Objective We investigated the effects of high-fat diet-induced obesity on vascular proinflammatory factors and oxidative stress on endothelium-dependent relaxation of the aorta. Methods Female Swiss mice were submitted to a high-fat diet for 16 weeks. At the end of the experimental period, we evaluated blood pressure, relaxation in response to acetylcholine in aortic rings in the absence and the presence of the superoxide anion scavenger, superoxide dismutase (SOD, 150 U/ml), and the nuclear factor (NF)-kappa B inhibitor, sodium salicylate (5 mmol/l). Aortic protein expression of endothelial nitric oxide synthase, Cu/Zn-SOD, NF-kappa B, I kappa B-alpha, and proinflammatory cytokines were also evaluated. Results Obese mice presented higher systolic and diastolic blood pressure than control mice (P<0.05). The relaxation of aortas to acetylcholine, but not to sodium nitroprusside, was significantly decreased in obese mice and was corrected by both SOD and sodium salicylate (P<0.05). The protein expression of endothelial nitric oxide synthase and Cu/Zn-SOD was significantly decreased in aorta from obese mice (P<0.05). Total p65 NF-kappa B subunit protein expression was not affected by obesity, but the protein expression of NF-kappa B inhibitor I kappa B-alpha was lower in aorta from obese mice (P<0.05). There were no significant differences in the interleukin (IL)-1 beta and IL-6 protein expression between groups. In contrast, the expression of TNF-alpha was significantly increased in aortas from obese mice. Conclusion Our resultssuggest that the reducedantioxidant defense and the local NF-kappa B pathway play an important role in the impairment of endothelium-dependent relaxation in aorta from obese mice. Scavenger or Fc gamma receptors are important for capture and clearance of modified LDL particles by monocytes/macrophages. Uptake via scavenger receptors is not regulated by intracellular levels of cholesterol and in consequence, macrophages develop into foam cells in the arterial intima. The levels of scavenger receptor CD36 are increased in atherosclerotic lesions and there is evidence that some components of oxLDL auto-regulate the expression of this receptor. Fc gamma receptor expression is increased in cardiovascular diseases but it is not known weather their expression is regulated by oxLDL. The biological properties of oxLDLs vary depending on the degree of oxidation. In the present study we investigated the effect of LDL particles showing extensive or low oxidation (HoxLDL and LoxLDL) on the expression of CD36 and Fc gamma RII in a human monocytic cell line (THP-1), differentiated or not to macrophage, and the involvement of PPAR gamma. It was found that both forms of oxLDL are able to increase the expression of CD36 and Fc gamma RII and that this effect is dependent on the degree of oxidation and of the stage of cell differentiation ( monocyte or macrophage). We also showed that the increased expression of Fc gamma RII is dependent on PPAR. whereas that of the CD36 is independent of PPAR gamma. Copyright (c) 2008 S. Karger AG, Basel

The new trinuclear gadolinium complex [Gd(3)L(2)(NO(3))(2)(H(2)O)(4)]NO(3)center dot 8H(2)O (1) with the unsymmetrical ligand 2-[N-bis-(2-pyridylmethyl)aminomethyl]-4-methyl-6-[N-bis(2-hydroxy-2-oxoethyl)aminomethyl] phenol (H(3)L) was synthesized and characterized. The new ligand H(3)L was obtained in good yield. Complex I crystallizes in an orthorhombic cell, space group Pcab. Kinetic studies show that complex 1 is highly active in the hydrolysis of the substrate 2,4-bis(dinitrophenyl)phosphate (K(m) = 4.09 mM, V(max) = 2.68 x 10(-2) mM s(-1), and k(cat) = V(max)/[1] = 0.67 s(-1)). Through a potentiometric study and determination of the kinetic behavior of 1 in acetonitrile/water solution, the species present in solution could be identified, and a trinuclear monohydroxo species appears to be the most prominent catalyst under mild conditions. Complex 1 displays high efficiency in DNA hydrolytic cleavage, and complete kinetic studies were carried out (K(m) = 4.57 x 10(-4) M, K(cat)` = 3.42 h(-1), and k(cat)`/K(m) = 7.48 x 10(3) M(-1) h(-1)). Studies with a radical scavenger (dimethyl sulfoxide, DMSO) showed that it did not inhibit the activity, indicating the hydrolytic action of 1 in the cleavage of DNA, and studies on the incubation of distamycin with plasmid DNA suggest that 1 is regio-specific, interacting with the minor groove of DNA.

