Nephrolepis exaltata L. Schott "Bostoniensis" family Davalliaceae and Equisetum giganteum L. family Equisetaceae, Phylum Pteridophyta, exhibit a strong mechanism of dominance in the areas in which they live. Have secondary compounds with allelopathic activity. The objective of this article was evaluate allelopathic potential of two ferns species, using bioassay applying aqueous extracts of dried fronds, in cucumber and lettuce seeds, and observing germination and initial development. To observe the influence on germination was analyze the percentage of germinated seeds and germination speed index (GSI). To observe initial development was analyzed shoot and root growth of the seedlings. The bioassays revealed that no concentration significantly inhibited the germination, but germination speed was delayed gradually in two species tested, as increased the extract concentration. In initial development, all the extracts showed a tendency to inhibit the growth, and an increase in extract concentration decreasing growth of radicle and hypocotyl axis. We conclude that the aqueous extract has inhibitory activity more pronounced in early development than in seed germination, affecting the primary structures of the tested plants, corroborating with the observations of occurrences of the species in natural places where dominate and suppress the growth of other species.

Child sexual abuse continues to be a prevalent and complex problem in today’s society as it poses serious and pervasive mental health risks to child victims and their non-offending parents. The main objectives of this study were (a) to elucidate the psychological symptoms and support needs of parents of child sexual abuse victims as they present to group treatment, (b) to examine changes in psychological symptoms and support needs and their relationship with child functioning over the course of a parallel group treatment, and (c) to examine the impact of these factors on completion of group treatment. Participants included 104 sexually abused youth and their non-offending parent presenting to Project SAFE Group Intervention, a 12-session cognitive-behavioral group treatment for sexually abused children and their non-offending parents. This project had a unique advantage of utilizing a variety of demographic, parent-, and child-report measures, allowing for a more comprehensive examination of change in symptomatology and needs over the course of treatment. Several significant findings were noted, including the identification of four clusters of youth at pre-treatment, which were maintained at post-treatment; elevations on the CTQ Sexual Abuse scale; parents of youth sexually abused by a non-family member had significantly higher PSI-Restriction of Role subscale scores; parental expectations of a negative impact on their child were worse for older children; several parent characteristics predicted client treatment retention (e.g., older parents, lower SCL-90-R GSI scores); and an early age of onset of abuse also increased treatment retention. Future directions, recommendations, and limitations were discussed.

Patterns of population dynamics of Loligo plei and Loligo sanpaidensis in Southeastern Brazil were investigated with samples obtained from commercial catches and research cruises from 1999 to 2000 and from 2002 to 2003. Size and maturity Structure of the two species varied according to depth and season. Body size decreased with depth for L plei, whereas for L. sanpaidensis size increased with depth LIP to 100 m and decreased again in deeper areas. GSI and incidence of mature animals decreased with depth in both species. L. plei females matured at a larger size in summer, while size at maturity in L. sanpaulensis was greater in autumn. For L. plei, reproductive events Occurred in the late winter and spring, in depths up to 40 m, and during summer in inshore waters. L. sanpaidensis increased reproductive activity ill Summer, winter and spring between depths of 30 and 80 Ill. The presence of high proportions of immature Squid offshore Suggests that juveniles might develop in these areas and, upon maturation, migrate back to inshore waters to spawn, particularly in spring and Summer. However, differences found in aggregation patterns in different depth strata, as well as the seasonal differences found in the size structure of L. plei and L. sanpaidensis, Could ultimately be due to Changes in the water Column related to food availability. Considering that L. plei and L. sanpaidensis are both caught as by-catch by shrimp trawlers throughout file year, we also discuss hypotheses on the life cycle of the species and its implications for fishery management. We recommend measures to protect squid spawning grounds by creating spring and summer protected areas where trawling would be prohibited until 60 m depth, i.e. in the area and time of year when mature individuals concentrate.

The neutron-rich lead isotopes, up to Pb-216, have been studied for the first time, exploiting the fragmentation of a primary uranium beam at the FRS-RISING setup at GSI. The observed isomeric states exhibit electromagnetic transition strengths which deviate from state-of-the-art shell-model calculations. It is shown that their complete description demands the introduction of effective three-body interactions and two-body transition operators in the conventional neutron valence space beyond Pb-208.

