Diferentes critérios utilizados para a escolha de parceiros entre homens e mulheres têm sido identificados. Essa diferença, provavelmente, origina-se pelos diferentes graus de investimento parental entre gêneros. Mulheres parecem ter predisposição a selecionar parceiros com características de investimento emocional, material e com bons indicativos de saúde. Já homens podem utilizar os mesmos critérios que as mulheres, porém dão mais importância que estas à aparência física e juventude. Em relacionamentos de curto e longo prazo a literatura indica que há uma diferença nas escolhas entre mulheres. No primeiro caso, elas têm demonstrando preferir características relacionadas à saúde física, comparado ao segundo tipo de relacionamento, no qual a ênfase tem sido voltada à parceiros bons provedores de recursos e com alto nível de investimento emocional. Há poucas pesquisas que investigaram os critérios que mulheres homossexuais utilizam na escolha de suas parceiras amorosas. Estudos que investigaram a origem da homossexualidade apontaram a possibilidade de influências biológicas. Em termos evolutivos, a homossexualidade poderia ter surgido, em parte, como subproduto da evolução do prazer característico das atividades sexuais. Se esta hipótese estiver correta, o potencial para o desenvolvimento de uma orientação homo, hetero ou bissexual pode ser potencializado por ambientes característicos dos indivíduos em particular. Tal hipótese pode sugerir que os mecanismos psicológicos para escolha de parceiros sejam semelhantes entre as mulheres de variadas orientações sexuais. Para testar esta hipótese, investigou-se as preferências na escolha de parceiras de 100 mulheres em período reprodutivo, entre 18 e 40 anos, que se auto-classificaram como “homossexual exclusivo” ou “homossexual, e às vezes heterossexual”. Para a coleta de dados foram utilizados dois instrumentos, um para seleção das participantes e outro para a coleta de informações. O instrumento de coleta de dados foi dividido em: 1) Dados Demográficos; 2) Dados da parceira; 3) Critérios valorizados na escolha de uma parceira; 4) Critérios valorizados na escolha de uma parceira de curto e longo prazo; 5) Variáveis relacionadas ao desempenho sexual. As participantes foram contatadas pelo método a) “snow ball”, b) bares frequentados por grupos homossexuais e c) associações GLBT. Especificamente, investigou-se as variáveis envolvidas na escolha de parceiras de curto e longo prazo e comparou-se os resultados com os dados coletados por Cruz (2009), com mulheres heterossexuais em período reprodutivo. Os resultados indicaram que há maior preferência por atributos físicos em relacionamentos de curto prazo entre mulheres homo e heterossexuais. Atributos referentes à formação de vínculo foram mais solicitados em relacionamentos de longo prazo, possivelmente porque 75,6% dessas mulheres têm renda e não dependem do parceiro(a) para o provimento na relação, diminuindo a necessidade de parceiro(a)s que invistam recursos materiais. Mulheres homossexuais parecem ter os mesmos padrões de escolha de parceiros que heterossexuais.

O artigo discute as contribuições da obra de Michel Foucault para a compreensão dos registros íntimos que o general brasileiro José Vieira Couto de Magalhães fez em seu próprio diário, escrito em sua maior parte em Londres, na segunda metade do século XIX, época supostamente marcada pelos rigores repressivos da moral vitoriana. Ao registrar detalhadamente sua intimidade, seus sonhos eróticos hetero e homossexuais, bem como condutas e paixões sexuais consideradas àquela época como desviadas da normalidade, o diário íntimo de Couto de Magalhães constitui um reforço da crítica à "hipótese repressiva" desenvolvida por Foucault em seu projeto de uma história da sexualidade. Por outro lado, evidencia-se a legitimidade dos diários íntimos enquanto fonte de pesquisa nas ciências sociais.

This article introduces the concepts of self-organization and autonomy as presented by the thought of Michel Debrun and Edgar Morin and their relationship to the development of a bioethical conscience by means of the school educational process. The proposal aims to contribute to the development of autonomy of students and thus be conducive to their self-organization as “responsible ethical subjects”.

