Obesity has been shown to impair myocardial performance. Nevertheless, the mechanisms underlying the participation of calcium (Ca2+) handling on cardiac dysfunction in obesity models remain unknown. L-type Ca2+ channels and sarcoplasmic reticulum (SR) Ca2+-ATPase (SERCA2a), may contribute to the cardiac dysfunction induced by obesity. The purpose of this study was to investigate whether myocardial dysfunction in obese rats is related to decreased activity and/or expression of L-type Ca2+ channels and SERCA2a. Male 30-day-old Wistar rats were fed standard (C) and alternately four palatable high-fat diets (Ob) for 15 weeks. Obesity was determined by adiposity index and comorbidities were evaluated. Myocardial function was evaluated in isolated left ventricle papillary muscles under basal conditions and after inotropic and lusitropic maneuvers. L-type Ca2+ channels and SERCA2a activity were determined using specific blockers, while changes in the amount of channels were evaluated by Western blot analysis. Phospholamban (PLB) protein expression and the SERCA2a/PLB ratio were also determined. Compared with C rats, the Ob rats had increased body fat, adiposity index and several comorbidities. The Ob muscles developed similar baseline data, but myocardial responsiveness to post-rest contraction stimulus and increased extracellular Ca2+ was compromised. The diltiazem promoted higher inhibition on developed tension in obese rats. In addition, there were no changes in the L-type Ca2+ channel protein content and SERCA2a behavior (activity and expression). In conclusion, the myocardial dysfunction caused by obesity is related to L-type Ca2+ channel activity impairment without significant changes in SERCA2a expression and function as well as L-type Ca2+ protein levels. INTRODUÇÃO: O objetivo deste trabalho foi analisar a prevalência do uso de drogas por estudantes da Faculdade de Medicina de Botucatu - Unesp, comparada com outras oito escolas médicas paulistas (uso na vida, nos últimos 12 meses e nos últimos 30 dias). A pesquisa foi realizada entre 1994 e 1995, com 5.227 estudantes do 1o ao 6o ano de graduação. MATERIAL E MÉTODO: Foi usado um questionário de auto-respostas, anônimo, incluindo o questionário da Organização Mundial da Saúde para levantamento de uso de drogas e álcool. Setenta e um por cento (3.725) dos alunos responderam ao mesmo, e destes, 421 eram de Botucatu. RESULTADOS: Não houve diferenças estatisticamente significantes entre escolas e, nos 30 dias anteriores ao preenchimento do questionário, a prevalência do uso de drogas para os estudantes de Botucatu foi a seguinte, com a variação entre outras escolas mostrada entre parênteses: álcool 50% (42-50%); tabaco 7% (7-13%); solventes 8% (7-12%); maconha 6% (6-16%); benzodiazepínicos (BZD) 3% (2-9%); cocaína 0,5% (0,2-4%); anfetaminas 1 % (0-1%). Embora tenha se encontrado um uso crescente de todas as drogas do 1o ao 6o ano, e em especial os BZD, os estudantes não aprovam este uso. A análise de regressão logística indicou que o uso de álcool e drogas foi favorecido por: a) ser homem; b) perder aulas sem razão e referir ou ter muito tempo livre nos finais de semana; e c) ter uma atitude favorável em relação ao uso de álcool e drogas. Diferentemente de outras escolas, na Unesp não houve diferenças estatisticamente significantes de gênero em relação ao uso de tranqüilizantes. No entanto, as mulheres iniciam uso mais precocemente e o fazem mais freqüentemente. Também as mulheres já faziam uso de maconha antes de entrar para a faculdade (30% mulheres X 10% homens), o contrário ocorrendo com solventes (50% homens X 2% mulheres), sendo essas diferenças estatisticamente significantes. CONCLUSÕES: Embora a pesquisa tenha focalizado o uso (não abuso ou dependência), os resultados sugerem a necessidade de as universidades estabelecerem uma política clara de orientação sobre uso de drogas e álcool para os estudantes, incluindo mudanças curriculares e programas de prevenção.

OBJETIVO: Analisar os efeitos da ingestão crônica de álcool sobre o nervo óptico em um modelo murino adulto. MÉTODOS: Doze ratos machos da raça Wistar, de 30 dias de idade, foram divididos por sorteio em 2 grupos experimentais: tratado (TG), com 8 animais, alimentados com ração-padrão para roedores de laboratório e uma mistura de água de torneira e etanol ad libitum; controle (CG), com 4 animais, alimentados com a mesma ração e água de torneira pura ad libitum. Após 40 semanas todos os ratos foram sacrificados, sendo os nervos ópticos de ambos os olhos preparados para microscopia óptica e eletrônica. A área de secção transversal de cada nervo a aumento de 500´, assim como número de fibras axonais dentro de 5 campos aleatoriamente selecionados a aumento de 2000´ foram medidos com auxílio de digitalizador de imagens acoplado ao microscópio óptico. Foram realizadas fotomicrografias de 10 campos aleatoriamente selecionados de cada nervo (5 centrais e 5 periféricos) a aumento de 4200´ em microscópio eletrônico de transmissão. RESULTADOS: A análise morfométrica não mostrou diferenças estatisticamente significativas entre os 2 grupos estudados. em contraste com o CG, o exame ultra-estrutural dos nervos ópticos do TG mostrou um intenso desarranjo das bainhas de mielina, que se tornaram espessadas, com separação de suas lamelas, apresentando, por vezes, degenerações interlamelares elétron-densas, além da presença de muitas organelas degeneradas. CONCLUSÃO: Os achados desse estudo mostram alterações ultra-estruturais no nervo óptico de ratos adultos após ingestão crônica de álcool, sem modificações morfométricas significativas.

