Two regulons, soxRS and marRAB, are associated with resistance to quinolones or multiple antibiotic in Salmonella enterica serovar Typhimurium. These regulons are activated by nitric oxide and redox-cycling drugs, such as paraquat and cause on activation of the acrAB-encoded efflux pump. In this study, we investigated the effect of nitric oxide (NO) alone and in combination with ofloxacin, ciprofloxacin, and pefloxacin against S. typhimurium clinical isolates and mutant strains in vitro. We did not observe synergistic effect against clinical isolates and SH5014 (parent strain of acr mutant), while we found synergistic effect against PP120 (soxRS mutant) and SH7616 (an acr mutant) S. typhimurium for all quinolones. Our results suggest that the efficiencies of some antibiotics, including ofloxacin, ciprofloxacin, and pefloxacin are decreased via activation of soxRS and marRAB regulons by NO in S. enterica serovar Typhimurium. Further studies are warranted to establish the interaction of NO with the genes of Salmonella and, with multiple antibiotic resistance.

The oxidation process of sulfur (IV) species (SO2, HSO3- e SO32-) by oxygen, catalysed by trace metal ion and complexes, can play an important role in atmospheric, analytical and bioinorganic chemistry. An overview of the most important reactions in these fields is presented. A fascinating redox cycling of the metal ions and complexes during such autoxidation process was revealed by the combination of kinetics and coordination chemistry studies.

Aerobic metabolism changes rapidly to glycolysis post-mortem resulting in a pH-decrease during the transformation of muscle in to meat affecting ligand binding and redox potential of the heme iron in myoglobin, the meat pigment. The "inorganic chemistry" of meat involves (i) redox-cycling between iron(II), iron(III), and iron(IV)/protein radicals; (ii) ligand exchange processes; and (iii) spin-equilibra with a change in coordination number for the heme iron. In addition to the function of myoglobin for oxygen storage, new physiological roles of myoglobin are currently being discovered, which notably find close parallels in the processes in fresh meat and nitrite-cured meat products. Myoglobin may be characterized as a bioreactor for small molecules like O2, NO, CO, CO2, H2O, and HNO with importance in bio-regulation and in protection against oxidative stress in vivo otherwise affecting lipids in membranes. Many of these processes may be recognised as colour changes in fresh meat and cured meat products under different atmospheric conditions, and could also be instructive for teaching purposes.

Apatone™, a combination of menadione (2-methyl-1,4-naphthoquinone, VK3) and ascorbic acid (vitamin C, VC) is a new strategy for cancer treatment. Part of its effect on tumor cells is related to the cellular pro-oxidative imbalance provoked by the generation of hydrogen peroxide (H2O2) through naphthoquinone redox cycling. In this study, we attempted to find new naphthoquinone derivatives that would increase the efficiency of H2O2 production, thereby potentially increasing its efficacy for cancer treatment. The presence of an electron-withdrawing group in the naphthoquinone moiety had a direct effect on the efficiency of H2O2 production. The compound 2-bromo-1,4-naphthoquinone (BrQ), in which the bromine atom substituted the methyl group in VK3, was approximately 10- and 19-fold more efficient than VK3 in terms of oxygen consumption and H2O2 production, respectively. The ratio [H2O2]produced / [naphthoquinone]consumed was 68 ± 11 and 5.8 ± 0.2 (µM/µM) for BrQ and VK3, respectively, indicating a higher efficacy of BrQ as a catalyst for the autoxidation of ascorbic acid. Both VK3 and BrQ reacted with glutathione (GSH), but BrQ was the more effective substrate. Part of GSH was incorporated into the naphthoquinone, producing a nucleophilic substitution product (Q-SG). The depletion of BrQ by GSH did not prevent its redox capacity since Q-SG was also able to catalyze the production of reactive oxygen species. VK3/VC has already been submitted to clinical trials for the treatment of prostate cancer and has demonstrated promising results. However, replacement of VK3 with BrQ will open new lines of investigation regarding this approach to cancer treatment.

Vários estudos destacam as espécies reativas de oxigênio e nitrogênio (ERONs) como importantes contribuintes na patogênese de numerosas doenças cardiovasculares, incluindo hipertensão, aterosclerose e falência cardíaca. Tais espécies são moléculas altamente bioativas e com vida curta derivadas, principalmente, da redução do oxigênio molecular. O complexo enzimático da NADPH oxidase é a maior fonte dessas espécies reativas na vasculatura. Sob condições fisiológicas, a formação e eliminação destas substâncias aparecem balanceadas na parede vascular. Durante o desbalanço redox, entretanto, há um aumento na atividade da NADPH oxidase e predomínio de agentes pró-oxidantes, superando a capacidade de defesa orgânica antioxidante. Além disso, tal hiperatividade enzimática reduz a biodisponibilidade do óxido nítrico, crucial para a vasodilatação e a manutenção da função vascular normal. Apesar de a NADPH oxidase relacionar-se diretamente à disfunção endotelial, foi primeiramente descrita por sua expressão em fagócitos, onde sua atividade determina a eficácia dos mecanismos de defesa orgânica contra patógenos. As sutis diferenças existentes entre as unidades estruturais das NADPH oxidases, a depender do tipo celular que as expressa, podem ter implicações terapêuticas, permitindo a inibição seletiva do desequilíbrio redox induzido pela NADPH oxidase, sem comprometer, entretanto, sua participação nas vias fisiológicas de sinalização celular que garantem a proteção contra microorganismos.

