Microbial degradation is a major determinant of the fate of pollutants in the environment. para-Nitrophenol (PNP) is an EPA listed priority pollutant with a wide environmental distribution, but little is known about the microorganisms that degrade it in the environment. We studied the diversity of active PNP-degrading bacterial populations in river water using a novel functional marker approach coupled with [13C6]PNP stable isotope probing (SIP). Culturing together with culture-independent terminal restriction fragment length polymorphism analysis of 16S rRNA gene amplicons identified Pseudomonas syringae to be the major driver of PNP degradation in river water microcosms. This was confirmed by SIP-pyrosequencing of amplified 16S rRNA. Similarly, functional gene analysis showed that degradation followed the Gram-negative bacterial pathway and involved pnpA from Pseudomonas spp. However, analysis of maleylacetate reductase (encoded by mar), an enzyme common to late stages of both Gram-negative and Gram-positive bacterial PNP degradation pathways, identified a diverse assemblage of bacteria associated with PNP degradation, suggesting that mar has limited use as a specific marker of PNP biodegradation. Both the pnpA and mar genes were detected in a PNP-degrading isolate, P. syringae AKHD2, which was isolated from river water. Our results suggest that PNP-degrading cultures of Pseudomonas spp. are representative of environmental PNP-degrading populations.

Background. Mesenchymal stem cells (MSCs) are an attractive source for generation of cells with beta-cell properties. Previous studies have demonstrated the ability of prolactin to induce an increase in beta-cell mass and maturation, which suggests beneficial effects of its use in MSC differentiation protocols. Objective. To evaluate the expression of endocrine differentiation markers in rat MSCs treated in vitro with prolactin. Methods. Mesenchymal stem cells from bone marrow of Wistar rats were isolated, expanded, and characterized. Differentiation of MSCs was induced in medium containing 23 mmol/L of glucose, and nicotinamide, 2-mercaptoethanol, and exendin-4, in the presence or absence of 500 ng/mL of rat recombinant prolactin. Expression of endocrine markers and prolactin receptor genes was evaluated using real-time polymerase chain reaction, and compared between culture stages and presence vs absence of prolactin in the culture medium. Expression of insulin, somatostatin, glucagon, and pancreatic and duodenal homeobox 1 was also evaluated at immunofluorescence microscopy. Results. Isolated cells were mostly MSCs, as confirmed at fluorescent-activated cell sorting and cytochemistry. Pax6, Ngn-3, Isl1, NeuroD1, Nkx2.2, and Nkx6.1 exhibited varied expression during culture stages. The long form of the prolactin receptor messenger RNA was induced in prolactin-treated cultures (P < .05). The somatostatin gene was induced in early stages of differentiation (P < .05), and its expression was induced by prolactin, as confirmed using immunofluorescence. Conclusion. Culture of rat bone marrow MSCs in differentiation medium induces expression of pancreatic endocrine-specific genes, and somatostatin and prolactin receptor expression was also induced by prolactin.

A very high level of theoretical treatment (complete active space self-consistent field CASSCF/MRCI/aug-cc-pV5Z) was used to characterize the spectroscopic properties of a manifold of quartet and doublet states of the species BeP, as yet experimentally unknown. Potential energy curves for 11 electronic states were obtained, as well as the associated vibrational energy levels, and a whole set of spectroscopic constants. Dipole moment functions and vibrationally averaged dipole moments were also evaluated. Similarities and differences between BeN and BeP were analysed along with the isovalent SiB species. The molecule BeP has a X (4)Sigma(-) ground state, with an equilibrium bond distance of 2.073 angstrom, and a harmonic frequency of 516.2 cm(-1); it is followed closely by the states (2)Pi (R(e) = 2.081 angstrom, omega(e) = 639.6 cm(-1)) and (2)Sigma(-) (R(e) = 2.074 angstrom, omega(e) = 536.5 cm(-1)), at 502 and 1976 cm(-1), respectively. The other quartets investigated, A (4)Pi (R(e) = 1.991 angstrom, omega(e) = 555.3 cm(-1)) and B (4)Sigma(-) (R(e) = 2.758 angstrom, omega(e) = 292.2 cm(-1)) lie at 13 291 and 24 394 cm(-1), respectively. The remaining doublets ((2)Delta, (2)Sigma(+)(2) and (2)Pi(3)) all fall below 28 000 cm(-1). Avoided crossings between the (2)Sigma(+) states and between the (2)Pi states add an extra complexity to this manifold of states.

