Imagiologia por Ressonância Magnética (IRM) é uma modalidade de imagem médica que está a recuperar o interesse como uma técnica não invasiva no estudo da pele. Tipicamente campos magnéticos de elevada densidade e quipamentos específicos são usados. Este facto limita o usos da técnica a laboratórios e centros de investigação especializados. Neste trabalho estudou-se a viabilidade do uso da IRM no estudo da pele e da sua vasculatura usando equipamento convencional disponível em contexto clínico. Sequências IRM para imagem estrutural e veascular foram optimizadas e testadas para obtenção de imagens da pele do punho de 6 voluntários saudáveis. As sequências observáveis dos vasos, razão sinal-ruído, e razão contraste-ruído. Foi observado que duas sequências volumétricas baseadas em eco de gradiente e com ponderações T1 e T2 forneciam informação complementar em respeito à vasculatura da pele com resoluções espaciais da ordem dos micrómetros, podendo ainda esta informação ser fundida com imagens estruturais das cadamas da pele. Foi igualmente observado que estas sequências fornecem informação útil usando equipamento convencional e perspectiva-se a sua utilização no estudo das vasculatura de tumores cutâneos e na doença vascular periférica.

Os vários estudos sobre a importância da água na hidratação da pele humana vêm sugerindo que o aumento da ingestão de água se revela positivamente na pele. Considerando que a contabilização da água consumida deve atender às suas várias origens, avaliámos em dois grupos com diferentes consumos de água total, o impacto do aumento do aporte de 2L/dia de água bebida. Os resultados sugerem que, existe uma melhoria da hidratação epidérmica e dérmica obtida por métodos de referência. Amagnitude do impacto sobre a hidratação cutânea é maior no grupo que regularmente consome menos água, sugerindo que o aumento do consumo de água tem mais efeitos benéficos na saúde cutânea dos indivíduos que consomem menos água por dia. Estes dados estão de acordo, com a literatura publicada, justificando o interesse no aprofundamento do tema.

Os modelos animais têm sido úteis para estudar mecanismos específicos que afetam a pele humana. É o caso do envelhecimento e das alterações micromecânicas que determinam o enrugamento (a formação de rugas) nos ratinhos irradiados com UV. Estes modelos permitiram perceber que o envelhecimento envolve muitas respostas mecânicas peculiares que não podem ser explicadas pela deformação homogénea da pele. No entanto, a diferença entre o tempo de vida destas espécies afeta também os processos e este é um aspeto principal a ter em conta. Este projeto é direcionado à comparação das propriedades cutâneas de dois grupos de ratos Wistar com diferentes idades – ratos jovens-adultos (n=7, 20-24 semanas) e adultos-velhos (n=5, 48-72 semanas). Medições não-invasivas de perda transepidérmica de água (PTEA), hidratação superficial (MoistureMeter) e biomecânica (Reviscometer e Cutometer por MPA80) foram realizadas nas zonas dorsais (pescoço, dorso-posterior) do animal durante 5 dias, permitindo comparações estatísticas entre grupos. Os resultados não revelaram diferenças significativas para a PTEA, firmeza ou viscoelasticidade entre os dois grupos. Contudo, foi notada uma diminuição significativa, próxima de 40%, da hidratação da pele no grupo dos ratos adultos-velhos. Estes resultados, apesar de preliminares, se fizermos a translação para os seres humanos poderiam ser úteis para avaliar mudanças relacionadas à idade em processos como a recuperação cutânea, ou eficácia / segurança de pensos, onde a hidratação é um factor determinante. Asua potencial utilização para a predição da eficácia e/ou segurança dos produtos de aplicação tópica para idosos em comparação aos adultos merecem ser exploradas.

Os vários estudos sobre a importância da água na Hidratação da pele humana vêm sugerindo que o aumento da ingestão de água pode ter um impacto positivo na fisiologia da pele normal. Mantendo os hábitos dietários, avaliados por Questionário de Frequência Alimentar e com base no consumo de água total , avaliámos as consequências do aumento de água bebida na bioimpedância e na hidratação da pele “in vivo”. Em dois grupos com diferentes consumos de água total, avaliamos, ao fim de um mês, o impacto do aumento do aporte de 2L/dia de água bebida. Este aumento não produziu alterações hemodinânicas ou ponderais. Os resultados sugerem uma melhoria da hidratação epidérmica. A magnitude do impacto sobre a hidratação cutânea é maior no grupo que Regularmente consome menos água, sugerindo que o aumento do consumo de água tem mais efeitos benéficos na saúde cutânea dos indivíduos que consomem menos água por dia. Estes dados estão de acordo, com a literatura publicada, justificando o interesse no aprofundamento do tema, de modo a contribuir para o conhecimento sobre a importância da água no contexto da fisiologia cutânea em especial no grupo de indivíduos com hábitos de menor consumo.