FUNDAMENTO: Estudos recentes descrevem a participação de espécies reativas de oxigênio e nitrogênio na hipertensão. OBJETIVO: Identificar o desbalanço redox em sangue de hipertensos. MÉTODOS: Superóxido dismutase (SOD), catalase (CAT), glutationa peroxidase (GPx), glutationa (GSH), vitamina C, transferrina, ceruloplasmina, malondialdeído (MDA) e o grupo carbonila, foram quantificados no sangue de 20 hipertensos e 21 controles. Os indivíduos tinham um Índice de Massa Corporal de > 18,5 e < 30 kg/m², glicemia < 100 mg/dL, colesterol sérico < 200 mg/dL, e eram mulheres não fumantes, não grávidas e não lactantes, não usuárias de alopurinol e probucol, e hipertensos em medicação anti-hipertensiva. Todos os indivíduos foram submetidos a um período preparatório de quatro semanas sem álcool, suplementos vitamínicos, dexametasona e paracetamol. RESULTADOS: Níveis reduzidos de CAT (p = 0,013), GSH ( p = 0,003) e MDA (p = 0,014), e altos níveis de GPx (p = 0,001) e ceruloplasmina (p = 0,015) foram obtidos no grupo de hipertensos, em comparação com os controles. Foi verificada uma correlação positiva entre a pressão sistólica e o MDA nos hipertensos e diastólica e CAT nos controles. CONCLUSÃO: Os dados obtidos são sugestivos de que os hipertensos apresentavam desequilíbrio em reações redox, a despeito do possível efeito atenuante de sua medicação anti-hipertensiva.

The Bacteroides fragilis ATCC strain was grown in a synthetic media with contrasting redox potential (Eh) levels [reduced (-60 mV) or oxidised (+100mV)] and their adhesion capacity to extracellular matrix components was evaluated. The strain was capable of adhering to laminin, fibronectin, fibronectin + heparan sulphate and heparan sulphate. A stronger adherence to laminin after growing the strain under oxidising conditions was verified. Electron microscopy using ruthenium red showed a heterogeneous population under this condition. Dot-blotting analyses confirmed stronger laminin recognition by outer membrane proteins of cells cultured at a higher Eh. Using a laminin affinity column, several putative laminin binding proteins obtained from the cultures kept under oxidising (60 kDa, 36 kDa, 25 kDa and 15 kDa) and reducing (60 kDa) conditions could be detected. Our results show that the expression of B. fragilis surface components that recognise laminin are influenced by Eh variations.

The precise sampling of soil, biological or micro climatic attributes in tropical forests, which are characterized by a high diversity of species and complex spatial variability, is a difficult task. We found few basic studies to guide sampling procedures. The objective of this study was to define a sampling strategy and data analysis for some parameters frequently used in nutrient cycling studies, i. e., litter amount, total nutrient amounts in litter and its composition (Ca, Mg, Κ, Ν and P), and soil attributes at three depths (organic matter, Ρ content, cation exchange capacity and base saturation). A natural remnant forest in the West of São Paulo State (Brazil) was selected as study area and samples were collected in July, 1989. The total amount of litter and its total nutrient amounts had a high spatial independent variance. Conversely, the variance of litter composition was lower and the spatial dependency was peculiar to each nutrient. The sampling strategy for the estimation of litter amounts and the amount of nutrient in litter should be different than the sampling strategy for nutrient composition. For the estimation of litter amounts and the amount of nutrients in litter (related to quantity) a large number of randomly distributed determinations are needed. Otherwise, for the estimation of litter nutrient composition (related to quality) a smaller amount of spatially located samples should be analyzed. The determination of sampling for soil attributes differed according to the depth. Overall, surface samples (0-5 cm) showed high short distance spatial dependent variance, whereas, subsurface samples exhibited spatial dependency in longer distances. Short transects with sampling interval of 5-10 m are recommended for surface sampling. Subsurface samples must also be spatially located, but with transects or grids with longer distances between sampling points over the entire area. Composite soil samples would not provide a complete understanding of the relation between soil properties and surface dynamic processes or landscape aspects. Precise distribution of Ρ was difficult to estimate.

