Societal concerns about environmental sustainability has lead to the development of ecologically-friendly alternatives to chemical insecticides for crop protection. One such alternative is biological pest control. In particular, baculoviruses are well suited as insect biopesticides due to their narrow host specificity and relative ease of propagation. In Brazil, the baculovirus Anticarsia gemmatalis nucleopolyhedrovirus (AgMNPV) is the main biological control agent employed for the soybean pest, Anticarsia gemmatalis. This baculovirus biopesticide is currently produced using caterpillars, but increasing market demand for the product has encouraged the development of an in vitro manufacturing process, which can be scaled up to much higher virus productivities. In this study, three wild-type AgMNPV isolates (AgMNPV-2D, AgMNPV-MP2 and AgMNPV-MP5) and a recombinant form (vAgEGT-LacZ) were characterised in terms of occlusion body (OB) production and infection kinetics, to enable future optimisation of the in vitro production process. These viruses were propagated using a Spodoptera frugiperda (IPLB-SF21) insect cell line grown in shaker-flask batch cultures. Among the virus isolates tested, AgMNPV-MP5 was found to be the best producer, yielding (5.3±0.85)x108 OB/mL after 8 days post infection. The characterisation of vAgEGT-LacZ propagation in suspension cell cultures has not been previously reported in the literature; hence it became the main focus for this thesis. In particular, it was carried out a study on the effect of the multiplicity of infection (MOI) on OB production. Five successive batches were performed getting a final production (8.9±1.42)x1014 occlusion bodies, considering that production is related for a bioreactor with final volume of 10m3. A low MOI associated with a fed-batch process for vAgEGT-LacZ production was found to support a 3-fold higher OB yield when compared to the default batch process (1.8x107 and 5.3x107 OB/mL, respectively). This yield is competitive with regards to the production process.

Embora não haja cultivos comerciais de milho geneticamente modificado no Brasil, o efeito de híbridos de milho Bt sobre inimigos naturais e artrópodos de solo deve ser avaliado antes da liberação aos produtores. Assim, ensaios foram conduzidos durante uma safra em duas localidades. Os híbridos de milho modificado geneticamente 7590-Bt11 e Avant-ICP4 foram comparados com seus respectivos isogênicos não transgênicos. Os artrópodes foram avaliados através de observação direta nas plantas e armadilhas de alçapão. de modo geral, não se observaram diferenças entre as populações de tesourinha (Dermaptera: Forficulidae), joaninhas (Coleptera: Coccinellidae), percevejo-pirata (Coleoptera: Anthocoridae), carabídeos (Carabidae), cicindelídeos (Cicindelidae) e aranhas (Araneae). Também não houve diferença no parasitismo de ovos de Helicoverpa zea (Boddie) por Trichogramma sp. (Hymenoptera: Trichogrammatidae). Assim, milho geneticamente modificado expressando as proteínas inseticidas Cry1A(b) e VIP 3A não causa redução nas populações dos principais predadores e parasitóides.

Spodoptera frugiperda (Smith, 1797) (Lepidoptera: Noctuidae) is considered to be the main pest of maize crops in Brazil. Entomopathogenic nematodes (EPN) may be used to control this pest and exhibit different, unique abilities to search for their hosts. The movement of EPN in relation to S. frugiperda was evaluated. To test for horizontal movement, a styrofoam enclosure filled with sand was divided into segments, nematodes were placed at the entrance to the enclosure and a larva was placed at the end of each division. The same approach was used to evaluate vertical movement; however, PVC pipes were used in this case. In general, the mortality was inversely proportional to the initial distance between host and nematodes. In the vertical displacement test, both nematodes were able to kill the larvae up to a distance of 25 cm. Therefore, the infective juveniles of H. amazonensis and S. arenarium can search out, infect and kill larvae of S. frugiperda at distances of up to 60 cm and 25 cm of horizontal and vertical displacement, respectively.

The present work was conducted at FCAV-UNESP, Campus de Jaboticabal, in late harvest crop time of 1991. One hundred interpopulational hybrids obtained from a top-cross between Dent Composite and Flint Composite populations were evaluated. The analyzed characteristics were: Spodoptera frugiperda damages, lodging, damaged ears percentage, and productivity. Estimates of heritability, variance between progenies and genetical gain for each character of available population were determined. Estimates of the progenies variance, like those from heritability for the different analysed characters, indicate that there is an adequate genetical variance for the utilization of that material on subsequent genetic breeding programs, though allowing genetical gains, on the following selection cycles for the characteristics: grain weight, lodging and damages caused by armyworm larvae, whereas for the other characteristics the obtained results were not satisfactory.

