Type I diabetes is a disease caused by autoimmune destruction of the beta cells in the pancreas that leads to a deficiency in insulin production. The aim of this study was to evaluate the prophylactic potential of a prime-boost strategy involving bacille Calmette-Guérin (BCG) and the pVAXhsp65 vaccine (BCG/DNAhsp65) in diabetes induced by streptozotocin (STZ) in C57BL/6 mice and also in spontaneous type 1 diabetes in non-obese diabetic (NOD) mice. BCG/DNAhsp65 vaccination in NOD mice determined weight gain, protection against hyperglycaemia, decreased islet inflammation, higher levels of cytokine production by the spleen and a reduced number of regulatory T cells in the spleen compared with non-immunized NOD mice. In the STZ model, however, there was no significant difference in the clinical parameters. Although this vaccination strategy did not protect mice in the STZ model, it was very effective in NOD mice. This is the first report demonstrating that a prime-boost strategy could be explored as an immunomodulatory procedure in autoimmune diseases. © 2013 British Society for Immunology.

Context. The Sun shows abundance anomalies relative to most solar twins. If the abundance peculiarities are due to the formation of inner rocky planets, that would mean that only a small fraction of solar type stars may host terrestrial planets. Aims. In this work we study HIP 56948, the best solar twin known to date, to determine with an unparalleled precision how similar it is to the Sun in its physical properties, chemical composition and planet architecture. We explore whether the abundances anomalies may be due to pollution from stellar ejecta or to terrestrial planet formation. Methods. We perform a differential abundance analysis (both in LTE and NLTE) using high resolution (R similar to 100 000) high S/N (600-650) Keck HIRES spectra of the Sun (as reflected from the asteroid Ceres) and HIP 56948. We use precise radial velocity data from the McDonald and Keck observatories to search for planets around this star. Results. We achieve a precision of sigma less than or similar to 0.003 dex for several elements. Including errors in stellar parameters the total uncertainty is as low as sigma similar or equal to 0.005 dex (1%), which is unprecedented in elemental abundance studies. The similarities between HIP 56948 and the Sun are astonishing. HIP 56948 is only 17 +/- 7 K hotter than the Sun, and log g, [Fe/H] and microturbulence velocity are only +0.02 +/- 0.02 dex, +0.02 +/- 0.01 dex and +0.01 +/- 0.01 km s(-1) higher than solar, respectively. Our precise stellar parameters and a differential isochrone analysis shows that HIP 56948 has a mass of 1.02 +/- 0.02 M-circle dot and that it is similar to 1 Gyr younger than the Sun, as constrained by isochrones, chromospheric activity, Li and rotation. Both stars show a chemical abundance pattern that differs from most solar twins, but the refractory elements (those with condensation temperature T-cond greater than or similar to 1000 K) are slightly (similar to 0.01 dex) more depleted in the Sun than in HIP 56948. The trend with T-cond in differential abundances (twins -HIP 56948) can be reproduced very well by adding similar to 3 M-circle plus of a mix of Earth and meteoritic material, to the convection zone of HIP 56948. The element-to-element scatter of the Earth/meteoritic mix for the case of hypothetical rocky planets around HIP 56948 is only 0.0047 dex. From our radial velocity monitoring we find no indications of giant planets interior to or within the habitable zone of HIP 56948. Conclusions. We conclude that HIP 56948 is an excellent candidate to host a planetary system like our own, including the possible presence of inner terrestrial planets. Its striking similarity to the Sun and its mature age makes HIP 56948 a prime target in the quest for other Earths and SETI endeavors.

