The FOB-3, anew type fiber optic biosensor, is designed to rapidly detect a variety of biological agents or analytes with better stability, sensitivity and specificity. In order to detect Y. Pestis, a sandwich immunoassay was developed by using the purified antibody against antigen FI immobilized on polystyrene probes as the capture antibody and the monoclonal antibody-Cy5 conjugate as the detector. After a series of optimization for the stability, sensitivity and specificity of the FOB-3, 50-1000 ng/ml of antigen FI and 6 x 10(1)-6 x 10(7) CFU/ml Y. pestis could be detected constantly in about 20 min, and Y pestis could be detected specifically from Y. pseudotuberculosis, Y. enterocolitica, B. anthracis and E. coli. Then, 39 blind samples, including 27 tissues of mice infected with Y pestis and 12 tissues of healthy mice as negative control, were detected with the FOB-3. 92.6% infected tissues were identified from the tissues of healthy mice and the tissues containing more than 100 CFU/ml bacteria could be detected by the biosensor. The results demonstrated the feasibility of the FOB-3 as an effective method to detect Y. pestis rapidly and directly from the infected animal specimens with the advantage of portability, simple-operation as well as high sensitivity and specificity. Objective: Although dobutamine is widely used in neonatal clinical practice, the evidence for its use in this specific population is not clear. We conducted a systematic review of the use of dobutamine in juvenile animals to determine whether the evidence from juvenile animal experiments with dobutamine supported the design of clinical trials in neonatal/ paediatric population. Methods: Studies were identified by searching MEDLINE (1946-2012) and EMBASE (1974-2012). Articles retrieved were independently reviewed by three authors and only those concerning efficacy and safety of the drug in juvenile animals were included. Only original articles published in English and Spanish were included. Results: Following our literature search, 265 articles were retrieved and 24 studies were included in the review: 17 focused on neonatal models and 7 on young animal models. Although the aims and design of these studies, as well as the doses and ages analysed, were quite heterogeneous, the majority of authors agree that dobutamine infusion improves cardiac output in a dose dependent manner. Moreover, the cardiovascular effects of dobutamine are influenced by postnatal age, as well as by the dose used and the duration of the therapy. There is inadequate information about the effects of dobutamine on cerebral perfusion to draw conclusions. Conclusion: There is enough preclinical evidence to ensure that dobutamine improves cardiac output, however to better understand its effects in peripheral organs, such as the brain, more specific and well designed studies are required to provide additional data to support the design of clinical trials in a paediatric population.

Background: There is growing evidence that microglia are key players in the pathological process of amyotrophic lateral sclerosis (ALS). It is suggested that microglia have a dual role in motoneurone degeneration through the release of both neuroprotective and neurotoxic factors. Results: To identify candidate genes that may be involved in ALS pathology we have analysed at early symptomatic age (P90), the molecular signature of microglia from the lumbar region of the spinal cord of hSOD1(G93A) mice, the most widely used animal model of ALS. We first identified unique hSOD1(G93A) microglia transcriptomic profile that, in addition to more classical processes such as chemotaxis and immune response, pointed toward the potential involvement of the tumour suppressor gene breast cancer susceptibility gene 1 (Brca1). Secondly, comparison with our previous data on hSOD1(G93A) motoneurone gene profile substantiated the putative contribution of Brca1 in ALS. Finally, we established that Brca1 protein is specifically expressed in human spinal microglia and is up-regulated in ALS patients. Conclusions: Overall, our data provide new insights into the pathogenic concept of a non-cell-autonomous disease and the involvement of microglia in ALS. Importantly, the identification of Brca1 as a novel microglial marker and as possible contributor in both human and animal model of ALS may represent a valid therapeutic target. Moreover, our data points toward novel research strategies such as investigating the role of oncogenic proteins in neurodegenerative diseases.

Background: Previous studies suggest that dietary protein might play a beneficial role in combating obesity and its related chronic diseases. Total, animal and plant protein intakes and their associations with anthropometry and serum biomarkers in European adolescents using one standardised methodology across European countries are not well documented. Objectives: To evaluate total, animal and plant protein intakes in European adolescents stratified by gender and age, and to investigate their associations with cardio-metabolic indicators (anthropometry and biomarkers). Methods: The current analysis included 1804 randomly selected adolescents participating in the HELENA study (conducted in 2006-2007) aged 12.5-17.5 y (47% males) who completed two non-consecutive computerised 24-h dietary recalls. Associations between animal and plant protein intakes, and anthropometry and serum biomarkers were examined with General linear Model multivariate analysis. Results: Average total protein intake exceeded the recommendations of World Health Organization and European Food Safety Authority. Mean total protein intake was 96 g/d (59% derived from animal protein). Total, animal and plant protein intakes (g/d) were significantly lower in females than in males and total and plant protein intakes were lower in younger participants (12.5-14.9 y). Protein intake was significantly lower in underweight subjects and higher in obese ones; the direction of the relationship was reversed after adjustments for body weight (g/(kg.d)). The inverse association of plant protein intakes was stronger with BMI z-score and body fat percentage (BF%) compared to animal protein intakes. Additionally, BMI and BF% were positively associated with energy percentage of animal protein. Conclusions: This sample of European adolescents appeared to have adequate total protein intake. Our findings suggest that plant protein intakes may play a role in preventing obesity among European adolescents. Further longitudinal studies are needed to investigate the potential beneficial effects observed in this study in the prevention of obesity and related chronic diseases.

