Two new sesquiterpenes, 15-hydroxy-T-muurolol (3d) and 11,15-dihydroxy-T-muurolol (3e), along with the plant cadinenes T-muurolol (3f) and 3 alpha-hydroxy-T-muurolol (3g), were isolated from the marine-derived Streptomyces sp. M491. Their absolute configuration was established via NMR spectroscopy and X-ray crystallography of 3-oxo-T-muurolol (3a), which was reisolated from this strain. In addition, the absolute configuration of further sesquiterpenes previously reported from this strain was revised. These products were tested for their cytotoxicity against 37 human tumor cell lines using the MTT method. Only 3d was cytotoxic against a range of human tumor cell lines with a mean IC50 of 6.7 mu g/mL.

In our screening of marine Streptomycetes for bioactive principles, two novel antitumor antibiotics designated as chinikomycins A (2a) and B (2b) were isolated together with manumycin A (1), and their structures were elucidated by a detailed interpretation of their spectra. Chinikomycins A (2a) and B (2b) are chlorine-containing aromatized manumycin derivatives of the type 64-pABA-2 with an unusual para orientation of the side chains. They exhibited antitumor activity against different human cancer cell lines, but were inactive in antiviral, antimicrobial, and phytotoxicity tests.

The chemical investigation of the crude extract of the marine-derived Streptomyces sp. M491 yielded three new sesquiterpenes, namely, 10 alpha,11-dihydroxyamorph-4-ene (4), 10 alpha,15-dihydroxyamorph-4-en-3-one (6), and 5 alpha,10 alpha,11-trihydroxyamorphan-3-one (7). In addition, the known compounds 10 alpha-hydroxyamorph-4-en-3-one (2), o-hydroxyacetanilide, genistein, prunetin, and indole-3-carbaldehyde and the macrolide antibiotic chalcomycin A were identified. The structures were determined on the basis of spectroscopic analysis, especially 1D and 2D NMR data. This is the first report of these sesquiterpenes from bacteria.

Marine Streptomyces are potential candidates for novel natural products and industrial catalysts. In order to set up biosynthesis approach for a holomycin-producing strain M095 isotated from Jiaozhou Bay, China, a genetic transformation system was established using intergeneric conjugation. The plasmid pIJ8600 consists of an origin of replication for Escherichia coli, a phage integrase directing efficient site-specific integration in bacterial chromosome, thiostrepton-induced promoter and an attP sequence. Using E. coli ET12567 (pUZ8002) carrying pIJ8600 as a conjugal donor, while it was mated with strain M095, pIJ8600 was mobilized to the recipient and the transferred DNA was also integrated into the recipient chromosome. The frequency of exconjugants was 1.9 +/- 0.13 x 10(-4) per recipient cell. Analysis of eight exconjugants showed pIJ8600 was stable integrated at a single chromosomal site (attB) of the Streptomyces genome. The DNA sequence of the attB was cloned and shown to be conserved. The results of growth and antimicrobial activity analysis indicated that the integration of pIJ8600 did not seem to affect the biosynthesis of antibiotics or other essential amino acids. To demonstrate the feasibility of above gene transfer system, the allophycocyanin gene (apc) from cyanobacterium Anacystis nidulans UTEX625 was expressed in strain M095, and the results indicated heterologous allophycocyanin could be expressed and folded effectively. In the course of a screening program, we have isolated the new natural product, 5,7-dihydroxy-5,6,7,8-tetrahydroazocin-2(IH)-one (1), from the staurosporine producing marine-derived Streptomyces sp. strain QD518. Here we report the isolation and structure elucidation of 1 and the artifacts 3 and 4 resulting from I by acid catalyzed intra- and inter-molecular reactions.

Allophycocyanin is one of the most important marine active peptides. Previous studies suggested that recombinant allophycocyanin (rAPC) could remarkably inhibit the S-180 carcinoma in mice, indicating its potential pharmaceutical uses. Based on intergeneric conjugal transfer, heterologous expression of rAPC was first achieved in marine Streptomyces sp. isolate M097 through inserting the apc gene into the thiostrepton-induced vector pIJ8600. The transformation frequency for this system was approximately 10(-4) exconjugants/recipient. In the transformed Streptomyces sp. isolate M097, the yield of purified rAPC could amount to about 38 mg/l using a simple purification protocol, and HPLC analysis showed that the purity of the protein reached about 91.5%. In vitro activity tests also revealed that the purified rAPC had effective scavenging abilities on superoxide and hydroxyl radicals. This would widen the usefulness of the marine Streptomyces as a host to express the rAPC and to offer industrial strain for the production of rAPC.

