Synthesis of 1,2,3-triazolylpyranosides through click chemistry reaction

We have developed an efficient method for the synthesis of functionalized C-glycosyl 1,2,3-triazoles through a Cu(1)-promoted azide-alkyne 1,3-dipolar cycloaddition between a TMS-protected C-alkynyl-glycoside and organic azides. The reaction was accelerated by ultrasound irradiation and the addition of a base was not necessary to obtain the 1,2,3-triazole product. Moreover, further manipulation of the products led to chiral molecules with a C-glycoside linkage. (C) 2012 Elsevier Ltd. All rights reserved.

4-Organochalcogenoyl-1H-1,2,3-triazoles: synthesis and functionalization by a nickel-catalyzed Negishi cross-coupling reaction

A general method for the synthesis of triazoles containing selenium and tellurium was accomplished via a CuCAAC reaction between organic azides and a terminal triple bond, generated by in situ deprotection of the silyl group. The reaction tolerates alkyl and arylazides, with alkyl and aryl substituents directly bonded to the chalcogen atom. The products were readily functionalized by a nickel-catalyzed Negishi cross-coupling reaction, furnishing the aryl-heteroaryl products at the 4-position in good yields. (C) 2012 Elsevier Ltd. Neue wasserlösliche Cystein-Protease-Inhibitoren mit Aziridin-2,3-dicarbonsäure-Baustein

Die Leishmaniose gehört zu den „vernachlässigten tropischen Erkrankungen“. Sie wird durch parasitäre Protozoen der Gattung Leishmania ausgelöst. Weltweit sind ca. 12 Mio. Menschen infiziert, ca. 70 Tausend erliegen ihr jährlich. Die aktuelle Therapie wird überschattet von Toxizitäts- und Teratogenitätsproblemen und von aufkommenden Resistenzen. Die von den Leishmanien exprimierten Cysteinproteasen spielen vielfältige Rollen bei Wachstum und Vermehrung der Erreger. Aufgrund der evolutionären Verwandtschaft der Enzyme sind die parasitären Cysteinproteasen strukturell den humanen sehr ähnlich. Die Herausforderung bei der Entwicklung antiparasitärer Wirkstoffe, basierend auf der Hemmung dieser Proteasen, besteht deshalb darin, sehr selektive Inhibitoren zu entwickeln, die die Wirtsproteasen nicht, oder nur in einem vertretbaren Rahmen, inhibieren. Das Ziel dieser Arbeit war die Weiterentwicklung der Aziridin-2,3-dicarbonsäure-basierten Cysteinproteaseinhibitoren RV122C bzw. CS09 hinsichtlich Selektivität und Aktivität gegenüber der parasitären Cathepsin-L-ähnlichen Cystein-Protease LmCPB2.8 durch Design, incl. Docking, Synthese und Testung. Neben der gezielten Variation nicht essenzieller Gruppen wurde molekulares Docking mittels AutoDock Vina an Cruzain als verwandtes Modellenzym durchgeführt, um durch Variationen K.O.-Kandidaten für die Differenzierung zwischen zwei postulierten Bindungsmodi zu finden. Die Ergebnisse der Enzymassays zeigen eine Verbesserung der Hemmeigenschaften bei gleichzeitig verbesserter Selektivität sowie erhöhter ligand efficiency und ligand lipophilic efficiency für Derivate mit sterisch anspruchsvolleren Ester-Resten und für Derivate mit einer freien Carbonsäurefunktion am Aziridin-Ring (Halbester).

Immunodeficiency and the risk of cervical intra-epithelial neoplasia 2/3 and cervical cancer: A nested case-control study in the Swiss HIV Cohort Study.

HIV-infected women are at increased risk of cervical intra-epithelial neoplasia (CIN) and invasive cervical cancer (ICC), but it has been difficult to disentangle the influences of heavy exposure to HPV infection, inadequate screening, and immunodeficiency. A case-control study including 364 CIN2/3 and 20 ICC cases matched to 1,147 controls was nested in the Swiss HIV Cohort Study (1985-2013). CIN2/3 risk was significantly associated with low CD4+ cell counts, whether measured as nadir (odds ratio (OR) per 100-cell/μL decrease=1.15, 95% CI: 1.08, 1.22), or at CIN2/3 diagnosis (1.10, 95% CI: 1.04, 1.16). An association was evident even for nadir CD4+ 200-349 versus ≥350 cells/μL (OR=1.57, 95% CI: 1.09, 2.25). After adjustment for nadir CD4+, a protective effect of >2-year cART use was seen against CIN2/3 (OR versus never cART use=0.64, 95% CI: 0.42, 0.98). Despite low study power, similar associations were seen for ICC, notably with nadir CD4+ (OR for 50 versus >350 cells/μL= 11.10, 95% CI: 1.24, 100). HPV16-L1 antibodies were significantly associated with CIN2/3, but HPV16-E6 antibodies were nearly exclusively detected in ICC. In conclusion, worsening immunodeficiency, even at only moderately decreased CD4+ cell counts (200-349 CD4+ cells/μL), is a significant risk factor for CIN2/3 and cervical cancer. (Figures 2,3) Lipid composition of liver and muscles of the daubed shanny (Leptoclinus maculatus), Kongsfjorden

A comparative study on the lipid composition of the liver and muscles has been performed in daubed shanny caught in summer (July) in Arctic waters at three different sites (biotopes) along the north-western coast of Spitsbergen. In marine organisms living at high latitudes, lipids play an especially important role, primarily as reserve substances and as a factor influencing adaptation to severe environmental conditions. Since the ecology of daubed shanny is poorly known, the data obtained may be considered novel.

Tab. 2+3: The results of the vertical component of tides

Part of the geophysical work at the German Georg von Neumayer Station is the recording of the tidal movement of the Ekström Ice Shelf. Measurements are performed with an earthtide gravity meter for the vertical component of the movement and two simple tiltmeters for the horizontal components. Gravity measurements were done continuously during the 1984/85 winter season at the observatory of the Georg von Neumayer Station. Tilt measurements were carried out at the station and at three locations on an ice-rise at about 10 km distance from the station. Gravity measurements provide the tidal movements of the ice shelf, which amounts to about 1 m at spring tide. The most important result of the tiltmeter measurements lies in the fact that the amplitudes of tilt are substantially larger at the ice-rlse than at the observatory. Results of tide-correlated ice quake activities are also presented.
