FUNDAMENTO: Tem sido demonstrado que diminuições na variabilidade da frequência cardíaca (VFC) estão relacionadas com o prognóstico na insuficiência cardíaca (IC). A administração crônica de trimetazidina, além da terapia convencional, tem melhorado a classe funcional e a função ventricular esquerda de pacientes com IC. OBJETIVO: Avaliar os efeitos da trimetazidina na VFC em pacientes com IC de origem isquêmica, recebendo tratamento otimizado. MÉTODOS: Trimetazidina 20 mg 3 vezes/dia foi adicionada à terapia de 30 pacientes com IC tratados com inibidores da enzima conversora de angiotensinogênio ou bloqueadores do receptor da angiotensina, carvedilol, espironolactona, digital e furosemida. A etiologia da IC era doença arterial coronariana em todos os pacientes. Os pacientes foram avaliados através de ecocardiografia e análise da VFC de 24-horas antes e 3 meses depois da adição de trimetazidina. RESULTADOS: A fração de ejeção ventricular esquerda (FEVE) média aumentou significantemente após a adição da trimetazidina (33,5±5,1% para 42,5±5,8%, p<0,001). Dos parâmetros da VFC, o DPNN (97,3±40,1 to 110,5±29,2 msegs, p=0,049) e DPTNN (80,5±29,0 to 98,3±30,5 msegs) aumentaram significantemente após o tratamento com trimetazidina. O DPNN basal apresentou uma correlação significante com o FEVE basal (r=0,445, p=0,023, p=0,008) e o incremento no DPNN correlacionou-se com o aumento na FEVE (r=0,518, p=0,007). CONCLUSÕES: A trimetazidina, quando adicionada ao tratamento médico otimizado de pacientes com IC de origem isquêmica, pode melhorar a VFC, além de melhorar a fração de ejeção ventricular esquerda.

O objetivo deste trabalho foi avaliar a conservação pós-colheita de manga 'Tommy Atkins' orgânica destinada à exportação, colhida com Boas Práticas Agrícolas (CBP) e sem Boas Práticas Agrícolas (SBP), sob recobrimentos bio-orgânicos. Manga colhida na maturidade comercial foi recoberta com extrato de erva-doce a 1,5% (CBP+ED, SBP+ED), com extrato de erva-doce adicionada à fécula de mandioca a 3,0% (CBP+ED+F, SBP+ED+F) e Controle (sem recobrimento, CBP e SBP), armazenada a 10±0,5 ºC e 85±2% UR. O uso de BPA manteve a qualidade. O recobrimento com ED comprometeu a aparência da manga pela incidência de manchas na casca; recobrimento associando F + ED manteve a qualidade e retardou o amadurecimento.

The cytotoxic activity of amino (3a-e), aza-1-antraquinone (4a-e) lapachol derivatives against Ehrlich carcinoma and human K562 leukemia cells was investigated. Cell viability was determined using MTT assay, after 48 (Ehrlich) or 96 h (K562) of culture, and vincristine (for K562 leukemia) and quercetin (for Ehrlich carcinoma) were used as positive controls. The results showed dose-dependent growth-inhibiting activities and that the amino derivatives were active against the assayed cells, whereas the 4a-e derivatives were not. The allylamine derivative 3a was the most active against Ehrlich carcinoma, with IC50 = 16.94 ± 1.25 µM, and against K562 leukemia, with IC50 = 14.11 ± 1.39 µM. The analogous lawsone derivative, 5a, was also active against Ehrlich carcinoma (IC50 = 23.89 ± 2.3 µM), although the 5d and 5e derivatives showed lower activity. The interaction between 3a-d and calf thymus DNA was investigated by fluorimetric titration and the results showed a hyperchromic effect indicating binding to DNA as presented of ethidium bromide, used as positive control. The inhibitory action on DNA-topoisomerase II-a was also evaluated by a relaxation assay of supercoiled DNA plasmid, and the etoposide (200 µM) was used as positive control. Significant inhibitory activities were observed for 3a-d at 200 µM and a partial inhibitory action was observed for lapachol and methoxylapachol.

The merozoite surface protein-1 (MSP-1) locus of Plasmodium falciparum codes for a major asexual blood-stage antigen currently proposed as a major malaria vaccine candidate. The protein, however, shows extensive polymorphism, which may compromise its use in sub-unit vaccines. Here we compare the patterns of allelic diversity at the MSP-1 locus in wild isolates from three epidemiologically distinct malaria-endemic areas: the hypoendemic southwestern Brazilian Amazon (n = 54), the mesoendemic southern Vietnam (n = 238) and the holoendemic northern Tanzania (n = 79). Fragments of the variable blocks 2, 4a, 4b and 6 or 10 of this single-copy gene were amplified by the polymerase chain reaction, and 24 MSP-1 gene types were defined as unique combinations of allelic types in each variable block. Ten different MSP-1 types were identified in Brazil, 23 in Vietnam and 13 in Tanzania. The proportion of genetically mixed infections (isolates with parasites carrying more than one MSP-1 version) ranged from 39% in Brazil to 44% in Vietnam and 60% in Tanzania. The vast majority (90%) of the typed parasite populations from Brazil and Tanzania belonged to the same seven most frequent MSP-1 gene types. In contrast, these seven gene types corresponded to only 61% of the typed parasite populations from Vietnam. Non-random associations were found between allelic types in blocks 4a and 6 among Vietnamese isolates, the same pattern being observed in independent studies performed in 1994, 1995 and 1996. These results suggest that MSP-1 is under selective pressure in the local parasite population. Nevertheless, the finding that similar MSP-1 type frequencies were found in 1994 and 1996 argues against the prominence of short-term frequency-dependent immune selection of MSP-1 polymorphisms. Non-random associations between MSP-1 allelic types, however, were not detected among isolates from Brazil and Tanzania. A preliminary analysis of the distribution of MSP-1 gene types per host among isolates from Tanzania, but not among those from Brazil and Vietnam, shows significant deviation from that expected under the null hypothesis of independent distribution of parasites carrying different gene types in the human hosts. Some epidemiological consequences of these findings are discussed

Especial conditions were developed for the amplification of five DNA segments from US region of BHV-1 by polymerase chain reaction. In order to eliminate most nonspecific products it was found that addition of three cosolvents DMSO, glycerol and NP 40 was a simple method for increasing the specificity of amplification.

The composite membranes prepared via incorporation of 12.5% of molecular sieves 3A, 4A, 5A and 13X into chitosan/poly(vinyl alcohol) (1:1). The composite membranes were immersed in the cross-linker sulfuric acid in order to acquire high proton conductivity for applications in electrolytes in PEMCF and DMF. The influence of the molecular sieves on the structural, optical, thermal, mechanical, morphologic and protonic conductivity properties and water/methanol swelling degree of membranes were investigated.
