Vulval differentiation in C. elegans is mediated by an Epidermal growth factor (EGF)- EGF receptor (EGFR) signaling pathway. I have cloned unc-101, a negative regulator of vulval differentiation of the nematode C. elegans. unc-101 encodes a homolog of AP47, the medium chain of the trans-Golgi clathrin-associated protein complex. This identity was confirmed by cloning and comparing sequence of a C. elegans homolog of AP50, the medium chain of the plasma membrane clathrin-associated protein complex. I provided the first genetic evidence that the trans-Golgi clathrin-coated vesicles are involved in regulation of an EGF signaling pathway. Most of the unc-101 alleles are deletions or nonsense mutations, suggesting that these alleles severely reduce the unc-101 activity. A hybrid gene that contains parts of unc-101 and mouse AP4 7 rescued at least two phenotypes of unc-101 mutations, the Unc and the suppression of vulvaless phenotype of let-23(sy1) mutation. Therefore, the functions of AP47 are conserved between nematodes and mammals.

unc-101 mutations can cause a greater than wild-type vulval differentiation in combination with certain mutations in sli-1, another negative regulator of the vulval induction pathway. A mutation in a new gene, rok-1, causes no defect by itself, but causes a greater than wild-type vulval differentiation in the presence of a sli-1 mutation. The unc-101; rok-1; sli-1 triple mutants display a greater extent of vulval differentiation than any double mutant combinations of unc-101, rok-1 and sli-1. Therefore, rok-1 locus defines another negative regulator of the vulval induction pathway.

I analyzed a second gene encoding an AP47 homolog in C. elegans. This gene, CEAP47, encodes a protein 72% identical to both unc-101 and mammalian AP47. A hybrid gene containing parts of unc-101 and CEAP47 sequences can rescue phenotypes of unc-101 mutants, indicating that UNC- 101 and CEAP47 proteins can be redundant if expressed in the same set of cells.

Several different methods have been employed in the study of voltage-gated ion channels. Electrophysiological studies on excitable cells in vertebrates and molluscs have shown that many different voltage-gated potassium (K+) channels and sodium channels may coexist in the same organism. Parallel genetic studies in Drosophila have identified mutations in several genes that alter the properties of specific subsets of physiologically identified ion channels. Chapter 2 describes molecular studies that identify two Drosophila homologs of vertebrate sodium-channel genes. Mutations in one of these Drosophila sodium-channel genes are shown to be responsible for the temperature-dependent paralysis of a behavioural mutant parats. Evolutionary arguments, based on the partial sequences of the two Drosophila genes, suggest that subfamilies of voltage-gated sodium channels in vertebrates remain to be identified.

In Drosophila, diverse voltage-gated K+ channels arise from alternatively spliced mRNAs generated at the Shaker locus. Chapter 3 and the Appendices describe the isolation and characterization of several human K+-channel genes, similar in sequence to Shaker. Each of these human genes has a highly conserved homolog in rodents; thus, this K+-channel gene family probably diversified prior to the mammalian radiation. Functional K+ channels encoded by these genes have been expressed in Xenopus oocytes and their properties have been analyzed by electrophysiological methods. These studies demonstrate that both transient and noninactivating voltage-gated K+ channels may be encoded by mammalian genes closely related to Shaker. In addition, results presented in Appendix 3 clearly demonstrate that independent gene products from two K+-channel genes may efficiently co-assemble into heterooligomeric K+ channels with properties distinct from either homomultimeric channel. This finding suggests yet another molecular mechanism for the generation of K+-channel diversity.