BACKGROUND: The use of Trichogramma species is a potential key strategy in integrated pest management. However, its effectiveness depends on the use of chemicals that do not interfere with parasitism and parasite population growth. Here, a study was made of the effects of synthetic insecticides on Trichogramma pretiosum and Trichogramma exiguum in different hosts (Ephestia kuehniella, Plutella xylostella and Spodoptera frugiperda) and the influence of International Organisation for Biological Control (IOBC/WPRS) methodology in selectivity studies using different Trichogramma species. The insecticides used were commercial formulations (triflumuron at a concentration of 0.2 mL L-1 water, etofenprox at a concentration of 0.47 mL L-1 water and endosulfan at a concentration of 7.5 mL L-1 water); the control treatment consisted of distilled water. Eggs attached to cardboard cards were offered to parasitoids inside glass cages. Parasitised eggs, parasitism and adult emergence rates and parasitism reduction were evaluated.RESULTS: Endosulfan and etofenprox, classified as class-4 toxic products, were extremely toxic to the parasitoids. Triflumuron, classified as a non-toxic product, was selective to the parasitoids in eggs of all hosts.CONCLUSIONS: The methodology recommended by IOBC/WPRS influenced results regarding the use of different species of parasitoids, and the use of a single parasitoid species in their experiment is questionable. The control of cotton pests may be accomplished using Bacillus thuringiensis Cry proteins. For this purpose, the objective of this work was to evaluate the insecticidal activity of a new Cry1Ia protein against neonatal larvae of Spodoptera frugiperda and Anthonomus grandis. The complete cry1Ia gene, previously obtained by PCR with oligonucleotide primers based on the sequenced gene, was cloned into the vector pET28a(+), introduced into Escherichia coli BL21(DE3) and expressed by induction with IPTG. The expression of the Cry1Ia protein was confirmed with molecular weight of approximately 81 kDa. The results demonstrated the efficiency of the bacterial system for the expression of B. thuringiensis Cry1Ia protein, which was subsequently used in quantitative bioassays against S. frugiperda and A. grandis larvae, resulting in an extremely toxic protein for both species. This characteristic is exceptionally important for obtaining transgenic cotton plants resistant to these pests.

The study aimed to evaluate the occurrence of pest and management of cotton genetically modified with the introduction of the bacterium Bacillus thunringiensis compared to conventional cotton in the 'Cerrado' region, installing the field experiment in 2007/2008 in Chapadão do Sul-MS. The experimental design was randomized blocks in a 2 × 3 factorial, with two managements of insecticides (with and without insecticides to control lepidopteran) and 3 modes of use of cultivars: 100% transgenic (NUOPAL), 100% non transgenic (Deltaopal) and interior area with transgenic (80% NUOPAL) with non-transgenic border (20% Deltaopal) with five replications. Evaluations were performed weekly of pest infestation, observing in 15 plants per plot the number of Alabama argillacea, Heliothis virescens and Spodoptera frugiperda. Based on these results it was concluded that: the occurrence of A. argillacea and H. virescens was lower in treated transgenic cultivar. There were no differences between transgenic and conventional farming on the occurrence of S. frugiperda.

Recent studies have shown that ingestion by the army worm Spodoptera frugiperda of Cry1Ac toxin from Bt cotton promotes histochemical and ultrastructural changes in the digestive cells of the predatory pentatomid bug Podisus nigrispinus. Therefore, mindful of the changes in the midgut of the predator, which represents the first line of defence in this insect, our aim was to test the hypothesis that the consumption of Bt cotton-fed S. frugiperda by P. nigrispinus might lead to alterations in components of the immune system of P. nigrispinus. The Cry1Ac toxin level in the leaves of Bt cotton, nitric oxide, phenoloxidase activity, and total proteins were quantified by ELISA. Total and differential hemocyte counts were evaluated, and hemocyte ultrastructure analysis was undertaken. We found that ingestion of the prey fed daily with approximately 23 ± 0.70 ng g-1 Cry1Ac by wet weight of leaves, and expressed by the Bt cotton, induces small ultrastructural changes in the predator's granulocytes and plasmatocytes. However, these changes did not affect the total number and differential and humoral variables analyzed for the bug's hemocytes. © 2013 Copyright Taylor and Francis Group, LLC.

We performed laboratory experiments to study maternal care in Doru lineare Eschs. (Dermaptera: Forficulidae), a predator of the maize pest Spodoptera frugiperda J.E. Smith (Lepidoptera: Noctuidae). Females laid 29.7 ± 8.43 (SD) eggs which took 7-10 days to hatch. Mothers searched for an appropriate nest, where they cleaned and defended eggs and nymphs. Females did not recognize eggs from other females as foreign and adopted them. Other evidence of maternal behavior was also observed, such as egg-cleaning, nest-cleaning, and a lower viability of eggs not cared for by females. © 2013 Springer Science+Business Media New York.