Para determinar as variáveis morfogênicas, estruturais e o fluxo de tecidos os tratamentos foram duas intensidades (baixa e moderada) e dois métodos de pastejo (pastejo com lotação contínua e rotacionada). No experimento 1 o bastão graduado apresentou a melhor correlação com a massa de forragem (r2=0,65). No 2 as melhores correlações foram obtidas quando avaliadas as faixas de pós-pastejo para o disco medidor (r2=0,47) e as de pré-pastejo para o bastão graduado (r2=0,36). Para as variáveis morfogênicas e estruturais as intensidades de pastejo foram responsáveis por diferenças na taxa de elongação de folhas (intensidade baixa resultou em maior taxa de elongação) e nas características estruturais (intensidade baixa resultou em menor densidade de perfilhos, maior comprimento e número de folhas vivas). Os métodos de pastejo influenciaram as características morfogênicas (lotação contínua resultou em maior taxa de elongação de folhas, maior taxa de surgimento e tempo de vida das folhas no ciclo de observação I) e estruturais (lotação contínua resultou em maior densidade de perfilhos); bem como foi obtida interação com as intensidades e com os ciclos de avaliação. O fluxo de crescimento (favorecido por lotação rotacionada a baixa intensidade) e de senescência (favorecido por lotação contínua a baixa intensidade) foram afetados pelos tratamentos, enquanto que o fluxo de consumo não foi alterado pelos tratamentos.

The keratin is not degraded by common enzyme, keratinases have the ability to degrade native keratin and others insoluble enzymes. In the present work was Studied keratinase produced by Streptomyces sp LMI-1 isolated from industrial plant of poultry processing. The enzyme degraded 87% of feathers after 120 h, it was stimulated by Ba(2+) and inhibited by Ca(2+), Mn(2+), EDTA and Hg(+). The optimum pH and temperature for the enzyme was 8.5 and 60 degrees C, respectively. The enzyme was stable after 2 hours at 50 degrees C. The culture broth analysis by thin layer chromatography showed presence of amino acids serine, methionine, proline, tyrosine and leucine after 72 hours of incubation. The microorganism showed potential for use in industrial process because of higher enzyme production and feathers degradation.

In a previous work a strain of microorganism was isolated in Araraquara region, SP, Brazil, and characterized as being Streptomyces sp. Ar386, producer of 26-deoxylaidlomycin and of more three polietheric antibiotics. In this paper there are presented studies about fermentation characteristics of the antibiotic performed in incubator agitator utilizing various cultivation technics and media. A medium containing part of energy source as free sugar and part as amilaceous material presented the best result. As the Streptomyces sp. Ar386 produced few spores, it was necessary special care to provide sufficient quantity of microorganisms in inoculation. The biosynthesis of antibiotics intensified between the forth and seventh day. The yields varied from 8 to 443 mg of antibiotic complex per litre of medium. As a polyether antibiotic it may be used as anticoccidal agent in poultry and as growth promoters in cattle and swine.

An actinomycete strain (Ar386) was isolated from the soil of the Araraquara regio, SP, Brazil. The strain, named Streptomyces jacareensis, formed irregular rayed, rugose, grayish-white mycelium with sinuous, branched hyphae carrying rare isolated spores; assimilated glucose, galactose, inositol, ribose, maltose, sucrose, melibiose and starch but not mannitol, rhamnose, arabinose, xylose, lactose and raffinose; and contained LL- diaminopimelic acid in its cell wall. An antibiotic active against Gram- positive bacteria, which was characterized as being 26-deoxylaidlomycin and which may have application against poultry coccidiosis, was isolated from cultures of the strain. This was the first isolation of this antibiotic from a microorganism of the genus Streptomyces and also the first isolation of this antibiotic in Brazil.

Chemical analysis of an Australian Streptomyces species yielded a range of known anthracyclines and biosynthetically related metabolites, including daunomycin (1), E-rhodomycinone (2), 11-hydroxyauramycinone (3), 11-hydroxysulfurmycinone (4), aklavinone (5), bisanhydro-gamma-rhodomycinone (6), and the anthraquinone 7, as well as the hitherto unreported blanchaquinone (8). The structure assigned to 8 was secured by detailed spectroscopic analysis and correlation to known analogues, such as the anthraquinone 7. This account also represents the first natural occurrence of 3, 4, and 7 and the first spectroscopic characterization of 11-hydroxysulfurmycinone (4).