The objective of this paper is to study selected components of the nutrient cycle of pure and mixed stands of native forest species of Atlantic Forest in southeastern Brazil. Tree diameter, height, above-ground biomass, and nutrient content were determined in 22-year-old stands. Litterfall, litter decomposition, and nutrient concentration were evaluated from August 1994 to July 1995. The following species were studied: Peltogyne angustiflora, Centrolobium robustum, Arapatiella psilophylla, Sclerolobium chrysophyllum, Cordia trichotoma, Macrolobium latifolium. The litter of a natural forest and a 40-year-old naturally regenerated second-growth forest was sampled as well. The mixed-species outmatched pure stands in height, stem volume and total biomass (29.4 % more). The greatest amount of forest litter was observed in the natural forest (9.3 Mg ha-1), followed by the mixed-species stand (7.6 Mg ha-1) and secondary forest (7.3 Mg ha-1), and least litterfall was measured in the pure C. robustum stand (5.5 Mg ha-1). Litterfall seasonality varied among species in pure stands (CV from 44.7 to 91.4 %), unlike litterfall in the mixed-tree stand, where the variation was lower (CV 31.2 %). In the natural and second-growth forest, litterfall varied by 57.8 and 34.0 %, respectively. The annual rate of nutrient return via litterfall varied widely among forest ecosystems. Differences were detected between forest ecosystems in both the litter accumulation and quantity of litterlayer nutrients. The highest mean nutrient accumulation in above-ground biomass was observed in mixed-species stands. The total nutrient accumulation (N + P + K+ Ca + Mg) ranged from 0.97 to 1.93 kg tree-1 in pure stands, and from 1.21 to 2.63 kg tree-1 in mixed-species stands. Soil fertility under mixed-species stands (0-10 cm) was intermediate between the primary forest and pure-stand systems. The litterfall rate of native forest species in a mixed-species system is more constant, resulting in a more continuous decomposition rate. Consequently, both nutrient availability and quantity of organic matter in the soil are higher and the production system ecologically more sustainable.

Dependendo das condições do sistema, os ácidos húmicos podem atuar como oxidantes ou redutores. Nos sistemas naturais, o fluxo de elétrons está diretamente associado à quantidade e à qualidade do húmus. O potencial do eletrodo e a capacidade de oxidação informam sobre os fatores intensidade e capacidade dos sistemas redox, respectivamente. Estudos prévios têm aventado a hipótese de que radicais livres nos ácidos húmicos participam dessas reações redox. No presente estudo, seis ácidos húmicos isolados de adubos orgânicos foram titulados com um oxidante (I2) em atmosfera inerte e condições especificadas. Os ácidos húmicos apresentaram valores do potencial formal-padrão do eletrodo semelhantes: entre 0,773 e 0,794 V a 25 ºC. A capacidade de oxidação dos ácidos húmicos variou de 3,88 a 4,39 mol c kg-1 a pH 5,0 e de 5,35 a 7,89 mol c kg-1 a pH 7,0. Foi observada correlação positiva e significativa entre a capacidade de oxidação dos ácidos húmicos e as suas concentrações de grupos funcionais fenólicos, quinonas e semiquinonas.

A matéria orgânica do solo é o maior reservatório de C nos sistemas naturais. Em tais sistemas a qualidade e a estabilidade do C podem ser estimadas pelo aumento da concentração das frações humificadas que, dentre outros fatores, está condicionada ao balanço entre as perdas e os ganhos que envolvem as reações de oxidação e de redução da matéria orgânica do solo. O objetivo deste estudo foi avaliar a eletroquímica, usando titulações redox iodimétricas, de ácidos húmicos isolados de solos cultivados continuamente com cana-de-açúcar submetida ou não à queima da palha para a colheita ou à adição anual de vinhaça. Os ácidos húmicos apresentaram valores do potencial formal padrão do eletrodo entre 0,760 e 0,779 V a 25 ºC. A capacidade de oxidação dos ácidos húmicos variou de 1,01 a 3,44 mol c kg-1 a pH 5,0 e de 1,64 a 6,44 molc kg-1 a pH 7,0. Observou-se correlação positiva e significativa entre a capacidade de oxidação dos ácidos húmicos e suas concentrações de grupos funcionais fenólicos, quinonas e semiquinonas.