Com o propósito de fornecer dados para a avaliação de características ergonômicas no posto do operador de quatro colhedoras combinadas presentes na agricultura brasileira, foram realizadas, na região de Uberaba - MG, determinações das distâncias, a partir do ponto de referência do assento até os órgãos de comandos das colhedoras. Dentre as máquinas analisadas, a colhedora NH TC 57 foi a que apresentou melhores características ergonômicas do projeto interno da cabine.

The rate coefficients for the formation of carbon monophosphide (CP) and silicon monophosphide (SiP) by radiative association are estimated for temperatures ranging from 300 to 14 100 K. In this temperature range, the radiative association rate coefficients are found to vary from 1.14 x 10(-18) to 1.62 x 10(-18) cm(3) s(-1) and from 3.73 x 10(-20) to 7.03 x 10(-20) cm(3) s(-1) for CP and SiP, respectively. In both cases, rate coefficients increase slowly with the increase in temperature.

In some supergravity models there are light weakly coupled scalar (S)-and pseudoscalar (P) particles. These particles arise following a superlight gravitino. In these models the decay SIP → γγ exists. We examine constraints on this process considering these photons as responsible by the extragalactic background light. We also consider the amount of SIP particles produced through the fusion of the cosmic background photons and contributing to the effective number of light neutrino species during primordial nucleosynthesis. We obtain bounds on the gravitino mass complementary to the existing ones.

A tecnologia DSL- Digital Subscriber Line permitem um acesso universal de banda larga, com redução de custo e tempo de implantação necessários, quando comparadas a outras redes de acesso. DSL pode ser considerada uma rede de banda larga de grande capilaridade, uma vez que utiliza uma combinação de infra-estrutura de telefonia existente e a tecnologia de transmissão. Não obstante, a rede DSL precisa ser amplamente compreendida, de maneira que suas principais vantagens não sejam suplantadas pela ineficiência geral do sistema. Esta tese apresenta uma abordagem baseada em estratégias que possibilitem o planejamento de redes de comunicação que a priori não foram implementadas para suportar fluxos Triple Play (pré-requisito obrigatório diante do perfil atual de usuários da Internet). Será mostrado que, a partir do uso de medidas reais e análises probabilísticas, é possível elaborar o planejamento de redes de comunicação, considerando os parâmetros físicos e lógicos, tais como: o caminho de transmissão, a influência do ruído na comunicação, os protocolos utilizados e tráfego de Triple Play (voz, vídeo e dados).

Recentemente a sociedade está vivendo em meio a um aglomerado de dispositivos móveis com múltiplas interfaces sem fio que permitem que clientes móveis domésticos e corporativos solucionem problemas rotineiros no menor espaço de tempo e em diversas localizações. Por outro lado, a disponibilidade de diversas tecnologias de acesso sem fio como: Wi-Fi (Wireless Fidelity), WiMAX (Worldwide Interoperability for Microwave Access) e LTE (Long Term Evolution), proporcionam um cenário heterogêneo com diversas oportunidades de conectividade para o usuário de dispositivos com múltiplas interfaces. Alguns dos desafios atuais no contexto da concepção da então denominada NGN (Next Generation Networks) são o desenvolvimento de arcabouços e mecanismos que viabilizem tanto o suporte adequado à Qualidade de Serviço (QoS – Quality of Service) e à Qualidade de Experiência (QoE – Quality of Experience) para aplicações multimídia, quanto a garantia de equidade entre vazões de diferentes aplicações em ambiente heterogêneo. Esta dissertação propõe um arcabouço para o aprovisionamento de QoS e QoE em redes sem fio heterogêneas formada por redes WiMAX e Wi-Fi. Especificamente, nossa solução provê mapeamento estático de QoS entre as classes de serviços WiMAX e categorias de acesso Wi-Fi. Além disso, nossa proposta também combina funcionalidades de equidade entre vazões com a solução de mapeamento, a fim de alcançar um bom compromisso tanto para o operador da rede, quanto para o usuário, através de uma nova metodologia de coleta e calculo de vazão agregada e algoritmo de decisão de handover vertical. A avaliação quantitativa da proposta foi realizada através de modelos de simulação no ns-2 (Network Simulator). Os resultados de desempenho demonstram a eficiência da arquitetura em termos de métricas de QoS (vazão, vazão média e atraso) e QoE (PSRN - Peak Signal to Noise Ratio, SSIM - Structural Similarity Index e VQM - Video Quality Metric).

