A bovine male-specific marker was identified in our laboratory through random amplified polymorphic DNA (RAPD) analysis. This fragment of 3216 bp was cloned, sequenced and mapped by fluorescent in situ hybridization (FISH) on the taurine Yq. Primers derived from this sequence were initially screened by polymerase chain reaction (PCR) for their ability to detect Y-specific segments in zebu and taurine genomic DNA. Two of these primers amplified a 655 bp Y-specific sequence present in taurine and zebu male genomic DNA. These primers were then used for detecting the 655 bp male sequence in DNA from 173 zebu and 30 taurine embryos, which had been previously sexed using primers for the sequence BC 1.2. From a 0.72-kb fragment universally generated in Paracoccidioides brasiliensis strains, primers were designed and tested on genomic DNA of this and other pathogenic fungi. They were specific and highly sensitive for P. brasiliensis DNA. Positive results were obtained when these were tested in clinical samples.

Background: The increasing number of genomic sequences of bacteria makes it possible to select unique SNPs of a particular strain/species at the whole genome level and thus design specific primers based on the SNPs. The high similarity of genomic sequences among phylogenetically-related bacteria requires the identification of the few loci in the genome that can serve as unique markers for strain differentiation. PrimerSNP attempts to identify reliable strain-specific markers, on which specific primers are designed for pathogen detection purpose.Results: PrimerSNP is an online tool to design primers based on strain specific SNPs for multiple strains/species of microorganisms at the whole genome level. The allele-specific primers could distinguish query sequences of one strain from other homologous sequences by standard PCR reaction. Additionally, PrimerSNP provides a feature for designing common primers that can amplify all the homologous sequences of multiple strains/species of microorganisms. PrimerSNP is freely available at http://cropdisease.ars.usda.gov/similar to primer.Conclusion: PrimerSNP is a high-throughput specific primer generation tool for the differentiation of phylogenetically-related strains/species. Experimental validation showed that this software had a successful prediction rate of 80.4 - 100% for strain specific primer design.

Randomly amplified polymorphic DNA (RAPD) analysis of 35 Paracoccidioides brasiliensis isolates was carried out to evaluate the correlation of RAPD profiles with the virulence degree or the type of the clinical manifestations of human paracoccidioidomycosis. The dendrogram presented two main groups sharing 64% genetic similarity. Group A included two isolates from patients with chronic paracoccidioidomycosis; group B comprised the following isolates showing 65% similarity: two non-virulent, six attenuated, five virulent, eight from patients with chronic paracoccidioidomycosis and two from patients with acute paracoccidioidomycosis. The virulent Pb18 isolate and six attenuated or non-virulent samples derived from it were genetically indistinguishable (100% of similarity). Thus, in our study, RAPD patterns could not discriminate among 35 P. brasiliensis isolates according to their differences either in the degree of virulence or in the type of the clinical manifestation of this fungal infection. © 2002 Federation of European Microbiological Societies. Path formulation can be used to classify and structure efficiently multiparameter bifurcation problems around fundamental singularities: the cores. The non-degenerate umbilic singularities are the generic cores for four situations in corank 2: the general or gradient problems and the ℤ 2-equivariant (general or gradient) problems. Those categories determine an interesting 'Russian doll' type of structure in the universal unfoldings of the umbilic singularities. One advantage of our approach is that we can handle one, two or more parameters using the same framework (even considering some special parameter structure, for instance, some internal hierarchy). We classify the generic bifurcations that occur in those cases with one or two parameters.

Since the use of air abrasion has grown in pediatric dentistry, the aim of this study was to evaluate, by means of shear bond strength testing, the need to use the total etching technique or self-etching primers on dentin of primary teeth after air abrasion. Twenty-five exfoliated primary molars had their occlusal dentin exposed by trimming and polishing. Specimens were treated by: Air abrasion + Scotchbond MultiPurpose adhesive (G1); 37% phosphoric acid + Scotchbond MP adhesive (G2); Clearfil SE (G3); Air abrasion ( 37% phosphoric acid + Scotchbond MP adhesive (G4); Air abrasion + Clearfil SE (G5). On the treated surface, a cylinder of 2 mm by 6 mm was made using a composite resin (Z100). Duncan's test showed that: (G2 = G3 = G5) > (G1 = G4). The use of a selfetching primer on air abraded dentin is recommended to obtain higher bond strengths.

We use a time-dependent dynamical mean-field-hydrodynamic model to study mixing-demixing in a degenerate fermion-fermion mixture (DFFM). It is demonstrated that with the increase of interspecies repulsion and/or trapping frequencies, a mixed state of a DFFM could turn into a fully demixed state in both three-dimensional spherically symmetric as well as quasi-one-dimensional configurations. Such a demixed state of a DFFM could be experimentally realized by varying an external magnetic field near a fermion-fermion Feshbach resonance, which will result in an increase of interspecies fermion-fermion repulsion, and/or by increasing the external trap frequencies. © 2006 The American Physical Society.

