Oat hull hemicellulosic hydrolysate obtained by diluted acid hydrolysis was employed as fermentation medium for Pichia stipitis cultivation. A comparison between the use of treated hydrolysate with 1% activated charcoal to reduce the toxic compounds generated during the hydrolysis process and untreated hydrolysate as a control was conducted. In the cultures using treated hydrolysate the total consumption of glucose, low xylose consumption and ethanol and glycerol formation were observed. The medium formulated with untreated hydrolysate showed morphological cell modifications with consequently cell death, no ethanol formation and formation of glycerol as byproduct of fermentative process, probably as a response to stressful conditions to yeast due to presence of high concentration of toxic compounds. Thus, further studies are suggested in order to determine the best conditions for hydrolysis and detoxification of the hydrolysate to improve the fermentative performance of P. stipitis.

This work was focused on the steam explosion pretreatment reproduction and alkaline delignification reactions on a pilot scale for the ethanol production, through different varieties of natural sugarcane bagasse, pretreated bagasse and delignified pretreated bagasse (cellulosic pulp). The possible chemical composition differences of the various types of bagasse, as well as the chemical composition variations of the materials in the 20 processes of pretreatment and delignification on the pilot scale were verified. The analytical results of the 20 samples of most diverse varieties and origins of natural sugarcane bagasse considering planting soils, planting periods and weather; show no significant chemical differences. It is evident that only with the chemical composition it is not possible to verify the differences between the varieties of sugarcane bagasses. The research results may offer some evidences of these varieties, but it is not a reliable parameter. The pilot process of steam explosion pretreatment and the alkaline delignification process of pretreated material showed through analytical results a good capacity of reproduction, as the standard differences were below 2.7. The average allowed in the pretreatment and alkaline delignification processes were 66.1 +/- 0.8 and 51.5 +/- 2.6 respectively, ensuring an excellent reproduction capacity of the processes obtained through chemical characterizations. Liquid biofuels can be produced from a variety of feedstocks and processes. Ethanol and biodiesel production processes based on conventional raw materials are already commercial, but subject to further improvement and optimization. Biofuels production processes using lignocellulosic feedstocks are still in the demonstration phase and require further R&D to increase efficiency. A primary tool to analyze the efficiency of biofuels production processes from an integrated point of view is offered by exergy analysis. To gain further insight into the performance of biofuels production processes, a simulation tool, which allows analyzing the effect of process variables on the exergy efficiency of stages in which chemical or biochemical reactions take place, were implemented. Feedstocks selected for analysis were parts or products of tropical plants such as the fruit and flower stalk of banana tree, palm oil, and glucose syrups. Results of process simulation, taking into account actual process conditions, showed that the exergy efficiencies of the acid hydrolysis of banana fruit and banana pulp were in the same order (between 50% and 60%), lower than the figure for palm oil transesterification (90%), and higher that the exergy efficiency of the enzymatic hydrolysis of flower stalk (20.3%). Addition of salts, especially perchlorates, to zwitterionic micelles of SB3-14, C(14)H(29)NMe(2)(+)(CH(2))(3)SO(3)(-), induces anionic character and uptake of H(3)O(+) by SB3-14 micelles. Thus, the addition of alkali metal perchlorates accelerates the acid hydrolysis of 2-(p-heptoxypheny1)-1,3-dioxolane, HPD, in the presence of SB3-14 micelles, which depends on the local proton concentration at the micelle surface. The addition of metal chlorides to solutions of such perchlorate-modified SB3-14 micelles decreases both the negative zeta potential of the micelles and the observed rate constant for acid hydrolysis of HPD. The effect of the monovalent cations Li(+), Na(+), and K(+) is smaller than that of the divalent cations Be(2+), Mg(2+), and Ca(2+), and much smaller than that of the trivalent cations Al(3+), La(3+), and Er(3+). The major factor responsible for this cation valence dependence of these effects is shown to be electrostatic in nature, reflecting the strong dependence of the micellar surface potential on the cation valence. The fact that the salt effects are not identical after correction for the electrostatic effects indicates that additional secondary nonelectrostatic effects may contribute as well.

