We aimed to evaluate the effectiveness and safety of bismuth-containing quadruple therapy plus postural change after dosing for Helicobacter pylori eradication in gastrectomized patients. We compared 76 gastric stump patients with H. pylori infection (GS group) with 50 non-gastrectomized H. pylori-positive patients who met the treatment indication (controls). The GS group was divided into GS group 1 and GS group 2. All groups were administered bismuth potassium citrate (220 mg), esomeprazole (20 mg), amoxicillin (1.0 g), and furazolidone (100 mg) twice daily for 14 days. GS group 1 maintained a left lateral horizontal position for 30 min after dosing. H. pylori was detected using rapid urease testing and histologic examination of gastric mucosa before and 3 months after therapy. Mucosal histologic manifestations were evaluated using visual analog scales of the updated Sydney System. GS group 1 had a higher prevalence of eradication than the GS group 2 (intention-to-treat [ITT]: P=0.025; per-protocol [PP]: P=0.030), and the control group had a similar prevalence. GS group 2 had a lower prevalence of eradication than controls (ITT: P=0.006; PP: P=0.626). Scores for chronic inflammation and activity declined significantly (P<0.001) 3 months after treatment, whereas those for atrophy and intestinal metaplasia showed no significant change. Prevalence of adverse reactions was similar among groups during therapy (P=0.939). A bismuth-containing quadruple therapy regimen plus postural change after dosing appears to be a relatively safe, effective, economical, and practical method for H. pylori eradication in gastrectomized patients.

PHEX est une protéine importante dans le processus de minéralisation osseuse. Des mutations ou la délétion d’une partie de ce gène causent l’hypophosphatémie liée au chromosome X (XLH). Cette maladie est caractérisée par une hypophosphatémie, accompagnée de défauts de minéralisation, de rachitisme et de lésions ostéomalaciques. Avec l’hypophosphatémie, les taux circulants de vitamine D devraient être augmentés, ce qui n’est pas le cas d’où une régulation anormale de la production de vitamine D a lieu. Cependant, malgré le fait que cette protéine soit une peptidase, aucun substrat physiologique n’a encore été répertorié pour PHEX. PHEX est une protéine membranaire de type II de la famille M13 des métalloendopeptidases à zinc possédant un court domaine N-terminal cytosolique, un segment transmembrannaire d’environ 20 acides aminés et une large portion C-terminale extracellulaire où se trouve le site actif de l’enzyme. PHEX est exprimée de façon majoritaire dans les os et dans les dents et elle apparaît à l’initiation de la minéralisation. Les patients souffrant de XLH et la souris Hyp, qui est un modèle animal de la maladie humaine, montrent des quantités importantes de la protéine FGF23. De plus, FGF23 est impliqué dans une autre maladie reliée au métabolisme du phosphate, l’hypophosphatémie rachitique autosomale dominante (ADHR) où des mutations de FGF23 causent sensiblement les mêmes symptômes que XLH. FGF23 est produit principalement par les ostéoblastes et les ostéocytes. FGF23 cause une hypophosphatémie par la diminution de l’expression du cotransporteur NaPi de type II, responsable de la réabsorption du phosphate rénal. L’hypothèse proposée dans la littérature serait que PHEX activerait ou inactiverait des peptides importants pour la minéralisation osseuse. Plus spécifiquement, l’activation ou l’inactivation de ces peptides aurait pour rôle de réguler les quantités de FGF23. Selon l’hypothèse mentionnée précédemment, la régulation de PHEX pourrait donc avoir un effet sur la minéralisation. Une quantité croissante de données sur la régulation de PHEX sont maintenant disponibles. Par exemple, la vitamine D diminue l’expression de PHEX tandis que les glucocorticoïdes et l’hormone de croissance augmentent son expression. Dans une première étude, nous avons voulu déterminer si un peptide relié à la minéralisation osseuse, le PTHrP1-34, pouvait réguler l’expression de PHEX. Nous avons déterminé que le PTHrP1-34 peut réguler de façon négative l’expression de PHEX dans les cellules UMR-106, une lignée cellulaire ostéoblastique. Cette régulation passe par la voie de l’AMPc/protéine kinase A. De plus, cette diminution d’expression est également observée au jour 7 dans des cultures primaires d’ostéoblastes de rat en minéralisation. Par la suite, nous avons étudié un mutant de PHEX, le mutant E4Q retrouvé chez un patient souffrant de XLH, où la mutation se retrouve dans le domaine cytosolique de PHEX. Cette mutation n’interfère pas avec le site catalytique de l’enzyme puisque ce mutant de PHEX peut tout aussi bien cliver un substrat synthétique que la protéine sauvage. Il a été déterminé que cette mutation annule un motif di-acide. Nous avons démontré que ce motif di-acide est responsable de la liaison de PHEX à COPII, responsable de la formation de vésicules de sécrétion. De plus, il semblerait que ce motif soit important, probablement par son interaction avec COPII, à l’incorporation de PHEX dans des vésicules de calcification, lesdites vésicules étant importantes dans le processus de minéralisation. Finalement, des essais de compétitions ont démontré que la minéralisation pouvait être perturbée lorsque l’on surexprimait la queue cytosolique sauvage de PHEX, contrairement à la queue mutée. Ceci suggère possiblement que l’interaction avec COPII menant à l’incorporation de PHEX dans les vésicules de calcification