Corynebacterium diphtheriae é um importante patógeno humano e agente causal da difteria. Embora seja observada uma redução mundial no número de casos da doença, a difteria permanece endêmica em muitos países e surtos são esporadicamente notificados. No Brasil, o último surto ocorreu no estado no Maranhão e revelou mudanças em aspectos clínico-epidemiológicos da doença. Diferentemente da maioria das cepas de difteria brasileiras, que pertencem ao biovar mitis, nesse surto dois diferentes pulsotipos de C. diphtheriae biovar intermedius foram isolados. Além disso, sinais patognomônicos da doença não foram relatados em parte dos casos. C. diphtheriae também vem sendo relacionado com quadros de infecções invasivas, apesar de ser reconhecido como patógeno tipicamente extracelular. Em conjunto, estas mudanças no perfil das infecções por C. diphtheriae sugerem a existência de outros fatores de virulência além da produção da toxina diftérica. Neste sentindo, foram realizadas análises de tipagem molecular e de genômica comparativa para avaliar a diversidade genética e o potencial de virulência de cepas de C. diphtheriae isoladas de difteria clássica e infecções invasivas. Os resultados obtidos demonstram a circulação de diferentes clones invasores no Brasil. Além disso, revelaram diferenças marcantes na presença e na composição de ilhas de patogenicidade entre as amostras, bem como nos genes sob regulação do DtxR e nas sequências dos corinefagos integradas ao cromossomo bacteriano. Uma vez que o potencial invasor bacteriano e a persistência no ambiente podem estar relacionados à tolerância ao estresse oxidativo, foram procurados nos genomas sequenciados, genes possivelmente envolvidos neste processo. Dentre estes, os genes DIP0906, predito como gene de resistência ao oxidante telurito (TeO32-), e DIP1421, codificador do regulador transcricional OxyR, foram caracterizados funcionalmente e tiveram seus papéis na patogenicidade investigados. Ensaios in vivo utilizando o nematódeo Caenorhabditis elegans demonstraram que ambos são importantes para a virulência de C. diphtheriae. Além da resistência ao TeO32, DIP0906 parece contribuir para a resistência ao peróxido de hidrogênio (H2O2) e para a viabilidade no interior de células respiratórias humanas. Já OxyR, além de controlar negativamente a resposta ao H2O2, parece estar envolvido com a ligação de C. diphtheriae a proteínas plasmáticas e de matriz extracelular. Adicionalmente, foi investigada resistência e a capacidade de adaptação de C. diphtheriae frente a agentes oxidantes, através da indução de resposta adaptativa e/ou resistência cruzada e da formação de biofilme. As cepas de C. diphtheriae apresentaram diferentes níveis de resistência e um comportamento heterogêneo na presença dos agentes oxidantes, o que sugere a existência de diferentes estratégias de sobrevivência e adaptação de C. diphtheriae nas condições de estresse oxidativo.

The origin of cytoskeleton and the origin of relevant intracellular transportation system are big problems for understanding the emergence of eukaryotic cells. The present article summarized relevant information of evidences and molecular traces on the origin of actin, tubulin, the chaperonin system for folding them, myosins, kinesins, axonemal dyneins and cytoplasmic dyneins. On this basis the authors proposed a series of works, which should be done in the future, and indicated the ways for reaching the targets. These targets are mainly: 1) the reconstruction of evolutionary path from MreB protein of archaeal ancestor of eukaryotic cells to typical actin; 2) the finding of the MreB or MreB-related proteins in crenarchaea and using them to examine J. A. Lake's hypothesis on the origin of eukaryote from "eocytes" (crenarchaea); 3) the examinations of the existence and distribution of cytoskeleton made of MreB-related protein within coccoid archaea, especially in amoeboid archaeon Thermoplasm acidophilum; 4) using Thermoplasma as a model of archaeal ancestor of eukaryotic cells; 5) the searching for the homolog of ancestral dynein in present-day living archaea. During the writing of this article, Margulis' famous spirochaete hypothesis on the origin of flagella and cilia was unexpectedly involved and analyzed from aspects of tubulins, dyneins and spirochaetes. Actually, spirochaete cannot be reasonably assumed as the ectosymbiotic ancestor of eukaryotic flagella and cilia, since their swing depends upon large amount of bacterial flagella beneath the flexible outer wall, but not depends upon their intracellular tubules and the assumed dyneins. In this case, if they had "evolved" into cilia and lost their bacterial flagella, they would immediately become immobile! In fact, tubulin and dynein-like proteins have not been found in any spirochaete.