Evaluation of: Rodriguez D, Gonzalez-Aseguinolaza G, Rodriguez JR et al. Vaccine efficacy against malaria by the combination of porcine parvovirus-like particles and vaccinia virus vectors expressing CS of Plasmodium. PLoS ONE 7(4), e34445 (2012). Recently, a vaccine against malaria was successfully tested in a human Phase III trial. The efficacy of this vaccine formulation, based on the Plasmodium falciparum circumsporozoite protein, was approximately 50% and correlated with the presence of antibodies specific to the infective stages of the malaria parasites. Different strategies are being pursued to improve vaccine efficacy levels. One such strategy is the induction of specific cytotoxic T cells that can destroy the intracellular hepatocyte stages of the malaria parasite. In this study, a novel vaccination protocol was developed to elicit strong immune responses mediated by CD8(+) cytotoxic cells specific to the circumsporozoite protein. As proof-of-concept, the authors used the rodent malaria Plasmodium yoelii parasite. The vaccination strategy consisted of a heterologous prime-boost vaccination regimen involving porcine parvovirus-like particles for priming and the modified vaccinia virus Ankara for the booster immunization, both of which expressed the immunodominant CD8 epitope of the P. yoelii circumsporozoite protein. Results from this experimental model were extremely meaningful. This vaccination strategy led to a significant T-cell immune response mediated by CD8(+) multifunctional T effector and effector-memory cells. However, most importantly for the malaria vaccine development was the fact that following a sporozoite challenge, immunized mice eliminated more than 97% of the malaria parasites during the hepatocyte stages. These results confirm and extend a vast body of knowledge showing that a heterologous prime-boost vaccination strategy can elicit strong CD8(+) T-cell-mediated protective immunity and may increase the efficacy of malaria vaccines.

The present work is part of a larger project aimed at obtaining compounds of industrial interest from renewable sources. The work is particularly aimed to investigate the reactivity of 2,5-bis-hydroxymethylfuran (BHMF), an important building block of organic nature easily obtainable from biomass, with acid catalysis. Through the study of the reactivity of BHMF in water, in the presence of an heterogeneous acid catalyst (Amberlyst 15), has been developed a new synthetic method for the preparation of α-6-hydroxy-6-methyl-4-enyl-2H-pyran-3-one a derivative whose molecular skeleton is similar to that of natural products which are used in pharmaceutical chemistry. The product is obtained in milder conditions and with better selectivity with respect to the strongly oxidizing conditions with which it is prepared in the literature starting from different precursors containing furan ring.

Il recupero dei materiali di scarto è un aspetto di grande attualità in campo stradale, così come negli altri ambiti dellâingegneria civile. Lâattenzione della ricerca e degli esperti del settore è rivolta allâaffinamento di tecniche di riciclaggio che riducano lâimpatto ambientale senza compromettere le prestazioni meccaniche finali. Tali indagini cercano di far corrispondere le necessità di smaltimento dei rifiuti con quelle dellâindustria infrastrutturale, legate al reperimento di materiali da costruzione tecnicamente idonei ed economicamente vantaggiosi. Attualmente sono già diversi i tipi di prodotti rigenerati e riutilizzati nella realizzazione delle pavimentazioni stradali e numerosi sono anche quelli di nuova introduzione in fase di sperimentazione. In particolare, accanto ai materiali derivanti dalle operazioni di recupero della rete viaria, è opportuno considerare anche quelli provenienti dallâesercizio delle attività di trasporto, il quale comporta ogni anno il raggiungimento della fine della vita utile per centinaia di migliaia di tonnellate di pneumatici di gomma. Lâobiettivo della presente analisi sperimentale è quello di fornire indicazioni e informazioni in merito alla tecnica di riciclaggio a freddo con emulsione bituminosa e cemento, valutando la possibilità di applicazione di tale metodologia in combinazione con il polverino di gomma, ottenuto dal recupero degli pneumatici fuori uso (PFU). La ricerca si distingue per una duplice valenza: la prima è quella di promuovere ulteriormente la tecnica di riciclaggio a freddo, che si sta imponendo per i suoi numerosi vantaggi economici ed ambientali, legati soprattutto alla temperatura dâesercizio; la seconda è quella di sperimentare lâutilizzo del polverino di gomma, nelle due forme di granulazione tradizionale e criogenica, additivato a miscele costituite interamente da materiale proveniente da scarifica di pavimentazioni esistenti e stabilizzate con diverse percentuali di emulsione di bitume e di legante cementizio.