Using the high-resolution performance of the fragment separator FRS at GSI we have discovered 60 new neutron-rich isotopes in the atomic number range of 60 <= Z <= 78. The new isotopes were unambiguously identified in reactions with a U-238 beam impinging on a Be target at 1 GeV/nucleon. The production cross-section for the new isotopes have been measured down to the pico-barn level and compared with predictions of different model calculations. For elements above hafnium fragmentation is the dominant reaction mechanism which creates the new isotopes, whereas fission plays a dominant role for the production of the new isotopes up to thulium. Based on the hypothesis that reproduction is a continuous process in tropical habitats, we analysed reproductive periodicity and egg production in the callianassid ghost shrimp Lepidophthalmus bocourti, one of the most common species in mangrove systems along the Pacific coast of Central America. During one year (May 2008 to April 2009), individuals of L. bocourti (N = 499) were collected nearshore Gulf of Nicoya, Pacific coast of Costa Rica. Observations were made on presence or absence of incubated embryos, and gonad activity of females was analysed as gonadosomatic index (GSI). Our results revealed that L. bocourti has a marked seasonal breeding period, which contradicts previous reports regarding coastal marine decapods from the tropics. Ovigerous females were found only from June to August, while high GSI values were obtained from March to July. The increase of GSI and appearance of ovigerous females were associated with a concomitant decrease of salinity, but not with temperature. We assume that reproduction of L. bocourti is adapted to local changes of environmental conditions, and that a decrease in salinity during rainy season may serve as a triggering factor for ovarian development. Compared to other ghost shrimps, L. bocourti produced on average more (2002 +/- 1365) and smaller (0.87 +/- 0.109 mm) eggs, which seems to suggest that this species does not have an abbreviated larval development as reported for other species of genus. The deviation from the generalization of constant reproduction in the tropics for shallow water marine invertebrates and its probable cause are adequately discussed.

Neutron-rich isotopes around lead, beyond N = 126, have been studied exploiting the fragmentation of an uranium primary beam at the FRS-RISING setup at GSI. For the first time beta-decay half-lives of Bi-219 and Tl-211,Tl-212,Tl-213 isotopes have been derived. The half-lives have been extracted using a numerical simulation developed for experiments in high-background conditions. Comparison with state of the art models used in r-process calculations is given, showing a systematic underestimation of the experimental values, at variance from close-lying nuclei. [EN] Preliminary data of the length-weight relationship and reproduction of the bogue Boops boops off Gran Canaria (Canary Islands, Central-east Atlantic) are provided. Two thousand and twenty-one individuals of bogue, ranging from 4 to 34 cm TL, were obtained from purse seine commercial landing. Reproduction parameters as sex determination, duration of spawning season, size at first maturity and GSI variation along time were determinated based on macroscopic evaluation of gonads. The results obtained suggest that bogue is a total spawner, with a long spawning season extending from January to May. Size at first maturity was 16.7 and 17.9 cm TL for males and females, respectively. The length-weight relationship obtained showed a positive allometry in both sexes

