Activation of the human complement system of plasma proteins during immunological host defense can result in overproduction of potent proinflammatory peptides such as the anaphylatoxin C5a. Excessive levels of C5a are associated with numerous immunoinflammatory diseases, but there is as yet no clinically available antagonist to regulate the effects of C5a. We now describe a series of small molecules derived from the C-terminus of C5a, some of which are the most potent low-molecular-weight C5a receptor antagonists reported to date for the human polymorphonuclear leukocyte (PMN) C5a receptor. H-1 NMR spectroscopy was used to determine solution structures for two cyclic antagonists and to indicate that antagonism is related to a turn conformation, which can be stabilized in cyclic molecules that are preorganized for receptor binding. While several cyclic derivatives were of similar antagonistic potency, the most potent antagonist was a hexapeptide-derived macrocycle AcF[OPdChaWR] with an IC50 = 20 nM against a maximal concentration of C5a (100 nM) on intact human PMNs. Such potent C5a antagonists may be useful probes to investigate the role of C5a in host defenses and to develop therapeutic agents for the treatment of many currently intractable inflammatory conditions.

Life histories are generally assumed to evolve via antagonistic pleiotropy (negative genetic correlations) among traits, and trade-offs between life-history traits are typically studied using either phenotypic manipulations or selection experiments. We investigated the trade-off between egg size and fecundity in Drosophila melanogaster by examining both the phenotypic and genetic relationships between these traits after artificial selection for large and small eggs, relative to female body size. Egg size responded strongly to selection in both directions, increasing in the large-egg selected lines and decreasing in the small-egg selected lines. Phenotypic correlations between egg size and fecundity in the large-egg selected lines were negative, but no relationship between these traits occurred in either the control or small-egg selected lines. There was no negative genetic correlation between egg size and fecundity. Total reproductive allocation decreased in the small-egg selected lines but did not increase in the large-egg lines. Our results have three implications. First, our selection procedure may have forced females selected for large eggs into a physiological trade-off not reflected in a negative genetic correlation between these traits. Second, the lack of a negative genetic correlation between egg size and number suggests that the phenotypic trade-off frequently observed between egg size and number in other organisms may not evolve over the short term via a direct genetic trade-off whereby increases in egg size are automatically accompanied by decreased fecundity. Finally, total reproductive allocation may not evolve independently of egg size as commonly assumed.

Disease resistance is associated with a plant defense response that involves an integrated set of signal transduction pathways. Changes in the expression patterns of 2.375 selected genes were examined simultaneously by cDNA microarray analysis in Arabidopsis thaliana after inoculation with an incompatible fungal pathogen Alternaria brassicicola or treatment with the defense-related signaling molecules salicylic acid (SA), methyl jasmonate (MJ), or ethylene, Substantial changes (up- and down-regulation) in the steady-state abundance of 705 mRNAs were observed in response to one or more of the treatments, including known and putative defense-related genes and 106 genes with no previously described function or homology, In leaf tissue inoculated with A. brassicicola, the abundance of 168 mRNAs was increased more than 2.5-fold, whereas that of 39 mRNAs was reduced. Similarly, the abundance of 192, 221, and 55 mRNAs was highly (>2.5-fold) increased after treatment with SA, MJ, and ethylene, respectively. Data analysis revealed a surprising level of coordinated defense responses, including 169 mRNAs regulated by multiple treatments/defense pathways. The largest number of genes coinduced (one of four induced genes) and corepressed was found after treatments with SA and MJ. In addition, 50% of the genes induced by ethylene treatment were also induced by MJ treatment. These results indicated the existence of a substantial network of regulatory interactions and coordination occurring during plant defense among the different defense signaling pathways, notably between the salicylate and jasmonate pathways that were previously thought to act in an antagonistic fashion.

