Propolis is mostly used as hydroalcoholic extract. Recently there has been a growing number of patents dealing with new solvents for preparing propolis extracts. This study aimed to prepare edible oil propolis extracts and compare their chemical composition and biological activity with ethanolic propolis extracts. ESI-MS and spectrophotometric methods were used for qualitative and quantitative analyses, respectively. Antibacterial activity was evaluated by diffusion in agar. Cytotoxicity was tested by MTT assay using tumor cell lines. The oil is able to extract bioactive compounds from propolis. Further studies are needed to improve extraction efficiency and to characterize the active components.

The aim of this work was to evaluate antioxidant activity of lemon seeds added to soybean oil, submitted to accelerated incubator-storage test and to determine its synergistic effect with the synthetic antioxidant TBHQ. The treatments Control, TBHQ (50 mg/kg), LSE (2,400 mg/kg Lemon Seed Extract), Mixture 1 (LSE + 50 mg/kg TBHQ) and Mixture 2 (LSE + 25 mg/kg TBHQ) were prepared and subjected to the accelerated incubator-storage test at 60 ºC for 12 days; samples were taken every 3 days and analyzed regarding peroxide value and conjugated dienes. The results showed that antioxidant activity of the tested treatments were: TBHQ = Mixture 1 = Mixture 2 > LSE > Control.

Three phloroglucinols were obtained from Hypericum brasiliense: japonicine A (1), isouliginosin B (2) and uliginosin B (3). Bioautography and disk diffusion methods were used to determine antibacterial activity of the hexanic extract. Strains of the Coagulase Negative Staphylococcus and American Methicillin Resistant Staphylococcus aureus clones showed a growth inhibition zone ranging from 10 to 12 mm and 7 to 15 mm, respectively. Minimal inhibitory concentration (MIC) values were used to measure antistaphylococcal activity for all phloroglucinols. Isouliginosin B and uliginosin B presented MIC values of 1.5 and 3.0 µg/mL, respectively, while japonicine A displayed MIC value of 50.0 µg/mL.

This work presents a detailed study about the sorption of crystal violet (CV) cationic dye onto polyether type polyurethane foam (PUF). The sorption process was based on the formation of an ionic-pair between cationic dye and dodecylsulfate anion (SDS), which presented high affinity by PUF. Set-up employed in the study was built up by adjusting a 200 mg cylinder of PUF to the arm of an overhead stirrer. The system was characterized in relation to equilibrium and kinetic aspects and it was modeled by employing Langmuir and Freundlich isotherms. Obtained results showed that the ratio between SDS and MB concentrations played an important role on the sorption process. According to results found it was possible to retain up to 3.4 mg of dye from 200 mL of a 5.0 x 10-5 mol L-1 CV solution containing 1.25 x 10-4 mol L-1 SDS, which represented a removal efficiency of around 92%.

The presence of non-aqueous phase liquids (NAPLs) in the subsurface is a threat to public health as well as a serious environmental issue. NAPLs may remain adsorbed or form lenses floating on aquifers causing long-term contaminations. Surfactants may increase NAPLs solubility, enhancing the pump-and-treatment performance. Size, shape, hydration and ionization degree of the micelles define the affinity and the space available for the solubilization of a particular contaminating agent. The tests carried out at laboratory scale, taking into account the NAPL to be removed and the medium characteristics were useful to select surfactants and evaluate their efficiency as NAPLs solubilizers.

The Neem tree, Azadirachta indica, provides many useful compounds that are used as pesticides. However, the efficiency in field of products like neem oil can be committed because they have not been observed reproductive content of secondary metabolic like azadirachtin. Based on reverse-phase high-performance liquid chromatography (HPLC) a new method was developed to permit the rapid quantitative analysis of azadirachtin from seeds, extracts and oil of Neem. In the present study it was evaluated the azadirachtin quantitative variation among various Neem's extracts and seeds showing the importance of quality control for reproduction of the insecticide efficiency, using S. frugiperda as target insect.

This work describes the isolation of an active flavonoid fraction and identification of isorhamnetin 3-O-β-D-(6''-acetyl)-galactopyranoside from flowers of B. perennis, and also the evaluation of anticholinesterase (AChE) activity of ethanolic extract from flowers (EEF) and the active fraction. The chemical structure of the flavonoid was defined on the basis of spectroscopic ¹H NMR, IR and UV data. EEF or flavonoid reduces AChE activity in vivo, while flavonoid also reduces AChE activity in vitro, showing a value of 1.49 mM for 50% inhibitory concentration (IC50), suggesting potential use as an insecticide or in the treatment of neurodegenerative diseases such as Alzheimer's disease.

This paper describes the chemical composition and acetylcholinesterase inhibition of an active chromatographic fraction of the EtOAc extract of Citrus limon leaves. The composition of the active fraction presented a mixture of two coumarins, 5,8-dimethoxy-psoralen and 5,7- dimethoxycoumarin, identified by ¹H and ¹³C NMR data analysis, including DEPT, COSY, HMBC and HSQC experiments. It was also demonstrated that this mixture presents qualitative and quantitative AChE inhibition. In vitro studies indicated a CE50 value of 340 µg/mL with 95% of confidence. In vivo studies (10 and 25 mg/kg) revealed inhibition of 30.09 and 30.06% of AChE activity in relation to neostigmine, respectively.

