The aim of this work is to study some of the density estimation tec- niques and to apply to the segmentation of medical images. Medical images are used to help the diagnostic of tumor diseases as well as to plan and deliver treatment. A computer image is an array of values representing colors in some scale. The smallest element of the image to which it is possible to assign a value is called pixel. Segmen- tation is the process of dividing the image in portions through the classi¯cation of each pixel. The simplest way of classi¯cation is by thresholding, given the number of portions and the threshold values. Another method is constructing a histogram of the pixel values and assign a portion to each pike. The threshold is the mean between two pikes. As the histogram does not form a smooth curve it is di±cult to discern between true pikes and random variation. Density estimation methods allow the estimation of a smooth curve. Image data can be considered as mixture of different densities. In this project parametric and nonparametric methods for density estimation will be addressed and some of them are applied to CT image data

From August 2005 to March 2007, the two seasons (with 12 and 10 episodes respectively) of the award winning miniseries HBO‟s ROME were aired by the Home Box Office (HBO) channel. With screenplay signed by various writers and directors, the TV series was a coproduction of HBO (USA) and BBC (UK) with support from RAI (Italy), and the show was filmed in multiple locations, but mainly in Cinecittà Film Studios in Rome, very famous for having been headquarters also for Federico Fellini‟s movies. In the first season, the miniseries depicts the conquest of Gaul, made by the military genius of Gaius Julius Caesar, and the political trajectory that made him accumulate power to such an extent that this divided Roman citizens into two factions, one supporting and the other opposing him, the latter focused mainly on the historic figure of General Gnaeus Pompey Magnus. The second season shows the period of civil war following the assassination of Caesar, and the future rise to power of his nephew, adopted son and sole heir, Gaius Octavian Augustus, who was destined to overcome his rivals as well as their allies in the triumvirate that had been formed to pursue and punish Caesar‟s assassins. These facts are well known and usually crowd the mind and imagination of every minimally educated person. The HBO series broke new ground not only for the talent of its writers, directors and actors, not only for its visual effects and locations nor for the vibrancy and grandeur of historical scenes – after all, “historical movies” in general do the same – but it has done so also by the (re)construction of historical events from the perspective of a pair of protagonists of whom too little is known: the centurions Titus Pullo and Lucius Vorenus, who are the only low-rank soldiers mentioned by Caesar in his book Commentaries on the Gallic War (Commentarii de Bello Gallico V.44). Thus, the fictionalization of events also took into account several Roman civilization data which were scattered through historical sources and also those that belong to the modern knowledge of material culture, resulting in a TV series whose filmic aesthetics has rare beauty and creativity. From the survey of textual, historical and cultural data put together in this film, as well as the distance featuring the creative space in the dimension of the gap between them, this paper aims to highlight two pivotal moments of visual and narrative strategies of the show: the opening credits footage and the final scenes of the first season of HBO's Rome.