This study aimed to investigate the influence of calf genetic group on the performance of Nellore lactating cows. The variables studied included milk ingestion, pasture intake, calf weight gain and cow body weight. A total of 13 Nellore calves and 8 crossbred Simental x Nellore calves were used, all born from Nellore mothers. During the experimental period of 210 days, calves were weighted at birth and at 30-day intervals, whereas cows were weighed every 14 days. Milk intake was estimated using the weigh-suckle-weigh method. Pasture intake was determined 6, 12, and 24 weeks after birth by using chromic oxide as an external marker and indigestible acid detergent fiber as an internal marker. Average weight of crossbred calves was 10.1 kg and their milk intake was 0.42 kg higher than in purebred calves. There was no difference, however, in pasture intake between genetic groups. In Nellore cows, body weight and pasture consumption (9.2 kg of DM/cow/day) were not influenced by calf heterosis.

This study evaluated the effect of Matricaria chamomilla and vaccination frequency on cattle immunization against rabies. Four groups (n = 15/group) were treated with or without Matricaria chamomilla CH12 and vaccinated with one or two doses of rabies vaccine (30 day interval). No effect of chamomile was found on cattle immunization against rabies; however, antibody titers were protective in cattle vaccinated twice, while 93.3% of cattle vaccinated only once had titers under 0.5 UI/ml after 60 days. In conclusion, the use of chamomile did not alter the Immoral immune response in cattle, and two vaccine doses are suggested for achieving protective antibody titers.

Objective: The aim of this study was to compare two methodologies used in the evaluation of tissue response to root-end filling materials in rats. Material and Methods: Forty rats were divided into 4 groups: in Groups I and II (control groups), empty polyethylene tubes were implanted in the extraction site and in the subcutaneous tissue, respectively; in Groups III and IV, polyethylene tubes filled with ProRoot MTA were implanted in the extraction site and in the subcutaneous tissue, respectively. The animals were killed 7 and 30 days after tube implantation, and the hemi-maxillas and the capsular subcutaneous tissue, both with the tubes, were removed. Specimens were processed and evaluated histomorphologicaly under light microscopy. The scores obtained were analyzed statistically by the Kruskal-Wallis test (p<0.05). Results: There were no statistically significant differences between the implantation methods (p=0.78033, p=0.72039). It was observed that the 30-day groups presented a more mature healing process due to smaller number of inflammatory cells. Conclusions: The present study showed no differences in tissue responses as far as the implantation site and the studied period were concerned. Alveolar socket implantation methodology represents an interesting method in the study of the biological properties of root-end filling endodontic materials due to the opportunity to evaluate bone tissue response.

The aim of this in vivo study was to evaluate the biocompatibility of three current bonding agents and calcium hydroxide cement. Sixty polyethylene tubes filled with the following materials: Group 1: Prime & Bond NT (PB - Dentsply, US; Group 2: Bond 1 (BO - Jeneric/Pentron, US); Group 3: Optibond Solo (OP - Kerr, US); and Group 4 (control): calcium hydroxide cement - Dycal (CH - Dentsply, US) were implanted into the connective tissue of 30 rats. After 15, 30 and 60 days, the implants were excised and the animals sacrificed. The biopsies were immersed in Karnovsky (pH, 7.2) fixative solution for 48 hours, and processed using routine histological technique. Six-micron-thick sections were cut and stained with hematoxilin and eosin and Masson's trichome technique. Microscopic evaluation was used to compare the connective tissue reactions caused by the experimental and control materials adjacent to the tube opening. At 15 days, the experimental and control materials triggered a moderate to intense inflammatory response which gave rise to a thick capsule adjacent to the tube opening. With time, the inflammatory reaction decreased. At 60 days, the connective tissue adjacent to the bonding agents exhibited a persistent inflammatory response mediated by macrophages and giant cells which were engulfing displaced resin components. on the other hand, for the control group (calcium hydroxide) no inflammatory response associated with a thin capsule adjacent to the material was observed even at the 30-day period. The hard-setting calcium hydroxide cement allowed complete healing and was considered more biocompatible than the bonding agents.

Objective. The present study evaluated the dynamics of the development of periapical lesions.Study design. Root canals from dogs' teeth were exposed to the oral environment, and then sealed for 7 days (Group A), 15 days (Group B), 30 days (Group C), and 60 days (Group D). After each experimental period, radiographs were taken to detect periapical bone resorption. In addition, histological sections from the periapical region were prepared. The radiographic and histological results were analyzed by ANOVA and Tukey's, Wilcoxon, and Pearson's tests. Significance level was set at 5%.Results. Lesions were radiographically visible at 15 and 30 days, and had similar size at these periods (P > .05). At 60 days, lesions were larger than in the other periods (P < .05). Bone resorption was detected histologically at 7 days. The greatest values of bone resorption were observed at the 30- and 60-day periods (P < .05). The results of the methods of evaluation were similar only at the 30-day period. There was no correlation between the radiographic and histological results.Conclusions. Periapical radiography did not provide detection of periapical lesion in its initial stages. The periapical lesions became more evident radiographically when the bone resorption area increased.