Self-incompatibility (SI) systems have evolved in many flowering plants to prevent self-fertilization and thus promote outbreeding. Pear and apple, as many of the species belonging to the Rosaceae, exhibit RNase-mediated gametophytic self-incompatibility, a widespread system carried also by the Solanaceae and Plantaginaceae. Pear orchards must for this reason contain at least two different cultivars that pollenize each other; to guarantee an efficient cross-pollination, they should have overlapping flowering periods and must be genetically compatible. This compatibility is determined by the S-locus, containing at least two genes encoding for a female (pistil) and a male (pollen) determinant. The female determinant in the Rosaceae, Solanaceae and Plantaginaceae system is a stylar glycoprotein with ribonuclease activity (S-RNase), that acts as a specific cytotoxin in incompatible pollen tubes degrading cellular RNAs. Since its identification, the S-RNase gene has been intensively studied and the sequences of a large number of alleles are available in online databases. On the contrary, the male determinant has been only recently identified as a pollen-expressed protein containing a F-box motif, called S-Locus F-box (abbreviated SLF or SFB). Since F-box proteins are best known for their participation to the SCF (Skp1 - Cullin - F-box) E3 ubiquitine ligase enzymatic complex, that is involved in protein degradation through the 26S proteasome pathway, the male determinant is supposed to act mediating the ubiquitination of the S-RNases, targeting them for the degradation in compatible pollen tubes. Attempts to clone SLF/SFB genes in the Pyrinae produced no results until very recently; in apple, the use of genomic libraries allowed the detection of two F-box genes linked to each S haplotype, called SFBB (S-locus F-Box Brothers). In Japanese pear, three SFBB genes linked to each haplotype were cloned from pollen cDNA. The SFBB genes exhibit S haplotype-specific sequence divergence and pollen-specific expression; their multiplicity is a feature whose interpretation is unclear: it has been hypothesized that all of them participate in the S-specific interaction with the RNase, but it is also possible that only one of them is involved in this function. Moreover, even if the S locus male and female determinants are the only responsible for the specificity of the pollen-pistil recognition, many other factors are supposed to play a role in GSI; these are not linked to the S locus and act in a S-haplotype independent manner. They can have a function in regulating the expression of S determinants (group 1 factors), modulating their activity (group 2) or acting downstream, in the accomplishment of the reaction of acceptance or rejection of the pollen tube (group 3). This study was aimed to the elucidation of the molecular mechanism of GSI in European pear (Pyrus communis) as well as in the other Pyrinae; it was divided in two parts, the first focusing on the characterization of male determinants, and the second on factors external to the S locus. The research of S locus F-box genes was primarily aimed to the identification of such genes in European pear, for which sequence data are still not available; moreover, it allowed also to investigate about the S locus structure in the Pyrinae. The analysis was carried out on a pool of varieties of the three species Pyrus communis (European pear), Pyrus pyrifolia (Japanese pear), and Malus × domestica (apple); varieties carrying S haplotypes whose RNases are highly similar were chosen, in order to check whether or not the same level of similarity is maintained also between the male determinants. A total of 82 sequences was obtained, 47 of which represent the first S-locus F-box genes sequenced from European pear. The sequence data strongly support the hypothesis that the S locus structure is conserved among the three species, and presumably among all the Pyrinae; at least five genes have homologs in the analysed S haplotypes, but the number of F-box genes surrounding the S-RNase could be even greater. The high level of sequence divergence and the similarity between alleles linked to highly conserved RNases, suggest a shared ancestral polymorphism also for the F-box genes. The F-box genes identified in European pear were mapped on a segregating population of 91 individuals from the cross 'Abbé Fétel' × 'Max Red Bartlett'. All the genes were placed on the linkage group 17, where the S locus has been placed both in pear and apple maps, and resulted strongly associated to the S-RNase gene. The linkage with the RNase was perfect for some of the F-box genes, while for others very rare single recombination events were identified. The second part of this study was focused on the research of other genes involved in the SI response in pear; it was aimed on one side to the identification of genes differentially expressed in compatible and incompatible crosses, and on the other to the cloning and characterization of the transglutaminase (TGase) gene, whose role may be crucial in pollen rejection. For the identification of differentially expressed genes, controlled pollinations were carried out in four combinations (self pollination, incompatible, half-compatible and fully compatible cross-pollination); expression profiles were compared through cDNA-AFLP. 28 fragments displaying an expression pattern related to compatibility or incompatibility were identified, cloned and sequenced; the sequence analysis allowed to assign a putative annotation to a part of them. The identified genes are involved in very different cellular processes or in defense mechanisms, suggesting a very complex change in gene expression following the pollen/pistil recognition. The pool of genes identified with this technique offers a good basis for further study toward a better understanding of how the SI response is carried out. Among the factors involved in SI response, moreover, an important role may be played by transglutaminase (TGase), an enzyme involved both in post-translational protein modification and in protein cross-linking. The TGase activity detected in pear styles was significantly higher when pollinated in incompatible combinations than in compatible ones, suggesting a role of this enzyme in the abnormal cytoskeletal reorganization observed during pollen rejection reaction. The aim of this part of the work was thus to identify and clone the pear TGase gene; the PCR amplification of fragments of this gene was achieved using primers realized on the alignment between the Arabidopsis TGase gene sequence and several apple EST fragments; the full-length coding sequence of the pear TGase gene was then cloned from cDNA, and provided a precious tool for further study of the in vitro and in vivo action of this enzyme.