ou d’autres protéines comprenant de tels motifs pourrait être importante pour la minéralisation. Finalement, la dernière étude porte sur la protéine FGF23. Nous avons démontré, par la surexpression de FGF23 dans la lignée MC3T3 d’ostéoblastes de souris, que cette surexpression a un effet sur la sénescence de ces cellules. En effet, des essais de sénescence ont montré l’augmentation de celle-ci lorsque FGF23 est surexprimé. Par contre, la prolifération n’est pas altérée. De plus, il semblerait que la différenciation soit plus rapide, tel qu’observé par une minéralisation survenant plus tôt, mais n’étant pas plus importante. Bref, la surexpression de FGF23 semblerait faire en sorte que les ostéoblastes se différencient plus rapidement et passent donc à un état de sénescence prématuré comparativement aux cellules sauvages. Ceci est en accord avec la littérature où KLOTHO, un cofacteur de FGF23 permettant sa liaison avec une plus grande affinité sur son récepteur, lorsqu’inactivé démontre un phénotype similaire au vieillissement incluant un phénotype de sénescence.

Background: The aim of this study is to verify the regenerative potential of particulate anorganic bone matrix synthetic peptide-15 (ABM-P-15) in class III furcation defects associated or not with expanded polytetrafluoroethylene membranes. Methods: Class III furcation defects were produced in the mandibular premolars (P2, P3, and P4) of six dogs and filled with impression material. The membranes and the bone grafts were inserted into P3 and P4, which were randomized to form the test and control groups, respectively; P2 was the negative control group. The animals were sacrificed 3 months post-treatment. Results: Histologically, the complete closure of class III furcation defects was not observed in any of the groups. Partial periodontal regeneration with similar morphologic characteristics among the groups was observed, however, through the formation of new cementum, periodontal ligament, and bone above the notch. Histologic analysis showed granules from the bone graft surrounded by immature bone matrix and encircled by newly formed tissue in the test group. The new bone formation area found in the negative control group was 2.28 +/- 2.49 mm(2) and in the test group it was 6.52 +/- 5.69 mm(2), which showed statistically significant differences for these groups considering this parameter (Friedman test P <0.05). There was no statistically significant difference among the negative control, control, and test groups for the other parameters. Conclusions: The regenerative potential of ABM-P-15 was demonstrated through new bone formation circumscribing and above the graft particles. The new bone also was accompanied by the formation of new cementum and periodontal ligament fibers. J Periodontol 2010;81:594-603.

Background: Recent studies in animals have shown pronounced resorption of buccal bone plate after immediate implantation. The sectioning of experimental material for histologic evaluation of the bone plates could provide valuable information about the possible effect of bone exposure in periodontal and implant surgeries. Methods: Twenty-four incisors were collected from dogs. After decalcification, the blocks were immersed in paraffin and bucco-lingual histologic sections were examined under light microscope. Some sections were reserved for immunohistochemical analysis. Results: The bone density, the width of the bone plates, and the percentage of vessels presented in the periodontal ligament and periosteum were analyzed in the buccal and lingual bone plates, which were divided corono-apically into thirds. The buccal bone plates showed statistically higher bone density compared to the lingual bone plates in the coronal thirds. The width of both bone plates increased from the coronal to the apical third, but all the buccal thirds were significantly thinner compared to the lingual thirds. No statistically significant differences were found between the bone plates for the percentage of area occupied by the blood vessels in the periodontal ligament or periosteum. Conclusion: It is reasonable to conclude that the higher bone density, represented by the lower number of marrow spaces, in association with the thinner aspect of the buccal bone plates made them more fragile to absorb compared to the lingual bone plates, especially during mucoperiosteal procedures. This study investigated the immunodetection of PCNA in epithelial components of dental follicles associated with impacted third molars without radiographical and morphological signs of pathosis. A total of 105 specimens of dental follicles associated with impacted third molars with incomplete rhizogenesis (between Nolla`s stage 6 and 9) were surgically removed from 56 patients. Epithelial cell proliferating was determined by using immunohistochemical labeling. Statistical analysis was performed using the Fisher exact test. Of the 105 dental follicles collected, 6 were PCNA-positive (approximate to 6%). The specimens with squamous metaplasia and epithelial hyperplasia had higher rates of positivity for PCNA, as well as those with proliferative remnants of odontogenic epithelium. In conclusion, this study shows that dental follicles at this stage of development have low proliferative potential, but suggests that squamous metaplasia, hyperplasia of the epithelial lining and presence of proliferative odontogenic epithelial rests in the connective tissue may be early signs of developing lesions of odontogenic origin.