As an alternative to the relatively complex and expensive spectroscopic methods, the redox properties of humic acids, determined by potentiometric titrations, have been used to evaluate the stability of soil organic C. The objective of the present study was to establish a Redox Index of C Stability (RICS) and to correlate it with some properties of the humic acids extracted from different modal soils in Brazil (distinct weathering stages or management) to facilitate system comparison. The RICS was efficient for soil comparison and variations were comparable to those of the chemical and spectroscopic methods used for humic acid characterization. The values of soil pH, point of zero salt effect, sum of bases, exchangeable Ca content, weathering index, as well as the humic acid O/C ratio, quinone and semiquinone free radical contents, aromatic C and fluorescence intensity were closely related with the RICS. The RICS was higher in less weathered soils, with more active clays and higher fertility. The RICS values of soils under long-term sugarcane management were ranked in decreasing order: unburned, burned with vinasse, burned without vinasse.

A análise conjunta do potencial redox com outras variáveis pode ser útil para o entendimento da dinâmica dos solos hidromórficos, ao longo de todo o ciclo hidrológico a que o Pantanal está submetido. Neste trabalho, foram estudados o potencial redox e algumas variáveis do solo (CO2, O2, temperatura, umidade e potencial matricial) pelo monitoramento contínuo e ininterrupto. Para isso, foram realizadas determinações nas profundidades de 10 e 30 cm num Planossolo Háplico alítico gleissólico, na fitofisionomia Cerrado sensu stricto. O trabalho foi realizado de fevereiro de 2010 a julho de 2011 na RPPN (Reserva Particular do Patrimônio Natural/Sistema Nacional de Unidades de Conservação - SNUC), Estância Ecológica SESC (Serviço Social do Comércio) Pantanal, município de Barão de Melgaço, nordeste do Pantanal de Mato Grosso. Os valores do potencial redox variaram de 636 mV (enchente) a -341 mV (cheia), caracterizando ambiente anaeróbico na estação da cheia e ambiente oxidado nas demais estações do ciclo hidrológico, o que indica que o potencial redox é ferramenta versátil para compreender as reações de oxirredução nos solos hidromórficos do Pantanal mato-grossense, pois os valores diminuíram quando o teor de umidade era elevado. Além disto, o estudo evidenciou também que o monitoramento contínuo de variáveis complementares pode ser importante por permitir análise mais aprofundada das condições de hipersazonalidade, em que esses solos estão submetidos. Neste contexto, o potencial matricial e a umidade foram as variáveis de maior importância para explicar a variação do conjunto de dados obtidos ao longo do ciclo hidrológico, o que indica que esses parâmetros físicos são determinantes nos processos biológicos desse solo tropical com hipersazonalidade hídrica.

In a no-tillage system, cover crops must be used that combine shoot dry matter production and nutrient recycling. The aim of this study was to evaluate shoot dry matter production, decomposition rate and macronutrient and silicon release from pigeonpea and pearl millet in monoculture and intercropping systems. A randomized block design was used with a 3 x 6 factorial arrangement, with four replications. The first factor consisted of three cover crops (pigeonpea, pearl millet and intercropping of these cover crops) and the second consisted of six sampling times [0, 18, 32, 46, 74 and 91 days after desiccation (DAD)]. Pearl millet produced greater amounts of shoot dry matter and content of N, P, K, Ca, Mg, S, C and Si and had a higher decomposition rate and macronutrient and Si release than the other cover crops. The rates of decomposition and daily nutrient release from shoot dry matter were highest in the first period of evaluation (0-18 DAD). Over time, the C/N, C/P and C/S ratios increased, while C/Si and the decomposition rate decreased. Potassium was the nutrient most quickly released to the soil, especially from pearl millet residue. Silicon had the lowest release rate, with 62, 82 and 74 % of the total content in the shoot dry matter remaining in the last evaluation of pearl millet, pigeonpea and in the intercrop system, respectively. The shoot dry matter from the intercrop system had a different decomposition rate than that from the pearl millet monoculture and pigeonpea. Plants with greater shoot dry matter production and lower C/Si ratio are more effective in a no-tillage system for providing a more complete and persistent soil cover.

Abstract:The objective of this work was to evaluate the effect of grazing intensity on the decomposition of cover crop pasture, dung, and soybean residues, as well as the C and N release rates from these residues in a long-term integrated soybean-beef cattle system under no-tillage. The experiment was initiated in 2001, with soybean cultivated in summer and black oat + Italian ryegrass in winter. The treatments consisted of four sward heights (10, 20, 30, and 40 cm), plus an ungrazed area, as the control. In 2009-2011, residues from pasture, dung, and soybean stems and leaves were placed in nylon-mesh litter bags and allowed to decompose for up to 258 days. With increasing grazing intensity, residual dry matter of the pasture decreased and that of dung increased. Pasture and dung lignin concentrations and C release rates were lower with moderate grazing intensity. C and N release rates from soybean residues are not affected by grazing intensity. The moderate grazing intensity produces higher quality residues, both for pasture and dung. Total C and N release is influenced by the greater residual dry matter produced when pastures were either lightly grazed or ungrazed.