To investigate germline development and germ cell specification, we identified a Dazl homolog (CagDazl) from gynogenetic gibel carp (Carassius auratus gibelio). Its cDNA sequence and BAC clone sequence analyses revealed the genomic organization conservation and conserved synteny of the Dazl family members and their neighborhood genes among vertebrates, especially in fish. Moreover, a polyclonal antibody specific to CagDazl was produced and used to examine its expression and distribution throughout germline development at protein level. Firstly, ovary-specific expression pattern of CagDazl was confirmed in adult tissues by RT-PCR and Western blot. In addition, in situ hybridization and immunofluorescence localization demonstrated its specific expression in germ cells, and both its transcript and protein were localized to germ plasm. Then, co-localization of CagDazl and mitochondrial cloud was found, confirming that CagDazl transcript and its protein are germ plasm component and move via METRO pathway during oogenesis. Furthermore, the CagDazl is abundant and continuous throughout germline development and germ cell specification including primordial germ cell (PGC) formation, oogonium differentiation, oocyte development, and embryogenesis, and the dynamic distribution occurs at different development stages. The data suggest that maternal CagDazl might play an important role in gibel carp PGC formation. Therefore, CagDazl is a useful and specific marker for tracing germ plasm and germ cell development in the gynogenetic gibel carp. In addition, in comparison with previous studies in sexual reproduction species, the continuous and dynamic distribution of CagDazl protein in the germ plasm throughout the life cycle seems to have significant implication in sex evolution of vertebrates. Surface sediments and bivalves were collected from the Changjiang Estuary in December 2003 and November 2004, respectively. Polychlorinated dibenzo-p-dioxins (PCDDs) and dibenzofurans (PCDFs) in these samples were measured with high-resolution chromatography (HRGC)/High Resolution Mass Spectrometer (HRMS). The concentrations of total PCDD/Fs and toxic equivalent (TEQ) were 169.83 +/- 119.63 and 0.81 +/- 0.36 pg/g dry weight (dw) in sediments, and 580.33 +/- 240.17 and 7.24 +/- 3.65 pg/g dw in bivalves. The homolog compositions of PCDD/Fs were similar among samples, the most abundant congener was octa-chlorinated dibenzo-p-dioxin (OCDD) and then octa-chlorinated dibenzofuran (OCDF) and 1,2,3,4,6,7,8-hepta-chlorinated dibenzo-p-dioxin (HOCDD). The herbicide pentachlorophenol (PCP) and sodium pentachlorophenol (Na-PCP) were proved the main source of PCDD/Fs in this area.

Hir/Hira (histone regulation) genes were first identified in yeast as negative regulators of histone gene expression. It has been confirmed that HIRA is a conserved family of proteins present in various animals and plants. In this paper, the cDNAs of the Hira homolog named CagHira and CaHira were isolated from gynogenetic gibel carp (gyno-carp) and gonochoristic color crucian carp (gono-carp) respectively. The full-length CagHira is 3,860 bp in length with an open reading frame (ORF) of 3,033 bp that encodes 1,011 amino acids, while the full-length CaHira is 3,748 bp in length and also has an ORF of 3,033 bp. The deduced amino acid sequences of both Hira homologs contain seven WD domains and show high identity with other HIRA family members. RT-PCR analyses revealed strong expression of Hira in the ovaries, whereas no expression was detected in the testes of either of the fishes. Hira transcription was not detected in the liver of gyno-carp, but a high level of Hira mRNA was observed in gono-carp. The temporal expression pattern showed that the Hira mRNA is consistently expressed during all embryonic development stages in gyno-carp. However, the abundance of CaHira mRNA significantly decreased (P < 0.05) shortly after fertilization and then increased again and remained stable from gastrula till hatching. The varying spatiotemporal expression patterns of Hira genes in gyno-carp and gono-carp may be associated with the differing reproductive modes used by these two closely related fishes. Our results suggest that Hira may play a role not only in the decondensation of sperm nucleus and the formation of pronucleus during fertilization, but also in gastrulation and the subsequent development of embryos.