The objective of he article is to research the dynamic capacities developed and used by WEG in its internationalization process and to explain how these capacities help the company defends and supports competitive advantage. The article presents an exploratory study of the internationalization process of WEG in Argentina and China. This article has as analysis approach the dynamic capacities, contributes to the literature of international management in two aspects. First, it adds the analytical look of the internationalization based on dynamic capacities that are still well restricted. Second, when working the dynamic capacities as central element of the analysis of the internationalization process, it Proposes one framework of integrative analysis of the economic and behavioral theories that are used to explain the process of companies`-internationalization, although they are dealt independently and sometimes antagonistic way. The result shows as the dynamic capacities are articulated in the base of WEG in its process of internationalization for Argentina and the subsequent movement for China. The developed dynamic capacities in Argentina were acquired for the Brazilian headquarter and could have been applied in the process of internationalization for China. However, a more complex organizational structure cannot be identified where the inter-subsidiary relationships could share dynamic capacities as proposed in framework.

The insulin/insulin-like signaling (IIS) pathway is an evolutionarily conserved module in the control of body size and correlated organ growth in metazoans. In the highly eusocial bees, the caste phenotypes differ not only in size and several structural features but also in individual fitness and life history. We investigated the developmental expression profiles of genes encoding the two insulin-like peptides (AmILP-1 and AmILP-2) and the two insulin receptors (AmInR-1 and AmInR-2) predicted in the honey bee genome. Quantitative PCR analysis for queen and worker larvae in critical stages of caste development showed that AmILP-2 is the predominantly transcribed ILP in both castes, with higher expression in workers than in queens. Expression of both InR genes sharply declined in fourth instar queen larvae, but showed little modulation in workers. On first sight, these findings are non-intuitive, considering the higher growth rates of queens, but they can be interpreted as possibly antagonistic crosstalk between the IIS module and juvenile hormone. Analyzing AmInR-1 and AmInR-2 expression in ovaries of queen and worker larvae revealed low transcript levels in queens and a sharp drop in AmInR-2 expression in fifth instar worker larvae, indicating relative independence in tissue-specific versus overall IIS pathway activity. We analyzed the effect of (+)alpha-tocopheryl succinate (alpha-TOS) alone or associated with arsenic trioxide (ATO) or all-trans retinoid acid (ATRA) in acute promyelocytic leukemia (APL). alpha-TOS-induced apoptosis in APL clinical samples and in ATRA-sensitive (NB4) and ATRA-resistant (NB4-R2) APL cell lines. The effective dose 50% (ED-50) was calculated to be 71 and 58 mu M, for NB4 and NB4-R2, respectively. a-TOS neither induced nor modified ATRA-induced differentiation of APL cells, and did not affect the proliferation and differentiation of normal CD34(+) hematopoietic progenitors in methylcellulose assays. alpha-TOS exerted a moderate antagonistic effect to ATO-induced apoptosis when treatment was done simultaneously but when alpha-TOS was added 24 h after ATO, an additive effect was observed. Our results support the concept of alpha-TOS as an anti-leukemic compound which spares normal hematopoiesis. (C) 2008 Elsevier Ltd. All rights reserved.

In temporal lobe epilepsy (TLE) seizures, tonic or clonic motor behaviors (TCB) are commonly associated with automatisms, versions, and vocalizations, and frequently occur during secondary generalization. Dystonias are a common finding and appear to be associated with automatisms and head deviation, but have never been directly linked to generalized tonic or clonic behaviors. The objective of the present study was to assess whether dystonias and TCB are coupled in the same seizure or are associated in an antagonistic and exclusive pattern. Ninety-one seizures in 55 patients with TLE due to mesial temporal sclerosis were analyzed. Only patients with postsurgical seizure outcome of Engel class I or II were included. Presence or absence of dystonia and secondary generalization was recorded. Occurrence of dystonia and occurrence of bilateral tonic or clonic behaviors were negatively correlated. Dystonia and TCB may be implicated in exclusive, non-coincidental, or even antagonistic effects or phenomena in TLE seizures. A neural network related to the expression of one behavioral response (e.g., basal ganglia activation and dystonia) might theoretically ""displace"" brain activation or disrupt the synchronism of another network implicated in pathological circuit reverberation and seizure expression. The involvement of basal ganglia in the blockade of convulsive seizures has long been observed in animal models. The question is: Do dystonia and underlying basal ganglia activation represent an attempt of the brain to block imminent secondary generalization? (C) 2007 Elsevier Inc. All rights reserved.

