La maladie de Crohn (MC) pédiatrique a des conséquences majeures sur la qualité de vie des patients atteints (troubles de croissance, absentéisme scolaire, etc). L’étiologie de la MC est inconnue. La théorie de l’hygiène (TH) stipule que les conditions de vie sanitaires des pays industrialisés préviennent l’exposition antigénique et empêchent le développement de la tolérance immunitaire chez les enfants. Ceci mènerait à une réaction excessive du système immunitaire lors d’expositions subséquentes et engendrerait le développement de maladies inflammatoires chroniques telles la MC. Objectif: Analyser l’association entre la fréquence, la temporalité et le type d’infections infantiles (indicateurs d’environnements pourvus d’antigènes) et le risque de MC pédiatrique. Une étude cas-témoin fût réalisée, les cas de MC provenant d’un centre hospitalier tertiaire montréalais. Les témoins, provenant des registres de la Régie d’assurance maladie du Québec (RAMQ), furent appariés aux cas selon leur âge, sexe et lieu de résidence. L’exposition aux infections fût déterminée grâce aux codes de diagnostic ICD-9 inscrits dans la base de données de la RAMQ. Un modèle de régression logistique conditionnelle fût construit afin d’analyser l’association entre infections et MC. Des ratios de cotes (RC) et intervalles de confiance à 95% (IC 95%) furent calculés. Résultats: 409 cas et 1621 témoins furent recrutés. Les résultats de l’analyse suggèrent un effet protecteur des infections infantiles sur le risque de MC (RC: 0,67 [IC: 0,48-0,93], p=0,018), plus particulièrement au cours des 5 premières années de vie (RC: 0.74 [IC: 0,57-0,96], p=0,025). Les infections rénales et urinaires, ainsi que les infections des voies orales et du système nerveux central (virale), semblent particulièrement associées à l’effet protecteur. Les résultats de l’étude appuient la théorie de l’hygiène: l’exposition aux infections infantiles pourrait réduire le risque de MC pédiatrique.

Problématique: Le virus du papillome humain (VPH) est présent dans près de 50% des cancers de l’oropharynx. Le potentiel oncogénique du VPH est encodé dans les oncoprotéines E6 et E7, qui agissent en modulant différents gènes, dont les gènes suppresseurs de tumeur p53 et pRb. Les cellules VPH positives démontrent une altération au niveau de la signalisation de la réponse aux dommages à l’ADN (RDA), un mécanisme de contrôle dans l’arrêt de la croissance des cellules ayant subit des dommages au niveau de leur ADN. Hypothèse et objectifs : Nous croyons que les défauts au niveau de la RDA des cancers VPH positifs peuvent être exploités afin de sensibiliser préférentiellement les cellules cancéreuses aux traitements de radiothérapie. Cette stratégie de recherche nécessite l’élaboration d’un modèle cellulaire de carcinogenèse isogénique pour le cancer de l’oropharynx que nous proposons de développer et de caractériser. L’étude vise à dériver des lignées isogéniques à partir de kératinocytes primaires et cellules épithéliales de l’oropharynx pour ensuite valider la carcinogenèse de notre modèle in vitro & in vivo Méthodologie : Des lignées cellulaires de kératinocytes primaires et de cellules épithéliales de l’oropharynx ont été successivement modifiées par transduction afin de présenter les mutations associées aux cancers de l’oropharynx induits par le VPH. Les cellules ont été modifiées avec des lentivirus codants pour la télomérase (hTERT), les oncogènes E6, E7 et RasV12. Afin de valider la cancérogenèse in vitro de notre modèle, des études d’invasion en matrigel et de croissance sans ancrage en agar mou ont été réalisées. Les populations cellulaires transformées ont été ensuite introduites dans des souris immunodéficientes afin d’évaluer leur tumorogénicité in vivo. Résultats : À partir des plasmides recombinés construits par méthodes de clonage traditionnelle et de recombinaison « Gateway », nous avons produit des lentivirus codants pour la télomérase humaine (hTERT), les oncogènes viraux E6 et E7 et l’oncogène Ras. Les kératinocytes primaires et cellules épithéliales de l’oropharynx ont été infectés successivement par transduction et sélectionnés. Nous avons validé l’expression de nos transgènes par méthode d’immunofluorescence, de Western Blot et de réaction de polymérisation en chaîne quantitative en temps réel (qRT-PCR). Nous avons établi trois lignées des cellules épithéliales de l’oropharynx (HNOE) à partir d’échantillons tissulaires prélevés lors d’amygdalectomie (HNOE42, HNO45, HNOE46). Les cellules transduites avec le lentivirus exprimant le promoteur fort CMV/TO de l’oncogène RasV12 ont présenté un changement morphologique compatible avec une sénescence prématurée induite par l’oncogène Ras. En exprimant des quantités plus faibles du RasV12 mutant, la lignée cellulaire HEKn hTERT-E6-E7 PGK RasV12 a réussi à échapper à la sénescence induite par l’oncogène Ras. La population cellulaire exprimant HEKn hTERT-E6-E7-PGK RasV12 a présenté un phénotype malin en culture et à l’étude d'invasion, mais n’a pas démontré de résultats positifs à l’étude de croissance sans ancrage en agar mou ni en xénogreffe en souris immunodéficientes. Conclusion : Nos résultats démontrent qu’en présence des oncogènes viraux E6 et E7, il y a un troisième mécanisme suppresseur de tumeur qui médie la sénescence induite par l’oncogène Ras. Nous avons identifié que la présence de E6 seule ne suffit pas à immortaliser les kératinocytes primaires humains (HEKn). Nous n’avons pas réussi à créer un modèle in vitro de carcinogenèse pour les cancers de l’oropharynx induits par le VPH.

