This paper examines the potential of international actors to contribute to conflict resolution by analysing the evolving role of the European Union (EU) in embedding Northern Ireland's peace process. Scrutiny of the EU's approach to conflict resolution in Northern Ireland offers useful insights into the scope and potential of soft power for facilitating behavioural change from governmental to grass-roots levels. This paper traces the development of the EU's approach to conflict resolution in Northern Ireland from one concentrated on encouraging state-level agreement, to nurturing peace through multilevel funding, through now to consolidating the peace by facilitating regional-level empowerment. The core argument is that, in sum, the most critical element of the EU's contribution to peace in Northern Ireland has been, quite simply, that of enduring commitment.

The SH gene and its flanking sequences have been analysed for 10 strains of mumps virus (MuV) and compared to 5 others. A new lineage has been identified among UK isolates. Changes in the transcription pattern could not be correlated with differences in the sequences of the F-SH and SH-HN intergenic regions of the genome.

The Irish Competition (Amendment) Act 2012 introduced court-endorsed commitment agreements to Irish competition law. The new section 14B of the principal Competition Act 2002 provides for making commitment agreements between the Irish Competition and undertakings an order of the Irish High Court. This piece, first, investigates the prior Irish practice regarding commitment or settlement agreements and its legal basis. It looks then into the newly introduced rules on court-endorsed commitment agreements. Finally, before concluding, it points to the first instance of their application — to an order issued by the High Court in the FitFlop case in December 2012, which came into effect in February 2013.

Finding a suitable cell source for endothelial cells (ECs) for cardiovascular regeneration is a challenging issue for regenerative medicine. In the paper we describe a novel mechanism regulating induced pluripotent stem cells (iPSC) differentiation into ECs, with a particular focus on miRNAs and their targets. We first established a protocol using collagen IV and VEGF to drive the functional differentiation of iPSCs into ECs and compared the miRNA signature of differentiated and undifferentiated cells. Among the miRNAs overrepresented in differentiated cells, we focused on microRNA-21 (miR-21) and studied its role in iPSC differentiation. Overexpression of miR-21 in pre-differentiated iPSCs induced EC marker upregulation and in vitro and in vivo capillary formation; accordingly, inhibition of miR-21 produced the opposite effects. Importantly, miR-21 overexpression increased TGF-β2 mRNA and secreted protein level, consistent with the strong upregulation of TGF-β2 during iPSC differentiation. Indeed, treatment of iPSCs with TGFβ-2 induced EC marker expression and in vitro tube formation. Inhibition of SMAD3, a downstream effector of TGFβ-2, strongly decreased VE-cadherin expression. Furthermore, TGFβ-2 neutralization and knockdown inhibited miR-21-induced EC marker expression. Finally, we confirmed the PTEN/Akt pathway as a direct target of miR-21 and we showed that PTEN knockdown is required for miR-21 mediated endothelial differentiation. In conclusion, we elucidated a novel signaling pathway that promotes the differentiation of iPSC into functional ECs suitable for regenerative medicine applications.

Donor lymphocyte infusions (DLI) have been shown to enhance the graft-versus-leukaemia (GVL) effect and induce haematological and molecular remission in patients with relapsed CML following allogeneic bone marrow transplantation (BMT). The potent donor cell-mediated cytolysis following DLI may lead to a short period of aplasia before the re-establishment of donor haematopoiesis. The absence of detectable donor cells in patients prior to DLI infusion may result in permanent aplasia in certain patients. We report on four patients who relapsed 1, 3, 6.5 and 7 years post-BMT for chronic phase CML and were treated with DLI from their original BMT donor. Polymorphic short tandem repeats (STRs) were used to assess haematological chimaerism both prior to and following DLI. At the time of relapse, STR-PCR indicated the presence of donor cells in all four patients, at levels ranging from 1-40%. A clinical and molecular response was seen in 4/4 patients following a short period of cytopenia and all patients remain in clinical remission with a follow-up of 2 months-3 years post-DLI. STR-PCR indicated that a response was occurring during the period of pancytopenia when metaphase analysis was unsuccessful. Lineage-specific analysis of the cellular response to DLI was monitored using STR-PCR of peripheral blood (PB) and bone marrow (BM) lymphocyte-enriched fractions and CD2-positive and -negative T cell fractions. In one patient BM and PB CD34-positive and -negative fractions were also assessed. A change in the ratio of donor:recipient cells in the PB lymphocyte fraction was the earliest molecular indication of an anti-leukaemic response. Subsequent conversion to donor chimaerism occurred in the other lineages and the granulocyte fraction was the last lineage to convert. In conclusion, lineage-specific STR-PCR permits detailed monitoring of subtle changes in donor/recipient cell dynamics in specific lineages following DLI during the crucial pancytopenic phase and may be a useful predictor of haematological response to DLI therapy.