Septins form a conserved family of filament forming GTP binding proteins found in a wide range of eukaryotic cells. They share a common structural architecture consisting of an N-terminal domain, a central GTP binding domain and a C-terminal domain, which is often predicted to adopt a coiled-coil conformation, at least in part. The crystal structure of the human SEPT2/SEPT6/SEPT7 heterocomplex has revealed the importance of the GTP binding domain in filament formation, but surprisingly no electron density was observed for the C-terminal domains and their function remains obscure. The dearth of structural information concerning the C-terminal region has motivated the present study in which the putative C-terminal domains of human SEPT2, SEPT6 and SEPT7 were expressed in E. coli and purified to homogeneity. The thermal stability and secondary structure content of the domains were studied by circular dichroism spectroscopy, and homo- and hetero-interactions were investigated by size exclusion chromatography, chemical cross-linking, analytical ultracentrifugation and surface plasmon resonance. Our results show that SEPT6-C and SEPT7-C are able to form both homo- and heterodimers with a high alpha-helical content in solution. The heterodimer is elongated and considerably more stable than the homodimers, with a K (D) of 15.8 nM. On the other hand, the homodimer SEPT2-C has a much lower affinity, with a K (D) of 4 mu M, and a moderate alpha-helical content. Our findings present the first direct experimental evidence toward better understanding the biophysical properties and coiled-coil pairings of such domains and their potential role in filament assembly and stability.

Septins are a conserved group of GTP-binding proteins that form hetero-oligomeric complexes which assemble into filaments. These are essential for septin function, including their role in cytokinesis, cell division, exocytosis and membrane trafficking. Septin 2 (SEPT2) is a member of the septin family and has been associated with neurofibrillary tangles and other pathological features of senile plaques in Alzheimer's disease. An in silico analysis of the amino acid sequence of SEPT2 identified regions with a significant tendency to aggregate and/or form amyloid. These were all observed within the GTP-binding domain. This was consistent with the experimental identification of a structure rich in beta-sheet during temperature induced unfolding transitions observed for both the full length protein and the GTP-binding domain alone. This intermediate state is characterized by irreversible aggregation and has the ability to bind Thioflavin-T, suggesting its amyloid nature. Under electron microscopy, fibers extending for several micrometers in length could be visualized. The results shown in this study support the hypothesis that single septins, when present in excess or with unbalanced stoichiometries, may be unstable and assemble into amyloid-like structures. Chromatin is a highly dynamic, regulatory component in the process of transcription, repair, recombination and replication. The BRG1 and SNF2H proteins are ATP-dependent chromatin remodeling proteins that modulate chromatin structure to regulate DNA accessibility for DNA-binding proteins involved in these processes. The BRG1 protein is a central ATPase of the SWI/SNF complexes involved in chromatin remodeling associated with regulation of transcription. SWI/SNF complexes are biochemically hetero-geneous but little is known about the unique functional characteristics of the various forms. We have shown that SWI/SNF activity in SW13 cells affects actin filament organization dependent on the RhoA signaling pathway. We have further shown that the biochemical composition of SWI/SNF complexes qualitatively affects the remodeling activity and that the composition of biochemically purified SWI/SNF complexes does not reflect the patterns of chromatin binding of individual subunits. Chromatin binding assays (ChIP) reveal variations among subunits believed to be constitutive, suggesting that the plasticity in SWI/SNF complex composition is greater than suspected. We have also discovered an interaction between BRG1 and the splicing factor Prp8, linking SWI/SNF activity to mRNA processing. We propose a model whereby parts of the biochemical heterogeneity is a result of function and that the local chromatin environment to which the complex is recruited affect SWI/SNF composition. We have also isolated the novel B-WICH complex that contains WSTF, SNF2H, the splicing factor SAP155, the RNA helicase II/Guα, the transcription factor Myb-binding protein 1a, the transcription factor/DNA repair protein CSB and the RNA processing factor DEK. The formation of this complex is dependent on active transcription and links chromatin remodeling by SNF2H to RNA processing. By linking chromatin remodeling complexes with RNA processing proteins our work has begun to build a bridge between chromatin and RNA, suggesting that factors in chromatin associated assemblies translocate onto the growing nascent RNA.