This study focused on estimating the economic losses resulting from cysticercosis at beef cattle farms that supply an export slaughterhouse located in the state of Sao Paulo, Brazil, and to identify the epidemiological risks factors involved in the disease to ascertain if these farms adopt Good Agricultural Practices (GAP). To this, we used data recorded in 2012 by Brazil's Federal Inspection Service (SIP) on the daily occurrence of the disease, according to the farm from which the animals originated. In addition, the associated risk factors were determined based on a case-control study at 48 farms. Cysticercosis was detected in 2.26% (95% CI 2.2-2.33) of the 190,903 bovines supplied by 556 farms in the following four states: 2.92% (95% CI 2.83-3.03) in Sao Paulo, 1.81% (95% CI 1.71-1.93) in Minas Gerais, 0.71% (95% CI 0.6-0.82) in Goias and 1.11% (95% CI 0.79-1.57) in Mato Grosso do Sul, with significant differences in the epidemiological indices of these states. Cysticercosis was detected at 58.45% (95% CI 54.36-62.55) of the farms of this study, representing estimated economic losses of US$312,194.52 for the farmers. Lower prevalence of this disease were found at the farms qualified for exports to the European Union, indicating a statistically significant difference from those not qualified to export to Europe. The access of cattle to non-controlled water sources, as well as sport fishing activities near the farms, was identified as risk factors. Cysticercosis causes considerable losses in Brazil's beef supply chain, with lower prevalence appearing only at farms qualified to export to the European Union. As for the access of cattle to non-controlled water sources, this is an indication that GAP are not implemented by some farms, demonstrating the violation of international agreements by the industry and the farms. Studies have suggested a relationship between drug abuse and compulsive behaviors. The present experiments investigated the relationship between schedule-induced polydipsia (SIP) and self-administration (SA) of ethanol and sucrose. SIP served as a model of compulsive behavior. and oral self-administration on a progressive-ratio (PR) schedule of reinforcement assessed the reinforcing value of either a 10% ethanol solution or an isocaloric sucrose solution. Rats first were exposed to PR sessions in which break points were the dependent variable and then switched to SIP sessions. with number of licks as the dependent variable. Results showed a positive relationship between PR and SIP for sucrose but not for ethanol: higher and lower PRs for sucrose were associated with higher and lower SIP levels. The order of the sessions then was reversed, such that SIP sessions were run before PR sessions. An opposite relationship was observed in which high and low SIP animals exhibited low and high PR break points, respectively. The relationship between SIP and SA was dependent on the reinforcing value of the substance and on prior SIP exposure. These results may reflect a common dopaminergic substrate and suggest that prior experience in coping with stress may reduce vulnerability to substance abuse behavior. Electronic states of a new molecular species, SiAs, correlating with the three lowest dissociation channels are characterized at a high-level of theory using the CASSCF/MRCI approach along with quintuple-xi quality basis sets. This characterization includes potential energy curves, vibrational energy levels, spectroscopic parameters, dipole and transition dipole moment functions, transition probabilities, and radiative lifetimes. For the ground state (X-2 Pi), an assessment of spin-orbit effects and the interaction with the close-lying A(2)Sigma(+) state is also reported. Similarities and differences with other isovalent species such as SiP and CAs are also discussed.