Saprophytic bacteria, yeasts and filamentous fungi were isolated from Geraldton waxflower flowers and screened to identify potential antagonism towards Botrytis cinerea. Isolates from other sources (e.g. avocado) were also tested. Isolates were initially screened in vitro for inhibition of B. cinerea conidial germination, germ tube elongation and mycelial growth. The most antagonistic bacteria, yeasts and fungi were selected for further testing on detached waxflower flowers. Conidia of the pathogen were mixed with conidia or cells of the selected antagonists, co-inoculated onto waxflower flowers, and the flowers were sealed in glass jars and incubated at 20 degreesC. The number of days required for the pathogen to cause flower abscission was determined. The most antagonistic bacterial isolate, Pseudomonas sp. 677, significantly reduced conidial germination and retarded germ tube elongation of B. cinerea. None of the yeast or fungal isolates tested was found to significantly reduce conidial germination or retard germ tube elongation, but several significantly inhibited growth of B. cinerea. Fusarium sp., Epicoccum sp. and Trichoderma spp. were the most antagonistic of these isolates. Of the isolates tested on waxflower, Pseudomonas sp. 677 was highly antagonistic towards B. cinerea and delayed waxflower abscission by about 3 days. Trichoderma harzianum also significantly delayed flower abscission. However, as with most of the fungal antagonists used, inoculation of waxflower flowers with this isolate resulted in unsightly mycelial growth.

The beta -amino acid, taurine, is a full agonist of the human glycine receptor al subunit when recombinantly expressed in a mammalian (HEK293) cell line, but a partial agonist of the same receptor when expressed in Xenopus oocytes. Several residues in the Ala101-Thr112 domain have previously been identified as determinants of beta -amino acid binding and gating mechanisms in Xenopus oocyte-expressed receptors. The present study used the substituted cysteine accessibility method to investigate the role of this domain in controlling taurine-specific binding and gating mechanisms of glycine receptors recombinantly expressed in mammalian cells. Asn102 and Glu103 are identified as taurine and glycine binding sites, whereas Ala101 is eliminated as a possible binding site. The N102C mutation also abolished the antagonistic actions of taurine, indicating that this site does not discriminate between the putative agonist- and antagonist-bound conformations of beta -amino acids. The effects of mutations from Lys104-Thr112 indicate that the mechanism by which this domain controls beta -amino acid-specific binding and gating processes differs substantially depending on whether the receptor is expressed in mammalian cells or Xenopus oocytes. Thr112 is the only domain element in mammalian cell-expressed GlyRs which was demonstrated to discriminate between glycine and taurine.

Selected isolates of Cladosporium tenuissimum were tested for their ability to inhibit in vitro aeciospore germination of the two-needle pine stem rusts Cronartium flaccidum and Peridermium pini and to suppress disease development in planta. The antagonistic fungus displayed a number of disease-suppressive mechanisms. Aeciospore germination on water agar slides was reduced at 12, 18, and 24 h when a conidial suspension (1.5 x 10(7) conidia per ml) of the Cladosporium tenuissimum isolates was added. When the aeciospores were incubated in same-strength conidial suspensions for 1, 11, 21, and 31 days, viability was reduced at 20 and 4 degreesC. Light and scanning electron microscopy showed that rust spores were directly parasitized by Cladosporium tenuissimum and that the antagonist had evolved several strategies to breach the spore wail and gain access to the underlying tissues. Penetration occurred with or without appressoria. The hyperparasite exerted a mechanical force to destroy the spore structures (spinules, cell wall) by direct contact, penetrated the aeciospores and subsequently proliferated within them. However, an enzymatic action could also be involved. This was shown by the dissolution of the host tell wall that comes in contact with the mycelium of the mycoparasite, by the lack of indentation in the host wall at the contact site, and by the minimal swelling at the infecting hyphal tip. Culture filtrates of the hyperparasite inhibited germination of rust propagules. A compound purified from the filtrates was characterized by chemical and spectroscopic analysis as cladosporol, a known beta -1,3-glucan biosynthesis inhibitor. Conidia of Cladosporium tenuissimum reduced rust development on new infected pine seedlings over 2 years under greenhouse conditions. Because the fungus is an aggressive mycoparasite, produces fungicidal metabolites, and can survive and multiply in forest ecosystems without rusts, it seems a promising agent for the biological control of pine stem rusts in Europe.