Three diets were formulated using locally available feed ingredients in Malawi to test the effect of replacing animal protein (fish meal, meat and bone meal) with soybean meal (10:0, 5:5, 0:10% of diet) as the protein source on growth and feed conversion of Oreochromis karongae. There were no significant differences in growth rate (GR), specific growth rate (SGR) and feed conversion ratios (FCR) among the three diets. It can be concluded that more expensive and limited animal protein sources can totally be replaced by cheaper soybean in order to get similar growth rates in O. karongae.

A experimentação animal é uma prática extremamente incorporada à ciência biomédica moderna, com raízes desde a Antiguidade, mas que foi cada vez mais se desenvolvendo, adquirindo características bem definidas. O animal não humano, no contexto da experimentação, é um ser vulnerado, enclausurado dentro de uma estrutura muito bem definida, que mantém um discurso focado na excelência do modelo do animal na pesquisa e testes, com o objetivo de garantir a manutenção dessas práticas. No entanto, ao analisarmos os dados científicos obtidos com a experimentação animal, que visa primariamente um possível benefício humano, assim como as argumentações éticas relativas ao respeito à vida e ao sofrimento do animal envoltas nesta situação, a argumentação ética prepondera sobre uma base científica cuja eficácia é também extremamente questionável. Partindo de uma análise crítica da aplicação do modelo animal como padrão da pesquisa científica e da concepção ética do reconhecimento da vulnerabilidade do animal não humano na experimentação, propomos que esta reflexão colabore com políticas públicas que incentivem e implementem as técnicas substitutivas ao uso de animais não humanos em pesquisa, testes da indústria e ensino na área biomédica no Brasil.

A obesidade atinge proporções epidêmicas em países industrializados e está relacionada a uma série de doenças metabólicas e circulatórias. Nesse contexto, a atividade física, tratamento não farmacológico da obesidade, acessível a diversas populações e está relacionada com a redução do risco cardiovasvascular mesmo. O objetivo deste trabalho foi avaliar, após mudança ou não da dieta, associação ou não a um programa de treinamento aeróbico (PTA) durante 8 semanas, a possível reversibilidade dos danos causados por uma dieta hiperlipídica por 12 semanas. Para tal, 120 hamsters machos da espécie Mesocricetus auratus, com massa corporal de 60 g, foram distribuídos em quatro grupos, cada um subdividido em três subgrupos, com dez animais para diferentes análises. Os grupos Obeso Controle (OBC) e Obeso Exercitado (OBEX) receberam a ração hiperlipídica por 20 semanas, com adição do PTA ao grupo OBEX nas últimas 8 semanas. Os Obeso Ração Padrão (OBRP) e Obeso Ração Padrão/Exercício (OBRP/EX) tiveram a ração modificada para comercial padrão e adição do PTA ao grupo OBRP/EX após as 12 semanas iniciais. Para as análises microcirculatórias, a bolsa da bochecha foi usada para determinação do número máximo de extravasamentos induzidos por 30 min de isquemia seguida de reperfusão e da reatividade microvascular após a aplicação tópica de acetilcolina e nitroprussiato de sódio. No sangue coletado foi avaliado o perfil lipídico, glicemias quinzenais e leptina. As expressões de eNOS e iNOS foram determinadas na aorta por imunoblotting e a composição corporal avaliada nos tecidos adiposos visceral, urogenital e retroperitoneal, retirados no dia do experimento. Os resultados foram analisados com os métodos o teste estatístico de análise de variância (One Way ANOVA - Teste de Kruskal-Wallis), seguido pelo pós-teste de Dunn. Resultados mostram que a modificação dietética, associada ou não ao PTA, reduziu significativamente a massa corporal (p<0,0001), comprimento naso-anal (p=0,0011) e tecido adiposo (visceral [p<0,0001], urogenital [p=0.0004] e retroperitoneal [p= 0,0083]). Nas análises sanguíneas não foram encontradas diferenças com relação ao perfil lipídico e glicemia, já na leptina houve uma redução significativa (p=0,0039). A análise da reatividade microvascular mostrou melhora significativa na vasodilatação endotélio-dependente nos grupos submetidos à modificação dietética associada ou não ao PTA. Nas medidas de permeabilidade a macromoléculas houve redução significativa no número de extravasamentos nos grupos submetidos à modificação dietética associada ou não ao PTA, (5 min [p= 0,0207] e 10 min [p= 0,0057]). Houve um aumento na expressão de eNOS nos grupos submetidos à modificação dietética associada ou não ao PTA, em comparação ao grupo OBC (p=0,0352). Os resultados mostraram que a modificação dietética, associada ao protocolo de treinamento aeróbico melhora a vasodilatação endotélio-dependente, aumenta a expressão da óxido nítrico sintase endotelial e reduz o número de extravasamentos induzidos por isquemia e reperfusão, mesmo sem melhoras nos marcadores bioquímicos tradicionais como glicemia e perfil lipídico.