A dermatite atópica é uma patologia cutânea crónica que requer cuidados intensivos da pele e tratamento farmacológico; contudo, os tratamentos disponíveis necessitam urgentemente de ser melhorados, especialmente quando utilizados por períodos longos ou em grupos específicos (ex: crianças). A nanotecnologia tem contribuído com sistemas de veiculação inovadores e pode oferecer terapias efectivas e direcionadas. Os objectivos deste estudo centraram-se na preparação caracterização das nanopartículas de policaprolactona carregadas com acetato de hidrocortisona em termos das propriedades físico-químicas, eficiência de encapsulação, ensaios de libertação in vitro e ensaios de segurança dos excipientes utilizados em voluntários humanos. As nanopartículas produzidas apresentaram um tamanho médio de 258,4 24,5 nm e um índice de polidispersão de 0,084. O potencial zeta foi -4,39 0,62 mV e a eficiência de encapsulação foi 36,32 0,03 %. A libertação in vitro do fármaco foi controlada ao longo do tempo. Além disso, os testes de segurança indicaram que os excipientes foram bem tolerados. Este estudo demonstra que as nanopartículas de policaprolactona são sistemas estáveis para veiculação de acetato de hidrocortisona que poderão conduzir a uma libertação prolongada do fármaco, com resultados promissores ao nível da sua segurança quando aplicados na pele humana.

A relatively simple, selective, precise and accurate high performance liquid chromatography (HPLC) method based on a reaction of phenylisothiocyanate (PITC) with glucosamine (GL) in alkaline media was developed and validated to determine glucosamine hydrochloride permeating through human skin in vitro. It is usually problematic to develop an accurate assay for chemicals traversing skin because the excellent barrier properties of the tissue ensure that only low amounts of the material pass through the membrane and skin components may leach out of the tissue to interfere with the analysis. In addition, in the case of glucosamine hydrochloride, chemical instability adds further complexity to assay development. The assay, utilising the PITC-GL reaction was refined by optimizing the reaction temperature, reaction time and PITC concentration. The reaction produces a phenylthiocarbarnyl-glucosamine (PTC-GL) adduct which was separated on a reverse-phase (RP) column packed with 5 mu m ODS (C-18) Hypersil particles using a diode array detector (DAD) at 245 nm. The mobile phase was methanol-water-glacial acetic acid (10:89.96:0.04 v/v/v, pH 3.5) delivered to the column at 1 ml min(-1) and the column temperature was maintained at 30 degrees C Using a saturated aqueous solution of glucosamine hydrochloride, in vitro permeation studies were performed at 32 +/- 1 degrees C over 48 h using human epidermal membranes prepared by a heat separation method and mounted in Franz-type diffusion cells with a diffusional area 2.15 +/- 0.1 cm(2). The optimum derivatisation reaction conditions for reaction temperature, reaction time and PITC concentration were found to be 80 degrees C, 30 min and 1 % v/v, respectively. PTC-Gal and GL adducts eluted at 8.9 and 9.7 min, respectively. The detector response was found to be linear in the concentration range 0-1000 mu g ml(-1). The assay was robust with intra- and inter-day precisions (described as a percentage of relative standard deviation, %R.S.D.) < 12. Intra- and inter-day accuracy (as a percentage of the relative error, %RE) was <=-5.60 and <=-8.00, respectively. Using this assay, it was found that GL-HCI permeates through human skin with a flux 1.497 +/- 0.42 mu g cm(-2) h(-1), a permeability coefficient of 5.66 +/- 1.6 x 10(-6) cm h(-1) and with a lag time of 10.9 +/- 4.6 h. In this paper, we report the synthesis and healing ability of a non-cytotoxic supramolecular polyurethane network whose mechanical properties can be recovered efficiently (> 99%) at the temperature of the human body (37 ºC). Rheological analysis revealed an acceleration in the drop of the storage modulus above 37 ºC, on account of the dissociation of the supramolecular polyurethane network, and this decrease in viscosity enables the efficient recovery of the mechanical properties. Microscopic and mechanical characterisation has shown that this material is able to recover mechanical properties across a damage site with minimal contact required between the interfaces and also demonstrated that the mechanical properties improved when compared to other low temperature healing elastomers or gel-like materials. The supramolecular polyurethane was found to be non-toxic in a cytotoxicity assay carried out in human skin fibroblasts (cell viability > 94% and non-significantly different compared to the untreated control). This supramolecular network material also exhibited excellent adhesion to pig skin and could be healed completely in situ post damage indicating that biomedical applications could be targeted, such as artificial skin or wound dressings with supramolecular materials of this type.

