In this report, we present cationic dimeric (gemini) lipids for significant plasmid DNA (pDNA) delivery to different cell lines without any marked toxicity in the presence of serum. Six gemini lipids based on alpha-tocopherol were synthesized, which differed in their spacer chain lengths. Each of these gemini lipids mixed with a helper lipid, 1,2-dioleoyl phosphatidyl ethanolamine (DOPE), was capable of forming stable aqueous suspensions. These co-liposomal systems were examined for their potential to transfect pEGFP-C3 plasmid DNA into nine cell lines of different origins. The transfection efficacies noticed in terms of EGFP expression levels using flow cytometry were well corroborated using independent fluorescence microscopy studies. Significant EGFP expression levels were reported using the gemini co-liposomes, which counted significantly better than one well known commercial formulation, Lipofectamine 2000 (L2 K). Transfection efficacies were also analyzed in terms of the degree of intracellular delivery of labeled plasmid DNA (pDNA) using confocal microscopy, which revealed an efficient internalization in the presence of serum. The cell viability assays performed using optimized formulations demonstrated no significant toxicity towards any of the cell lines used in the study. We also had a look at the lipoplex internalization pathway to profile the uptake characteristics. A caveolae/lipid raft route was attributed to their excellent gene transfection capabilities. The study was further advanced by using a therapeutic p53-EGFP-C3 plasmid and the apoptotic activity was observed using FACS and growth inhibition assay.

Objectives: The ability to target conventional drugs efficiently inside cells to kill intraphagosomal bacteria has been a major hurdle in treatment of infective diseases. We aimed to develop an efficient drug delivery system for combating infection caused by Salmonella, a well-known intracellular and intraphagosomal pathogen. Chitosan dextran sulphate (CD) nanocapsules were assessed for their efficiency in delivering drugs against Salmonella. Methods: The CD nanocapsules were prepared using the layer-by-layer method and loaded with ciprofloxacin or ceftriaxone. Antibiotic-loaded nanocapsules were analysed in vitro for their ability to enter epithelial and macrophage cells to kill Salmonella. In vivo pharmacokinetics and organ distribution studies were performed to check the efficiency of the delivery system. The in vivo antibacterial activity of free antibiotic and antibiotic loaded into nanocapsules was tested in a murine salmonellosis model. Results: In vitro and in vivo experiments showed that this delivery system can be used effectively to clear Salmonella infection, CD nanocapsules were successfully employed for efficient targeting and killing of the intracellular pathogen at a dosage significantly lower than that of the free antibiotic. The increased retention time of ciprofloxacin in the blood and organs when it was delivered by CD nanocapsules compared with the conventional routes of administration may be the reason underlying the requirement for a reduced dosage and frequency of antibiotic administration. Conclusions: CD nanocapsules can be used as an efficient drug delivery system to treat intraphagosomal pathogens, especially Salmonella infection, This delivery system might be used effectively for other vacuolar pathogens including Mycobacteria, Brucella and Legionella.

This paper presents the development and application of a stochastic dynamic programming model with fuzzy state variables for irrigation of multiple crops. A fuzzy stochastic dynamic programming (FSDP) model is developed in which the reservoir storage and soil moisture of the crops are considered as fuzzy numbers, and the reservoir inflow is considered as a stochastic variable. The model is formulated with an objective of minimizing crop yield deficits, resulting in optimal water allocations to the crops by maintaining storage continuity and soil moisture balance. The standard fuzzy arithmetic method is used to solve all arithmetic equations with fuzzy numbers, and the fuzzy ranking method is used to compare two or more fuzzy numbers. The reservoir operation model is integrated with a daily-based water allocation model, which results in daily temporal variations of allocated water, soil moisture, and crop deficits. A case study of an existing Bhadra reservoir in Karnataka, India, is chosen for the model application. The FSDP is a more realistic model because it considers the uncertainty in discretization of state variables. The results obtained using the FSDP model are found to be more acceptable for the case study than those of the classical stochastic dynamic model and the standard operating model, in terms of 10-day releases from the reservoir and evapotranspiration deficit. A short-term real-time operation model with fuzzy state variables is developed for irrigation of multiple crops based on earlier work on long-term steady-state policy. The features of the model that distinguish it from the earlier work are (1) apart from inclusion of fuzziness in reservoir storage and in soil moisture of crops, spatial variations in rainfall and soil moisture of crops are included in the real-time operation model by considering gridded command area with a grid size of 0.5 degrees latitude by 0.5 degrees longitude; (2) the water allocation model and soil moisture balance equations are integrated with the real-time operation model with consideration of ponding water depth for Paddy crop; the model solution specifies reservoir releases for irrigation in a 10-day time period and allocations among the crops on a daily basis at each grid by maintaining soil moisture balance at the end of the day; and (3) the release policy is developed using forecasted daily rainfall data of each grid and is implemented for the current time period using actual 10-day inflow and actual daily rainfall of each grid. The real-time operation model is applied to Bhadra Reservoir in Karnataka, India. The results obtained using the real-time operation model are compared with those of the standard operating policy model. Inclusion of fuzziness in reservoir storage and soil moisture of crops captures hydrologic uncertainties in real time. Considerations of irrigation decisions on a daily basis and the gridded command area result in variations in allocating water to the crops, variations in actual crop evapotranspiration, and variations in soil moisture of the crops on a daily basis for each grid of the command area. This paper describes a new technology for solonchak soil reclamation in which surface flood irrigation of fresh water and pumped wells drainage of salty groundwater are combined. The comprehensive investigation of water and salt movement has been conducted through field test, laboratory simulation and numerical calculation. The dependence of desalination on irrigation water quantity, drainage quantity, leaching time and other parameters is obtained based on the field tests. The entire desalination process under the flood-irrigation and well-drainage operations was experimentally simulated in a vertical soil column. The water and salt movement has been numerically analysed for both the field and laboratory conditions. The present work indicates that this new technology can greatly improve the effects of desalination.

