sensibilise le lecteur en montrant le caractère violent et offensif d’appareils uniquement nommés. Celle-ci devient finalement, dans La Vie électrique, synonyme d’un pessimisme total quant à l’implication de la machine dans la société et à la puissance du savoir scientifique dans l’avenir, qui s’affiche dans des hors-textes sombres et maussades. Dans ce cadre, la machine illustrée exige une lecture iconotextuelle, une importance accordée au détail, aux éléments présents ou absents, aux modalités de passage d’un mode de présentation à l’autre, à la place anticipée ou tardive de l’illustration, au rapport entre le texte, le dessin et sa légende, aux mots qui migrent vers le dessin et surtout au reste du décor incomplet. Chez Robida, les louanges qui passent à la critique et l’humour qui se fait cynisme, sont assez représentatifs des espoirs et des craintes suscités par la découverte et la mise en application de l’électricité, par ses vertus, mais aussi par son aspect incontrôlable.

Le jeu vidéo est un produit qui ne cesse de gagner en popularité alors que les expériences ludiques tendent de plus en plus à se diversifier. Les recherches académiques sur l’objet vidéoludique se sont multipliées dans les dernières années afin de comprendre les particularités du nouveau média, surtout en ce qui concerne l’analyse du produit lui-même et sa réception, mais laissant peu de place à sa créa-tion et sa production. Montréal est un lieu idéal pour étudier le médium : en peu de temps, l’industrie du jeu vidéo est devenue l’un des fleurons industriels québécois. La présente étude s’est intéressée aux développeurs de Montréal, ville où se situe la plus grande partie des studios au Québec, afin de connaître leur perception du produit vidéoludique et de l’industrie. Au travers d’une perspective phénoménologique, un séjour ethno-graphique a été effectué dans un studio de production vidéoludique où plusieurs développeurs ont été observés et interviewés. Ce travail s’inscrit dans une anthropologie du travail et rend compte de la com-plexité qui émerge lorsqu’un travail essentiellement créatif vient se heurter à des motifs de production strictes. Plus encore, il rend compte d’un paradigme opposant directement la création et la production dans un milieu qui se présente comme une avenue prometteuse pour une jeunesse désirant vivre d’un travail créatif. Cette condition est attribuable à la nature du jeu vidéo lui-même qui se situe, selon Kline, Dyer-Witheford et De Peuter (2003), à mi-chemin entre la culture, la technologie et les visées commerciales (marketing). Les développeurs se trouvent donc entre deux eaux : d’un côté ils sont influencés par la culture du jeu, relevant de leurs pratiques, leurs préférences et des commu-nautés de développeurs et, de l’autre côté, par l’industrie qui dicte les façons de faire et viennent selon eux minimiser leur potentiel créatif.

Esta investigación enfoca su análisis al proceso de liquidación obligatoria, por el cual tuvo que atravesar la empresa colombiana de calzado Croydon S.A. Este proceso de liquidación se enmarca dentro de la ley 222 de 1995, demostrando algunas etapas como lo son; aceptación a la adhesión a la ley 222, inicio del proceso de liquidación obligatoria, entre otros. Dentro de esta investigación, se determinan las causas por las cuales Croydon S.A inicia el proceso de liquidación, resaltando motivos tales como; la problemática que vivió el país hacia los años 90s, a causa de la apertura económica, falta de preparación por parte de las empresas nacionales para afrontar la competencia extranjera, poco apoyo por parte del gobierno a empresas exportadoras, problemas de alto nivel de contrabando de productos de consumo, como el calzado, y sin dejar atrás el problema de sindicatos en el país, y la falta de conocimiento por parte de las empresas para iniciar un proceso de exportación.

En la depresión unipolar el estado de ánimo predominante es la tristeza y la experiencia corporal es de un cuerpo que se siente más cómodo con la quietud que con la actividad, parecido a la pereza de la vida cotidiana. En la depresión bipolar el estado de ánimo predominante es de apagamiento emocional. La experiencia corporal es de un cuerpo pesado, cansado, un elemento que se interpone entre los deseos de actuar y la realización de las acciones y que se vuelve un obstáculo para el movimiento. Además, en la depresión bipolar hay mayor bradipsíquia, dificultad para concentrarse y desesperanza que en la unipolar. Se realizó una investigación de tipo cualitativo, exploratorio, de las experiencias subjetivas (de primera persona) de un grupo de pacientes con depresión unipolar y bipolar. Se utilizó entrevistas en profundidad de orientación fenomenológica.

Los desórdenes musculo esqueléticos (DME) son entidades comunes y potencialmente discapacitantes, pero aun así prevenibles que comprenden un amplio número de entidades clínicas específicas (1); se pueden presentar durante la jornada laboral y en muchas ocasiones no son detectados en los exámenes ocupacionales. Se calculó la prevalencia de síntomas osteomusculares de miembro superior en una fábrica de calzado.