The thesis reports the synthesis, and the chemical, structural and spectroscopic characterization of a series of new Rhodium and Au-Fe carbonyl clusters. Most new high-nuclearity rhodium carbonyl clusters have been obtained by redox condensation of preformed rhodium clusters reacting with a species in a different oxidation state generated in situ by mild oxidation. In particular the starting Rh carbonyl clusters is represented by the readily available [Rh7(CO)16]3- 9 compound. The oxidized species is generated in situ by reaction of the above with a stoichiometric defect of a mild oxidizing agents such as [M(H2O)x]n+ aquo complexes possessing different pKa’s and Mn+/M potentials. The experimental results are roughly in keeping with the conclusion that aquo complexes featuring E°(Mn+/M) < ca. -0.20 V do not lead to the formation of hetero-metallic Rh clusters, probably because of the inadequacy of their redox potentials relative to that of the [Rh7(CO)16]3-/2- redox couple. Only homometallic cluster s such as have been fairly selectively obtained. As a fallout of the above investigations, also a convenient and reproducible synthesis of the ill-characterized species [HnRh22(CO)35]8-n has been discovered. The ready availability of the above compound triggered both its complete spectroscopic and chemical characterization. because it is the only example of Rhodium carbonyl clusters with two interstitial metal atoms. The presence of several hydride atoms, firstly suggested by chemical evidences, has been implemented by ESI-MS and 1H-NMR, as well as new structural characterization of its tetra- and penta-anion. All these species display redox behaviour and behave as molecular capacitors. Their chemical reactivity with CO gives rise to a new series of Rh22 clusters containing a different number of carbonyl groups, which have been likewise fully characterized. Formation of hetero-metallic Rh clusters was only observed when using SnCl2H2O as oxidizing agent because. Quite all the Rh-Sn carbonyl clusters obtained have icosahedral geometry. The only previously reported example of an icosahedral Rh cluster with an interstitial atom is the [Rh12Sb(CO)27]3- trianion. They have very similar metal framework, as well as the same number of CO ligands and, consequently, cluster valence electrons (CVEs). .A first interesting aspect of the chemistry of the Rh-Sn system is that it also provides icosahedral clusters making exception to the cluster-borane analogy by showing electron counts from 166 to 171. As a result, the most electron-short species, namely [Rh12Sn(CO)25]4- displays redox propensity, even if disfavoured by the relatively high free negative charge of the starting anion and, moreover, behaves as a chloride scavenger. The presence of these bulky interstitial atoms results in the metal framework adopting structures different from a close-packed metal lattice and, above all, imparts a notable stability to the resulting cluster. An organometallic approach to a new kind of molecular ligand-stabilized gold nanoparticles, in which Fe(CO)x (x = 3,4) moieties protect and stabilize the gold kernel has also been undertaken. As a result, the new clusters [Au21{Fe(CO)4}10]5-, [Au22{Fe(CO)4}12]6-, Au28{Fe(CO)3}4{Fe(CO)4}10]8- and [Au34{Fe(CO)3}6{Fe(CO)4}8]6- have been isolated and characterized. As suggested by concepts of isolobal analogies, the Fe(CO)4 molecular fragment may display the same ligand capability of thiolates and go beyond. Indeed, the above clusters bring structural resemblance to the structurally characterized gold thiolates by showing Fe-Au-Fe, rather than S-Au-S, staple motives. Staple motives, the oxidation state of surface gold atoms and the energy of Au atomic orbitals are likely to concur in delaying the insulator-to-metal transition as the nuclearity of gold thiolates increases, relative to the more compact transition-metal carbonyl clusters. Finally, a few previously reported Au-Fe carbonyl clusters have been used as precursors in the preparation of supported gold catalysts. The catalysts obtained are active for toluene oxidation and the catalytic activity depends on the Fe/Au cluster loading over TiO2.