In all higher nonhuman primates, species survival depends upon safe carrying of infants clinging to body hair of adults. In this work, measurements of mechanical properties of ape hair (gibbon, orangutan, and gorilla) are presented, focusing on constraints for safe infant carrying. Results of hair tensile properties are shown to be species-dependent. Analysis of the mechanics of the mounting position, typical of heavier infant carrying among African apes, shows that both clinging and friction are necessary to carry heavy infants. As a consequence, a required relationship between infant weight, hair-hair friction coefficient, and body angle exists. The hair-hair friction coefficient is measured using natural ape skin samples, and dependence on load and humidity is analyzed. Numerical evaluation of the equilibrium constraint is in agreement with the knuckle-walking quadruped position of African apes. Bipedality is clearly incompatible with the usual clinging and mounting pattern of infant carrying, requiring a revision of models of hominization in relation to the divergence between apes and hominins. These results suggest that safe carrying of heavy infants justify the emergence of biped form of locomotion. Ways to test this possibility are foreseen here.

The objective of this thesis work, is to propose an algorithm to detect the faces in a digital image with complex background. A lot of work has already been done in the area of face detection, but drawback of some face detection algorithms is the lack of ability to detect faces with closed eyes and open mouth. Thus facial features form an important basis for detection. The current thesis work focuses on detection of faces based on facial objects. The procedure is composed of three different phases: segmentation phase, filtering phase and localization phase. In segmentation phase, the algorithm utilizes color segmentation to isolate human skin color based on its chrominance properties. In filtering phase, Minkowski addition based object removal (Morphological operations) has been used to remove the non-skin regions. In the last phase, Image Processing and Computer Vision methods have been used to find the existence of facial components in the skin regions.This method is effective on detecting a face region with closed eyes, open mouth and a half profile face. The experiment’s results demonstrated that the detection accuracy is around 85.4% and the detection speed is faster when compared to neural network method and other techniques.

Mammography is a diagnostic imaging method in which interpretation depends on knowledge of radiological aspects as well as the clinical exam and pathophysiology of breast diseases. In this work a mammography phantom was developed to be used for training in the operation of mammographic x-ray equipment, image quality evaluation, self-examination and clinical examination of palpation. Polyurethane was used for the production of the phantoms for its physical and chemical properties and because it is one of the components normally used in prostheses. According to the range of flexibility of the polyurethane, it was possible to simulate breasts with higher or lower amount of adipose tissue. Pathologies such as areolar necrosis and tissue rejection due to surgery reconstruction after partial mastectomy were also simulated. Calcifications and nodules were simulated using the following materials: polyethylene, poly (methyl methacrylate), polyamide, polyurethane and poly (dimethyl silicone). Among these, polyethylene was able to simulate characteristics of calcification as well as breast nodules. The results from mammographic techniques used in this paper for the evaluation of the phantoms are in agreement with data found in the literature. The image analyses of four phantoms indicated significant similarities with the human skin texture and the female breast parenchyma. It was possible to detect in the radiographic images produced regions of high and low radiographic optical density, which are characteristic of breasts with regions of different amount of adipose tissue. The stiffnesses of breast phantoms were adjusted according to the formulation of the polyurethane which enabled the production of phantoms with distinct radiographic features and texture similar to human female breast parenchyma. Clinical palpation exam of the phantoms developed in this work indicated characteristics similar to human breast in skin texture, areolar region and parenchyma