There is growing interest in cellulose nanofibres from renewable sources for several industrial applications. However, there is a lack of information about one of the most abundant cellulose pulps: bleached Eucalyptus kraft pulp. The objective of the present work was to obtain Eucalyptus cellulose micro/nanofibres by three different processes, namely: refining, sonication and acid hydrolysis of the cellulose pulp. The refining was limited by the low efficiency of isolated nanofibrils, while sonication was more effective for this purpose. However, the latter process occurred at the expense of considerable damage to the cellulose structure. The whiskers obtained by acid hydrolysis resulted in nanostructures with lower diameter and length, and high crystallinity. Increasing hydrolysis reaction time led to narrower and shorter whiskers, but increased the crystallinity index. The present work contributes to the different widespread methods used for the production of micro/nanofibres for different applications. (C) 2012 Elsevier Ltd. All rights reserved.

A series of 3(2H)-furanones, based on side-chain modifications of a parent 3(2H)-furanone, was synthesized in good yield. The parent compound was prepared by hydrogenolysis, and subsequent acid hydrolysis, of isoxazole derivatives. The isoxazole was prepared by a [3+2] 1,3-dipolar cycloaddition reaction between 3-butyn-2-ol and nitrile oxide.

Abstract Background The use of lignocellulosic constituents in biotechnological processes requires a selective separation of the main fractions (cellulose, hemicellulose and lignin). During diluted acid hydrolysis for hemicellulose extraction, several toxic compounds are formed by the degradation of sugars and lignin, which have ability to inhibit microbial metabolism. Thus, the use of a detoxification step represents an important aspect to be considered for the improvement of fermentation processes from hydrolysates. In this paper, we evaluated the application of Advanced Oxidative Processes (AOPs) for the detoxification of rice straw hemicellulosic hydrolysate with the goal of improving ethanol bioproduction by Pichia stipitis yeast. Aiming to reduce the toxicity of the hemicellulosic hydrolysate, different treatment conditions were analyzed. The treatments were carried out according to a Taguchi L16 orthogonal array to evaluate the influence of Fe+2, H2O2, UV, O3 and pH on the concentration of aromatic compounds and the fermentative process. Results The results showed that the AOPs were able to remove aromatic compounds (furan and phenolic compounds derived from lignin) without affecting the sugar concentration in the hydrolysate. Ozonation in alkaline medium (pH 8) in the presence of H2O2 (treatment A3) or UV radiation (treatment A5) were the most effective for hydrolysate detoxification and had a positive effect on increasing the yeast fermentability of rice straw hemicellulose hydrolysate. Under these conditions, the higher removal of total phenols (above 40%), low molecular weight phenolic compounds (above 95%) and furans (above 52%) were observed. In addition, the ethanol volumetric productivity by P. stipitis was increased in approximately twice in relation the untreated hydrolysate. Conclusion These results demonstrate that AOPs are a promising methods to reduce toxicity and improve the fermentability of lignocellulosic hydrolysates.