Das Studium chemischer Eigenschaften der schwersten Elemente, der Transactiniden, erfordert immer leistungsfähigere Apparaturen. Wegen ihrer kurzen Halbwertszeiten müssen diese Elemente schnell von der Targetkammer mittels Gasjet zur Apparatur transportiert und dort in die wässrige Phase überführt werden. Das sollte zur Vermeidung von Verlusten durch vorzeitigen Zerfall möglichst online betrieben werden, d.h. die in der Kernreaktion gebildeten Atome werden kontinuierlich aus der Targetkammer ausgespült und in der Apparatur in die wässrige Phase überführt. Dabei stellt sich das Problem, die kleinen Aerosol-Partikel (100-200 nm und etwa 1010 Teilchen pro Liter) bei einem Gasfluss von 2 bis 3 Liter Jet-Gas pro min in eine mit 1 bis 2 mL/min fließende wässrige Lösung zu überführen. Hierzu wurden zwei verschiedene Systeme am TRIGA-Reaktor Mainz entwickelt und in Experimenten an den Schwerionenbeschleunigern am PSI und der GSI erfolgreich eingesetzt. Da die diskontinuierlich arbeitende Chemieapparatur ARCA, mit der bisher chemische Eigenschaften der Elemente 101, 103, 104, 105 und 106 bestimmt wurden, wegen niedriger Produktionsraten bei Element 106 an ihre technologischen Grenzen stößt, bestand eine weitere Aufgabe der vorliegenden Arbeit darin, ein kontinuierlich arbeitendes Chromatographiesystem aufzubauen, mit dem Verteilungskoeffizienten des Elements 106 gemessen werden können. Bei der in dieser Arbeit entwickelten und in Versuchen mit kurzlebigen Hafnium-, Wolfram- und Rutherfordium-Isotopen getesten, kontinuierlich arbeitenden Mehrsäulentechnik wird der direkte Nachweis des Transactinids aufgegeben, und die Retentionszeit in dem chromatographischen System über die Menge der während der Retention zerfallenen Atome bestimmt. Neben der apparativen Entwicklung dieser Technik, die geringste Totvolumina im System erfordert, mussten bei der Auswahl des geeigneten chemischen Systems die engen Grenzen dieser Methode beachtet werden, was umfangreiche Kenntnisse zur trägerfreien Chemie der homologen Elemente erfordert. Neben Batchexperimenten mit trägerfrei produzierten Nukliden wurde für offline-Experimente auch ARCA erfolgreich eingesetzt. Der Vergleich von Kd-Werten, die in Batchexperimenten, mit ARCA und mit der prinzipiell neuen Methode der Mehrsäulentechnik bestimmt wurden, zeigten dabei gute Übereinstimmungen. Für die Anwendbarkeit der Mehrsäulentechnik sind aber auch geeignete radioaktive Zerfallsketten notwendig, die in einem langlebigen Isotop enden sollten, welches über lange Experimentierzeiten akkumuliert werden kann. Dabei ist die Diskriminierung von einzelnen Atomen langlebiger Actiniden als Endglieder der Zerfallskette der Transactiniden gegen den natürlichen und elektronischen Untergrund sehr anspruchsvoll und möglicherweise der limitierende Faktor dieser Technik. Neben Beiträgen zur Fluoridkomplexierung von Elementen der 4. Nebengruppe (inklusive Rutherfordium) und der 6. Nebengruppe, wurde die Hydrolyse von Elementen der 6. Nebengruppe untersucht. Hier zeigen sich bei Verwendung von trägerfreien Aktivitäten abweichende Resultate gegenüber der Literatur.Auf der Grundlage dieser Daten wurde ein Mehrsäulenexperiment für 7,4-s 265Sg (Element 106) vorbereitet, für dessen Einsatz sowohl ein drehendes Targetradsystem für eine erhöhte Produktionsrate getestet wurde, als auch der Einsatz von speziell funktionalisierten Ionenaustauscherharzen.

In this work the flux line dynamics in High-Temperature Superconductor (HTSC) thin films in the presence of columnar defects was studied using electronic transport measurements. The columnar defects which are correlated pinning centers for vortices were generated by irradiation with swift heavy ions at the Gesellschaft für Schwerionenforschung (GSI) in Darmstadt. In the first part, the vortex dynamics is discussed within the framework of the Bose-glass model. This approach describes the continuous transition from a vortex liquid to a Bose-glass phase which is characterized by the localization of the flux lines at the columnar defects. The critical behavior of the characteristic length and time scales for temperatures in the vicinity of this phase transition were probed by scaling properties of experimentally obtained current-voltage characteristics. In contrast to the predicted universal properties of the critical behavior the scaling analysis shows a strong dependence of the dynamic critical exponent on the experimentally accessible electric field range. In addition, the predicted divergence of the activation energy in the limit of low current densities was experimentally not confirmed.The dynamic behavior of flux lines in spatially resolved irradiation geometries is reported in the second part. Weak pinning channels with widths between 10 µm and 100 µm were generated in a strong pinning environment with the use of metal masks and the GSI microprobe, respectively. Measurements of the anisotropic transport properties of these structures show a striking resemblance to the results in YBCO single crystals with unidirected twin boundaries which were interpreted as a guided vortex motion effect. The use of two additional test bridges allowed to determine in parallel the resistivities of the irradiated and unirradiated parts as well as the respective current-voltage characteristics. These measurements provided the input parameters for a numerical simulation of the potential distribution in the spatially resolved irradiation geometry. The results are interpreted within a model that describes the hydrodynamic interaction between a Bose-glass phase and a vortex liquid. The interface between weakly pinned flux lines in the unirradiated channels and strongly pinned vortices leads to a nonuniform vortex velocity profile and therefore a variation of the local electric field. The length scale of these interactions was estimated for the first time in measuring the local variation of the electric field profile in a Bose-glass contact.Finally, a method for the determination of the true temperature in HTSC thin films at high dissipation levels is described. In this regime of electronic transport the occurrence of a flux flow instability is accompanied by heating effects in the vortex system. The heat propagation properties of the film/substrate system are deduced from the time dependent voltage response to a short high current density pulse of rectangular shape. The influence of heavy ion irradiation on the heat resistance at the film/substrate interface is studied.

