O presente trabalho teve por objetivo quantificar, comparativamente, a área de preenchimento de dois materiais obturadores sólidos, cones de guta-percha (GP) e cones de Resilon (R), no terço apical de incisivos inferiores humanos, ex vivo, obturados pela técnica da onda contínua de condensação. Os espécimes foram submetidos a um protocolo, desde a cirurgia de acesso até o final do preparo químico-mecânico e divididos aleatoriamente em dois grupos, de 21 dentes cada, de acordo com o material utilizado. Não foi utilizado cimento endodôntico em nenhuma das amostras. Após a obturação, as amostras foram seccionadas transversalmente em dois níveis, a 3 e a 5mm do ápice, e subdivididas em grupos de acordo com a altura de corte e do material obturador, sendo estabelecido: GP3 (guta-percha com corte a 3mm), GP5 (guta-percha com corte a 5mm), R3 (Resilon com corte a 3mm) e R5 (Resilon com corte a 5mm). Posteriormente, as amostras foram submetidas a um processo de lixamento e polimento e examinadas em microscópio óptico por reflexão com aumento de 50x a 100x. Para a análise e processamento digital das imagens, foi utilizado o sistema de imagens Axio Vision 4.6 para Windows, sendo obtidas as medidas para cada área observada em micrômetros (μm); uma da área da cavidade, e outra da área de material obturador. Foi aferido o grau de circularidade de cada amostra, por uma fórmula matemática utilizada automaticamente pelo programa, onde 1 (um) é considerado o círculo perfeito e, quanto mais achatado o canal, mais tendente a 0 (zero) nesta escala. Obteve-se a área do canal, a circularidade de 0 a 1, a área preenchida pelo material obturador e, a porcentagem da área de preenchimento do material obturador em relação à área do canal. Foi realizado o cruzamento dos grupos dois a dois pelo teste t de Student, sendo verificada diferença estatisticamente significante entre os grupos GP3 e R3, tendo o grupo R3 apresentado maior porcentagem de área do canal radicular preenchida pelo material obturador em suas amostras (p<0,05). Na relação da circularidade com a quantidade de preenchimento, com o teste de Correlação de Pearson, não foi observada forte correlação entre a forma final do canal (relação de circularidade) e a quantidade de preenchimento do canal radicular pelos materiais obturadores testados. Conclui-se que houve grande variação de preenchimento mínimo e máximo em todos os grupos testados e o Resilon apresentou maior porcentagem de preenchimento de área do canal radicular em suas amostras.

O objetivo desse trabalho foi avaliar a biocompatibilidade de um material a base de guta-percha autoadesiva (Bio-Gutta) em tecido subcutâneo de camundongos. A guta-percha convencional foi utilizada como referência para comparação. Para isso, foram utilizados 30 camundongos, os quais foram distribuídos aleatoriamente em dois grupos (Bio-Gutta e guta-percha convencional). Os materiais avaliados foram implantados na região dorsal dos camundongos. Após 7, 21 e 63 dias, os animais foram sacrificados e 5 amostras por grupo por tempo experimental foram obtidas. As amostras histológicas foram seccionadas em 5 m de espessura e coradas convencionalmente com hematoxilina e eosina. Um escore de I-IV foi utilizado para graduar a reação inflamatória. O teste U de Mann-Whitney com correção de Bonferroni foi utilizado para análise estatística (p < 0,05). Os resultados mostraram que, após 7 dias, a Bio-Gutta induziu uma reação tecidual de leve a moderada (II) e a guta-percha convencional apresentou uma resposta inflamatória de moderada a intensa (III). Após 21 dias, a Bio-Gutta induziu uma leve reação tecidual (I), enquanto que a guta-percha convencional ainda apresentava uma resposta inflamatória moderada (III). Uma reação tecidual de leve a ausente (I) foi observada na Bio-Gutta após 63 dias, ao passo que na guta-percha convencional observou-se uma resposta inflamatória de leve a moderada (II). Houve diferença significativa na mediana do grau de inflamação entre os grupos em cada tempo experimental (p = 0,005, aos 7 dias; p = 0,011, aos 21 dias; e p = 0,003, aos 63 dias). Concluiu-se então que a Bio-Gutta foi mais biocompatível que a guta-percha convencional em tecido subcutâneo de camundongos apresentando uma boa resposta tecidual.

Single chain and pauci chain single crystals of gutta percha in nanometer size were prepared by a dilute solution spraying method. A new crystal modification of gutta percha was found. The unit cell of the new modification of gutta percha was determined by electron diffraction crystal structure analysis to be a hexagonal form with cell dimensions: a = b = 0.695 nm, c = 0.661 nm, alpha = beta = 90 degrees, gamma =120 degrees; the space group is P6. The molecular packing in the unit cell was determined by computer modelling with Cerius(2) 2.0 software. (C) 1998 Elsevier Science Ltd. All rights reserved.

