ABSTRACT - Meta-cinema can depict film viewers’ attitudes towards cinema and their type of devotion for films. One subcategory of viewers - which I call meta-spectators - is highly specialized in its type of consumption, bordering on obsession. I contend that there are two main varieties of meta-cinematic reception, not altogether incompatible with one another, despite their apparent differences. As both of them are depicted on meta-cinematic products, the films themselves are the best evidence of my typology. My categories of film viewers are the ‘cinephile’, an elite prone to artistic militancy and the adoration of filmic masters; and the ‘fan’, a low culture consumer keen on certain filmic universes and their respective figures and motifs. I will base my rationale on four films that portray such reception practices: Travelling Avant (Jean-Charles Tacchella, 1987, FRA), The Dreamers (Bernardo Bertolucci, 2003, UK/FRA/ITA); Free Enterprise (Robert Mayer Burnett, 1998, USA); Fanboys (2009, Kyle Newman, USA).

Optimization of Carnobacterium divergens V41 growth and bacteriocin activity in a culture medium deprived of animal protein, needs for food bioprotection, was performed by using a statistical approach. In a screening experiment, twelve factors (pH, temperature, carbohydrates, NaCl, yeast extract, soy peptone, sodium acetate, ammonium citrate, magnesium sulphate, manganese sulphate, ascorbic acid and thiamine) were tested for their influence on the maximal growth and bacteriocin activity using a two-level incomplete factorial design with 192 experiments performed in microtiter plate wells. Based on results, a basic medium was developed and three variables (pH, temperature and carbohydrates concentration) were selected for a scale-up study in bioreactor. A 23 complete factorial design was performed, allowing the estimation of linear effects of factors and all the first order interactions. The best conditions for the cell production were obtained with a temperature of 15°C and a carbohydrates concentration of 20 g/l whatever the pH (in the range 6.5-8), and the best conditions for bacteriocin activity were obtained at 15°C and pH 6.5 whatever the carbohydrates concentration (in the range 2-20 g/l). The predicted final count of C. divergens V41 and the bacteriocin activity under the optimized conditions (15°C, pH 6.5, 20 g/l carbohydrates) were 2.4 x 1010 CFU/ml and 819200 AU/ml respectively. C. divergens V41 cells cultivated in the optimized conditions were able to grow in cold-smoked salmon and totally inhibited the growth of Listeria monocytogenes (< 50 CFU g-1) during five weeks of vacuum storage at 4° and 8°C.

In Zimbabwe, the average sweet potato yield (6 t/ha) is relatively low when compared to Asian counterparts (17 t/ha). These low crop yields have been blamed on weevil infestations and viral infections which account for 60-90% of sweet potato yield losses in Africa. Meristem tip culture, a Centre for Potato Improvement (CIP) initiated tissue culture technique, has been widely used to eradicate viruses from clonally propagated crops and has been noted to be one of the instrumental techniques that helped China to increase sweet potato yields. In an effort to adopt the meristem tip culture technique for the production of virus-free planting material of a local sweet potato (cv Brondal), a study was conducted to evaluate the effect of Benzylamino purine (BAP), 1-Naphthaleneacetic acid (NAA) and Gibberellic acid (GA3) (either alone or in combination) on cultured Brondal meristems. The different hormonal treatments were assessed on the following parameters: plantlet regenerative capacity, multiple plantlet production, shoot height, average leaf number per shoot and average node number per shoot, ten weeks after meristem culture. All treatments containing a combination of BAP (1 mg-L) and GA3 (at either 5 mg-L, 10 mg-L, or 20 mg-L) had a significantly (p<0.01) higher plantlet regenerative capacity of 33-66% when compared to other treatment combinations. Only treatments, 10 mg-L GA3 + 1 mg-L BAP and 20 mg-L GA3 + 1 mg-L BAP were capable of inducing multiple plantlet formation, producing an average of three plantlets/meristem and two plantlets/meristem respectively. Overall, treatment 10 mg-L GA3 + 1 mg-L BAP gave rise to significantly (p<0.01) taller shoots (20 mm) compared to the rest of the treatments used. For average leaf number per shoot, all GA3 treatments (5 mg-L, 10 mg-L, or 20 mg-L) supplemented with 1 mg-L BAP gave significantly (p<0.01) higher numbers of leaves (six leaves/shoot) than the rest of the treatments. Treatments 10 mg-L GA3 + 1 mg-L BAP and 20 mg-L GA3 + 1 mg-L BAP gave rise to the highest number of nodes per shoot, producing an average of three nodes per shoot. In sharp contrast to treatments containing a combination of BAP and GA3, all treatments containing a combination of BAP and NAA performed poorly in all parameters tested for plant regeneration of Brondal sweet potato variety. In conclusion, the best hormonal treatment for culturing Brondal meristems proved to be 10 mg-L GA3 + 1 mg-L BAP.