Upregulated gene 19 (U19)/ELL-associated factor 2 (Eaf2) is a potential human tumor suppressor that exhibits frequent allelic loss and downregulation in high-grade prostate cancer. U19/Eaf2, along with its homolog Eaf1, has been reported to regulate transcriptional elongation via interaction with the eleven-nineteen lysine-rich leukemia (ELL) family of proteins. To further explore the tumor-suppressive effects of U19/Eaf2, we constructed and characterized a murine U19/Eaf2-knockout model. Homozygous or heterozygous deletion of U19/Eaf2 resulted in high rates of lung adenocarcinoma, B-cell lymphoma, hepato cellular carcinoma and prostate intraepithelial neoplasia. Within the mouse prostate, U19/Eaf2 defficiency enhanced cell proliferation and increased epithelial cell size. The knockout mice also exhibited cardiac cell hypertrophy. These data indicate a role for U19/Eaf2 in growth suppression and cell size control as well as argue for U19/Eaf2 as a novel tumor suppressor in multiple mouse tissues. The U19/Eaf2 knockout mouse also provides a unique animal model for three important cancers: lung adenocarcinoma, B-cell lymphoma and hepatocellular carcinoma.

Tumor necrosis factor receptor-associated factor 2 (TRAF2) is a crucial component of almost the entire tumor necrosis factor receptor superfamily signaling pathway. In the present study, a TRAF2 gene has been cloned from grass carp (Ctenopharyngodon idella) by reverse transcription-polymerase chain reaction and rapid amplification of cDNA ends. The full-length cDNA is 3162 bp, including a 60 bp 5' untranslated region (UTR), a 1611 bp open reading frame, and a 1491 bp 3' UTR. The polyadenylation signal (AATAAA) and the mRNA instability motifs (ATTTTA, ATTTA) were followed by a poly(A) tail in the 3' UTR. No signal peptide or transmembrane region has been found in the putative amino acids of grass carp TRAF2 (gcTRAF2). Phylogenetic tree analysis clearly showed that gcTRAF2 is nearest to the TRAF2 gene of goldfish. The identity of gcTRAF2 with its homologs in other vertebrates ranges from 56% to 97%. It is characterized by one RING-type signature at the N-terminus, one zinc finger in the middle part, and one conserved TRAF domain consisting of a C-proximal (TRAF-C) subdomain and a N-proximal (TRAF-N) subdomain. The identity of TRAF-C among all TRAF2 homologs in vertebrates varies from 78% to 97%, whereas the identity of TRAF-N ranges from 56% to 100%. The recombinant gcTRAF2 has been expressed in Escherichia coli using pET-32a expression vector. The rabbit anti-gcTRAF2 polyclonal antibody was obtained. The expression of gcTRAF2 in different organs was examined by real-time quantitative polymerase chain reaction and Western blot analysis. It was widely distributed in heart, head kidney, thymus, brain, gill, liver, spleen, and trunk kidney. This is the first report of a TRAF2 homolog molecule in fish.

The 3 beta-hydroxysteroid dehydrogenase (3 beta-HSD) isoenzymes play a key role in cellular steroid hormone synthesis. Here, a 3 beta-HSD gene homolog,was cloned from Rana grylio virus (RGV), a member of family Iridoviridae. RGV 3 beta-HSD gene has 1068 bp, encoding a 355 aa predicted protein. Transcription analyses showed that RGV 3 beta-HSD gene was transcribed immediate-early during infection from an initiation site 19 nucleotides upstream of the translation start site. Confocal microscopy revealed that the 3 beta-HSD-EGFP fusion protein was exclusively colocalized with the mitochondria marker (pDsRed2-Mito) in EPC cells. Upon morphological observation and MTT assay, it was revealed that overexpression of RGV 3 beta-HSD in EPC cells could apparently suppress RGV-induced cytopathic effect (CPE). The present studies indicate that the RGV immediate-early 3 beta-HSD gene encodes a mitochondria-localized protein, which has a novel role in suppressing virus-induced CPE. All these suggest that RGV 3 beta-HSD might be a protein involved in host-virus interaction. @ 2006 Elsevier Inc. All rights reserved.