Im Rahmen meiner Arbeit wurden erstmals die Intermediärfilament-Proteine (IF-Proteine) des Sibirischen Störs Acipenser baeri (Strahlenflosser, Knorpelganoid) kloniert und sequenziert. Aus einer cDNA-Bank konnten die Sequenzen von 13 IF-Proteine gewonnen werden. Von insgesamt zehn Keratinen codieren sieben für Typ I-Keratine und drei für Typ II. Zusätzlich konnten noch Desmin, Vimentin und ein Lamin identifiziert werden. Je einem Typ I- (K13) und einem Typ-II-Keratin (K2) fehlen wenige Aminosäuren in der Head-Domäne.Cytoskelett-Präparationen aus Epidermis, Mitteldarm, Magen und Kieme wurden mittels 2D-PAGE aufgetrennt. Durch Einsatz des CKBB-Test und Immunoblots wurden die verschiedenen Typ I und II-Keratine sowie Desmin und Vimentin identifiziert. Die gewebsspezifische Expression der Keratine ermöglichte zumeist ihre Einteilung in 'E' (epidermal) und 'S' ('simple epithelial').Die MALDI-MS-Analyse einer 2D-PAGE-Koelektrophorese von Seitenflosse und Mitteldarm zeigte, daß die 34 vorhandenen Proteinflecke auf nur 13 verschiedene IF-Proteine zurückgehen. Neun dieser Flecke konnten Sequenzen zugewiesen werden. Zusammen mit den verbleibenden vier Proteinflecken ergeben sich für den Stör nunmehr insgesamt 17 bekannte IF-Proteine. Von drei biochemisch identifizierten IS-Keratinen kommt eines nur im Mitteldarm vor und nur einem konnte eine Sequenz zugeordnet werden (K18). Dem einzigen Typ IIS-Keratin konnte keine Sequenz zugeordnet werden, wahrscheinlich handelt es sich um dabei um das K8-Orthologe. Jedem der fünf Typ IE-Proteine konnte eine Sequenz zugeordnet werden (K10 bis K14), ebenso wie dem einzigen identifizierten Typ IIE-Keratin (K2). Von den Typ III-Proteinen wurden Desmin und Vimentin ihren Proteinflecken zugeordnet. Die nicht zugeordnete Sequenz aba-k1 codiert möglicherweise für ein IIE-Keratin, während aba-k15 vermutlich die Sequenz für ein IE-Keratin enthält. Bei den Proteinflecken, denen eine Sequenz zugeordnet werden konnten, kann für Aba-K2 die Zugehörigkeit zum IIE-Typ angenommen werden, während es sich bei Aba-K10 wahrscheinlich um ein IE-Keratin handelt.Durch Datenbankvergleiche und molekulare Stammbäume konnte die Zugehörigkeit der identifizierten Lamin-Sequenz zum B3-Subtyp der Vertebraten gezeigt werden.Die Daten der Biochemie und indirekten Immunfluoreszenzmikroskopie zeigen, daß Keratine in Epithelien und Vimentin in mesenchymalen Geweben vorkommen. Es existieren starke Hinweise, daß im letzten Gewebetyp Keratine auch koexprimiert werden. Desmin kommt in großen Mengen im Magen und im Mitteldarm vor und stellt dort das prominenteste Protein.Mit den gewonnenen Sequenzdaten wurden molekulare Stammbäume und Sequenzidentitäten berechnet. Die daraus resultierenden Konsequenzen für die Verwandtschaftsverhältnisse der verschiedenen IF-Proteine sowie der Wirbeltiere werden diskutiert.