Nanosecond laser flash photolysis has been used to investigate injection and back electron transfer from the complex [(Ru-(bpy)(2)(4,4`-(PO(3)H(2))(2)bpy)](2+) surface-bound to TiO(2) (TiO(2)-Ru(II)). The measurements were conducted under conditions appropriate for water oxidation catalysis by known single-site water oxidation catalysts. Systematic variations in average lifetimes for back electron transfer, - were observed with changes in pH, surface coverage, incident excitation intensity, and applied bias. The results were qualitatively consistent with a model involving rate-limiting thermal activation of injected electrons from trap sites to the conduction band or shallow trap sites followed by site-to-site hopping and interfacial electron transfer, TiO(2)(e(-))-Ru(3+) -> TiO(2)-Ru(2+). The appearance of pH-dependent decreases in the efficiency of formation of TiO(2)-Ru(3+) and in incident-photon-to-current efficiencies with the added reductive scavenger hydroquinone point to pH-dependent back electron transfer processes on both the sub-nanosecond and millisecond-microsecond time scales, which could be significant in limiting long-term storage of multiple redox equivalents.

The tripeptide, glutathione (glutamylcysteinylglycine) is the primary endogenous free radical scavenger in the human body. When glutathione (GSH) levels are reduced there is an increased potential for cellular oxidative stress, characterised by an increase and accruement of reactive oxygen species (ROS). Oxidative stress has been implicated in the pathology of schizophrenia and bipolar disorder. This could partly be caused by alterations in dopaminergic and glutamatergic activity that are implicated in these illnesses. Glutamate and dopamine are highly redox reactive molecules and produce ROS during normal neurotransmission. Alterations to these neurotransmitter pathways may therefore increase the oxidative burden in the brain. Furthermore, mitochondrial dysfunction, as a source of oxidative stress, has been documented in both schizophrenia and bipolar disorder. The combination of altered neurotransmission and this mitochondrial dysfunction leading to oxidative damage may ultimately contribute to illness symptoms. Animal models have been established to investigate the involvement of glutathione depletion in aspects of schizophrenia and bipolar disorder to further characterise the role of oxidative stress in psychopathology. Stemming from preclinical evidence, clinical studies have recently shown antioxidant precursor treatment to be effective in schizophrenia and bipolar disorder, providing a novel clinical angle to augment often suboptimal conventional treatments.

Landscape planning in many countries is predicated upon on fulfilling the functions for human living objectives. Many land use practices have been plotted for living, busines~, trading, industrial, farming as well as providing places for dead people primarily through cemeteries. Research in Palm Beach County, FL, has demonstrated the need to plan for 30 years of demand of land use functions to service death (Coutts, Basmaj ian et al. 20 I I). Coutts et al assert that planners are required and responsible for the planning of funeral necessities. Therefore, the protection of landscapes of death is an important consideration in the planning of landscapes. Bali is popular with its beautiful landscape, hospitality, and traditional architecture as demonstrating the integrity between human, environment and God, as expressed in the Balinese Tri Hita Karana concept. Balinese commemorate life from birth to death through their traditional ceremonies which informs their traditional cultural landscape. One of the most important landscapes, which cannot be separated fi·om Balinese life are graveyards which are used for deceased ceremonies. This landscape is an integral part of traditional village patterns across Bali. Culturally, Balinese people have their own traditional cremation ceremony which is call the Ngaben Ceremony. The Ceremony takes place in graveyards and thereupon ashes are placed in the sea waters surrounding Bali. An interesting point of planning in Bali is how to enable eco-friendly interment extensions to villages. This is occurring because of the increasing number of corpses that require cremation thus necessitating no accretions in land provision of graveyards. This research investigates the landscape of death in Bali expressed in its traditional values in the area of planning which implicate sustainable environments and land conservation topics. Other functions of graveyards, as noted by Strangstad ( 1988), include ceremonial and their role as educational tools for history lessons, art, sociology, geology, English lessons, as well as for scavenger hunts.