Sabe-se que o refluxo crônico pode induzir lesão mucosa, estimular a proliferação de células e promover tumorigênese no esôfago distal. Ainda não é sabido por que apenas uma parcela dos pacientes com refluxo esofágico progrediram para uma metaplasia intestinal (Esôfago de Barrett) e adenocarcinoma. O estresse oxidativo parece exercer um papel importante nessa progressão . Assim sendo, examinamos o padrão de expressão da enzima Heme Oxigenase-1 (HO-1), enzima indutora do estresse oxidativo, em peças de esôfago obtidos de um estudo experimental com ratos que avaliou o papel do refluxo gástrico e duodenoesofágico na carcinogênese esofágica. Métodos: Uma amostra de três (3) peças de esôfago de cada grupo de ratos submetidos a tratamentos diferentes tiveram a expressão da enzima Heme Oxigenase-1 avaliada através de imunohistoquímica. Os ratos foram divididos nos seguintes grupos: (1) cardioplastia para induzir refluxo predominantemente gástrico, (2) anastomose esofagoduodenal para induzir refluxo duodenal, (3) sem tratamento, (4) cardioplastia+dietilnitrosamina (DEN), (5) anastomose esofagoduodenal +DEN, (6) DEN. Resultados: Não houve desenvolvimento de câncer ou metaplasia intestinal nos animais que não foram expostos ao refluxo de conteúdo duodenal. A expressão de HO-1 foi observada apenas em ratos submetidos à anastomose esofagoduodenal (Grupos 2 e 5) e a análise da média de intensidade de fluorescência demonstrou uma diferença significante de expressão de HO-1 (4,8 e 4,6 vezes respectivamente) comparando-se ao controle (Grupo 3) (p<0,05). O alvo principal para expressão da HO-1 foram as células inflamatórias dentro do tumor ou em áreas subepiteleiais. Os ratos expostos ao refluxo gástrico não desenvolveram tumores ou expressaram a HO-1. Conclusões: A esofagite de refluxo induzida por refluxo esofágico com conteúdo duodenal provocou estresse oxidativo considerável e pode desempenhar um papel importante na carcinogênese esofágica. O refluxo gástrico não foi suficiente para induzir estresse oxidativo neste modelo experimental.