Single chain single crystals (SCSC) of gutta percha (GP) were prepared by a dilute-solution spraying method. Electron diffraction (ED) patterns revealed that the single chain single crystal was of a new crystalline modification, the delta form. The images of SCSC of GP obtained with a high resolution electron microscope (HREM) showed a two dimensional periodic structure. Most of the images consisted of lattice fringes derived from the (001) zone. This is the first time that the single chain single crystal images of GP have been observed at a molecular level. Micrographs were image processed using optical filtering methods to improve the signal-to-noise ratio, and were compared with computer-generated simulations of the images. From the viewpoint of the defects seen in high resolution images, the crystal formation and melting processes are discussed. (C) 1998 Elsevier Science Ltd. All rights reserved.

Single-chain single crystals of gutta-percha have been observed by transmission electron microscopy of atomizer-sprayed particles deposited from a very dilute solution in chloroform onto a carbon film which had a filter paper wetted with ethanol in contact with its under side. Selected-area electron diffraction patterns of crystals having no definite crystal profiles showed that the crystals were of the low-melting crystalline form and that the chain segments in the single crystals were standing up from the substrate during crystallization. In cases of single crystals showing sharply defined crystal profiles, electron diffraction patterns showed that they were neither of the low-melting form nor the high-melting form. The structure of this new crystalline modification needs further studies.

The aim of this study was to compare the percentage of gutta-percha (PGP) in mesial root canals of mandibular molars obturated with LC (Lateral Compaction) or SC (Single Cone) ProTaper Universal System techniques at different levels of the root. Mesial root canals of 20 human permanent molars with similar anatomical characteristics were instrumented using the ProTaper Universal rotary system technique until the F2 instrument, with 20 canals filled by SC ProTaper Universal technique and 20 canals by the LC technique. The mesial roots were sectioned transversely to 3, 5, and 7 mm from the root apex. Digital images of specimens were obtained at MIC-D digital microscope in increases of 30 to 35X. The gutta-percha area was measured using ImageTool software. Data were analyzed using two-way ANOVA and Bonferroni test (a = 0.05). The SC technique provided greater PGP than the LC technique in the apical third (3 mm) (P < 0.001). In the other thirds (5 and 7 mm) there was no statistical difference between the two techniques regarding the PGP (P > 0.05). There was no statistically significant difference between thirds of the root canal for both techniques (P > 0.05). It was concluded that SC technique provided greater PGP than the LC technique in the apical third of mesial root canals of mandibular molars. International Endodontic Journal, 44, 1019-1023, 2011.Aim To analyse a method used to evaluate the thermomechanical properties of gutta-percha and Resilon at different temperatures and compression loads.Methodology Two hundred and seventy specimens measuring 10 mm in diameter and 1.5 mm in height were made from the following materials: conventional gutta-percha (GCO). thermoplastic gutta-percha (GTP) and Resilon (R) cones (RE). After 24 h, the specimens were placed in water at 50 degrees C. 60 degrees C or 70 degrees C for 60 s. After that, specimens were placed between two glass slabs, and loads weighing 1.0, 3.0 or 5.0 kg were applied. Images of the specimens were digitized before and after the test and analysed using imaging software to determine their initial and final areas. The thermomechanical property of each material was determined by the difference between the initial and final areas of the specimens. Data were subjected to ANOVA and SNK tests at 5% significance. To verify a possible correlation between the results of the materials, linear regression coefficients (r) were calculated.Results Data showed higher flow area values for RE under all compression loads at 70 degrees C and under the 5.0 kg load at 60 degrees C (P < 0.05). Regarding gutta-percha, GTP showed higher flow under loads weighing 3.0 and 5.0 kg. at 60 and 70 degrees C (P < 0.05). GCO presented higher flow at 70 degrees C with a load of 5.0 kg. Regression analyses showed a poor linear correlation amongst the results of the materials under the different experimental conditions.Conclusion Gutta-percha and Resilon (R) cones require different compression loads and temperatures for evaluation of their thermomechanical properties. For all materials, the greatest flow occurred at 70 degrees C under a load of 5.0 kg: therefore. these parameters may be adopted when evaluating endodontic tilling materials.