Viewpoints are a structural approach to training and directing for theatre. Originating from the innovative, inventive and exploratory approach of Mary Overlie and the self- confessed scavenger approach of Anne Bogart, Viewpoints offers a practical philosophy of working. As a training approach it begins with a disciplined engagement  of the body in space and time. The tangible elements of the Viewpoints provide a set of tasks on which the student can focus, thus freeing the imagination and spirit to  create. Yet at the same time the systematic logic of Viewpoints supports novice practitioners to begin to question their perception, invest in creative practice that demands action and exploration, and to deconstruct, re-organise and rebuild scores and sequences in the pursuit of theatre that is visceral and visual. This essay reports on undergraduate student experiences of learning Viewpoints. It interrogates the demands of embodied learning of the movement/structural system on non- ancers and examines student-actor experiences of embodied learning from multiple of subject positions – observer/participant/creator/reflector/actor.

Mitochondrial dysfunction and oxidative stress are increasingly implicated in the pathophysiology of schizophrenia. The brain is the body's highest energy consumer, and the glutathione system is the brain's dominant free radical scavenger. In the current paper, we review the evidence of central and peripheral nervous system anomalies in the oxidative defences of individuals with schizophrenia, principally involving the glutathione system. This is reflected by evidence of the manifold consequences of oxidative stress that include lipid peroxidation, protein carboxylation, DNA damage and apoptosis - all potentially part of the process of neuroprogression in the disorder. Importantly, oxidative stress is amenable to intervention. We consider the clinical potential of some possible interventions that help reduce oxidative stress, via augmentation of the glutathione system, particularly N-acetyl cysteine. We argue that a better understanding of the mechanisms and pathways underlying oxidative stress will assist in developing the therapeutic potential of this area.

On most developed coastlines, dunes backing ocean beaches constitute an urbanised landscape mosaic containing remnant pockets of small conservation areas. Urbanised beaches are also prime sites for domestic dogs, known to be environmentally harmful in many other settings. It is unknown, however, whether small, protected parcels of dune are adequate for biological conservation and whether dogs compromise their functional conservation objectives. Here we examine, for two small (2 km ocean boundary) reserves in Eastern Australia abutting an urban area, whether such small reserves can continue to function as effective conservation instruments on ocean beaches, using scavenger community composition and efficiency to assess ecosystem function. Two non-native species of canids—domestic dogs (Canis lupus familiaris) and red foxes (Vulpes vulpes)—were ubiquitous and numerous inside conservation areas, to the point of having become the most abundant vertebrate scavengers at the beach-dune interface, outcompeting native scavengers for wave-cast carrion. Dogs and foxes have effectively supplanted raptors, normally abundant on non-urban beaches in the region, and other avian scavengers, as the principal consumers of animal carcasses both inside the declared reserves and at the urban beach. Whilst the ecological threats posed by foxes are widely and intensively addressed in Australia in the form of fox-control programs, dog controls are less common and stringent. Our data emphasize, however, that managing domestic dogs may be required to the same extent in order to maintain key forms and functions in coastal reserves situated close to urban areas.

Road-killed animals are easy and inexpensive to survey, and may provide information about species distributions, abundances, and mortality rates. As with any sampling method, however, we need to explore methodological biases in such data. First, how does an animal's behavior (e.g., use of the center vs. periphery of the road) influence its vulnerability to vehicular traffic? Second, how rapidly do post-mortem processes (scavenging by other animals, destruction or displacement by subsequent vehicles) change the numbers and locations of roadkills? Our surveys of anurans on a highway in tropical Australia show that different anuran species are distributed in different ways across the width of the road, and that locations of live versus dead animals sometimes differ within a species. Experimental trials show that location on the road affects the probability of being hit by a vehicle, with anurans in the middle of the road begin hit 35% more often than anurans on the edges; thus, center-using species are more likely to be hit than edge-using taxa. The magnitude of post-mortem displacement and destruction by subsequent vehicles depended on anuran species and body size. The mean parallel displacement distance was 122.7 cm, and carcasses of thin-skinned species exhibited greater post-mortem destruction. Scavenging raptors removed 73% of carcasses, most within a few hours of sunrise. Removal rates were biased with respect to size and species. Overall, our studies suggest that investigators should carefully evaluate potential biases before using roadkill counts to estimate underlying animal abundances or mortality rates.