Existem fortes evidências de que os programas de rastreamento baseados em citologia resultaram em diminuição significativa da incidência e mortalidade por câncer do colo do útero, no entanto, um excesso substancial de tratamento de lesões intraepiteliais de baixo grau que dificilmente progrediriam para carcinoma cervical resulta da baixa especificidade do tradicional rastreio citológico. A detecção precoce das lesões através do rastreamento citológico e a avaliação do grau histológico em espécimes cervicais são fundamentais, entretanto não permitem identificar quais pacientes terão maior probabilidade de progressão para lesões de alto grau e carcinoma invasivo. A busca de potenciais marcadores de prognóstico; objetivando o entendimento da progressão das lesões intraepiteliais é de suma importância. Acredita-se que fatores imunoregulatórios, imunogenéticos e proteínas do ciclo celular estejam intimamente envolvidos no processo de carcinogênese. Considerando o exposto, a proposta do projeto foi inicialmente avaliar a eficácia da citologia oncológica no rastreamento do câncer cervical, foi investigado ainda o polimorfismo do gene do fator de transcrição FOXP3 e a expressão da proteína do ciclo celular P63 (P63) associados respectivamente a diagnóstico e prognóstico das lesões cervicais. Em um primeiro momento foi realizado estudo transversal que envolveu 3194 mulheres. As participantes foram submetidas à citologia e biópsia de colo dirigida por colposcopia e os resultados foram comparados para verificar-se a acurácia do teste de Papanicolaou na detecção de lesões intraepiteliais e câncer cervical. Posteriormente, realizou-se estudo comparativo do tipo observacional estratificado em três grupos: Grupo 1: 16 casos com diagnóstico histopatológico de metaplasia/cervicite, considerados normais, Grupo 2: 11 casos com lesão de baixo grau (LSIL) e Grupo 3: 15 casos com lesão de alto grau (HSIL) ou carcinoma epidermoide de colo. Um total de 42 participantes respondeu a um questionário epidemiológico padronizado sobre as características demográficas, hábitos pregressos, história reprodutiva e de comportamento sexual. Após exame colposcópico, foram coletados fragmentos de espécimes cervicais para a pesquisa da expressão proteica da P63 por imunohistoquímica. Amostras de sangue periférico foram coletadas para extração do DNA e detecção do polimorfismo do gene FOXP3. No primeiro estudo em que se avaliou a acurácia do teste de Papanicolaou, encontrou-se sensibilidade de 0,83, valor preditivo positivo (VPP) de 0,77 e especificidade de 0,23 no rastreamento das lesões cervicais e câncer de colo. viii Melhores resultados foram observados quando se avaliou a acurácia diagnóstica para lesões de alto grau e carcinoma com VPP de 0,99 e especificidade de 0,84.No estudo subsequente onde se comparou a expressão da proteína P63 observou-se maior número de núcleos marcados no grupo com lesões intraepiteliais de alto grau e câncer quando comparado ao grupo com biópsias negativas (p=0,0004). No último estudo pesquisou-se a associação do polimorfismo do gene FOXP3 com lesões intraepiteliais cervicais sendo evidenciada maior prevalência do genótipo heterozigoto, CT, no grupo com lesões de colo na histopatologia (p=0,027). Mulheres com lesões intraepiteliais de baixo ou alto grau e câncer de colo de útero apresentam maior expressão da proteína P63 e maior prevalência de genótipo heterozigoto do gene FOXP3 em comparação com as sem lesões cervicais. A associação da pesquisa da expressão da proteína e do polimorfismo do gene pode tornar os exames utilizados atualmente para a avaliação diagnóstica e prognóstica das lesões de colo uterino mais efetivos em detectar quais as mulheres com maior risco para progressão para câncer

Avaliou-se histologicamente a próstata de 30 cães adultos e idosos sexualmente intactos que apresentavam ou não sintomatologia clínica de doença prostática, e verificou-se a incidência de possíveis alterações da glândula. Dentre as alterações encontradas, a hiperplasia prostática benigna constituiu o diagnóstico mais comum, 85,6% (n=24), seguida por prostatite crônica, 64,3% (n=18), displasia do epitélio glandular, 42,8% (n=12), atrofia do epitélio glandular, 39,3% (n=11), infiltrado inflamatório focal, 25% (n=7), dilatação glandular focal, 21,4% (n=6), prostatite aguda, 7,1% (n=2), metaplasia escamosa, 3,6%, (n=1), metástase de neoplasia sistêmica, 3,6% (n=1) e abscesso prostático, 3,6% (n=1). Como em muitos casos os cães são assintomáticos, ressalta-se a importância da realização rotineira de exames clínicos específicos, como o toque retal e a ultrassonografia, para o diagnóstico precoce e o tratamento das afecções prostáticas.

This paper reports the occurrence of osteochondrosis in feedlot cattle submitted to a high-caloric diet. About 11.6% of the animals showed symptoms that included lameness, tarsal hyperflexion, muscle fasciculations, reluctance to move and delayed recumbency. Necropsy and radiologic examination showed typical lesion of osteochondrosis, including cartilage discontinuity and osseous injury. The diet, age and high grow rate of the animals were considered to he factors in the development of osteochondrosis.