Pigment epithelium-derived factor (PEDF) is acknowledged to be a non-inhibitory member of the serine protease inhibitor (serpin) superfamily, with antiangiogenesis, and neuroprotective and immumoregulatory function, mainly in the tissues of nervous system. Here, A PEDF gene homolog, Paralichthys olivaceus PEDF (PoPEDF), was isolated from flounder embryonic cells (FEC) treated with UV-inactivated Grass carp hemorrhage virus (GCHV) and subsequently identified as a differentially expressed gene. The full length of PoPEDF cDNA is 1803 bp with an open reading frame of 1212 bp encoding a 403-amino-acid protein. This deduced protein contains an N-terminal signal peptide, a glycosylation site, a consensus serpin motif, and a 34-mer and a 44-mer fragment, all of which are very conserved in the PEDF family. PoPEDF gene exhibits a conserved exon-intron arrangement with 8 exons and 7 introns. This conserved evolutionary relationship was further confirmed by a phylogenetic analysis, where fish PEDFs and mammalian members formed a well-supported clade. Constitutive expression of PoPEDF was widely detected in many tissues. In response to UV-inactivated GCHV or poly(I:C), PEDF mRNA was upregulated in FEC cells with time. The RNA helicase Vasa is a germ cell marker in animals, and its homolog in vertebrates to date has been limited to bisexual reproduction. We cloned and characterized CagVasa, a Vasa homolog from the gibel carp, a fish that reproduces bisexually or gynogenetically. CagVasa possesses 14 RGG repeats and eight conserved motifs of Vasa proteins. In bisexually reproducing gibel carp, vasa is maternally supplied and its zygotic expression is restricted to gonads. By in situ hybridization on testicular sections, vasa is low in spermatogonia, high in primary spermatocytes, reduced in secondary spermatocytes, but disappears in spermatids and sperm. In contrast, vasa persists throughout oogenesis, displaying low-high-low levels from oogonia over vitellogenic oocytes to maturing oocytes. A rabbit anti-Vasa antibody (alpha Vasa) was raised against the N-terminal CagVasa for fluorescent immunohistochemistry. On testicular sections, Vasa is the highest in spermatogonia, reduced in spermatocytes, low in spermatids, and absent in sperm. In the ovary, Vasa is the highest in oogonia but persists throughout oogenesis. Subcellular localization of vasa and its protein changes dynamically during oogenesis. The aVasa stains putative primordial germ cells in gibel carp fry. It detects gonadal germ cells also in several other teleosts. Therefore, Cagvasa encodes a Vasa ortholog that is differentially expressed in the testis and ovary. Interestingly, the alpha Vasa in combination with a nuclear dye can differentiate critical stages of spermatogenesis and oogenesis in fish. The cross-reactivity and the ability to stain stage-specific germ cells make this antibody a useful tool to identify fish germ cell development and differentiation. (c) 2005 Wiley-Liss, Inc.