Em um contexto de ampliação dos lugares públicos participativos no Brasil há de se considerar expectativas de despertar valores sociopolíticos nos estudantes universitários em seu processo de qualificação cidadã e profissional, diante das críticas à formação dos administradores. Portanto, este trabalho visa compreender a dinâmica da consciência política dos estudantes da graduação em administração de uma universidade pública federal no sudeste do Brasil em sua relação com a participação cidadã nos lugares públicos participativos no estado e municípios. Adota-se o modelo analítico de consciência política para a compreensão da participação em ações coletivas de Sandoval (2001) como marco teórico, associado à literatura sobre participação cidadã. Trata-se de uma pesquisa qualitativa, cujos dados foram coletados através de documentos, aplicação de 30 questionários e 17 entrevistas semiestruturadas, com 30 estudantes universitários da graduação em administração matriculados em 2014/1. Os dados foram submetidos à análise de conteúdo (BARDIN, 2004). Os resultados revelam 12 estudantes que não participam nos lugares públicos participativos e 18 estudantes que participam em pelo menos um destes lugares. O interesse em exercer a cidadania, melhorar as políticas públicas, gostar de implicar-se com os assuntos públicos e defender seus interesses em circunstâncias de conflito são as justificativas citadas pelos que participam. Evidenciam-se nos estudantes com participação mais ativa, crenças, valores e expectativas societais, articuladas à eficácia política, identidade coletiva, interesses antagônicos, sentimentos de justiça e injustiça, favorecendo a vontade de agir coletivamente, devido à percepção de conexão de seus interesses com as metas e ações coletivas dos movimentos que se envolvem. Os estudantes que não participam desconfiam dos lugares públicos participativos e demonstram desinteresse pelos assuntos públicos, embora apontem um desconforto em não participar. Suas crenças, valores e expectativas societais, associadas aos sentimentos de ineficácia política dificultam o desenvolvimento da consciência política. Conclui-se que estes estudantes possuem uma consciência política de senso comum, demonstrando valores sociais e políticos inerentes aos modismos presentes na vida cotidiana das pessoas. Já os estudantes com participação mais ativa apresentam uma consciência política de conflito, motivando-os à participação nos lugares avaliados como eficazes às suas proposições. Entretanto, o Centro Acadêmico Livre de Administração Honestino Guimarães (CALAD), principal lugar de representação e participação dos interesses dos estudantes no curso, encontra-se sem direção e participação nas instâncias institucionalizadas na universidade.