Weizenstroh als erneuerbare Ressource zur Produktion von Biopolymeren und wichtigen Grundchemikalien stellt eine ökologisch sinnvolle Alternative dar. Durch die vom PFI durchgeführte Thermodruckhydrolyse konnte das Weizenstroh und die darin enthaltenen Zucker fast vollständig mobilisiert werden. Ein umfangreiches Screening nach Organismen, welche die Zucker des Weizenstrohs verwerten konnten, ergab, dass einige wenige Stämme zur PHB-Bildung aus Xylose befähigt waren (10 %). Zur PHB-Synthese aus Glucose waren indes ca. doppelt so viele Organismen in der Lage (20 %). Zwei der insgesamt 118 untersuchten Organismen zeigten besonders gute PHB-Bildung sowohl mit Xylose als auch mit Glucose als Substrat. Dabei handelte es sich um die hauseigenen Stämme Bacillus licheniformis KHC 3 und Bacillus megaterium KNaC 2. Nach Enttoxifizierung der hemicellulosischen Fraktion konnte diese als C-Quelle im Mineral Medium eingesetzt werden. Burkholderia sacchari DSM 17165 und Hydrogenophaga pseudoflava DSM 1034, sowie die hauseigenen Isolate Bacillus licheniformis KHC 3 und Bacillus megaterium KNaC 2 wurden für die Synthese von PHB aus der hemicellulosischen Fraktion verwendet. Die Zucker der hemicellulosischen Fraktion (Xylose, Glucose, Arabinose) konnten durch diese Organismen zur PHB-Synthese genutzt werden. Hierbei stellte sich heraus, dass die beiden Bacillus-Stämme besser zur Produktion von PHB aus dem hemicellulosischen Hydrolysat geeignet waren als die Stämme der DSMZ. Die alternative Umsetzung der im hemicellulosischem Hydrolysat enthaltenen Zucker (Xylose, Glucose und Arabinose) in die wichtigen Grundchemikalien Lactat und Acetat konnte durch die Verwendung von heterofermentativen Milchsäurebakterien verwirklicht werden. Die Bildung dieser wichtigen Grundchemikalien stellt eine interessante Alternative zur PHB-Synthese dar. Die Menge an teuren Zusätzen wie Tomatensaft, welcher für das Wachstum der MSB essentiell war, konnte reduziert werden. Die Glucose der zweiten Fraktion des Weizenstrohs, der cellulosischen Fraktion, konnte ebenfalls durch den Einsatz von Mikroorganismen in PHB umgewandelt werden. Kommerzielle Cellulasen der Firma Novozymes konnten große Mengen an Glucose (≥10 g/l) aus der cellulosischen Fraktion freisetzen. Diese freie Glucose wurde mit Hilfe von Cupriavidus necator DSM 545, Cupriavidus necator NCIMB 11599, Bacillus licheniformis KHC 3 und Bacillus megaterium KNaC 2 zu PHB fermentiert. Wie auch beim hemicellulosischen Hydrolysat konnten hier die beiden Bacillus-Stämme die besten Ergebnisse erzielen. Bei ihnen machte die PHB mehr als die Hälfte der Trockenmasse aus. Die Abtrennung des Zielprodukts ohne die Verwendung von umweltschädlichen Lösungsmitteln wurde durch die Lyse der Zielzellen durch eigens isolierte Enzyme aus Streptomyceten verwirklicht. Die Zelllyse durch die Enzyme aus Streptomyces globisporus subsp. caucasius DSM 40814 und Streptomyces albidoflavus DSM 40233 war erfolgreich und zeigte vor allem bei den Bacillen hohe Wirkung (83 % und 99 % Zelllyse). Bei dem Gram-negativen Organismus Cupriavidus necator DSM 428 konnte die anfangs niedrige Zelllyse von 38 % durch Ultraschallbehandlung auf ca. 75 % erhöht werden.