蝰科蛇毒中含有丰富的具有血管通透增强活性的蛋白组分,本论文结合生物化学与分子生物学手段对几种毒蛇的蛇毒血管内皮生长因子进行了研究。其中,从云南产菜花烙铁头(Trimeresurus jerdonii)蛇毒中分离得到一个血管内皮生长因子,TjsvVEGF,并研究了其活性与受体结合特性的关系。同时,对圆斑蝰蛇、蝮蛇、山烙铁头和竹叶青等几种蛇的svVEGFs进行了分子生物学研究。此外,我们还分离到了一个蛇毒丝氨酸蛋白酶类似物,TjsvSPH,还对该蛋白的理化性质进行了初步研究。 TjsvVEGF是一个表观分子量为29 kDa的二聚体蛋白,由两个相同大小的亚基组成。活性实验表明,它在10ng的剂量下即可明显提高血管通透性,活性强度与VEGF165相当。虽然TjsvVEGF的氨基酸序列与TfsvVEGF和Pm-VEGF具有较高的相似性,但是它们的受体结合特性却有很大差异。TjsvVEGF与VEGFR-1的结合能力很弱,而对VEGR-2有很高的亲和力。这说明,TjsvVEGF的活性主要是VEGFR-2介导的。最后,我们对导致TjsvVEGF对VEGFR-1低亲和力的原因进行了探讨。 利用PCR方法,我们从圆斑蝰蛇毒腺中得到了三种svVEGFs蛋白编码区长短不一的cDNA序列,其差异是由cDNA链中一段富含AG(3’拼接接受位点)的区段发生了核苷酸缺失产生的。蛋白编码区核苷酸的缺失导致其编码的三种svVEGF蛋白N末端的氨基酸序列和长短均产生较大差异。因此我们推测,蝰属svVEGFs蛋白N末端普遍较短可能是编码它们的mRNA前体对3’拼接点的不同选择产生的。同时,通过对cDNAs推导的氨基酸序列分析发现,蝮蛇svVEGF和山烙铁头svVEGF在与受体结合相关的多个重要位点上发生了氨基酸替换,提示它们是研究svVEGFs与VEGFR结合机制的良好材料。 通过分子筛、离子交换和亲和层析等方法,我们还从菜花烙铁头蛇毒中得到了一个不具有酶活性的丝氨酸蛋白酶类似物,命名为:TjsvSPH。内肽序列测定结果表明,TjsvSPH三联体结构中的组氨酸已突变为精氨酸,这很可能是导致其失去蛋白水解酶活性的主要原因。活性检测验表明,与许多已发现的蛇毒活性组分不同,TjsvSPH不具有蛋白水解酶活性,不能引起血小板聚集,也不抑制ADP和凝血酶诱导的血小板聚集。它对人血浆的复钙时间也没有影响。TjsvSPH的氨基酸序列与典型蛇毒丝氨酸蛋白酶Halystase和Calobin有约77%的一致性。它与同科属来源的蛇毒丝氨酸蛋白酶类似物氨基酸序列相似性高达92%以上,与不同科属的也有约74%以上的相似性。通过对Genbank中六种毒蛇所有丝氨酸蛋白酶及其类似物进化分析,我们推测,蛇毒丝氨酸蛋白酶类似物很可能是由蛇毒丝氨酸蛋白酶进化而来,并在进化过程中形成了一类独特的蛋白质。

