Une étude récente auprès de 302 mathématiciens canadiens révèle un écart intriguant : tandis que 43% des sondés utilisent la programmation informatique dans leur recherche, seulement 18% indiquent qu'ils emploient cette technologie dans leur enseignement (Buteau et coll., 2014). La première donnée reflète le potentiel énorme qu'a la programmation pour faire et apprendre des mathématiques. La deuxième donnée a inspiré ce mémoire : pourquoi existe-t-il un tel écart ? Pour répondre à cette question, nous avons mené une étude exploratoire qui cherche à mieux comprendre la place de la programmation dans la recherche et la formation en mathématiques au niveau universitaire. Des entrevues semi-dirigées ont été conduites avec 14 mathématiciens travaillant dans des domaines variés et à différentes universités à travers le pays. Notre analyse qualitative nous permet de décrire les façons dont ces mathématiciens construisent des programmes informatiques afin d'accomplir plusieurs tâches (p.e., simuler des phénomènes réels, faire des mathématiques « expérimentales », développer de nouveaux outils puissants). Elle nous permet également d'identifier des moments où les mathématiciens exposent leurs étudiants à certains éléments de ces pratiques en recherche. Nous notons toutefois que les étudiants sont rarement invités à concevoir et à écrire leurs propres programmes. Enfin, nos participants évoquent plusieurs contraintes institutionnelles : le curriculum, la culture départementale, les ressources humaines, les traditions en mathématiques, etc. Quelques-unes de ces contraintes, qui semblent limiter l'expérience mathématique des étudiants de premier cycle, pourraient être revues.

Aquest projecte tracta d’un estudi tècnic, disseny i càlcul de les instal∙lacions d’un edifici mixte d’habitatges, local comercial i zona d’aparcament, complint la normativa que li pertoca segons l’ús a que es destina l’espai. L’edifici objecte d’aquest projecte es troba situat a la cruïlla Platja d’Aro s/n de la localitat de Santa Cristina d’Aro, i es tracta com un edifici de nova construcció. L’abast del projecte contempla les instal∙lacions elèctriques, tèrmiques, contra incendis, sanejament i de salubritat, per tal d’assolir les fases executives del projecte, així com les legalitzacions de les diferents activitats que es desenvolupen a l’edifici (aparcament de vehicles, comercial, taller de reparació de vehicles a motor i d’ús administratiu). Es pretén fer un estudi integral de les instal∙lacions i mesures correctores que es poden realitzar en un edifici amb les característiques esmentades

BACKGROUND: In order to maintain the most comprehensive structural annotation databases we must carry out regular updates for each proteome using the latest profile-profile fold recognition methods. The ability to carry out these updates on demand is necessary to keep pace with the regular updates of sequence and structure databases. Providing the highest quality structural models requires the most intensive profile-profile fold recognition methods running with the very latest available sequence databases and fold libraries. However, running these methods on such a regular basis for every sequenced proteome requires large amounts of processing power.In this paper we describe and benchmark the JYDE (Job Yield Distribution Environment) system, which is a meta-scheduler designed to work above cluster schedulers, such as Sun Grid Engine (SGE) or Condor. We demonstrate the ability of JYDE to distribute the load of genomic-scale fold recognition across multiple independent Grid domains. We use the most recent profile-profile version of our mGenTHREADER software in order to annotate the latest version of the Human proteome against the latest sequence and structure databases in as short a time as possible. RESULTS: We show that our JYDE system is able to scale to large numbers of intensive fold recognition jobs running across several independent computer clusters. Using our JYDE system we have been able to annotate 99.9% of the protein sequences within the Human proteome in less than 24 hours, by harnessing over 500 CPUs from 3 independent Grid domains. CONCLUSION: This study clearly demonstrates the feasibility of carrying out on demand high quality structural annotations for the proteomes of major eukaryotic organisms. Specifically, we have shown that it is now possible to provide complete regular updates of profile-profile based fold recognition models for entire eukaryotic proteomes, through the use of Grid middleware such as JYDE.