This Ph.D. research is comprised of three major components; (i) Characterization study to analyze the composition of defatted corn syrup (DCS) from a dry corn mill facility (ii) Hydrolysis experiments to optimize the production of fermentable sugars and amino acid platform using DCS and (iii) Sustainability analyses. Analyses of DCS included total solids, ash content, total protein, amino acids, inorganic elements, starch, total carbohydrates, lignin, organic acids, glycerol, and presence of functional groups. Total solids content was 37.4% (± 0.4%) by weight, and the mass balance closure was 101%. Total carbohydrates [27% (± 5%) wt.] comprised of starch (5.6%), soluble monomer carbohydrates (12%) and non-starch carbohydrates (10%). Hemicellulose components (structural and non-structural) were; xylan (6%), xylose (1%), mannan (1%), mannose (0.4%), arabinan (1%), arabinose (0.4%), galatactan (3%) and galactose (0.4%). Based on the measured physical and chemical components, bio-chemical conversion route and subsequent fermentation to value added products was identified as promising. DCS has potential to serve as an important fermentation feedstock for bio-based chemicals production. In the sugar hydrolysis experiments, reaction parameters such as acid concentration and retention time were analyzed to determine the optimal conditions to maximize monomer sugar yields while keeping the inhibitors at minimum. Total fermentable sugars produced can reach approximately 86% of theoretical yield when subjected to dilute acid pretreatment (DAP). DAP followed by subsequent enzymatic hydrolysis was most effective for 0 wt% acid hydrolysate samples and least efficient towards 1 and 2 wt% acid hydrolysate samples. The best hydrolysis scheme DCS from an industry's point of view is standalone 60 minutes dilute acid hydrolysis at 2 wt% acid concentration. The combined effect of hydrolysis reaction time, temperature and ratio of enzyme to substrate ratio to develop hydrolysis process that optimizes the production of amino acids in DCS were studied. Four key hydrolysis pathways were investigated for the production of amino acids using DCS. The first hydrolysis pathway is the amino acid analysis using DAP. The second pathway is DAP of DCS followed by protein hydrolysis using proteases [Trypsin, Pronase E (Streptomyces griseus) and Protex 6L]. The third hydrolysis pathway investigated a standalone experiment using proteases (Trypsin, Pronase E, Protex 6L, and Alcalase) on the DCS without any pretreatment. The final pathway investigated the use of Accellerase 1500® and Protex 6L to simultaneously produce fermentable sugars and amino acids over a 24 hour hydrolysis reaction time. The 3 key objectives of the techno-economic analysis component of this PhD research included; (i) Development of a process design for the production of both the sugar and amino acid platforms with DAP using DCS (ii) A preliminary cost analysis to estimate the initial capital cost and operating cost of this facility (iii) A greenhouse gas analysis to understand the environmental impact of this facility. Using Aspen Plus®, a conceptual process design has been constructed. Finally, both Aspen Plus Economic Analyzer® and Simapro® sofware were employed to conduct the cost analysis as well as the carbon footprint emissions of this process facility respectively. Another section of my PhD research work focused on the life cycle assessment (LCA) of commonly used dairy feeds in the U.S. Greenhouse gas (GHG) emissions analysis was conducted for cultivation, harvesting, and production of common dairy feeds used for the production of dairy milk in the U.S. The goal was to determine the carbon footprint [grams CO2 equivalents (gCO2e)/kg of dry feed] in the U.S. on a regional basis, identify key inputs, and make recommendations for emissions reduction. The final section of my Ph.D. research work was an LCA of a single dairy feed mill located in Michigan, USA. The primary goal was to conduct a preliminary assessment of dairy feed mill operations and ultimately determine the GHG emissions for 1 kilogram of milled dairy feed.

Proto-kerogens were isolated, by extraction and HF/HC1 treatment, from core samples of Holocene sediments of the Cariaco Trench, with interpolated ages of 900, 2850 and 6000 years, and examined via a combination of microscopic, spectroscopic and pyrolytic methods. It appears that these proto-kerogens were chiefly formed from phytoplanktonic components via the degradation-recondensation pathway. The natural sulfurisation pathway only afforded a minor contribution, in spite of the conditions prevailing in the water column and sediments that correspond to those generally considered as especially favourable for the formation of sulfurised organic matter. Proto-kerogen formation via sulfurisation, i.e. the endpoint of the continuum leading to insoluble high molecular weight structures cross-linked by sulfur and resistant to acid hydrolysis, is therefore a rather slow process under these conditions. However, the contribution of sulfurised moieties to the total proto-kerogen substantially increased with depth due to continuous sulfurisation in the time/depth interval, whereas formation through degradation-recondensation is almost complete for the 900 years old sample onwards. Proto-kerogen formation via carbohydrate sulfurisation is faster than lipid sulfurisation and only sulfurised carbohydrates were detected in the shallowest sample. In contrast, sulfurised lipids occur in the other two proto-kerogens. Moreover, their contribution relative to sulfurised carbohydrates increases with depth, probably due to the higher resistance of lipids to mineralisation compared to carbohydrates.