Nanopartikel durch Strukturfixierung mizellarer Assoziate aus amphiphilen, endgruppenfunktionalisierten Diblockcopolymeren Zwei unterschiedliche Diblockcopolymersysteme mit Molmassen unterhalb von Mw = 10 000 g/mol wurden über anionische Polymerisation synthetisiert. Ein hetero-telecheles a,w-Poly(dimethylsiloxan)-b-Poly(ethylenoxid) (PDMS-PEO) Diblockcopolymer wurde mit einer Methacrylatendgruppe am PDMS und entweder einer Benzyl-, Hydroxy- oder Carboxylatendgruppe am PEO funktionalisiert. Ein Poly(butadien)-b-Poly(ethylenoxid) (PB-PEO) Diblockcopolymer wurde am PEO ebenfalls entweder mit einer Benzyl-, Hydroxy- oder Carboxylatendgruppe funktionalisiert. In selektiven Lösungsmitteln wie Wasser oder Methanol bilden beide Diblockcopolymersysteme supramolekulare Strukturen mit sphärischer, zylindrischer oder toroider Geometrie aus, die mit statischer und dynamischer Lichtstreuung in Lösung und mit Rasterkraftmikroskopie (AFM) und Transmissionselektronenmikroskopie (TEM) auf der Oberfläche untersucht wurden. Durch Zusatz eines Vernetzers und Initiators wurden die selbstassoziierenden Mizellen des PDMS-PEO Diblockcopolymers permanent durch radikalische Polymerisation mit UV-Licht fixiert. Mizellen des PB-PEO Diblockcopolymers wurden über Bestrahlung mit gamma-Strahlen permanent fixiert. Die Untersuchung der resultierenden Nanopartikel beider Diblockcopolymersysteme mit AFM und TEM zeigte, daß diese sogar in nicht selektiven Lösungsmitteln wie Tetrahydrofuran formstabil bleiben.

Gekreuzt konjugierte Oligomere aus Benzolringen und Heterocyclen Oligomere aus Arylen- oder/und Hetarylen-Bausteinen stellenein aktuelles Arbeitsgebiet in der organischen Synthese undder Materialwissenschaft dar. Die Enoneinheit der Chalkonerepräsentiert ein bifunktionelles Elektrophil, das für denAufbau von unterschiedlichen Heterocyclen hervorragendgeeignet ist. Im Rahmen der vorliegenden Arbeit wurden dreineue Reihen von heterocyclischen Oligomeren hergestellt:1H-Pyrrole, 1H-Pyrazole und Pyridine, die über eine alter-nierende Sequenz von Phenylen- oder Benzoylen-Einheitenverbunden sind. Ihre Herstellung basierte auf Michael-Additionen von den kettenförmigen, gekreuzt konjugiertenOligochalkonen und den jeweiligen geeigneten N-haltigenBausteinen: Tosylmethylisocyanid, Methylhydrazin undPhenacylpyridiniumbromid in der Reihe der Pyrrole, Pyrazoleund Pyridine. Die UV/Vis-Untersuchung aller Reihen der Oligoheterocyclenzeigte als allgemeinen Trend im Vergleich mit dem UV/Vis-Verhalten der Oliogochalkone eine ausgeprägte hypsochromeVerschiebung der langwelligen Absorption. Dieser beobachtetehypsochrome Shift geht auf die veränderten Chromophore zurück und deutet außerdem darauf hin, dass die effektiveKonjugation bei wachsender Moleküllänge geringer sein sollte. In den einzelnen Reihen der Oligoheterocyclenerkennt man jeweils bei Vergrößerung der Chromophore einenkleinen bathochromen Shift der langwelligen Absorption.
