No presente trabalho, foram comparados dois métodos de extração de cobre (Cu) e zinco (Zn) em solos com teor de matéria orgânica (MO) maior que 50g kg-1. Os métodos de extração comparados foram: Extração com HCl 0,1mol L-1 (método padrão para os estados do Rio Grande do Sul e Santa Catarina) e Mehlich 3. Este trabalho foi realizado no Laboratório de Química e Fertilidade do Solo da Universidade do Estado de Santa Catarina (UDESC), no ano de 2009. As duas metodologias de extração foram aplicadas em 286 amostras de solo provenientes do Laboratório de Análise do Solo da UDESC, todas com mais de 50g kg-1 de MO. Os teores de cobre e zinco nos extratos foram determinados por espectrometria de absorção atômica com chama. Os resultados mostraram que houve correlação significativa entre os métodos, Cu (r=0,80) e Zn (r=0,93). A solução de Mehlich 3 extraiu mais cobre e menos zinco, quando comparada à solução de HCl 0,1mol L-1. O Mehlich 3 demonstrou ser eficiente na extração de cobre e zinco em solos com alto teor de MO, podendo substituir o método atual.

A simple and fast method for the determination of Ca, Cu, Fe, Mg, Mn, Se and Zn in bovine semen by quadrupole inductively coupled plasma spectrometry (q-ICP-MS) is described. Prior to analysis, samples (200 mu L) were diluted 1:50 in a solution containing 0.01% v/v Triton (R) X-100 and 0.5% v/v nitric acid and directly analyzed by ICP-MS. The limits of detection of the method are 0.3, 0.03, 0.2, 0.04, 0.04, 0.03 and 0.03 mu g L-1 for Ca-44, Cu-63, Fe-57, Mg-24, Zn-64, Se-82 and Mn-55, respectively. For purposes of comparison and method validation, four ordinary bovine semen samples were directly analyzed by ICP-MS and by flame atomic absorption spectrometry (FAAS) or graphite furnace atomic absorption spectrometry (GF AAS), with no statistical difference between the techniques at the 95% level when applying the t-test. Then, the proposed method was applied in the determinations of Ca, Cu, Fe, Mg, Mn, Se and Zn in collected samples of bovine semen from different breeds, which are used in reproduction programs and artificial insemination.

An intensive mineralogic and geochemical investigation was conducted on sediments recovered during Ocean Drilling Program Leg 166 from the western Great Bahama Bank at Sites 1006, 1008, and 1009. Pleistocene through middle Miocene sediments recovered from Site 1006, the distal location on the Leg 166 transect, are a mixture of bank-derived and pelagic carbonates with lesser and varying amounts of siliciclastic clays. A thick sequence of Pleistocene periplatform carbonates was recovered near the platform edge at Sites 1008 and 1009. Detailed bulk mineralogic, elemental (Ca, Mg, Sr, and Na), and stable isotopic (d18O and d13C) analyses of sediments are presented from a total of 317 samples from all three sites.

Geochemical and palynological data from an annually laminated core sequence (Lake Belau, Schleswig-Holstein) are interpreted with respect to vegetation and settlement history on the basis of a chronostratigraphical model and archaeological evidence. Most settlement periods indicated by pollen and archaeological data can be geochemically identified in the sediment sequence using tracer elements such as K, Rb, Zr and the K/Zr ratio. Whilst air-borne pollen carry a more regional signal, the sedimentary flux of these trace elements is determined by the allogenic input from the catchment area of the lake and, therefore, provides information about the local history of settlement and agricultural land use in the lake's vicinity. This is exemplified for the period of the middle Neolithic Funnel Beaker Culture ('Iversen landnam'), where a time offset of 250 years between both signals has been detected. In contrast, both geochemical and pollen signals are highly synchronous during the Early Migration Period and the High Medieval Period. Additionally, the Fe/Ca and/or U/Fe ratio may serve as a sensitive tracer for human impact on the trophic state of the lake. The suggested impact of the Romans and the High Medieval civilization can clearly be seen (and quantified) from elevated lead input into Lake Belau sediments at this time. Effects of secular climatic changes on the sedimentary chemistry have not been detected and, if present, seem to have been obliterated by anthropogenic activity.

The presence of calcium, iron, and zinc bound to human milk secretory IgA (sIgA) was investigated. The sIgA components were first separated by two-dimensional polyacrylamide gel electrophoresis and then identified by electrospray ionization-tandem mass spectrometry (ESI MS MS). The metal ions were detected by flame atomic absorption spectrometry after acid mineralization of the spots. The results showed eight protein spots corresponding to the IgA heavy chain constant region. Another spot was identified as the transmembrane secretory component. Calcium was bound to both the transmembrane component and the heavy chain constant region, while zinc was bound to the heavy chain constant region and iron was not bound with the identified proteins. The association of a metal ion with a protein is important for a number of reasons, and therefore, the findings of the present study may lead to a better understanding of the mechanisms of action and of additional roles that sIgA and its components play in human milk.

An analytical procedure based on microwave-assisted digestion with diluted acid and a double cloud point extraction is proposed for nickel determination in plant materials by flame atomic absorption spectrometry. Extraction in micellar medium was successfully applied for sample clean up, aiming to remove organic species containing phosphorous that caused spectral interferences by structured background attributed to the formation of PO species in the flame. Cloud point extraction of nickel complexes formed with 1,2-thiazolylazo-2-naphthol was explored for pre-concentration, with enrichment factor estimated as 30, detection limit of 5 mu g L(-1) (99.7% confidence level) and linear response up to 80 mu g L(-1). The accuracy of the procedure was evaluated by nickel determinations in reference materials and the results agreed with the certified values at the 95% confidence level.