Hydroponic isotope labelling of entire plants (HILEP) is a cost-effective method enabling metabolic labelling of whole and mature plants with a stable isotope such as N-15. By utilising hydroponic media that contain N-15 inorganic salts as the sole nitrogen source, near to 100% N-15-labelling of proteins can be achieved. In this study, it is shown that HILEP, in combination with mass spectrometry, is suitable for relative protein quantitation of seven week-old Arabidopsis plants submitted to oxidative stress. Protein extracts from pooled N-14- and N-15-hydroponically grown plants were fractionated by SDS-PAGE, digested and analysed by liquid chromatography electrospray ionisation tandem mass spectrometry (LC-ESI-MS/MS). Proteins were identified and the spectra of N-14/N-15 peptide pairs were extracted using their m/z chromatographic retention time, isotopic distributions, and the m/z difference between the N-14 and N-15 peptides. Relative amounts were calculated as the ratio of the sum of the peak areas of the two distinct N-14 and N-15 peptide isotope envelopes. Using Mascot and the open source trans-proteomic pipeline (TPP), the data processing was automated for global proteome quantitation down to the isoform level by extracting isoform specific peptides. With this combination of metabolic labelling and mass spectrometry it was possible to show differential protein expression in the apoplast of plants submitted to oxidative stress. Moreover, it was possible to discriminate between differentially expressed isoforms belonging to the same protein family, such as isoforms of xylanases and pathogen-related glucanases (PR 2). Quantitative analysis by mass spectrometry (MS) is a major challenge in proteomics as the correlation between analyte concentration and signal intensity is often poor due to varying ionisation efficiencies in the presence of molecular competitors. However, relative quantitation methods that utilise differential stable isotope labelling and mass spectrometric detection are available. Many drawbacks inherent to chemical labelling methods (ICAT, iTRAQ) can be overcome by metabolic labelling with amino acids containing stable isotopes (e.g. 13C and/or 15N) in methods such as Stable Isotope Labelling with Amino acids in Cell culture (SILAC). SILAC has also been used for labelling of proteins in plant cell cultures (1) but is not suitable for whole plant labelling. Plants are usually autotrophic (fixing carbon from atmospheric CO2) and, thus, labelling with carbon isotopes becomes impractical. In addition, SILAC is expensive. Recently, Arabidopsis cell cultures were labelled with 15N in a medium containing nitrate as sole nitrogen source. This was shown to be suitable for quantifying proteins and nitrogen-containing metabolites from this cell culture (2,3). Labelling whole plants, however, offers the advantage of studying quantitatively the response to stimulation or disease of a whole multicellular organism or multi-organism systems at the molecular level. Furthermore, plant metabolism enables the use of inexpensive labelling media without introducing additional stress to the organism. And finally, hydroponics is ideal to undertake metabolic labelling under extremely well-controlled conditions. We demonstrate the suitability of metabolic 15N hydroponic isotope labelling of entire plants (HILEP) for relative quantitative proteomic analysis by mass spectrometry. To evaluate this methodology, Arabidopsis plants were grown hydroponically in 14N and 15N media and subjected to oxidative stress.

Although Huntington's disease (HD) is a neurodegenerative disease characterized by motor, cognitive and behavioural disturbances, there has been little empirical data examining what patients are most concerned about throughout the different stages of disease, which can span many years. Semi-structured face-to-face interviews were individually conducted with 31 people living with different stages of Huntington's, from pre-clinical gene carriers to advanced stage. We examined how often participants raised issues and concerns regarding the impact of Huntington's on everyday life. The Physical/functional theme hardly featured pre-clinically, but was strongly present from Stage 1, rose steadily and peaked at Stage 5. There were no significant changes between stages for the Emotional, Social, and Self themes that all featured across all stages, indicating that these issues were not raised more frequently over the course of the disease. Likewise, the more rarely mentioned Financial and Legal themes also remained similar across stages. However, the Cognitive theme only featured between Stages 1 and 4, and hardly at all pre-clinically and at Stage 5. These findings provide insight into patients' important and unique perspective and have implications for the management and development of interventions across the spectrum of HD stages.

The ten-volume edition of The Collected Works of Thomas Heywood, forthcoming from Oxford University Press from 2015 to 2022, will attempt to place Heywood’s plays, poetry, and prose back where they belong: at the centre of the study of early modern English literature, drama, and theatre history. Especially as an actor, playwright, reviser, editor, and historical chronicler, Heywood had the longest and widest-ranging career of his contemporaries and thus can reveal how sixteenth- and seventeenth-century authors and theatrical and literary audiences came to see the practice and production of drama.

Motivated by the recent solution of Karlin's conjecture, properties of functions in the Laguerre-Polya class are investigated. The main result of this paper establishes new moment inequalities fur a class of entire functions represented by Fourier transforms. The paper concludes with several conjectures and open problems involving the Laguerre-Polya class and the Riemann xi -function.