A Escherichia coli de aderência difusa (DAEC), um patotipo diarreiogênico de E. coli, corresponde a um grupo heterogêneo sem marcador de virulência comum a todos os isolados e de papel controverso na diarreia infantil. O objetivo deste estudo foi caracterizar genotipica e fenotipicamente amostras de DAEC, portadoras e não portadoras de adesinas Afa/Dr, isoladas de crianças com e sem diarreia. Em 70 amostras de DAEC, PCR foi realizado para pesquisa de genes descritos em DAEC, EAEC ou UPEC, que codificam: (i) oito adesinas fimbriais e afimbriais (fimH, papC, sfa, aggA, aafA, agg3A, aidA/aah, afaC); (ii) cinco toxinas (pet, astA, set1A, sat, hlyA); (iii) três proteínas captadoras/receptora de ferro (irp2, iucA, chuA/shuA); (iv) invasina (daaD) e; antígeno 43 (agn43). Ensaio de formação de biofilme foi realizado a partir da bactéria cultivada em caldo Luria-Bertani e inoculada em placas de poliestireno com DMEM suplementado com 0,4% glicose. A leitura da densidade ótica (DO490) foi realizada após coloração com safranina. Soroaglutinação para 23 antígenos O (Probac do Brasil) foi realizada em 50% das DAEC. Método de difusão de disco foi realizado para testar a suscetibilidade a 13 antimicrobianos. A presença de pelo menos um gene que codifica adesinas, toxinas, proteínas captadoras/receptora de ferro, invasina ou antígeno 43 foram encontrados em 58,6%, 51,4%, 80%, 48,6% e 57,1%, respectivamente, com os genes fimH, irp2, agn43, iucA, chuA/shuA, presentes em mais de 50% das amostras. Gene afaC+ (PCR) e/ou sonda afaBC+ (hibridização de colônias) classificou 50% das DAEC como Afa/Dr, sendo pet, sat, irp2, iucA, chuA/shuA e agn43 significantes nessas amostras (p<0,05). Do total das DAEC, 44,3% foram formadoras de biofilme, igualmente distribuídas entre as Afa/Dr e não Afa/Dr, e nenhum gene foi associado com esse fenótipo. Sorologia de 35 amostras evidenciou os seguintes sorogrupos: 1 O29, 2 O125, 2 O127 e 7 O86. Todas as O86 foram de DAEC Afa/Dr. Maiores frequências de resistência antimicrobiana foram encontradas para ampicilina (55,7%), sulfametoxazol/trimetoprim (35,7%) e tetraciclina (28,6%) e o perfil resistente/intermediário para amoxicilina/ácido clavulânico, ampicilina, sulfametoxazol/trimetoprim foi significante nas DAEC Afa/Dr, assim como a multi-droga resistência (p<0,05). Em conclusão, observou-se: (i) alta frequência de fimH e pet e presença de agn43, até então não descrito em DAEC, em frequências similares àquelas encontradas em EAEC, UPEC e EAEC/UPEC, respectivamente; (ii) que as amostras de DAEC Afa/Dr e não Afa/Dr constituíram grupos com perfis genéticos diferenciados entre si; (iii) poucos sorogrupos foram encontrados entre as DAEC; (iv) frequências de resistência menores quando comparado com as poucas descrições em DAEC, sugerindo uma menor pressão seletiva da população do presente estudo e; (v) amostras de DAEC Afa/Dr podem representar um importante reservatório de genes de resistência a antimicrobianos, além de diversos fatores de virulência.

OBJETIVO: O objetivo deste estudo é avaliar a microbiologia dos abscessos periamigdalianos. MATERIAIS E MÉTODOS: Trinta pacientes com diagnóstico de abscesso periamigdaliano, idade média de 24,2 anos, foram submetidos à punção na região periamigdaliana de maior abaulamento com aspiração do material purulento (volume maior que 3mL). O material foi separado para realização das culturas aeróbicas e anaeróbicas. RESULTADOS: Houve um índice de positividade das culturas de 86,7%. Em 23,3% aspirados houve crescimento apenas de bactérias aeróbicas ou facultativas, 3,3% apenas de bactérias anaeróbicas e por fim em 60% aspirados houve crescimento de bactérias aeróbicas e anaeróbicas. Um total de 69 bactérias foi isolado (34 aeróbios e 35 anaeróbios). Os aeróbios predominantes foram Streptococcus sp, sendo o Streptococcus pyogenes em 23% dos casos. Os anaeróbios predominantes foram Prevotella sp e Peptostreptococcus sp. Pacientes receberam antibiótico prévio em 63% dos casos. Neste grupo foram isolados 1,8 bactérias por aspirado, menor número que nos paciente que não utilizaram antibiótico (3,0 bactérias por aspirado). Não houve diferença significativa no tipo de bactéria isolada nestes dois grupos. CONCLUSÃO: Os abscessos periamigdalianos apresentam na maioria dos casos infecções polimicrobianas, sendo os organismos anaeróbicos agentes importantes. O número de agentes isolados é maior nos pacientes que não utilizaram antibioticoterapia prévia, mas o uso de antimicrobiano não interferiu no tipo de bactéria isolada.