Introduction: A growing biological research field is the cellular senescence, a mechanism that has been associated, under certain circumstances, with malignant transformation. Given the high incidence of ovarian cancer and its main origin from the ovarian surface epithelium, as well as the possibility that an epithelial-mesenchymal transition occurs, we evaluated both the in vitro growth of stromal fibroblasts from the ovarian cortex and their β-galactosidase activity at pH 6, enzyme whose expression is considered as a marker of replicative senescence. Methods: 48 samples of ovarian cortical fibroblasts from donors without a history of cancer were serially cultured until the end of their replicative life. β-galactosidase activity at pH 6 was quantified in each passage by the chemiluminiscent method. As control, we used ovarian epithelial cell cultures from the same donors. The enzyme activity was also evaluated in fibroblasts previously induced to senescence by exposure to hydrogen peroxide. Results: The analysis of the enzyme activity and the replicative capacity taken together showed that the fibroblast cultures reached the senescent state at passages 4-5, as what happened with the control epithelial cells. Fibroblasts induced to senescence showed high variability in the values of enzymatic activity. Conclusions: The similarity between both types of cells in reaching the senescent state deserves to be taken into account in relation to the epithelialmesenchymal transition that has been proposed to explain their behavior in the genesis of cancer arising from ovarian surface epithelium. Low β-galactosidase activity values at pH 6 would suggest possible inactivation of the response pathways to oxidative stress.

We present an Overlapping Generations Model with two final goods: tradable goods are produced with a standard Cobb-Douglas production function and non-tradable goods are produced with linear production function where the only factor is labor. We maintain the fundamental assumption of factor mobility between sectors so model is consistent with the Balassa-Samuelson hypothesis. Given the general equilibrium structure of our model we can examine the effect of the saving rate on migration and non-tradable relative prices. Under this setting, we find that the elderly have incentives to migrate from economies where productivity is high to economies with low productivity because of the lower cost of living. In more general terms the elderly migration is likely to go from rich to poor countries. We also find that, for poor countries, the elderly migration has a positive effect in wages and capital accumulation.