人LMBR1(Limb region 1 homolog (mouse)) 基因位于染色体7q36区域,全长约210.2 kb,含17个外显子,编码一个由490个氨基酸构成的跨膜蛋白。研究表明,LMBR1 基因的表达活性与脊椎动物四肢的手指或脚趾数目变化有关;另外,发生在其重要元件——intron 5 内的许多变异与多种表型的轴前多指症((PPD, Preaxial polydactyly)存在相关性,这主要是因为LMBR1 intron 5 内含有一个与骨骼系统发育有关的基因(SHH(Sonic hedgehog)基因)的远程顺式调控元件。本研究旨在探究LMBR1基因 intron 5 内的遗传多样性,进而评估HapMap计划的样本选择策略,并检测该区域是否受自然选择的作用。 国际人类基因组单体型图计划(HapMap Project,The International Haplotype Map Project) 于2002年10月正式启动,该计划旨在构建人类基因组中常见变异的遗传图式。自其数据发布以来,广泛应用于生物医学、群体遗传学等领域,在复杂疾病的遗传机理研究、自然选择的检测等方面做出了前所未有的贡献;但是HapMap计划中样本的代表性有待评估。 本研究中,我们综合考虑地理来源信息及线粒体单倍型类群 (Haplogroup)信息选择了41个东亚人作为样本(以保证样本的代表性),测定位于LMBR1 基因intron 5 内的目的片段中存在的单核苷酸多态性(SNP, Singe nucleotide polymorphism)位点,通过所得数据与HapMap数据的比较,发现二者之间差异显著且HapMap数据不能覆盖所有我们得到的常见变异,因而我们认为:HapMap计划中国部分的样本选择策略有待进一步完善。 关于自然选择的研究不仅可以使我们了解生物的进化机制,同时还对复杂疾病的遗传机理研究具有重要的提示作用,因而,对于自然选择的检测,一直以来都是生物学研究的重点。平衡选择是一种维持遗传多态性的自然选择方式,现已发现很多与特定疾病或性状相关的基因或调控序列受平衡选择的作用,如 G6PD 基因、PTC 基因、FMO3 基因、FSHB 基因及 CCR5 基因5’端顺式调控区等我们对41个东亚样本中LMBR1 intron 5 内一段长为9256 bp (Chr7: 156280954-156271699 (Build36))的序列进行以 Tajima’s D 检验为主的群体遗传学分析,发现该区域在进化历程中受到平衡选择的作用。LMBR1 intron 5 内的多态位点与多种表型的多指症存在相关性,受其调控的 SHH 基因在骨骼系统发育中具有重要作用,人类骨骼系统的适应性进化等三方面的因素为该区域受平衡选择的作用提供了进一步的佐证。 总之,本研究对HapMap计划的样本选择策略和数据应用提供了一定的参考;同时还发现一个与骨骼系统发育有关的基因调控元件受平衡选择的作用。

土壤重金属污染是影响农业可持续发展和生态环境的重要问题,而通过生物标记物方式对污染土壤进行早期诊断已成为环境科学领域的研究热点。本文以M&S营养液为培养介质,以拟南芥为供试材料,以错配修复基因MutS 2 homolog (atMSH2),atMSH3,atMSH7,细胞增殖核抗原1 和2 (atPCNA1和atPCNA2)为检测目的基因,分别采用半定量反转录-聚合酶链式反应(RT-PCR)技术、克隆及测序技术研究了Cd在不同胁迫水平(0,0.125,0.25,1.0,3.0 mg•L-1 )上对上述错配修复相关基因表达和atMSH2基因突变的影响,并与拟南芥幼苗形态、生理指标的毒性效应进行比较分析,筛选出对Cd污染胁迫敏感的生物标记物。主要结果如下: 1. 不同浓度(0,0.125,0.25,1.0,3.0 mg•L-1 )Cd处理7天后,拟南芥幼苗叶片数、地上部鲜重变化与对照相比差异均不显著;而根长随Cd胁迫强度的增加明显降低; 2. 不同浓度(0,0.125,0.25,1.0,3.0 mg•L-1 )Cd处理7天后,叶绿素含量变化与对照相比差异均不显著; 地上部可溶性蛋白含量随Cd浓度的增加变化明显,0.125 mg•L-1 Cd处理下,地上部可溶性蛋白含量显著增加,而在0.25,1.0和3.0 mg•L-1 Cd时降低,但仍高于对照; 3. 地上部atMSH2,atPCNA1,atPCNA2基因表达量的变化与Cd胁迫浓度呈明显的倒U字型关系,分别在0.125mg•L-1,0.25mg•L-1和0.125mg•L-1 Cd时达到最大值。地上部可溶性蛋白含量变化趋势与atMSH2,atPCNA1,atPCNA2基因表达量的变化相似,均可作为对Cd污染胁迫敏感的潜在生物标记物。 4. 对不同浓度(0,0.125,0.25,1.0,3.0 mg•L-1 )Cd处理7天后,拟南芥atMSH2基因PCR后的扩增产物进行回收、纯化、克隆和测序。测序结果表明,0.25 mg•L-1 Cd处理拟南芥atMSH2基因在第8个和第9个外显子之间的内含子有一个碱基转换;在1.0 mg•L-1 Cd处理下,拟南芥在第10个外显子有一个碱基转换。