Visando obter subsídios para um estudo da qualidade da água em tanques de piscicultura, realizou-se um experimento de 166 dias com uma espécie nativa, pacu (Piaractus mesopotamicus). Nos tanques foram testados dois níveis diferentes de proteína na dieta (16% e 34% de proteína bruta) e três densidades de estocagem (0,25; 0,50 e 0,77 peixes/m²). Dos resultados obtidos foi observado que a interação entre a densidade de estocagem e a duração do experimento interferiram nas variáveis bicarbonato e alcalinidade e a interação entre a densidade de estocagem e a porcentagem de proteína interferiram nas concentrações de CO2 livre e total, condutividade e pH (P < 0,05). A temperatura da água nos tanques variou significativamente ao longo do período estudado (P < 0,05), diminuindo gradativamente do verão para o inverno. Não houve diferença significativa no tempo de residência da água nos tanques (P > 0,05) durante a duração do experimento. Os demais parâmetros não sofreram interferência dos tratamentos ao longo do período de estudo.

Ceruloplasmin (Cp) is a multicopper ferroxidase that is considered to be an important source of copper in milk for normal neonatal development. We investigated the expression, subcellular localization and secretion of Cp in PMC42-LA cell culture models representative of resting, lactating and suckled human mammary epithelia. Both secreted Cp (sCp) and plasma membrane associated glycosylphosphatidylinositol-linked Cp (GPI-Cp) were expressed in PMC42-LA cells. In all three epithelial models (resting, lactating and suckled), the expression and secretion of copper-bound, ferroxidase active, Cp (holo-Cp) was dependent on media copper concentration. In low copper (bathocuproinedisulphonic acid/d-penicillamine treated models) there was greater than a 2-fold decrease in holo-Cp expression and secretion, which was mirrored by a 2-fold increase in the expression and secretion of copper-free Cp protein (apo-Cp). Cell surface biotinylation studies revealed that the state of PMC42-LA cell differentiation (functionality), and the level of extracellular copper, had no significant effect on the level of plasma membrane bound GPI-Cp. Quantitative real time PCR analyses determined that there was no significant (P > 0.05) difference in Cp mRNA levels across all copper conditions investigated (0, 5, 50 μM). However, there was a significant (P < 0.05) increase (∼2-fold) in Cp mRNA in both the lactating and suckled models in comparison to the resting model. Furthermore, the Cp mRNA increase in response to PMC42-LA differentiation corresponded with more secreted Cp protein, both apo and holo forms, indicating a link between function and Cp requirement. Our results provide significant insight on the regulation of Cp expression and secretion in lactation and copper incorporation into milk.

Three Instant Algae® (Reed Mariculture Inc., Campbell, CA, USA, 95008) products: (1) mono-cultured Isochrysis sp. (Haptophyceae) (Isochrysis 1800®); (2) mono-cultured Pavlova sp. (Haptophyceae) (Pavlova 1800®); and (3) mono-cultured Thalassiosira weissflogii (Bacillariophyceae) (TW 1200®) were used to feed sandfish (Holothuria scabra) larvae both singly and in ternary combination to assess their nutritional efficacy. Two-day auriculariae were held at a starting density of 0.3 mL-1 and were initially fed a daily ration equivalent to the dry weight of 10,000 cells mL-1 of Isochrysis 1800®. This ration was increased by the dry weight equivalent of 1000 cells mL-1 of Isochrysis 1800® per day as larval development proceeded. Post-settled larvae fed TW 1200® were significantly larger than those fed the ternary diet, Isochrysis 1800® or Pavlova 1800®. There were significant differences in the mean (±SE) survival of auriculariae and post-settled larvae between treatments and survival to settlement was significantly higher (P < 0.05) for larvae fed TW 1200® (13.7 ± 0.7%) alone. Laval development, competency and survival were significantly correlated with dietary levels of total protein, lipid and nitrogen-free extract (NFE), and with total polyunsaturated fatty acid (PUFA) content of the diets, and the levels of some specific fatty acids (FA). The proportion of late auriculariae with hyaline spheres (day 13), numbers of competent doliolariae (day 15) and the total length of post-settled larvae (day 21) were all positively correlated with dietary NFE and palmitic acid (16:0) contents, as well as dietary EPA: DHA ratio. This study is the first comprehensive assessment of the nutritional value of micro-algae concentrates for sandfish larvae based on their nutrient compositions. Our study confirms the feasibility of using commercially available microalgae concentrates as a sole food source for hatchery culture of sandfish, and is the first to report successful hatchery culture of H. scabra without using live micro-algae. All micro-algae concentrates used in this study proved nutritious for H. scabra larvae and supported normal growth and development and relatively high survival, through settlement. Use of commercially available micro-algae concentrates as a replacement for live micro-algae in sandfish hatcheries supports development of cheaper, simpler larval rearing protocols for this species. Statement of relevance: Hatchery culture of sandfish using micro-algae concentrates, without the use of live micro-algae, is a significant finding that supports development of cheaper, simpler larval rearing protocols for this species. Our study provides detailed nutritional compositional data for the products used and on this basis we were able to deduce new information relating to key nutrients for sea cucumber larvae.