Intact amyloplasts from potato (Solanum tuberosum L.) were used to study starch biosynthesis and phosphorylation. Assessed by the degree of intactness and by the level of cytosolic and vacuolar contamination, the best preparations were selected by searching for amyloplasts containing small starch grains. The isolated, small amyloplasts were 80% intact and were free from cytosolic and vacuolar contamination. Biosynthetic studies of the amyloplasts showed that [1-14C]glucose-6-phosphate (Glc-6-P) was an efficient precursor for starch synthesis in a manner highly dependent on amyloplast integrity. Starch biosynthesis from [1-14C]Glc-1-P in small, intact amyloplasts was 5-fold lower and largely independent of amyloplast intactness. When [33P]Glc-6-P was administered to the amyloplasts, radiophosphorylated starch was produced. Isoamylase treatment of the starch followed by high-performance anion-exchange chromatography with pulsed amperometric detection revealed the separated phosphorylated α-glucans. Acid hydrolysis of the phosphorylated α-glucans and high-performance anion-exchange chromatography analyses showed that the incorporated phosphate was preferentially positioned at C-6 of the Glc moiety. The incorporation of radiolabel from Glc-1-P into starch in preparations of amyloplasts containing large grains was independent of intactness and most likely catalyzed by starch phosphorylase bound to naked starch grains.

Devido à preocupação com o meio ambiente e o volume crescente de resíduos plástico em aterros sanitários, os polímeros biodegradáveis estão sendo estudados extensivamente. Um deles é o PLA. Apesar de possuir propriedades comparáveis a polímeros commodities e polímeros de engenharia, ainda é necessário melhorar certas características do PLA, como resistência ao impacto. Para isso, a nanocelulose (NC) pode ser usada sem alterações significativas na biodegradação polimérica. Este estudo teve como objetivo obter a nanocelulose, caracteriza-la e incorpora-la ao poli(ácido láctico) (PLA), assim como, estudar as propriedades térmicas, morfológicas e mecânicas do compósito obtido. A NC foi obtida por hidrólise ácida utilizando ácido fosfórico e posteriormente foi silanizada com três silanos distintos. As nanopartículas foram caracterizadas por Birrefringência, Microscopia Eletrônica de Transmissão (MET), Termogravimetria (TG), Potencial Zeta, Espectroscopia Vibracional de Absorção no Infravermelho com Transformada de Fourier (FTIR) e Difração de Raio X (DRX). Com as imagens obtidas pelo MET foi possível medir o tamanho das partículas de NC. E então obter a razão de aspecto de 82 e o limite de percolação de 1,1% em massa, confirmando a morfologia de nanofibra. De acordo as analises TG\'s, a presença de NC silanizada aumentou o início da degradação térmica. Os compósitos, contendo 3% em massa de NC, foram obtidos por fusão em câmara de mistura e moldados por injeção. Os compósitos foram caracterizados por FTIR, Cromatografia de Permeação em Gel (GPC), TG, Calorimetria Exploratória Diferencial (DSC), Microscopia Eletrônica de Varredura (MEV-FEG), Impacto e Tração. As análises dos compósitos mostraram que a NC atuou como agente de nucleação, facilitando a cristalização do PLA, além de a NC ter atuado como reforço na matriz polimérica melhorando as propriedades mecânicas.