Purpose: This paper presents the application of MAGIC-f gel in a three-dimensional dose distribution measurement and its ability to accurately measure the dose distribution from a tomotherapy unit. Methods: A prostate intensity-modulated radiation therapy (IMRT) irradiation was simulated in the gel phantom and the treatment was delivered by a TomoTherapy equipment. Dose distribution was evaluated by the R2 distribution measured in magnetic resonance imaging. Results: A high similarity was found by overlapping of isodoses of the dose distribution measured with the gel and expected by the treatment planning system (TPS). Another analysis was done by comparing the relative absorbed dose profiles in the measured and in the expected dose distributions extracted along indicated lines of the volume and the results were also in agreement. The gamma index analysis was also applied to the data and a high pass rate was achieved (88.4% for analysis using 3%/3 mm and of 96.5% using 4%/4 mm). The real three-dimensional analysis compared the dose-volume histograms measured for the planning volumes and expected by the treatment planning, being the results also in good agreement by the overlapping of the curves. Conclusions: These results show that MAGIC-f gel is a promise for tridimensional dose distribution measurements. (C) 2012 American Association of Physicists in Medicine. Purpose Cediranib is a highly potent inhibitor of vascular endothelial growth factor (VEGF) signaling with activity against all three VEGF receptors. HORIZON II [Cediranib (AZD2171, RECENTIN) in Addition to Chemotherapy Versus Placebo Plus Chemotherapy in Patients With Untreated Metastatic Colorectal Cancer] assessed infusional fluorouracil, leucovorin, and oxaliplatin/capecitabine and oxaliplatin (FOLFOX/CAPOX) with or without cediranib in patients with previously untreated metastatic colorectal cancer (mCRC). Patients and Methods Eligible patients were initially randomly assigned 1:1:1 to receive cediranib (20 or 30 mg per day) or placebo plus FOLFOX/CAPOX. In an early analysis of this and two other cediranib studies (HORIZON I [Cediranib Plus FOLFOX6 Versus Bevacizumab Plus FOLFOX6 in Patients With Previously Treated Metastatic Colorectal Cancer] and HORIZON III [Cediranib Plus FOLFOX6 Versus Bevacizumab Plus FOLFOX6 in Patients With Untreated Metastatic Colorectal Cancer]), the 20-mg dose met the predefined criteria for continuation. Subsequent patients were randomly assigned 2: 1 to the cediranib 20 mg or placebo arms. Progression-free survival (PFS) and overall survival (OS) were coprimary end points. Results In all, 860 patients received cediranib 20 mg (n = 502) or placebo (n = 358). The addition of cediranib to FOLFOX/CAPOX resulted in PFS prolongation (hazard ratio [HR], 0.84; 95% CI, 0.73 to 0.98; P = .0121; median PFS, 8.6 months for cediranib v 8.3 months for placebo) but had no impact on OS (HR, 0.94; 95% CI, 0.79 to 1.12; P = .5707; median OS, 19.7 months for cediranib v 18.9 months for placebo). There were no significant differences in the secondary end points of objective response rate, duration of response, or liver resection rate. Median chemotherapy dose-intensity was decreased by approximately 10% in patients treated with cediranib. Adverse events (AEs) associated with cediranib were manageable. Conclusion Addition of cediranib 20 mg to FOLFOX/CAPOX resulted in a modest PFS prolongation, but no significant difference in OS. The cediranib AE profile was consistent with those from previous studies. Because of the lack of improvement in OS, cediranib plus an oxaliplatin-based regimen cannot be recommended as a treatment for patients with mCRC. J Clin Oncol 30:3596-3603. Der Stamm der Apicomplexa ist eine artenreiche Gruppe, der einzellige, meist obligat intrazelluläre Parasiten angehören, darunter auch erstzunehmende Krankheitserreger wie Plasmodium sp. sowie tierpathogene Vertreter wie Eimeria sp. und Theileria sp. Eimeria sp. verursacht die Kokzidiose beim Huhn. Diese Krankheit bedingt weltweite Verluste in der Geflügelindustrie von etwa 3 Milliarden US$ pro Jahr [DALLOUL & LILLEHOJ, 2006; SHIRLEY et al., 2007; LUCIUS & LOOS-FRANK, 2008]. Die Parasiten weisen eine hohe Resistenzbildungsrate gegen vorhandene Wirkstoffe auf. Zudem ist der Einsatz von Vakzinen mit Nebenwirkungen verbunden und für hohe Produktionskosten verantwortlich. Daher ist die Entwicklung von neuen, kostengünstigen und effektiven