In this paper we study the local codimension one, two and three Hopf bifurcations which occur in the classical Chua's differential equations with cubic nonlinearity. A detailed analytical description of the regions in the parameter space for which multiple small periodic solutions bifurcate from the equilibria of the system is obtained. As a consequence, a complete answer for the challenge proposed in [Moiola & Chua, 1999] is provided. © 2009 World Scientific Publishing Company.

The objective of this study was to evaluate the influence of different primers on the microtensile bond strength (μT BS) between a feldspathic ceramic and two composites. Forty blocks (6.0 × 6.0 × 5.0 mm 3) were prepared from Vita Mark II . After polishing, they were randomly divided into 10 groups according to the surface treatment: Group 1, hydrofluoric acid 10% (HF) + silane; Group 2, CoJet + silane; Group 3, HF + Metal/Zirconia Primer; Group 4, HF + Clearfil Primer; Group 5, HF + Alloy Primer; Group 6, HF + V-Primer; Group 7, Metal/Zirconia Primer; Group 8, Clearfil Primer; Group 9, Alloy Primer; Group 10, V-Primer. After each surface treatment, an adhesive was applied and one of two composite resins was incrementally built up. The sticks obtained from each block (bonded area: 1.0 mm2 ± 0.2 mm) were stored in distilled water at 37°C for 30 days and submitted to thermocycling (7,000 cycles; 5°C/55°C ± 1°C). The μT BS test was carried out using a universal testing machine (1.0 mm/min). Data were analyzed using ANOVA and a Tukey test (α = 0.05). The surface treatments significantly affected the results (P < 0.05); no difference was observed between the composites (P > 0.05). The bond strength means (MPa) were as follows: Group 1a = 29.6; Group 1b = 33.7; Group 2a = 28.9; Group 2b = 27.1; Group 3a = 13.8; Group 3b = 14.9; Group 4a = 18.6; Group 4b = 19.4; Group 5a = 15.3; Group 5b = 16.5; Group 6a = 11; Group 6b = 18; Groups 7a to 10b = 0. While the use of primers alone was not sufficient for adequate bond strengths to feldspathic ceramic, HF etching followed by any silane delivered higher bond strength.

Trapped degenerate dipolar Bose and Fermi gases of the cylindrical symmetry with the polarization vector along the symmetry axis are only stable for the strength of dipolar interaction below a critical value. In the case of bosons, the stability of such a dipolar Bose-Einstein condensate (BEC) is investigated for different strengths of contact and dipolar interactions using a variational approximation and a numerical solution of a mean-field model. In the disc shape, with the polarization vector perpendicular to the plane of the disc, the atoms experience an overall dipolar repulsion and this fact should contribute to the stability. However, a complete numerical solution of the dynamics leads to the collapse of a strongly disc-shaped dipolar BEC due to the long-range anisotropic dipolar interaction. In the case of fermions, the stability of a trapped single-component degenerate dipolar Fermi gas is studied including the Hartree-Fock exchange and Brueckner-Goldstone correlation energies in the local-density approximation valid for a large number of atoms. Estimates for the maximum allowed number of polar Bose and Fermi molecules in the BEC and degenerate Fermi gas are given. © 2013 IOP Publishing Ltd.

Objectives: To investigate the adhesive potential of novel zirconia primers and universal adhesives to surface-treated zirconia substrates.Methods: Zirconia bars were manufactured (3.0 mm x 3.0 mm x 9.0 mm) and treated as follows: no treatment (C); air abrasion with 35 mu m alumina particles (S); air abrasion with 30 mu m silica particles using one of two systems (Rocatec or SilJet) and; glazing (G). Groups C and S were subsequentially treated with one of the following primers or adhesives: ZP (Z-Prime Plus), AZ (AZ Primer); MP (Monobond Plus); SU (ScotchBond Universal) and; EA (an Experimental Adhesive). Groups Rocatec and SilJet were silanized prior to cementation. Samples form group G were further etched and silanized. Bars were cemented (Multilink) onto bars of a silicate-based ceramic (3.0 mm x 3.0 mm x 9.0 mm) at 90 degrees angle, thermocycled (2.500 cycles, 5-55 degrees C, 30 s dwell time), and tested in tensile strength test. Failure analysis was performed on fractured specimens to measure the bonding area and crack origin.Results: Specimens from group C did not survive thermocycling, while CMP, CSU and CEA groups survived thermocycling but rendered low values of bond strength. All primers presented a better bond performance after air abrasion with Al2O3 particles. SilJet was similar to Rocatec, both presenting the best bond strength results, along with SMP, SSU and CEA. G promoted intermediate bond strength values. Failure mode was predominately adhesive on zirconia surface combined to cohesive of the luting agent.Conclusions: Universal adhesives (MP, SU, EA) may be a considerable alternative for bonding to zirconia, but air abrasion is still previously required. Air abrasion with silica particles followed by silane application also presented high bond strength values. (C) 2013 Elsevier Ltd. All rights reserved.
