Aedes (Stegomyia) aegypti is a species of mosquitoes known to be the vector of diseases such as dengue and yellow fever, and a better understanding of aspects of their biology can help in the establishment of control strategies for the same. Several previous studies showed that temperature significantly affects the development of immature stages of insects. In general, higher temperatures (up to a threshold) accelerate the development of insects, and lower temperature retards the same. This rule also applies to mosquitoes, including Ae. aegypti. But not still know the effects of daily variation of temperature on the developmental stages of mosquitoes. And this detail is very important, since in natural breeding or artificial, The mosquitoes usually face temperature variations over a single day, which should interfere with its development until the emergence of the adult forms. For this reason, the objective of this study is to analyze the effect of alternating temperatures on the development of Ae. aegypti. To conduct the study, adults were collected active in the neighborhood Bela Vista Campus of UNESP - Rio Claro, SP, using a sweep net or using ovitraps for immatures, and the active search for breeding. Individuals collected were kept under experimental conditions in the laboratory. The adult samples were identified to species level, were considered for the experiments, only samples of Ae. aegypti. The insects were housed in plastic cages, suitable for creating flies. These were fed with sugar solution and blood meal on alternate days. The eggs obtained were used in the experiment with four different temperature regimes. The data collected were analyzed by evaluating whether the different treatments influenced the development of immature to adult, performing the Kruskal-Wallis test and the statistical software BioEstat. Chitinases are enzymes involved in degradation of chitin and are present in a range of organisms, including those that do not contain chitin, such as bacteria, viruses, plants and animals, and play important physiological and ecological roles. Chitin is hydrolyzed by a chitinolytic system classified as: endo-chitinases, exo-chitinases and N-acetyl-b-D-glucosaminidases. In this study a Litochitinase1 extracted from the cephalotorax of the shrimp Litopenaeus Schmitt was purified 987.32 times using ionexchange chromatography DEAE-Biogel and molecular exclusion Sephacryl S-200. These enzyme presented a molecular mass of about 28.5 kDa. The results, after kinetic assay with the Litochitinase1 using as substrate p-nitrophenyl-N-acetyl-b-Dglucosaminideo, showed apparent Km of 0.51 mM, optimal activity at pH ranging from 5.0 to 6.0, optimum temperature at 55°C and stability when pre-incubated at temperatures of 25, 37, 45, 50 and 55°C. The enzyme showed a range of stability at pH 4.0 to 5.5. HgCl2 inhibited Litochitinase1 while MgCl2 enhances its activity. Antimicrobial tests showed that Litochitinase1 present activity against gram-negative bacterium Escherichia coli in the 800 μg/mL concentration. The larvicidal activity against Aedes aegypti was investigated using crude extracts, F-III (50-80%) and Litochitinase1 at 24 and 48 hours. The results showed larvicidal activity in all these samples with EC50 values of 6.59 mg/mL for crude extract, 5.36 mg/mL for F-III and 0.71 mg/mL for Litochitinase1 at 24 hours and 3.22 and 0.49 mg/mL for the F-III and Litochitinase1 at 48 hours, respectively. Other experiments confirmed the presence of chitin in the midgut of Aedes aegypti larvae, which may be suffering the action of Litochitinase1 killing the larvae, but also the absence of contaminating proteins as serine proteinase inhibitors and lectins in the crude extract, F-III and Litochitinase1, indicating that the death of the larvae is by action of the Litochitinase1. We also observed that the enzymes extracted from intestinal homogenate of the larvae no have activity on Litochitinase1. These results indicate that the enzyme can be used as an alternative to control of infections caused by Escherichia coli and reducing the infestation of the mosquito vector of dengue.

The endosymbiotic bacterium Wolbachia pipientis infects many species of insects and has been transinfected into the mosquito Aedes aegypti (L.), the primary vector of dengue virus (DENV). Recently, it has been shown that Wolbachia blocks the replication and transmission of RNA viruses, such as DENV, in a number of mosquito species including Ae. aegypti and Aedes albopictus (Skuse), which is naturally infected with Wolbachia and considered a secondary vector for DENV. The mosquito species Aedes notoscriptus (Skuse) is highly prevalent in Australia, including in areas where DENV outbreaks have been recorded. The mosquito has been implicated in the transmission of Ross River and Barmah Forest viruses, but not DENV. We investigated whether Wolbachia naturally infects this mosquito species and whether it has an impact on the ability of Ae. notoscriptus to transmit DENV. We show, for the first time, that Ae. notoscriptus is naturally infected with a strain of Wolbachia that belongs to supergroup B and is localized only in the ovaries. However, Wolbachia infection in Ae. notoscriptus did not induce resistance to DENV and had no effect on overall DENV infection rate or titer. The presence of a native Wolbachia in Ae. notoscriptus cannot explain why this mosquito is an ineffective vector of DENV.

O presente estudo aborda a introdução de ações de regulação na gestão de saúde de um município do interior do estado do Rio de Janeiro, de 2009 a 2012. Mais especificamente, são analisadas a responsabilização sanitária do ente municipal a partir da aquisição da gestão plena do sistema (Pacto de Gestão) e as ações desencadeadas pela implantação do Controle, Avaliação e Auditoria, e pela criação do Complexo Regulador, tendo como referência a Política Nacional de Regulação. A metodologia empregada envolveu análise documental e entrevistas com informantes-chave. Os resultados demonstram o desenvolvimento de ações em todas as dimensões da regulação e o amadurecimento da função reitora da secretaria municipal de saúde sobre o sistema, através do direcionamento qualitativo e quantitativo dos prestadores privados.