Kokzidiostatika eine dringend notwendige Herausforderung [KINNAIRD et al., 2004]. rnAuf Grund ihrer essentiellen, regulatorischen Funktion im eukaryotischen Zellzyklus sind Zyklin-abhängige Kinasen (CDKs) validierte Zielproteine [LEHNINGER et al., 2005]. Auch Eimeria tenella CDC2-related kinase 2 (EtCRK2) wurde bereits mittels des bekannten CDK-Inhibitors Flavopiridol als Zielprotein chemisch validiert [ENGELS et al., 2010]. Wie bei allen CDKs ist die Aktivität von EtCRK2 abhängig von der Bindung eines Aktivators, der zur Zyklin-Proteinfamilie gehört. Dieser natürliche EtCRK2-Aktivator war jedoch bislang nicht bekannt. Deshalb war ein Teil dieser Arbeit die Identifizierung des natürlichen EtCRK2-Aktivators. Bioinformatische Analysen identifizierten vier E. tenella Zyklin-ähnliche Proteine (EtCYC1, EtCYC3a, EtCYC3b und EtCYC4), die nah verwandt zu den Plasmodium falciparum-Zyklinen sind [ENGELS et al., 2010; SUÁREZ FERNÁNDEZ et al., bislang unveröffentlichte Daten]. Im Rahmen dieser Arbeit konnten zwei neue Aktivatoren identifiziert und biochemisch charakterisiert werden: der bekannte CDK-Aktivator XlRINGO und das neue E. tenella-Zyklin EtCYC3a. Nachdem der nicht-radioaktive TR-FRET-Assay für die EtCRK2 etabliert und optimiert wurde, konnte die EtCRK2-Aktivität im Komplex mit beiden Aktivatoren und weitere wichtige kinetische Parameter bestimmt werden.rnZusätzlich wurde dieser Assay zum in vitro Screening einer kommerziellen Chemikalienbibliothek auf die EtCRK2 eingesetzt, um potentielle Inhibitoren für EtCRK2 zu identifizieren. Dieses in vitro Screening gefolgt von einer in silico Hit-Anreicherung identifizierte 19 aktive Verbindungen für die durch EtCYC3a und XlRINGO aktivierte EtCRK2. Zudem wurden drei Struktur-Cluster definiert: Naphthoquinone, 8-Hydroxyquinoline und 2-Pyrimidinyl-aminopiperidin-propan-2-ole. rnDie aktivsten Vertreter von jedem Cluster wurden als Leitstrukturen ausgewählt und auf EtCRK2 und HsCDK2 getestet. Aufgrund ihrer inhibierenden Wirkung auf EtCRK2 stellen diese Verbindungen viel versprechende Leitstrukturen für die Entwicklung eines neuen Antikokzidiums dar. Hiermit konnte auch gezeigt werden, dass BES124764, der Vertreter des 2-Pyrimidinyl-aminopiperidin-propan-2-ol-Clusters, in der Lage ist, die EtCRK2 selektiv zu inhibieren. rnDaher wird BES124764 sowie einige Derivate in den Leitstruktur-Optimierungsprozess für die Auffindung eines neuen Arzneimittelkandidaten gegen Kokzidiose eingehen.rn

A laboratory model system with the rosy apple aphid (Dysaphis plantaginea Pass.) on apple seedlings was developed to study the effects of homeopathic preparations on this apple pest. The assessment included the substance Lycopodium clavatum and a nosode of the rosy apple aphid. Each preparation was applied on the substrate surface as aqueous solution of granules (6c, 15c, or 30c). Controls were aqueous solutions of placebo granules or pure water. In eight independent, randomized, and blinded experiments under standardized conditions in growth chambers, the development of aphids on treated and untreated apple seedlings was observed over 17 days, each. Six experiments were determined to assess the effects of a strict therapeutic treatment; two experiments were designed to determine the effects of a combined preventative and therapeutic treatment. After application of the preparations, the number of juvenile offspring and the damage on apple seedlings were assessed after 7 and 17 days, respectively. In addition, after 17 days, the seedling weight was measured. In the final evaluation of the six strictly therapeutic trials after 17 days, the number of juvenile offspring was reduced after application of L. clavatum 15c (-17%, p = 0.002) and nosode 6c (-14%, p = 0.02) compared to the pure water control. No significant effects were observed for leaf damage or fresh weight for any application. In the two experiments with combined preventative and therapeutic treatment, no significant effects were observed in any measured parameter. Homeopathic remedies may be effective in plant-pest systems. The magnitude of observed effects seems to be larger than in models with healthy plants, which renders plant-pest systems promising candidates for homeopathic basic research. For successful application in agriculture, however, the effect is not yet sufficient. This calls for further optimization concerning homeopathic remedy selection, potency level, dosage, and application routes.