Tumour classification has traditionally focused on differentiation and cellular morphology, and latterly on the application of genomic approaches. By combining chromatin immunoprecipitation with expression array, it has been possible to identify direct gene targets for transcription factors for nuclear hormone receptors. At the same time, there have been great strides in deriving stem and progenitor cells from tissues. It is therefore timely to propose that pairing the isolation of these cell subpopulations from tissues and tumours with these genomics approaches will reveal conserved gene targets for transcription factors. By focusing on transcription factors (lineage-survival oncogenes) with roles in both organogenesis and tumourigenesis at multiple organ sites, we suggest that this comparative genomics approach will enable developmental biology to be used more fully in relation to understanding tumour progression and will reveal new cancer markers. We focus here on neurogenesis and neuroendocrine differentiation in tumours.

Vertreter der Generation Y, Personen, die in den 1980er Jahren geboren wurden, drängen verstärkt in den Arbeitsmarkt. Ihre Wert- und Erwartungshaltung ist deshalb von großem Interesse für Unternehmen, die mit Themen wie dem Fachkräftemangel und demografischen Wandel konfrontiert sind und die Leistungsträger langfristig binden müssen. Ziel dieser Hausarbeit ist herauszufinden, wie ausgeprägt die emotionale Bindung der Generation Y in Unternehmen allgemein ist und unter welcher Voraussetzung affektives Commitment bei der Generation Y entsteht. Ein weiteres Ziel ist die Erarbeitung von Handlungsempfehlungen bezüglich des Personalmanagements von Generation Y in Unternehmen. Generation Y hat hohe Erwartungen an Arbeitsinhalt, Selbstverwirklichungsmöglichkeiten, Aus- und Weiterbildungsmaßnahmen, Unternehmenswerte und -ethik, Sinnhaftigkeit der Tätigkeit und Teamarbeit. Vertreter dieser Generation wünschen sich Wertschätzung, eine gute Beziehung zum Vorgesetzten, viele Partizipationsmöglichkeiten, regelmäßiges Feedback und Flexibilität. Weiterhin sind Work-Life-Balance, Karriereoptionen, Einkommen, digitale Vernetzung und Arbeitsplatzsicherheit wichtige Themen für diese Generation. Affektives Commitment entsteht durch Erfüllung von Erwartungen. Generation Y ist nur bedingt affektiv gebunden, tendiert zu häufigen Unternehmenswechseln und ist durch eine schwache Loyalität gekennzeichnet. Dies liegt hauptsächlich daran, dass die Erwartungen dieser Generation nicht erfüllt werden. Es wird empfohlen HR-Maßnahmen zu implementieren, die auf Erfüllung der Erwartungen von Generation Y abzielen.

Die Erwartungen der Generation Y zu kennen, ist wichtig für Unternehmen, die Leistungsträger aus dieser Generation langfristig binden wollen. Die affektive Bindung der Generation Y gegenüber einer Organisation, der Zusammenhang zwischen der Erfüllung arbeitsbezogener Erwartungen und affektivem Commitment sowie der Fluktuationsneigung der Generation Y werden empirisch überprüft. Die Generation Y hat hohe Erwartungen an Unternehmenswerte und Unternehmenskultur. Es wurden Zusammenhänge zwischen der Erfüllung von Erwartungen und der affektiven Bindung gegenüber einer Organisation gefunden. Es wird empfohlen Personalmanagementmaßnahmen zu implementieren, die passgenau auf die Erfüllung der Erwartungen der Generation Y ausgerichtet sind.