The control of the spread of dengue fever by introduction of the intracellular parasitic bacterium Wolbachia in populations of the vector Aedes aegypti, is presently one of the most promising tools for eliminating dengue, in the absence of an efficient vaccine. The success of this operation requires locally careful planning to determine the adequate number of mosquitoes carrying the Wolbachia parasite that need to be introduced into the natural population. The latter are expected to eventually replace the Wolbachia-free population and guarantee permanent protection against the transmission of dengue to human. In this paper, we propose and analyze a model describing the fundamental aspects of the competition between mosquitoes carrying Wolbachia and mosquitoes free of the parasite. We then introduce a simple feedback control law to synthesize an introduction protocol, and prove that the population is guaranteed to converge to a stable equilibrium where the totality of mosquitoes carry Wolbachia. The techniques are based on the theory of monotone control systems, as developed after Angeli and Sontag. Due to bistability, the considered input-output system has multivalued static characteristics, but the existing results are unable to prove almost-global stabilization, and ad hoc analysis has to be conducted.

INTRODUÇÃO: O vírus da dengue é transmitido pela picada do mosquito Aedes aegypti e, o atual programa de controle não atinge o objetivo de impedir sua transmissão. Este trabalho objetivou analisar a relação entre a distribuição espaço-temporal de casos de dengue e os indicadores larvários no município de Tupã, de janeiro de 2004 a dezembro de 2007. MÉTODOS: Foram construídos indicadores larvários por quarteirão e totalidade do município. Utilizou-se o método cross-lagged correlation para avaliar a correlação entre casos de dengue e indicadores larvários. Foi utilizado estimador kernel para análise espacial. RESULTADOS: A correlação cruzada defasada entre casos de dengue e indicadores larvários foi significativa. Os mapas do estimador Kernel da positividade de recipientes indicam uma distribuição heterogênea, ao longo do período estudado. Nos dois anos de transmissão, a epidemia ocorreu em diferentes regiões. CONCLUSÕES: Não ficou evidenciada relação espacial entre infestação larvária e ocorrência de dengue. A incorporação de técnicas de geoprocessamento e análise espacial no programa, desde que utilizados imediatamente após a realização das atividades, podem contribuir com as ações de controle, indicando os aglomerados espaciais de maior incidência.

This study started from the hypothesis of the existence of a relation between the type of the urban occupation concerning to the sustainability conditions at the proximity of Environment Protected Zones and the occurrence of vectors insects in Natal, Rio Grande do Norte, Brazil. This research, which used data available by the City Administration Health and Urbanization Secretaries (respectively SMS and SEMURB), in the time period of 2006 to 2008, aimed to characterize the study site in terms of urban occupation, relating it to social environmental aspects of land occupation and the occurrence of vectors insects. This study is presented in two papers, the first one linking the occurrence of vectors insects and sustainable development indicators and the second relating the incidence of reported cases of Dengue and Dengue Hemorrhagic Fever (DHF) and the occurrence of larvae infection indexes of Aedes aegypti, in Natal, Rio Grande do Norte State. In the first paper, was made a correlation between Dengue Fever vectors and Visceral and Tegumentar Leishmaniasis vectors and sustainable development indicators, selected from IDS Brasil- 2008. Through factorial analysis a Sustainability Index (SI) was acquired for each region, the northern region of the municipality obtained lower numbers than southern region, which, in its turn, presented better sustainability conditions. Linking this index to vector infestation parameters shows a high significant correlation between the SI and the Breteau Index of Aedes aegypti (p=0,028) as well as with SI and sand flies infestation index (p=0,01). Higher rates in vectors infestation in regions with a lower Sustainable Development Index demonstrates that this index can be used to determine the increasing of probability of Aedes and sand flies occurrence in urban environment. The second paper analyzed the occurrence of the main vector of Dengue and DHF, the Aedes aegypti mosquito, and the relation between larvae infection indexes of this insect and reported cases of the diseases. This study revealed unexpected relation where areas with higher Breteau s Indexes showed lower infection rates of Dengue Fever, although showing high incidence of DHF

One of the main problems with dengue is the control of Aedes aegypti, its major vector. In Brazil, the current control program for Ae. aegypti and Aedes albopictus populations includes larval density surveys. An interesting alternative is the use of a distinct index, the Premise Condition Index (PCI). This tool relates conditions of property, such as houses and yards, and the degree of shade with the occurrence of Aedes sp. oviposition, and is calculated as scores from 3 to 9. The lowest score indicates property in good condition and an unfavorable breeding environment, while the highest score indicates property at high risk for infestation by Aedes sp. The present study is based on the application of the PCI in an urban area of Botucatu, Brazil to confirm its effectiveness.

A proliferação do Aedes aegypti é propiciada pelo habito de permitir a formação de criadouros em vários tipos de recipientes. Uma das formas de controla-lo é a disseminação do conhecimento sobre o vetor, por conduzir a conscientização e a tomada de medidas contra a sua proliferação. Para avaliar um método de ensino sobre o vetor e a dengue, foram comparando alunos de 5ª e 6ª séries antes e após a intervenção didática. Os alunos com intervenção foram mais aptos em reconhecerem as fases do ciclo e tiveram um discernimento maior sobre a importância dos mosquitos para a saúde. Eles também foram mais aptos em reconhecerem quais medidas de controle são mais eficientes e viáveis, repercutindo em suas residências que apresentaram duas vezes menos criadouros em relação aos que não tiveram intervenção.