A contribuição dos radicais livres na hipertensão está relacionada com a produção do ânion radical superóxido e sua influência na ativação da enzima conversora da angiotensina (ECA). Os estrogênios têm um potencial antioxidante bastante relevante, uma vez que esse hormônio esteróide pode interferir no processo de iniciação da lipoperoxidação (LPO), atuando como “scavenger” de radicais livres. Foram objetivos deste trabalho avaliar a influência dos estrogênios na LPO, na capacidade antioxidante total (TRAP), na atividade das enzimas antioxidantes e no metabolismo do NO em coração e rins de ratas hipertensas. Procurou-se, ainda, avaliar a progressão temporal do estresse oxidativo sistêmico. Foram utilizadas 60 ratas Wistar, divididas em 4 grupos: normotenso controle (NCO), hipertenso controle (HCO), normotenso castrado (NCA) e hipertenso castrado (HCA). Foi induzida a hipertensão renovascular (modelo Goldblatt 2) por 21 dias e, concomitantemente, realizada a ovariectomia. Num grupo de animais, amostras de sangue foram coletadas no 3º, 10º e 20º dia. Em outros animais, os corações e rins foram homogeneizados, no 21° dia. Na avaliação sistêmica, a quimiluminescência (QL) aumentou do 3º para o 21º dia nos grupos HCO, NCA e HCA em relação ao grupo NCO. A atividade da superóxido dismutase (SOD) seguiu o mesmo padrão de oscilação. Os níveis de nitratos também aumentaram no 21° dia nos grupos HCO, NCA e HCA. No entanto, no 3° dia mostraram-se menores que os do grupo NCO.A atividade da catalase (CAT) mostrou-se aumentada no 10º dia nos grupos HCO e HCA em relação aos grupos NCO e NCA. Já o TRAP apresentou-se diminuído no 10° dia nos grupos HCO, NCA e HCA em relação ao 3° dia e ao grupo NCO. Nos tecidos, a LPO apresentou-se aumentada no grupo NCA em relação ao grupo NCO e o grupo HCA apresentou-se aumentado em relação aos grupos HCO e NCA . A atividade da SOD em homogeneizado cardíaco apresentou-se aumentada nos grupos hipertensos. A atividade da glutationa peroxidase (GPx) em tecido cardíaco apresentou-se aumentada no grupo HCO em relação aos grupos NCO e HCA. A atividade da CAT em homogeneizado cardíaco apresentou-se aumentada nos grupos HCO e NCA em relação ao grupo NCO. No rim a CAT apresentou-se aumentada nos grupos castrados. A atividade da glutationa S-transferase (GST), em coração e rins, apresentou-se aumentada com a hipertensão. O TRAP apresentou-se, em tecido cardíaco, diminuído no grupo HCO e aumentado no grupo HCA. No tecido renal, apenas o grupo NCA apresentou TRAP aumentado. O nível de nitratos em tecido cardíaco apresentou-se menor no grupo HCO e maior no grupo NCA em relação ao grupo NCO. O grupo HCA apresentou-se menor em relação ao grupo NCA. Nos homogeneizados de rins a SOD, a GPx e os nitratos não apresentaram diferenças significativas entre os grupos. Pode-se observar que tanto a hipertensão quanto a castração induziram um aumento de estresse oxidativo sistêmico que progride com o passar do tempo, assim como adaptações do sistema antioxidante enzimático e não enzimático. Nos tecidos, o estresse oxidativo é aumentado pela retirada dos estrogênios, efeito este potencializado quando concomitante à hipertensão. Os dados encontrados neste trabalho confirmam a ação antioxidante dos estrogênios.

