This study evaluated the influence of bone marrow aspirate (BMA), low-level laser therapy (LLLT) and their combination on bone healing in surgically created critical-size defects (CSDs) in rat calvaria. 40 rats were divided into four groups: C (control), BMA, LLLT and BMA/LLLT. A 5 mm diameter CSD was created in the calvarium of each animal. In Group C, the defect was filled by blood clot only. In Group BMA, the defect was filled with BMA. In groups LLLT and BMA/LLLT, the defect received laser irradiation (InGaAlP laser), was filled with blood clot or BMA respectively, and irradiated again. Animals were euthanized 30 days postoperatively. Histomorphometric and immunohistochemical analyses were performed. Newly formed bone area (NFBA) was calculated as percentage of the total area of the original defect. Proliferating cell nuclear antigen (PCNA), runt-related transcription factor 2 (Runx2) and osteocalcin (OCN) immunohistochemical staining were performed. PCNA-positive, Runx2-positive and OCN-positive cells were quantified. Data were statistically analyzed. Group BMA/LLLT had significantly greater NFBA than groups C, BMA or LLLT. Group BMA presented significantly greater NFBA than control, while group LLLT did not. Group BMA/LLLT presented a significantly higher number of PCNA-positive and OCN-positive cells than any of the other groups. Groups BMA/LLLT and BMA showed a significantly lower number of Runx2-positive cells than groups C or LLLT. The combination of BMA/LLLT yielded significantly greater bone formation in surgically created CSD in rat calvaria when compared to control, or either treatment alone. © 2013 Elsevier B.V. This study aimed to evaluate the dynamic factor applied to the results obtained by dynamic resistance formulas Janbu and Hiley, would lead to the results obtained by static strength analysis CAPWAPC ®. The evaluation was done by backcalculation dynamic loading tests (ECD), taking into consideration the type of soil in which the cutting base precast concrete were settled, and using the information on the effective energy transmitted to the stakes by hammers, obtained by CAPWAPC ®. The results are shown in tables and graphs, and showed that the use of these formulations and their dynamic factors can become an efficient and economical field, assisting the engineer in making decisions regarding the staking of the work

Osteosarcoma is the most common primary bone cancer in dogs. It affects most commonly dogs of big or giant breeds with 7 to 8 years and the etiology is unknown. Osteosarcoma is defined as a bone matrix-producing malignant mesenchymal tumor and has a predilection for the metaphyseal region of appendicular skeleton, however, it can affect axial skeleton and soft tissues. Distal radius is the most commonly affected site. The definitive diagnosis of osteosarcoma can be obtained with history, physical examination, radiographs and biopsy. Lung is the most common organ for metastatic disease. The mainly treatment for osteosarcoma is limb amputation and systemic chemotherapy for metastatic disease control. Limb-sparing surgery is a viable alternative to amputation for dogs with concomitant conditions that impede limb amputation. Palliative treatments for osteosarcoma have been studied such as local and systemic radiotherapy, immunotherapy and biphosphonates. This study has the objective of presenting the aspects of diagnosis and treatment for appendicular osteosarcoma

The bone mineral density increments in patients with sporadic primary hyperparathyroidism after parathyroidectomy have been studied by several investigators, but few have investigated this topic in primary hyperparathyroidism associated with multiple endocrine neoplasia type 1. Further, as far as we know, only two studies have consistently evaluated bone mineral density values after parathyroidectomy in cases of primary hyperparathyroidism associated with multiple endocrine neoplasia type 1. Here we revised the impact of parathyroidectomy (particularly total parathyroidectomy followed by autologous parathyroid implant into the forearm) on bone mineral density values in patients with primary hyperparathyroidism associated with multiple endocrine neoplasia type 1. Significant increases in bone mineral density in the lumbar spine and femoral neck values were found, although no short-term (15 months) improvement in bone mineral density at the proximal third of the distal radius was observed. Additionally, short-term and medium-term calcium and parathyroid hormone values after parathyroidectomy in patients with primary hyperparathyroidism associated with multiple endocrine neoplasia type 1 are discussed. In most cases, this surgical approach was able to restore normal calcium/parathyroid hormone levels and ultimately lead to discontinuation of calcium and calcitriol supplementation.