A celulose é o polímero natural renovável disponível em maior abundância atualmente. Por possuir estrutura semicristalina, é possível extrair seus domínios cristalinos através de procedimentos que ataquem sua fase amorfa, como a hidrólise ácida, obtendo-se assim partículas cristalinas chamadas nanopartículas de celulose (NCs). Estas nanopartículas têm atraído enorme interesse científico, uma vez que possuem propriedades mecânicas, como módulo de elasticidade e resistência à tração, semelhantes a várias cargas inorgânicas utilizadas na fabricação de compósitos. Além disso, possuem dimensões nanométricas, o que contribui para menor adição de carga à matriz polimérica, já que possuem maior área de superfície, quando comparadas às cargas micrométricas. Nanocompósitos formados pela adição destas cargas em matrizes poliméricas podem apresentar propriedades comerciais atraentes, como barreira a gases, melhores propriedades térmicas e baixa densidade, quando comparados aos compósitos tradicionais. Como se trata de uma carga com dimensões nanométricas, obtida de fontes renováveis, uma das principais áreas de interesse para aplicação deste reforço é em biopolímeros biodegradáveis. O poli(ácido lático) (PLA), é um exemplo de biopolímero com propriedades mecânicas, térmicas e de processamento superiores a de outros biopolímeros comerciais. No presente trabalho foram obtidas nanopartículas de celulose (NCs), por meio de hidrólise ácida, utilizando-se três métodos distintos, com o objetivo de estudar o método mais eficiente para a obtenção de NCs adequadas à aplicação em compósitos de PLA. Os Métodos I e II empregam extração das NCs por meio do H2SO4, diferenciando-se apenas pela neutralização, a qual envolve diálise ou neutralização com NaHCO3, respectivamente. No Método III a extração das NCs foi realizada com H3PO4. As NCs foram caracterizadas por diferentes técnicas, como difração de raios X (DRX), análise termogravimétrica (TG), espectroscopia vibracional de absorção no infravermelho (FTIR), microscopia eletrônica de transmissão (MET) e microscopia de força atômica (MFA). Os resultados de caracterização das NCs indicaram que, a partir de todos os métodos utilizados, há formação de nanocristais de celulose (NCCs), entretanto, apenas os NCCs obtidos pelos Métodos II e III apresentaram estabilidade térmica suficiente para serem empregados em compósitos preparados por adição da carga no polímero em estado fundido. A incorporação das NCs em matriz de PLA foi realizada em câmara de mistura, com posterior moldagem por prensagem a quente. Compósitos obtidos por adição de NCs obtidas pelo Método II foram caracterizados por calorimetria exploratória diferencial (DSC), análise termogravimétrica, microscopia óptica, análises reológicas e microscopia eletrônica de varredura (MEV). A adição de NCs, extraídas pelo Método II, em matriz de PLA afetou o processo de cristalização do polímero, o qual apresentou maior grau de cristalinidade. Além disso, a adição de 3% em massa de NCs no PLA foi suficiente para alterar seu comportamento reológico. Os resultados reológicos indicaram que a morfologia do compósito é, predominantemente, composta por uma dispersão homogênea e fina da carga na fase matriz. Micrografias obtidas por MEV corroboram os resultados reológicos, mostrando, predominantemente a presença de partículas de NC em escala nanométrica. Compósitos de PLA com NCs obtidas pelo Método III apresentaram aglomerados de partículas de NC em escala micro e milimétrica, ao longo da fase matriz, e não foram extensivamente caracterizados.

A chromatographic method was developed for the determination of tryptophan content in food and feed proteins. The method involves separation and quantitation of tryptophan (released from protein by alkaline hydrolysis with NaOH) by isocratic ion-exchange chromatography with O-phthalaldehyde derivatization followed by fluorescence detection. In this procedure, chromatographic separation of the tryptophan and alpha-methyl tryptophan, the internal standard, was complete in 15 min, without any interference from other compounds. The precision of the method was 1-4%, relative standard deviation. Accuracy was validated by agreement with the value for chicken egg white lysozyme, a sequenced protein, and by quantitative recoveries after spiking with lysozyme. The method allows determination in a range of feed proteins, containing varied concentrations of tryptophan, and is applicable to systems used for routine amino acid analysis by ion-exchange chromatography.