Spondias sp. (Anacardiaceae), popularly known as cajá-umbu, is an endemic plant from Northeastern Brazil, where their leaves are widely used in folk medicine to treat inflammatory processes, while their fruits have a great agro industrial potential. This study was designed to evaluate hepatoprotective, antinociceptive, antioxidant, antimicrobial and anti-inflammatory properties, as well as the acute toxicity and repeated dose 28, using a methanolic extract (MES), a fraction rich in flavonoids (FRF) and a precipitate from Spondias sp.leaves. The antioxidant activity of them was valued to evaluate their free radical scavenger capacity by DPPH test, whereas MES and FRF were used to evaluate while the preventive action on carbon tetrachloride (CCl4)-induced hepatotoxicity. Seven groups (n=5) of female Wistar rats were used as follows: control group, CCl4-intoxicated group treated with EMS (500 mg/kg) for 7 days, three CCl4-intoxicated groups treated with FRF (25, 50 and 75 mg/kg) for 7 days and the CCl4-intoxicated group treated with Legalon ® (silimarina; (phytotherapeutic reference) (50 mg/kg; 7 days). MES and FRF showed a protective action against liver injury induced by CCl4, being observed a significant reduction of serum enzyme activity marker of liver damage (alanine transaminase and aspartate transaminase). On the other hand, the lipid peroxidation (SRAT) decrease, as well as the increase of glutathione content and enzyme activity of antioxidant defense system (SOD, CAT, GPx) toward near normal values indicated the ability of EMS to restore the oxidative imbalance induced by CCl4. The histological analysis confirmed the hepatoprotection, compared to degenerative changes in CCl4-treated group. This hepatoprotetor effect was similar to that shown by Legalon®. The in vitro high antioxidant capacity of extract (93.16 ± 1.00%) showed analogous results to those obtained by Carduus marianus BHT (reference standard). This fact explains the obtained results in vivo. Although no antimicrobial activity was detected, EMS and FRF promoted the antinociceptive effect induced in the second phase by the intraplantar formalin test, evidencing the anti-inflammatory action; confirmed by the carrageenan-induced peritonitis model. The evaluation of the mechanical allodynia (CFA a 80%) demonstrated the involvement of the Spondias sp. chemical composition in the anti-inflammatory activity toward the acute processes. The acute exposure and repeated dose during 28 days did not produce significant changes in the parameters that evaluate toxicity. Together the experimental results reveal, that Spondias sp. leaf extracts have a promising potential in pharmaceutical area, and due to its non-toxic condition present efficiency and security

A capacidade antioxidante e a qualidade pós-colheita do abacate 'Fuerte' foram avaliadas. Os frutos foram mantidos sob temperatura ambiente (24 ± 1 ºC) e sob refrigeração em 10 ± 1 ºC e 90±5% UR e avaliados durante 15 dias. Determinaram-se a perda de massa e a taxa respiratória. As características físico-químicas avaliadas foram acidez total titulável (AT), sólidos solúveis (SS), ratio, pH, firmeza, atividade da enzima polifenoloxidase (PPO), cor (L, a* e b*), fenólicos totais e atividade antioxidante (AA) por DPPH·. Nas duas condições de armazenamento, o pico respiratório ocorreu no 9º dia.Os frutos mantidos sob temperatura refrigerada apresentaram menor perda de massa e maior firmeza em relação àqueles em temperatura ambiente. O comportamento respiratório refletiu na redução dos valores de AT e no conteúdo de SS que diminuíram a partir do pico de respiração. Os valores de pH aumentaram como consequência da diminuição da AT. Os valores de luminosidade mantiveram-se elevados, observando-se diminuição do componente de cor verde na polpa com o armazenamento. A atividade da PPO diminuiu com o armazenamento. Os valores de AA variaram de 17,6 a 68,7%. O conteúdo de fenólicos totais diminuiu após o pico respiratório, e valores crescentes de AA foram observados mesmo após o 9º dia.

The protective effect of gallic acid and its esters, methyl, propyl, and lauryl gallate, against 2,2'-azobis(2-amidinopropane)hydrochloride (AAPH)-induced hemolysis and depletion of intracellular glutathione (GSH) in erythrocytes was studied. The inhibition of hemolysis was dose-dependent, and the esters were significantly more effective than gallic acid. Gallic acid and its esters were compared with regard to their reactivity to free radicals, using the DPPH and AAPH/pyranine free-cell assays, and no significant difference was obtained. Gallic acid and its esters not only failed to inhibit the depletion of intracellular GSH in erythrocytes induced by AAPH but exacerbated it. Similarly, the oxidation of GSH by AAPH or horseradish peroxidase/H(2)O(2) in cell-free systems was exacerbated by gallic acid or gallates. This property could be involved in the recent findings on proapoptotic and pro-oxidant activities of gallates in tumor cells. We provide evidence that lipophilicity and not only radical scavenger potency is an important factor regarding the efficiency of antihemolytic substances.

Maytenus ilicifolia is an important plant with potential on cancer treatment and has been largely used in Brazil and other countries. We have evaluated the crude ethanolic extract of M. ilicifolia as a potential antioxidant source using an assay based on the bleaching of the radical monocation 2,2'-azinobis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS(center dot+)) and by HOCl scavenger capacity. Trolox and uric acid were used as positive controls. The results indicated M. ilicifolia root bark as a great source of antioxidants based on its potential as scavenger of radicals.