Introduction: The aim of this study was to evaluate the ability of Resilon (Resilon Research, LLC, North Branford, CT) and 2 types of gutta-percha to fill simulated lateral canals when using the Obtura II system (Model 823-700; Obtura Spartan, Fenton, MO). Methods: Forty-five human single-rooted teeth were selected and subjected to root canal preparation. After that, simulated lateral canals were made at 2, 5, and 8 mm from the working length (WL). The specimens were divided into 3 groups (n = 15) according to the filling material used: Obtura Flow 150 gutta-percha (Obtura flow), Odous Endo Flow gutta-percha (Odous; Odous de Deus Ind e Corn. Ltda Belo Horizonte, MG, Brazil), and Resilon pellets (Resilon). Root canals were filled using the Obtura II system with the tip inserted to 3 mm from the WL. No sealer was used for root canal obturation. Specimens were subjected to a tooth decalcification and clearing method, and filling of the lateral canals was analyzed by digital radiography and photographs. The measurement of lateral canal filling was done using Image Tool software (UTHSCSA Image Tool for Windows version 3.0, San Antonio, TX). Data were statistically analyzed with the Kruskal-Wallis test at 5% significance. Results: All materials showed an ability to penetrate into the simulated lateral canals, with a minimum percentage of 73% in all thirds of the root canal. Conclusions: It was concluded that gutta-percha and Resilon are solid core materials with a lateral canal filling ability when used with the Obtura II system. (J Endod 2012;38:676-679)

Aim To analyse the thermoplasticity of several endodontic filling materials using the Obtura II System at different temperature settings.Methodology The following materials based on gutta-percha: Regular Obtura (OBT), Obtura Flow 150 (OBT F), Endo Flow (EDF), Odous (ODO) and the synthetic thermoplastic polymer material Resilon (RE) were heated using the Obtura II System at three temperature settings (140, 170 and 200 degrees C). Samples of the heated materials were placed on the sensor of a digital thermometer (THR-140; Instrutherm, São Paulo, Brazil) to determine their real temperature (RT) when the system was set at 140 degrees C (from 64.5 to 69 degrees C), 170 degrees C (from 73.8 to 77.5 degrees C) and 200 degrees C (from 83.6 degrees C for EDF and 100 degrees C for RE). Specimens (n = 30) were made by placing samples of each material in metallic ring moulds and compressing them between two glass slabs. After 24 h, specimens (n = 10) were heated at the different settings (RT) and submitted to compression under a 5-kg load. Plasticization was assessed by calculating the differences between the post-compression and initial diameters of each specimen. Data were submitted to ANOVA and Tukey's test at 5% significance.Results At 140 degrees C, Obtura Flow presented the highest thermoplasticity values and Regular Obtura, the lowest. At 170 degrees C, Obtura Flow and Resilon demonstrated greater plasticization. Resilon had the highest mean thermoplasticity values at 200 degrees C.Conclusions Thermoplasticity values were influenced both by the temperature settings on the Obtura II System and by the type of material analysed. Obtura Flow and Resilon had the highest mean thermoplasticity values.

The aim of this study was to evaluate the effectiveness of 2% peracetic acid for the disinfection of gutta-percha cones contaminated in vitro with Escherichia coli, Staphylococcus aureus, Streptococcus mutans, Candida albicans and Bacillus subtilus (in spore form). Two hundred and twenty-five gutta-percha cones were contaminated with standardized suspensions of each microorganism and incubated at 37 degrees C for 24 h. The cones were divided into 10 experimental groups (n = 15), according to the microorganism tested and disinfection testing times. The disinfection procedure consisted of immersing each cone in a plastic tube containing the substance. The specimens remained in contact with the substance for 1 or 2.5 minutes. Afterwards, each cone was transferred to a 10% sodium thiosulphate solution (Na2S2O3) to neutralize the disinfectant. Microbial biofilms adhering to the cones were dispersed by agitation. Aliquots of 0.1 ml of the suspensions obtained were plated on Sabouraud dextrose agar, or brain and heart infusion agar, and incubated at 37 degrees C for 24 h. The results were expressed in colony forming units (CFU/ml) and the data were submitted to the Wilcoxon Signed Rank Test (level of significance at 0.05). A significant reduction was observed, after 1 minute of exposure, in the test solution for C. albicans (p = 0.0190), S. aureus (p = 0.0001), S. mutans (p = 0.0001), B. subtilis (p = 0.0001), and E. coli (p = 0.0001). After 2.5 minutes of exposure, 100% of the microbial inocula were eliminated. It was concluded that the 2% peracetic acid solution was effective against the biofilms of the tested microorganisms on gutta-percha cones at 1 minute of exposure.

Foi propósito deste trabalho observar se o uso de cones de guta-percha contendo Ca(OH)2, promove melhora no selamento marginal apical e, também, se apenas o cone principal contendo essa droga produz esse efeito. Assim, dentes humanos extraídos foram preparados biomecânicamente e obturados pela técnica da condensação lateral com OZE e cones de guta-percha contendo ou não Ca(OH)2. Após imersão dos espécimes em azul de metileno a 2%, em ambiente com vácuo, observou-se diferença estatisticamente significante entre os espécimes obturados com cones contendo Ca(OH)2, comparativamente aos casos obturados com cones de guta percha comuns (p=0.01). Os resultados obtidos permitiram concluir que esses cones tornam as obturações mais herméticas e que esse efeito também pode ser obtido com o emprego do cone principal da mesma fórmula, aliado a cones acessórios comuns (p=0.05).