OBJETIVO: Avaliar a biocompatibilidade do cimento de fosfato de cálcio, para verificar sua eficácia como possível substituto ósseo. MÉTODOS: No presente trabalho, foi utilizado cimento de fosfato de cálcio em rádio de 8 coelhos, separados em dois grupos (GI e GII), referentes aos tempos de observação de 12 e 26 semanas pós-operatórias, a fim de se observar as reações entre este biomaterial e o tecido ósseo do animal. Foram feitas análises radiográficas e de densitometria óptica, além de microscopia óptica e eletrônica de varredura. RESULTADOS: Observou-se, ao final do experimento, que o cimento à base de fosfato de cálcio foi parcialmente reabsorvido durante o tempo de observação de 26 semanas, apresentando biocompatibilidade, com ausência de reações indesejáveis que pudessem ser atribuídas aos implantes. CONCLUSÕES: O cimento à base de fosfato de cálcio foi biocompatível e parcialmente reabsorvido no período de 26 semanas de observação. Tempos maiores de observação são necessários para a avaliação da reabsorção.

A quantitative, descriptive, cross-sectional and retrospective study, using technical procedures of document consultation from secondary sources and health household survey with application form for face to face inter views, with the assent nº.039/2011 from the Ethics Committee of the Federal University of Rio Grande do Norte. The aim of this study was to analyze the cervix cancer control in the area47 of the Health Family Centre Nova Natal II. The cancer cervix is the second most common cancer among women worldwide. In Brazil screening for early detection and treatment of disease has been poorly done and follow-up to reduce mortality has not been executed. From a total of1170women belonging to area 47, who under went screening by the Pap test in the period from 2005 to 2010,was elected a sample of 38 women with positive cervical changes, over 18 years old. The calculation of frequency analysis of socio demographic and clinical and epidemiological selected variables with the results of cervical changes, using the X2 test and taking as significance level of p<0.05 was not statistically significant. The predominant age range was 25 to 64 years (68.9%), most no white women (60.5%), predominantly with primary education (57.9%), most married (68.4%) and housewives (68.4%) with early age of sexual activity (86.8%), the minority smokers (13.2%), with a sexual partner (36.8%). At the time of interview, 42.1% of the women voiced complaint of discharge, while only 2.6% reported bleeding. In relation to the occurrence of STDs (including HPV), 10.5% of women reported being a carrier. The use of oral contraceptives was 32.3% of women, from 2 to 4 years (44.4%). The result of the last screening test performed, showed prevalence of immature squamous metaplasia (55.3%), followed by intraepithelial low- grade lesion (including the cytopathologic HPV effect and cervical intra epithelial neoplasia grade I) (31.6%); intraepithelial high-grade lesion (including cervical intraepithelial neoplasia grade II and III) (7.9%), atypical squamous non neoplastic cells (5.3%). There was no squamous cell carcinoma and adenocarcinoma. Most women received information about the action that should be done after the last screening test result (55.3%), but how to perform follow, most women did not report having done so (55.3%). The follow-up group of women studied, with varying degrees of cervical abnormalities, should only be completed with the discharge by cure, established inconsecutive negative cytology, a goal that is not being achieved in the area 47 of the Health Family Centre of Nova Natal II

Cells from rat bone marrow exhibit the proliferation-differentiation sequence of osteoblasts, form mineralized extracellular matrix in vitro and release alkaline phosphatase into the medium. Membrane-bound alkaline phosphatase was obtained by method that is easy to reproduce, simpler and fast when compared with the method used to obtain the enzyme from rat osseous plate. The membrane-bound alkaline phosphatase from cultures of rat bone marrow cells has a MWr of about 120 kDa and specific PNPP activity of 1200 U/tng. The ecto-enzyme is anchored to the plasma membrane by the GPI anchor and can be released by PIPLC (selective treatment) or polidocanol (0.2 mg/mL protein and 1% (w/v) detergent). The apparent optimum pH for PNPP hydrolysis by the enzyme was pH 10. This fraction hydrolyzes ATP (240 U/mg), ADP (350 U/ mg), glucose 1-phosphate (1100 U/mg), glucose 6-phosphate (340 Wing), fructose 6-phosphate (460 U/mg), pyrophosphate (330 U/mg) and (3glycerophosphate (600 U/mg). Cooperative effects were observed for the hydrolysis of PPi and beta-glycerophosphate. PNPPase activity was inhibited by 0.1 mM vanadate (46%), 0.1 mM ZnCl2 (68%), 1 mM levamisole (66%), 1 mM arsenate (44%), 10 mM phosphate (21%) and 1 mM theophylline (72%). We report the biochemical characterization of membrane-bound alkaline phosphatase obtained from rat bone marrow cells cultures, using a method that is simple, rapid and easy to reproduce. Its properties are compared with those of rat osseous