Vários materiais são propostos para reconstrução nasal, não havendo consenso sobre qual o melhor. A manta de celulose produzida por bactéria pode ser mais um elemento para adição cartilaginosa. Não há estudos deste material no dorso nasal. OBJETIVO: Avaliar a resposta tecidual à presença da celulose bacteriana no dorso nasal de coelhos. MATERIAL E MÉTODO: Foram utilizados 22 coelhos Nova Zelândia, sendo que em 20 deles foi implantada a manta de celulose no dorso nasal e em 2 controles nada foi feito. Foram acompanhados por um período de três e seis meses, sendo então retirados as regiões do dorso nasal e narinas dos coelhos e realizado estudo histopatológico levando em consideração parâmetros definidos de condição inflamatória como congestão vascular, intensidade do processo inflamatório e presença de exsudato purulento. RESULTADOS: O processo inflamatório manteve-se estável, demonstrando sua relação com o procedimento cirúrgico, e não com a presença da manta de celulose. Nos demais parâmetros estudados não houve diferença estatisticamente significante. CONCLUSÃO: A manta de celulose de Acetobacter xylinum mostrou boa biocompatibilidade e manteve-se estável no decorrer do tempo de estudo, podendo ser considerada um bom material para uso na elevação do dorso nasal.

A associação de bactérias a plantas tem sido estudada como uma possível tecnologia emergente, para fitorremediação de contaminantes, entre eles os herbicidas, que, por sua recalcitrância, ameaçam a qualidade do ambiente. O objetivo deste trabalho foi verificar a tolerância de mucuna-anã (Stizolobium deeringianum Bort) e mucuna-preta (Stizolobium aterrimum Piper & Tracy), inoculadas e não inoculadas com Rhizobium sp., ao herbicida atrazina. Os tratamentos foram: plantas com inoculante + 0,1 g/m², 0,2 g/m² atrazina e sem atrazina (T1, T2 e T3, respectivamente), sem inoculante + 0,1 g/m², 0,2 g/m² atrazina e sem atrazina (T4, T5 e T6, respectivamente). O delineamento experimental utilizado foi inteiramente casualizado, com três repetições. Foram avaliados germinação, sobrevivência, número de nódulos, altura, biomassa verde, biomassa seca da parte aérea, após o crescimento das plantas por 50 dias em casa de vegetação. Nos tratamentos com inoculante, avaliou-se a porcentagem de germinação de plantas bioindicadoras (Bidens pilosa L.). Mucuna-preta e mucuna-anã demonstraram maior tolerância ao herbicida quando associadas ao Rhizobium. Os valores de sobrevivência de mucuna-preta, nas doses 0,1 e 0,2 g/m² de atrazina (T1 e T2), foram de 34 a 24% superiores aos observados nas mesmas doses, mas sem o inoculante (T4 e T5). Para mucuna-anã, T1 e T2 foram de 17 e 8% superiores a T4 e T5, respectivamente. As alturas médias de mucuna-anã em T1, T2 e T3 foram mais elevadas que em T4, T5 e T6, reforçando a importância do simbionte à resistência ao herbicida. Os resultados encontrados para as variáveis altura, biomassa verde e seca para mucuna-preta não apresentaram diferença estatística entre os tratamentos com e sem inoculante, mostrando uma resistência natural à atrazina e a possibilidade de atuar como planta remediadora. A germinação de B. pilosa indica uma possível degradação da atrazina no solo com ambas as espécies de mucunas inoculadas com Rhizobium sp.