O objetivo principal deste trabalho é a análise do sermão Pecados capitais e hipocrisia, simonia, detração e adulação, circunscrito no conjunto dos escritos antonianos que se reportam ao universo dos bestiários medievais. Na obra em questão, os respectivos vícios humanos são evocados, cada qual, a partir da analogia estabelecida com o comportamento de determinados animais. Santo Antônio de Lisboa (1192-1231) viveu a maior parte de sua vida durante as primeiras décadas do período conhecido como Baixa Idade Média (século XIII ao XV). Data do início dessa era o estabelecimento de uma arte de pregar medieval, que toma como referência as idéias propaladas no princípio do Cristianismo, a filosofia dos Padres da Igreja e, por fim, os diversos preceptores do século XIII, que espelhavam o novo contorno do qual havia se revestido a teoria da prédica, em que se sublinhava também a forma de pregar, até então preterida em função do conteúdo da oratória. É também nessa época que tomam vulto as ordens mendicantes, dentre as quais se destacam os franciscanos. Santo Antônio, como pregador dos Frades Menores, valeu-se dos conhecimentos adquiridos durante o período em que integrou a Ordem dos Cônegos Regrantes de Santo Agostinho; da filosofia propalada pelo fundador da Ordem instituída por São Francisco de Assis; e, por fim, de todos os artifícios que lhe oferecia a oratória de seu tempo. Dirigiu sua prédica, sobretudo, aos cátaros, hereges que desprezavam as coisas materiais, inclusive a natureza. Ao utilizar o simbolismo dos bestiários medievais, estima-se que Santo Antônio conciliou três fatores fundamentais na eficácia de seu discurso. O primeiro deles se refere à plena comunhão entre os recursos da ars praedicandi e a alegoria do bestiário medieval. Em seguida, ressalta-se a própria utilização do mundo animal como fator de eficaz eloqüência, considerando-se a concepção, em voga na sociedade medieval, de que a natureza seria um espelho codificado do universo espiritual, e nela estariam subjacentes os propósitos de Deus para com os homens. O terceiro fator a ser destacado é a harmonia entre a utilização do bestiário medieval como subsídio retórico e a filosofia dos franciscanos, que confere valor especial à natureza, sobretudo aos animais

An experiment was conducted with Labeo rohita fingerlings in an indoor static fish rearing water system of glass made aquaria. Five experimental diets A, B, C, D and E were formulated containing 33% dietary protein level in five treatments each having two replicates containing 12 fingerlings of mean total initial weight of 13.00±0.2g. Sixty days of feeding trial in this experiment showed that fish fed on diet 'A' containing fish meal and diet 'E' containing mixed plant sources protein had significantly highest and lowest growth respectively. However, no significant difference of growth was found in fish fed on diets C and D containing meat and bone meal, and mix of animal protein source diets respectively. The result showed that the apparent protein digestibility (APD) of diets 'A' and 'E' had significantly best and least values respectively. Food conversion ratio (FCR) and protein efficiency ratio (PER) ranged between 1.37 to 2.17 and 1.38 to 2.18 respectively. On the basis of observed FCR and PER diets 'A' and 'E' produced significantly highest and lowest growth respectively.

Tripura is a densely populated small state with meagre water resources. 47.51% of the population is constituted by socio-economically backward, illiterate, orthodox, tribal and scheduled castes. Some of them are nomad and a majority of the rest of the population is refugees from Bangladesh, but almost 100% is fish eater. Settlement of tribes in villages, provision of nutritious food and employment therefore calls for proper utilization of every resource they have. The State is poor in water resources but recently has created 21,636.23 ha of new water area. Tripura is rich in pig population, besides poultry birds. Paddy is the main crop cultivated in arable lands. An integration of livestock raising and land based agriculture with pisciculture practices around mini barrages will help in solving the problems to a great extent. The paper is an attempt to outline the prospects of integration of aquaculture with animal husbandry and land based crop culture in Tripura State.

Although shrimp head meal alone does not provide for good growth and survival, fish meal can provide high survival rate. The addition of shrimp head improves this diet. It is suggested that cholesterol present in shrimp could have caused this difference. Composition of the test diets is tabulated, as are proximate chemical analysis of the diets, and the mean initial weights, final weights, weight gains, survival rate, feed consumed, protein consumed, of Penaeus monodon postlarvae, feed conversion and protein efficiency ratio.