Trichophyton rubrum is a dermatophyte, which can cause infections in human skin, hair and nail. Pothomorphe umbellata (L.) Miq. (Piperaceae) is a native Brazilian plant, in which phytochemical studies have demonstrated the presence of steroids, 4-nerolidylcatechol, sesquiterpenes and essential oils. The objective of this study was to analyze the in vitro activity of extracts and fractions of P. umbellata on resistant strains of T. rubrum. The microdilution plate method was utilized to test Tr1, H6 and Delta TruMDR2 strains of T rubrum; Delta TruMDR2 strain was obtained from H6 by TruMDR2 gene rupture, which is involved in multiple drugs resistance. The highest antifungal activity to all strains was observed for dichloromethane and hexane fractions of the 70% ethanolic extract which showed minimal inhibitory concentration (MIC) and minimal fungicide concentration (MFC) of 78.13 mu g/mL. This antifungal activity was also obtained by 70% ethanolic extract, which presented MIC and MFC of 78.13 mu g/mL to Delta TruMDR2, whereas the MIC values for Tr1 and H6 were 78.13 and 156.25 mu g/mL, respectively. Our results suggest the potential for future development of new antifungal drugs from P umbellata, especially to strains presenting multiple resistance. (C) 2012 Elsevier Masson SAS. O autor desenvolveu em silicona uma escala de tonalidades de pele humana. Foram confeccionados vinte e sete corpos-de-prova em silicona acética (Silastic 732 RTV), pigmentados com óxidos de ferro e dióxido de titânio. A quantidade de silicona acética manteve-se constante (dois gramas) em todos os corpos-de-prova, e os pigmentos foram misturados a ela em várias proporções até a obtenção de vinte e sete diferentes tonalidades. Através da comparação da cor dos corpos-de-prova com a cor da pele de quarenta e um indivíduos, foram selecionados os cinco corpos-de-prova com as tonalidades que mais se igualavam à cor da pele dos pacientes, compondo, assim, um guia de tonalidades. Com a metodologia empregada, foi possível desenvolver uma escala de tonalidades de pele que poderá facilitar a definição do tom da pele do paciente quando da confecção de próteses faciais em silicona, permitindo economia de tempo e de material no momento da seleção da cor.

A purified glycoprotein of 43 000 daltons from Paracoccidioides brasiliensis (gp43) was tested as paracoccidioidin in delayed-type hypersensitivity (DTH) tests in both experimental animals (guinea pig and mice) and patients with paracoccidioidomycosis (PCM). The gp43 paracoccidioidin was compared with the traditional Fava Netto antigen (AgFN). In guinea pigs, the intradermal injection of 2 μg of gp43 showed a similar response to those obtained with AgFN, showing in histological sections a population of lymphoid cells that participate in DTH. In mice, gp43 at a dose of 3.75μg showed positive DTH response. The use of gp43 as paracoccidioidin in humans showed that this molecule can be used to evaluate the DTH response in patients with PCM. Of 25 PCM patients studied, 48% were positive to gp43 while only 28% were positive to AgFN; 12 PCM patients were completely anergic to both antigens. Considering only those 13 PCM patients who were responsive to gp43 and/or to AgFN, 92.3% reacted against gp43 and 53.8% reacted against AgFN (P < 0.05). Gp43 skin test responses (13.67 ± 9.56 mm) were significantly larger than those obtained with AgFN (8.43 ±3.69 mm). Immunohistochemical study of the human skin showed a perivascular inflammatory response constituted predominantly by T lymphocytes, macrophages and polymorphonuclear leukocytes. © 1996 ISHAM.

Based on the lack of electromyographic researches on sport and programmes of physical conditioning, we can say that it is necessary to reexamine some exercises routinely used in the programmes of physical conditioning. Thus, the trapezius and serratus anterior muscles were studied electromyographically so that we could evaluate the validity in some ways of execution of the frontal-lateral cross, dumbbells exercises for the development of these muscles. We analyzed 24 male volunteers, 18 to 25 years old, using a 2-channel TECA TE 4 electromyograph and Hewlett Packard surface electrodes. For the execution of the exercise it was used a supine bench, a straight board and two bars of 40 cm made of light wood. The results showed that TS acted preferentially in standing modality and in the inclined supine modality, however with activity levels that do not justify its inclusion in physical fitness programmes.