链霉菌是十分重要的一类放线菌,绝大多数的抗生素都由该类细菌产生。毛壳属真菌是一类重要的丝状真菌,从中也发现有很多结构新颖、活性独特的活性物质。因此本论文对两株链霉菌的活性成分及一株金毛壳菌的次生代谢产物进行了研究。 1.从吸水链霉菌(Streptomyces hygroscopicus 1.358)液态发酵产物(乙酸乙酯提取物)中分离得到3个化合物,通过波谱方法鉴定为RK955A (1)、Nigericin(2)、Elaiophylin(3)。以青霉素耐药-金黄色葡萄球菌作为指示菌的抗菌活性测定表明,三者均具有较强抗菌活性。 2.通过抗肿瘤体外活性筛选模型筛选得到得到一株链霉属土壤放线菌,从中分离得到六个化合物:苯乙酰胺(4)、苯丙酰胺(5)、肉桂酰胺(6)、3-(N-(甲酰胺基)乙酰基)吲哚(7)、鸟苷磷酸(8)、鸟苷(9)。 3.从金色毛壳菌(Chaetomium aureus)的固态培养物中分离得到13个化合物,利用波谱方法将其鉴定为:金毛壳菌素A(10)、金毛壳菌素B(11)、Eugenetin(12)、Eugenitol(13)、Chaetoquadrin A(14)、Chaetoquadrin B(15)、Chaetoquadrin G(16)、Chaetoquadrin H(17)、Chaetochromin A(18)、Sterigmatocystin(19)、O-methylsterigmatocystin(20)、3β-羟基-麦角甾-5,7,22-三烯(21)和过氧麦角甾醇(22)。 4.综述了聚醚类抗生素的结构、生物合成、生物活性及作用机理。 The genus Streptomyces (Actinomycetes) is an important group of microbe. Most antibiotics known nowdays are discovered from species of Streptomyces. The fungi of the genus Chaetomium have attracted much attention because various kinds of secondary metabolites with diverse bioactivities have been found from them. Thus, the bioactive compounds from two strains of Streptomyces and the secondary metabolites of Chaetomium aureus were investigated. 1. Three compounds were isolated from the ethyl acetate extract of the fermentation broth of Streptomyces hygroscopicus. They are identified to be elaiophylin (1), nigericin (2), and antibiotic RK955A (3) on the basis of their spectroscopic data. Compounds 1-3 possess antibacterial activities against Staphyloccocus aureus. 2. It was found that the extract of the fermented broth of a strain of Actinomycetes could inhibit some tumor cel lines. Separation of the bioactive fraction led to the isolation of six compounds. They were characterized to be phenylacetamide (4), phenylpropylamide (5), trans-cinnamamide (6), 3- (N- (formylmethyl) acetamide) indole (7), guanylicacid (8), and guanosine (9). 3. From the fermented broth of Chaetomium aureus, 13 compounds were isolated for the first time. They were determined to be chaetomiumycin A (10), chaetomiumycin B (11), eugenetin (12), eugenitol (13), chaetoquadrin A (14), chaetoquadrin B (15), chaetoquadrin G (16), chaetoquadrin H (17), chaetochromin A (18), sterigmatocystin (19), O-methylsterigmatocystin (20), 3β-hydroxyergosta-5, 7, 22-triene (21) and peroxy-ergosterol (22). Compounds 10 and 11 are new ones. 4. Structure, biosynthesis, biological activity, and mechanisms of polyether antibiotics were reviewed.

Streptomyces clavuligerus é uma importante espécie industrial que produz múltiplos compostos de interesse clínico, destacando-se o antibiótico cefamicina C (CefC) e o ácido clavulânico (AC), um potente inibidor de b-lactamases produzidas por bactérias resistentes a penicilinas e cefalosporinas. Sabe-se que situações limite como subculturas sucessivas, forma inadequada de armazenamento das células, variações bruscas de condições de cultura, podem causar deleções de mais de 25% dos genes de Streptomyces sp que, embora muitas vezes não comprometam a sobrevivência do micro-organismo, acarretam perdas na capacidade produtiva. Neste trabalho, investigou-se a estabilidade da linhagem selvagem S. clavuligerus ATCC 27064 quanto à produção de CefC variando-se condições do inóculo utilizado nos cultivos submersos de obtenção do antibiótico. As fermentações foram realizadas em frascos agitados (28ºC, 260 rpm) e as amostras coletadas em 48 e 72 horas do caldo fermentativo foram analisadas quanto ao pH, biomassa (massa seca a 105ºC por 24 h) e CefC foi determinada por bioensaio de difusão em ágar (bactéria-teste Escherichia coli ESS 2235). Foi selecionado o micro-organismo melhor produtor por meio de screening, e o que apresentou a melhor produção de CefC e os menores desvios com relação à média foi utilizado para preparar novos lotes de células: ampolas de micélios liofilizados e criotubos contendo 108 esporos.mL-1 . Observou-se que a capacidade produtiva foi afetada quando foram utilizadas subculturas do micro-organismo original. A realização do screening mostrou-se eficiente na seleção de colônias superiores em termos de produção volumétrica, sendo recomendada a realização de tal procedimento periodicamente para manutenção de uma linhagem com maior potencial produtivo...