La tesi tratta di strumenti finalizzati alla valutazione dello stato conservativo e di supporto all'attività di manutenzione dei ponti, dai più generali Bridge Management Systems ai Sistemi di Valutazione Numerica della Condizione strutturale. Viene proposto uno strumento originale con cui classificare i ponti attraverso un Indice di Valutazione Complessiva e grazie ad esso stabilire le priorità d'intervento. Si tara lo strumento sul caso pratico di alcuni ponti della Provincia di Bologna. Su un ponte in particolare viene realizzato un approfondimento specifico sulla determinazione approssimata dei periodi propri delle strutture da ponte. Si effettua un confronto dei risultati di alcune modellazioni semplificate in riferimento a modellazioni dettagliate e risultati sperimentali.

In der vorliegenden Arbeit wurde die Druckabhängigkeit der molekularen Dynamik mittels 2H-NMR und Viskositätsmessungen untersucht. Für die Messungen wurde der niedermolekulare organische Glasbildner ortho-Terphenyl (OTP) ausgewählt, da dieser aufgrund einer Vielzahl vorliegender Arbeiten als Modellsubstanz angesehen werden kann. Daneben wurden auch Messungen an Salol durchgeführt.Die Untersuchungen erstreckten sich über einen weiten Druck- und Temperaturbereich ausgehend von der Schmelze bis weit in die unterkühlte Flüssigkeit. Dieser Bereich wurde aufgrund experimenteller Voraussetzungen immer durch eine Druckerhöhung erreicht.Beide Substanzen zeigten druckabhängig ein Verhalten, das dem der Temperaturvariation bei Normaldruck sehr ähnelt. Auf einer Zeitskala der molekularen Dynamik von 10E-9 s bis zu 10E+2 s wurde daher am Beispiel von OTP ein Druck-Temperatur-Zeit-Superpositionsprinzip diskutiert. Zudem konnte eine Temperatur-Dichte-Skalierung mit rho T-1/4 erfolgreich durchgeführt werden. Dies entspricht einem rein repulsiven Potentialverlauf mit rho -12±3 .Zur Entscheidung, ob die Verteilungsbreiten der mittleren Rotationskorrelationszeiten durch Druckvariation beeinflußt werden, wurden auch Ergebnisse anderer experimenteller Methoden herangezogen. Unter Hinzuziehung aller Meßergebnisse kann sowohl eine Temperatur- als auch Druckabhängigkeit der Verteilungsbreite bestätigt werden. Zur Auswertung von Viskositätsdaten wurde ein Verfahren vorgestellt, das eine quantitative Aussage über den Fragilitätsindex von unterkühlten Flüssigkeiten auch dann zuläßt, wenn die Messungen nicht bis zur Glasübergangstemperatur Tg durchgeführt werden. Die Auswertung der druckabhängigen Viskositätsdaten von OTP und Salol zeigt einen sehr differenzierten druckabhängigen Verlauf des Fragilitätsindexes für beide Glasbildner. OTP zeigt zunächst eine leichte Abnahme und danach wieder eine Zunahme des Fragilitätsindexes, dieses Ergebnis wird auch von Simulationsdaten, die der Literatur entnommen wurden, unterstützt. Salol hingegen zeigt zunächst eine deutliche Zunahme und danach eine Abnahme des Fragilitätsindexes. Das unterschiedliche Verhalten der beiden Glasbildner mit ähnlichem Fragilitätsindex bei Normaldruck wird auf die Wasserstoffbrückenbindungen innerhalb von Salol zurückgeführt.

