A sequential system for fractionation by ultrafiltration (SSFU) equipped with advanced membranes filters (molecular size cut-off: 5, 10, 30, 50 and 100 kDalton) of the polyethersulfone type was developed for analytical fractionation of humic substances (HS) extracted from aquatic systems or soils. The device consists of five membrane filters (Sartocon® Micro) operated by a multi-channel peristaltic pump, enabling an easy handling, working in a closed system and with simple collection of the six obtained fractions (F1>100; F2: 100-50; F3: 50-30; F4:30-10; F5: 10-5 and F6 <5 kDalton). Then, the HS sample (250 mL solution 1.0 mg/mL, pH 5.0 to 6.0) to be fractionated is pumped by pump through the series of membrane filters with a tangential flow of 85 mL/min, initial pressure 0.2 to 0.3 bar and permeation flux through the membranes of 0.8 to 1.4 mL/min. The overall time for fractionation and cleaning of the device is about 10 h and 25 mL of each fraction is obtained.

A method for determination of lead and cadmium in aqueous samples using solvent microextraction and dithizone as complexing agent with FAAS was developed. Solvent microextraction parameters were optimized. The effect of foreign ions on the extraction yields was studied. The extraction was carried out until the aqueous to organic phase ratio achieved a 250 fold preconcentration of metals. For preconcentration times of 4 min the 3sigma detection limits, relative standard deviations (n=7) and linear calibration ranges were 1.6 mug L-1, 5.8% and 10.0 -- 80.0 mug L-1 for lead and 11.1 ng L-1, 5.9% and 0.3 -- 3.0 mug L-1 for cadmium, respectively. The solvent microextraction procedure presented here was applied to the determination of lead and cadmium in natural waters.

Multi-element analyses of sediment samples from the Santos-Cubatão Estuarine System were carried out to investigate the spatial and seasonal variability of trace-element concentrations. The study area contains a rich mangrove ecosystem that is a habitat for tens of thousands of resident and migratory birds, some of them endangered globally. Enrichments of metals in fine-grained surface sediments are, in decreasing order, Hg, Mn, La, Ca, Sr, Cd, Zn, Pb, Ba, Cu, Cr, Fe, Nb, Y, Ni and Ga, relative to pre-industrial background levels. The maximum enrichment ranged from 49 (Hg) to 3.1 (Ga). Mercury concentrations were greater in the Cubatão river than in other sites, while the other elements showed greater concentrations in the Morrão river. Concentrations of Mn were significantly greater in winter and autumn than in summer and spring. However, other elements (e.g. Cd and Pb) showed the opposite, with greater concentrations in summer and spring. This study suggests that seasonal changes in physical and chemical conditions may affect the degree of sediment enrichment and therefore make the assessment of contamination difficult. Consequently, these processes need to be considered when assessing water quality and the potential contamination of biota.

The present work deals with the design and construction of an equipment for muti-cell accelerated stability test measurements (AST) of dimensionally stable anodes (DSA). The equipment was built using only components that were available in the laboratory. Measurements of three electrochemical cells can be performed using the developed software. The acquisition time interval and the cut-off potencial can be set by the user. Experimental data for RuO2 electrodes obtained with the built equipment are in agreement with the literature.

Epoxidation of soybean oil was investigated using 1-n-butyl-3-methylimidazolium hexafluorophosphate [bmim][PF6] ionic liquid as biphasic medium with molybdenum(VI) acetylacetonate complex and tert-butyl hydroperoxide TBHP as oxidizing agent. Reaction conditions were molar ratio TBHP:number of double bonds of oil:catalyst of 100:100:1, reaction temperature of 60 ºC and reaction time between 2 and 24 h. The proposed system showed catalytic activity for epoxidation reactions under tested conditions. Reuse of ionic liquid/catalyst system for epoxidation reactions was also investigated. Evaluation of epoxidation observed in this catalytic system was done by quantitative ¹H NMR data.

A multi-commuted flow system was developed to determine propylthiouracil (PTU) based on the reaction of its thiol form with iminoquinone radical generated by the oxidation of N,N-dimethyl-p-phenylenediamine in an alkaline medium. PTU can be found in tautomeric forms and the tautomeric equilibrium was displaced to enhance the thiol form. However, the reaction product is unstable and its residence time in the flow path was carefully investigated. The proposed procedure showed good precision (< 1.5%) and a limit of detection (3s) of 0.11 mg L-1. High recoveries were obtained in the validation test. The procedure was employed for propylthiouracil determination in medications.

Nos últimos anos cresce a preocupação dos consumidores quanto à qualidade do leite e às condições de produção e bem-estar dos animais. Simultaneamente, aumenta o interesse e o consumo de produtos e subprodutos de origem animal produzidos no sistema orgânico, com destaque para o leite e derivados. O presente estudo investigou a presença de microrganismos patogênicos, a sensibilidade e a multi-resistência dos isolados aos antimicrobianos, a celularidade e a presença de resíduos de drogas no leite de vacas, com e sem mastite, produzido no sistema orgânico. Foram amostradas 148 vacas no período médio de lactação, das quais duas com mastite clínica, 72 com mastite subclínica e 74 sem mastite (controles), provenientes de quatro pequenas propriedades do interior do Estado de São Paulo, certificadas como orgânicas. Staphylococcusaureus (25,7%), Streptococcus spp. (21,4%), Corynebacterium bovis (12,9%), Streptococcus agalactiae (4,3%) e Staphylococcus spp. (4,3%) foram os microrganismos mais frequentemente isolados nos animais com mastite. Aspergillus spp. foi isolado de um animal. Ceftiofur (95,2%), oxacilina (84,2%), gentamicina (76,3%) e cefoperazona (70,3%) foram os antimicrobianos mais efetivos frente aos isolados. As maiores taxas de resistência das linhagens foram constatadas para penicilina (53,5%), ampicilina (41,6%) e neomicina (38,6%). Multi-resistência a três ou mais fármacos foi encontrada em 40 (39,6%) linhagens. A média da contagem de células somáticas das propriedades para animais com mastite foi 175.742,67 céls/mL, enquanto que para os animais controles foi 58.227,6 céls/mL. A presença de resíduos de antimicrobianos foi observada no leite de quatro (2,7%) animais. Os resultados revelaram baixa celularidade média nos animais, indicativo de boa qualidade do leite. Entretanto, apontam para a necessidade da adoção de medidas de controle para microrganismos contagiosos e maior rigor na proibição do uso de antimicrobianos em fazendas de leite orgânico.