Research has shown that aquatic weeds, particularly hydrilla ( Hydrilla verticillata , (L.F.) Royle), can be controlled with exposure of 8 to 12 weeks with concentrations of 10 to 15 ppb of fluridone (1-methyl-3-phenyl-5-[3-trifluoromethyl) phenyl]-4(1 H )- pyridinone) (Haller et al. 1990 and Fox et al. 1994). Fluridone label recommendations restrict the use of the treated waters for irrigation of turf or newly seeded crops and seed beds for 30 days following the last application of the herbicide. The objective of this research was to determine the effects of 10 weeks of irrigation with fluridone containing water on a common Florida residential turfgrass.

American pondweed ( Potamogeton nodosus Poir.) is commonly found in northern California irrigation canals. The purpose of this study was to test the hypothesis that exposure of American pondweed winter buds to dilute acetic acid under field conditions would result in reduced subsequent biomass.

The aims of this paper are twofold. Firstly to characterise rural poverty and to give a broad overview of the agro-ecological, climatic and socio-economic conditions in Sri Lanka which shape poverty. Secondly to present the methodology employed to screen suitable field research areas and the techniques subsequently used to carry out Rapid Rural Appraisal in two upper-watersheds villages. Also presented are details of a concurrent stakeholder analysis that aimed to investigate the capacity of secondary stakeholders to promote sustainable aquatic resource development and to invite their participation in the formulation of a participatory research agenda.[PDF contains 58 pages]

RRAs were carried out in two Small Tank Cascade systems (STCs) of North West Province, Sri Lanka (less than 1000 ha total watershed area). A total of 21 tanks and 7 villages were investigated with primary emphasis on two upper watershed communities. The two systems differ primarily in their resource base; namely rainfall, natural forests and proximity to large scale perennial irrigation resources. [PDF contains 86 pages]

The visit highlighted the vital contribution of the inland fisheries sector to provision of basic food security within the uncleared area (farmers report very low consumption frequencies for all other fish or meat protein substitutes). A 30-mile system of Brackish water lagoons which demarcates the cleared and uncleared areas is the main source of retailed fish in the uncleared area. Second in importance is the inland tank fishery, where the bulk of production emanates from 17 major irrigation reservoirs. [PDF contains 29 pages]

One of the critical problems currently being faced by agriculture industry in developing nations is the alarming rate of groundwater depletion. Irrigation accounts for over 70% of the total groundwater withdrawn everyday. Compounding this issue is the use of polluting diesel generators to pump groundwater for irrigation. This has made irrigation not only the biggest consumer of groundwater but also one of the major contributors to green house gases. The aim of this thesis is to present a solution to the energy-water nexus. To make agriculture less dependent on fossil fuels, the use of a solar-powered Stirling engine as the power generator for on-farm energy needs is discussed. The Stirling cycle is revisited and practical and ideal Stirling cycles are compared. Based on agricultural needs and financial constraints faced by farmers in developing countries, the use of a Fresnel lens as a solar-concentrator and a Beta-type Stirling engine unit is suggested for sustainable power generation on the farms. To reduce the groundwater consumption and to make irrigation more sustainable, the conceptual idea of using a Stirling engine in drip irrigation is presented. To tackle the shortage of over 37 million tonnes of cold-storage in India, the idea of cost-effective solar-powered on-farm cold storage unit is discussed.

The neurotransmitter serotonin (5-HT) has a multifaceted function in the modulation of information processing through the activation of multiple receptor families, including G-protein-coupled receptor subtypes (5-HT1, 5-HT2, 5-HT4-7) and ligand-gated ion channels (5-HT3). The largest population of serotonergic neurons is located in the midbrain, specifically in the raphe nuclei. Although the medial and dorsal raphe nucleus (DRN) share common projecting areas, in the basal ganglia (BG) nuclei serotonergic innervations come mainly from the DRN. The BG are a highly organized network of subcortical nuclei composed of the striatum (caudate and putamen), subthalamic nucleus (STN), internal and external globus pallidus (or entopeduncular nucleus in rodents, GPi/EP and GPe) and substantia nigra (pars compacta, SNc, and pars reticulata, SNr). The BG are part of the cortico-BG-thalamic circuits, which play a role in many functions like motor control, emotion, and cognition and are critically involved in diseases such as Parkinson's disease (PD). This review provides an overview of serotonergic modulation of the BG at the functional level and a discussion of how this interaction may be relevant to treating PD and the motor complications induced by chronic treatment with L-DOPA.

The viability of integrating rice farming with fish culture was studied in ten (10) rice plots. The on-farm research was done during one rice-growing season starting May 2003. The rice variety used was IR 2793-80-1 while the fish species was the Nile tilapia, Oreochromis niloticus. The fish culture period lasted 77 days. An average fish production of 132.4 kg/ha was obtained. The mean recovery rate of tilapia was 43 per cent. Total rice yield from the fields stocked with fish was lower than from unstocked fields. The net returns were not significantly different.