Delayed fracture healing and non-unions represent rare but severe complications in orthopedic surgery. Further knowledge on the mechanisms of the bone repair process and of the development of a pseudoarthrosis is essential to predict and prevent impaired healing of fractures. The present study aimed at elucidating differences in gene expression during the repair of rigidly and non-rigidly fixed osteotomies. For this purpose, the MouseFix™ and the FlexiPlate™ systems (AO Development Institute, Davos, CH), allowing the creation of well defined osteotomies in mouse femora, were employed. A time course following the healing process of the osteotomy was performed and bones and periimplant tissues were analyzed by high-resolution X-ray, MicroCT and by histology. For the assessment of gene expression, Low Density Arrays (LDA) were done. In animals with rigid fixation, X-ray and MicroCT revealed healing of the osteotomy within 3 weeks. Using the FlexiPlate™ system, the osteotomy was still visible by X-ray after 3 weeks and a stabilizing cartilaginous callus was formed. After 4.5 weeks, the callus was remodeled and the osteotomy was, on a histological level, healed. Gene expression studies revealed levels of transcripts encoding proteins associated with inflammatory processes not to be altered in tissues from bones with rigid and non-rigid fixation, respectively. Levels of transcripts encoding proteins of the extracellular matrix and essential for bone cell functions were not increased in the rigidly fixed group when compared to controls without osteotomy. In the FlexiPlate™ group, levels of transcripts encoding the same set of genes were significantly increased 3 weeks after surgery. Expression of transcripts encoding BMPs and BMP antagonists was increased after 3 weeks in repair tissues from bones fixed with FlexiPlate™, as were inhibitors of the WNT signaling pathways. Little changes only were detected in transcript levels of tissues from rigidly fixed bones. The data of the present study suggest that rigid fixation enables accelerated healing of an experimental osteotomy as compared to non-rigid fixation. The changes in the healing process after non-rigid fixation are accompanied by an increase in the levels of transcripts encoding inhibitors of osteogenic pathways and, probably as a consequence, by temporal changes in bone matrix synthesis.

OBJECTIVES: The aim of the here described case series was to develop and evaluate the minimally invasive percutaneous osteosynthesis for the plate fixation of tibial fractures in dogs and cats. METHODS: Six dogs and four cats with shaft fractures of the tibia were treated using minimally invasive percutaneous osteosynthesis. Follow-up radiographs four to six weeks after fracture fixation were evaluated for fracture healing. For the long-term follow-up (minimum 2.4 years), owners were contacted by phone to complete a questionnaire. RESULTS: All fractures healed without the need for a second procedure. Follow-up radiographs obtained after four to six weeks in seven cases showed advanced bony healing with callus formation and filling of the fracture gaps with calcified tissue in all seven. All the patients had a good to excellent long-term result with full limb function. The time needed for regaining full limb use was two to three months. CLINICAL SIGNIFICANCE: Minimally invasive percutaneous osteosynthesis seems to be a useful technique for the treatment of tibial shaft fractures in dogs and cats.

INTRODUCTION: Angiogenesis is known to be a critical and closely regulated step during bone formation and fracture healing driven by a complex interaction of various cytokines. Delays in bone healing or even nonunion might therefore be associated with altered concentrations of specific angiogenic factors. These alterations might in turn be reflected by changes in serum concentrations. METHOD: To determine physiological time courses of angiogenic cytokines during fracture healing as well as possible changes associated with failed consolidation, we prospectively collected serum samples from patients who had sustained surgical treatment for a long bone fracture. Fifteen patients without fracture healing 4 months after surgery (nonunion group) were matched to a collective of 15 patients with successful healing (union group). Serum concentrations of angiogenin (ANG), angiopoietin 2 (Ang-2), basic fibroblast growth factor (bFGF), platelet derived growth factor AB (PDGF-AB), pleiotrophin (PTN) and vascular endothelial growth factor (VEGF) were measured using enzyme linked immunosorbent assays over a period of 24 weeks. RESULTS: Compared to reference values of healthy uninjured controls serum concentrations of VEGF, bFGF and PDGF were increased in both groups. Peak concentrations of these cytokines were reached during early fracture healing. Serum concentrations of bFGF and PDGF-AB were significantly higher in the union group at 2 and 4 weeks after the injury when compared to the nonunion group. Serum concentrations of ANG and Ang-2 declined steadily from the first measurement in normal healing fractures, while no significant changes over time could be detected for serum concentrations of these factures in nonunion patients. PTN serum levels increased asymptotically over the entire investigation in timely fracture healing while no such increase could be detected during delayed healing. CONCLUSION: We conclude that fracture healing in human subjects is accompanied by distinct changes in systemic levels of specific angiogenic factors. Significant alterations of these physiologic changes in patients developing a fracture nonunion over time could be detected as early as 2 (bFGF) and 4 weeks (PDGF-AB) after initial trauma surgery.