This paper discusses the dangers inherent in allempting to simplify something as complex as development. It does this by exploring the Lynn and Vanhanen theory of deterministic development which asserts that varying levels of economic development seen between countries can be explained by differences in 'national intelligence' (national IQ). Assuming that intelligence is genetically determined, and as different races have been shown to have different IQ, then they argue that economic development (measured as GDP/capita) is largely a function of race and interventions to address imbalances can only have a limited impact. The paper presents the Lynne and Vanhanen case and critically discusses the data and analyses (linear regression) upon which it is based. It also extends the cause-effect basis of Lynne and Vanhanen's theory for economic development into human development by using the Human Development Index (HDI). It is argued that while there is nothing mathematically incorrect with their calculations, there are concerns over the data they employ. Even more fundamentally it is argued that statistically significant correlations between the various components of the HDI and national IQ can occur via a host of cause-effect pathways, and hence the genetic determinism theory is far from proven. The paper ends by discussing the dangers involved in the use of over-simplistic measures of development as a means of exploring cause-effect relationships. While the creators of development indices such as the HDI have good intentions, simplistic indices can encourage simplistic explanations of under-development. (c) 2005 Elsevier B.V. Development geography has long sought to understand why inequalities exist and the best ways to address them. Dependency theory sets out an historical rationale for under development based on colonialism and a legacy of developed core and under-developed periphery. Race is relevant in this theory only insofar that Europeans are white and the places they colonised were occupied by people with darker skin colour. There are no innate biological reasons why it happened in that order. However, a new theory for national inequalities proposed by Lynn and Vanhanen in a series of publications makes the case that poorer countries have that status because of a poorer genetic stock rather than an accident of history. They argue that IQ has a genetic basis and IQ is linked to ability. Thus races with a poorer IQ have less ability, and thus national IQ can be positively correlated with performance as measured by an indicator like GDP/capita. Their thesis is one of despair, as little can be done to improve genetic stock significantly other than a programme of eugenics. This paper summarises and critiques the Lynn and Vanhanen hypothesis and the assumptions upon which it is based, and uses this analysis to show how a human desire to simplify in order to manage can be dangerous in development geography. While the attention may naturally be focused on the 'national IQ' variables as a proxy measure of 'innate ability', the assumption of GDP per capita as an indicator of 'success' and 'achievement' is far more readily accepted without criticism. The paper makes the case that the current vogue for indicators, indices and cause-effect can be tyrannical.

Comparative analyses of survival senescence by using life tables have identified generalizations including the observation that mammals senesce faster than similar-sized birds. These generalizations have been challenged because of limitations of life-table approaches and the growing appreciation that senescence is more than an increasing probability of death. Without using life tables, we examine senescence rates in annual individual fitness using 20 individual-based data sets of terrestrial vertebrates with contrasting life histories and body size. We find that senescence is widespread in the wild and equally likely to occur in survival and reproduction. Additionally, mammals senesce faster than birds because they have a faster life history for a given body size. By allowing us to disentangle the effects of two major fitness components our methods allow an assessment of the robustness of the prevalent life-table approach. Focusing on one aspect of life history - survival or recruitment - can provide reliable information on overall senescence.

Winter wheat was grown in three field experiments, each repeated over two or three seasons, to investigate effects of extending flag leaf life by fungicide application on the concentration, kg ha(-1) and mg grain(-1) of nitrogen (N) and sulphur (S) as well as N:S ratio and sodium dodecyl sulphate (SDS) sedimentation volume. The experiments involved up to six cultivars and different application rates, timings and frequencies of azoxystrobin and epoxiconazole. For every day the duration to 37 % green flag leaf area (m) was extended, N yield was increased by 2.58 kg ha(-1), N per grain by 0.00957 mg, S yield by 0.186 kg ha(-1) and S per grain by 0.000718 mg. The N:S ratio decreased by 0.0135 per day. There was no evidence that these responses varied with cultivar. In contrast, the relationship between flag leaf life and N or S concentration interacted with cultivar. The N and S concentrations of Shamrock, the cultivar that suffered most from brown rust (Puccinia rccondita), increased with the extension of flag leaf life whereas the concentrations of N and S in Malacca, a cultivar more susceptible to Septoria tritici, decreased as flag leaf senescence was delayed. This was because the relationships between m and N and S yields were much better conserved over cultivars than those between m and thousand grain weight (TGW) and grain yield ha(-1).