plate enzyme and revealed that the alkaline phosphatase obtained has some kinetics and structural behaviors with higher levels of enzymatic activity, facilitating the comprehension of the mineralization process and its function. Alkaline phosphatase is required for the mineralization of bone and cartilage. This enzyme is localized in the matrix vesicle, which plays a role key in calcifying cartilage. In this paper. we standardize a method for construction an alkaline phosphatase liposome system to mimic matrix vesicles and examine a some kinetic behavior of the incorporated enzyme. Polidocanol-solubilized alkaline phosphatase, free of detergent, was incorporated into liposomes constituted from dimyristoylphosphatidylcholine (DMPC), dilaurilphosphatidylcholine (DLPC) or dipalmitoylphosphatidylcholine (DPPC). This process was time-dependent and >95% of the enzyme was incorporated into the liposome after 4 h of incubation at 25 degreesC. Although, incorporation was more rapid when vesicles constituted from DPPC were used, the incorporation was more efficient using vesicles constituted from DMPC. The 395 nm diameter of the alkaline phosphatase-liposome system was relatively homogeneous and more stable when stored at 4 degreesC.Alkaline phosphatase was completely released from liposome system only using purified phosphatidylinositol-specific phospholipase C (PIPLC). These experiments confirm that the interaction between alkaline phosphatase and lipid bilayer of liposome is via GPI anchor of the enzyme, alone. An important point shown is that an enzyme bound to liposome does not lose the ability to hydrolyze ATP, pyrophosphate and p-nitrophenyl phosphate (PNPP), but a liposome environment affects its kinetic properties, specifically for pyrophosphate.The standardization of such system allows the study of the effect of phospholipids and the enzyme in in vitro and in vivo mineralization, since it reproduces many essential features of the matrix vesicle. Alkaline phosphatase is required for the mineralization of bone and cartilage. This enzyme is localized in the matrix vesicle, which plays a role key in calcifying cartilage. In this paper we standardize a method to construction a resealed ghost cell-alkaline phosphatase system to mimic matrix vesicles and examine the kinetic behavior of the incorporated enzyme. Polidocanol-solubilized alkaline phosphatase, free of detergent, was incorporated into resealed ghost cells. This process was time-dependent and practically 50% of the enzyme was incorporated into the vesicles in 40 h of incubation, at 25 degreesC. Alkaline phosphatase-ghost cell systems were relatively homogeneous with diameters of about 300 nm and were more stable when stored at -20 degreesC.Alkaline phosphatase was completely released from the resealed ghost cell-system using only phospholipase C. These experiments confirm that the interaction between alkaline phosphatase and the lipid bilayer of resealed ghost cell is exclusively via glycosylphosphatidylinositol (GPI) anchor of the enzyme.An important point shown is that an enzyme bound to resealed ghost cell does not lose the ability to hydrolyze ATP, pyrophosphate and p-nitrophenyl phosphate (PNPP), but the presence of a ghost membrane, as a support of the enzyme, affects its kinetic properties. Endochondral calcification involves the participation of matrix vesicles (MVs), but it remains unclear whether calcification ectopically induced by implants of demineralized bone matrix also proceeds via MVs. Ectopic bone formation was induced by implanting rat demineralized diaphyseal bone matrix into the dorsal subcutaneous tissue of Wistar rats and was examined histologically and biochemically. Budding of MVs from chondrocytes was observed to serve as nucleation sites for mineralization during induced ectopic osteogenesis, presenting a diameter with Gaussian distribution with a median of 306 ± 103 nm. While the role of tissue-nonspecific alkaline phosphatase (TNAP) during mineralization involves hydrolysis of inorganic pyrophosphate (PPi), it is unclear how the microenvironment of MV may affect the ability of TNAP to hydrolyze the variety of substrates present at sites of mineralization. We show that the implants contain high levels of TNAP capable of hydrolyzing p-nitrophenylphosphate (pNPP), ATP and PPi. The catalytic properties of glycosyl phosphatidylinositol-anchored, polidocanol-solubilized and phosphatidylinositol-specific phospholipase C-released TNAP were compared using pNPP, ATP and PPi as substrates. While the enzymatic efficiency (k cat/Km) remained comparable between polidocanol-solubilized and membrane-bound TNAP for all three substrates, the k cat/Km for the phosphatidylinositol-specific phospholipase C-solubilized enzyme increased approximately 108-, 56-, and 556-fold for pNPP, ATP and PPi, respectively, compared to the membrane-bound enzyme. Our data are consistent with the involvement of MVs during ectopic calcification and also suggest that the location of TNAP on the membrane of MVs may play a role in determining substrate selectivity in this micro-compartment.