Phenylpropanoid glycoside verbascoside was isolated and identified from the ethyl acetate fraction of the aerial parts of Buddleja stachyoides Cham. & Schltdl. by 1H-NMR. A method using high-performance liquid chromatography has been developed and validated for determination of verbascoside in alcoholic crude extract of the aerial parts of B. stachyoides. Analysis was performed on a Phenomenex® Gemini-NX C18 analytical column (250 mm × 4.6 mm; 5 µm) using a mobile phase (pump A - aqueous solution containing H2SO4 (0.01 M), H3PO4 (0.4%), and (C2H5)2NH (0.4%); pump B - methanol:aqueous (95:5) solution containing H2SO4 (0.05 M), H3PO4 (2%), and (C2H5)2NH (0.2%); pump C - acetonitrile:aqueous (90:10) solution containing H2SO4 (0.05 M) and H3PO4 (2%)) and a diode array detector at 325 nm. The method was validated in accordance with ANVISA guidelines and may be applied to quality control of herbal medicine with aerial parts of B. stachyoides.

An enzymatic spectrophotometric method for the determination of methyldopa in a dissolution test of tablets was developed using peroxidase from radish (Raphanus sativus). The enzyme was extracted from radish roots using a phosphate buffer of pH 6.5 and partially purified through centrifugation. The supernatant was used as a source of peroxidase. The methyldopachrome resulting from the oxidation of methyldopa catalyzed by peroxidase was monitored at 480 nm. The enzymatic activity was stable for a period of at least 25 days when the extract was stored at 4 or -20 ºC. The method was validated according to RDC 899 and ICH guidelines. The calibration graph was linear in the range 200-800 µg mL-1, with a correlation coefficient of 0.9992. The limits of detection and quantification in the dissolution medium were 36 and 120 µg mL-1, respectively. Recovery was greater than 98.9%. This method can be applied for the determination of methyldopa in dissolution tests of tablets without interference from the excipients.

Inicialmente buscou-se identificar plantas com extratos metanólicos ativos in vitro contra o nematóide Panagrellus redivivus. Das 24 espécies vegetais estudadas, constatou-se que os melhores resultados foram obtidos com Leucaena leucocephala, que causou mortalidade de 93% dos indivíduos expostos ao seu extrato por 24 h. Em seguida, submeteu-se o referido extrato a fracionamento direcionado pelos testes in vitro com P. redivivus, o que possibilitou isolar uma substância nematicida que, segundo análises preliminares, correspondia a um alcalóide.

Uma formulação natural (VLAF) obtida da extração aquosa a frio de pó de tecido necrótico de lobeira (Solanum lycocarpum), infectado por Crinipellis perniciosa (Stahel) Singer, promoveu redução significativa no progresso da mancha foliar bacteriana (Xanthomonas campestris pv. vesicatoria), quando previamente pulverizado em folhas de tomateiro. Duas frações obtidas por precipitação salina, F0/30 e F30/60, apresentaram a maior parte das proteínas do extrato VLAF e foram submetidas à cromatografia de troca catiônica para separação das proteínas contidas nas frações. Os picos não retidos dessa cromatografia foram então submetidos à cromatografia de troca aniônica. Todos os picos, retidos e não retidos das duas cromatografias, foram amostrados e pulverizados sobre plantas de tomate cv. Santa Cruz Kada. Respostas diferenciais de atividade de peroxidases foram obtidas 14 horas após pulverizações. As amostras que induziram os maiores aumentos na atividade de peroxidases nas plantas foram o pico retido em CM-celulose da F0/30 (F0-30CMR) e o pico retido em DEAE-celulose da F30/60 (F30-60DEAER). Os resultados deste estudo indicaram a viabilidade da purificação e da caracterização de proteínas ou carboidratos eliciadores provenientes de VLAF.

Voltamogramas de redissolução catódica de cefaloglicina em meio ácido exibem uma única onda de redução em potenciais próximos da descarga do eletrólito (-1.0 V). Entretanto, o fármaco pode ser determinado indiretamente em meio alcalino (pH 10) através do complexo formado entre cefaloglicina e Hg(I) gerado na superfície do eletrodo sob potencial de acúmulo positivo (Eac= +0.20 V). Uma curva de calibração linear no intervalo de 0.122 a 1.22 mg/mL foi obtida para cefaloglicina em tampão B-R pH 10 utilizando-se tempo de acúmulo de 30 seg e potencial de acúmulo de +0.20 V. O método mostra alta reprodutibilidade e sensibilidade com desvio padrão relativo de 1,4 % para 5 determinações de 5x10-7 mol/L de cefaclor e limite de detecção de 5,17 ng/mL.

Este trabalho descreve a construção de um eletrodo de segunda espécie, Pt|Hg|Hg2Ox|Grafite, sensível ao íon oxalato (Ox). O eletrodo é construído facilmente, apresenta um rápido tempo de resposta, é de baixo custo, e tem um tempo de vida útil superior a 12 meses. O potencial molal padrão do eletrodo a 25ºC é 417,3 ± 1,0 mV. A validação do eletrodo foi conseguida pela determinação do produto de solubilidade,T K PS (I=0), do Hg2C2O4 (T K PS = (2,53 ± 0,21) x 10-13 mol². L-2). Uma característica favorável consiste no fato de que o eletrodo pode ser utilizado para determinar a concentração de íons oxalato livres em sistemas complexos de íons metálicos.
