Diante dos riscos e das limitações advindos do cultivo exclusivo de abacaxizeiros Pérola e Smooth Cayenne e visando à variabilidade genética, o Estado da Paraíba estuda material genético diversificado: os híbridos Emepa-01, Imperial e MD-2. objetivando a caracterização destes novos híbridos, avaliaram-se alguns parâmetros químicos e a atividade da bromelina nas polpas e nas cascas de todas as cultivares. o abacaxi Smooth Cayenne apresentou o menor pH (3,44 para polpa e 3,91 para a casca) e a maior acidez titulável (0,56g de ácido cítrico para 100g de polpa e 0,52g de ácido cítrico para 100g de casca), sendo a cultivar mais ácida. Dentre as polpas, as dos abacaxis Pérola e Smooth Cayenne contiveram níveis de açúcares redutores, não redutores e totais significativamente maiores (p<0,05) do que os híbridos, apesar de estes terem valores mais elevados (maior que 30,00) da relação sólidos solúveis e acidez titulável e, consequentemente, maior aceitação pelo consumidor. o extrato bruto da cultivar Imperial apresentou maior atividade proteolítica e teor de proteína tanto na polpa (56,41 U/ml e 7,50 mg de proteína/ml) quanto na casca (68,56 U/ml e 10,40 mg de proteína/mL). Para a atividade específica, a casca do abacaxi Pérola (10,01 U/mg de proteína) contém valor significativamente mais elevado que os encontrados nas demais amostras. No entanto, dentre as polpas, a da cultivar Imperial (7,49 U/mg de proteína) apresenta atividade específica significativamente superior (p<0,05) às demais.

O objetivo do trabalho foi avaliar o efeito do ácido giberélico (GA3) e anelamento de ramos sobre os teores de carboidratos nas folhas, a produção e o peso de frutos de limeira ácida 'Tahiti'. O delineamento experimental foi em blocos casualizados, com cinco repetições, em esquema fatorial 4x3. O primeiro fator foi a aplicação de GA3 em quatro concentrações (0; 7; 14 e 21 mg/L-1), e o segundo, duas épocas de anelamento, no início e um mês após o florescimento, mais a testemunha sem anelamento. Verificou-se que o GA3 não teve efeito sobre a produção e o anelamento de ramos, independentemente da época em que foi realizado, aumentaram a produção das plantas e os teores de carboidratos nas folhas. A maior produção e relação frutos/m-3 de copa causou redução no peso médio dos frutos de limeira ácida 'Tahiti'.

RESUMO A podridão floral dos citros (PFC), causada por Colletotrichum acutatum Simmons e C. gloeosporioides, é a doença fúngica mais importante em limeira-ácida ‘Tahiti’, pois, leva à queda prematura de flores e frutos, acarretando a redução da produção. Avaliou-se a suscetibilidade à PFC dos clones de lima-ácida ‘Tahiti’ “IAC 5”, “IAC 5-1”, “CNPMF/EECB”, “CNPMF 2000” e “CNPMF 2001”, em Bebedouro-SP. Todos os clones são suscetíveis à doença. A maior incidência de PFC em plantas do clone “IAC 5-1” indica que este clone apresenta maior suscetibilidade. Mesmo com sintomas, plantas do clone “CNPMF/EECB” apresentam maior fixação de frutos.

OBJETIVO: Avaliar, no coração, por imuno-histoquímica, a localização das proteínas TGFbeta1 latente e TGFbeta1 ativa, se ocorre ativação radioinduzida da proteína TGFbeta1 latente, e a distribuição das fibras colágenas em diversos períodos de tempo após irradiação. MATERIAIS E MÉTODOS: Trinta e dois camundongos isogênicos (C57BL) foram divididos em dois grupos: GI (não irradiado), com 12 animais, e GII (irradiado), com 20 animais. Os animais do GII receberam radiação gama (telecobaltoterapia, 60Co, com rendimento de 0,97 Gy/min., dose única de 7 Gy em corpo inteiro). Os camundongos dos grupos I e II foram sacrificados por estiramento cervical nos períodos de 1, 14, 30 e 90 dias após irradiação. RESULTADOS: Os corações irradiados apresentaram: 1) alterações nucleares e diminuição das estriações das células musculares cardíacas; 2) aumento significante da deposição de fibras colágenas aos 90 dias depois da irradiação; 3) ativação da proteína TGFbeta1 latente em cardiomiócitos e células do conjuntivo depois da irradiação. CONCLUSÃO: Nossos resultados mostram a importância da proteína TGFbeta1 no processo de fibrose cardíaca radioinduzida e sugerem que células do parênquima (cardiomiócitos) e do conjuntivo podem participar deste mecanismo atuando como fontes da proteína TGFbeta1 ativa.

Some aspects of the chemistry involved in the preparation and characterization of functionalized silicon oxide by sol-gel method are considered in this work. The synthesis was performed with different silicon alcoxide precursors and the influence of the acid and basic catalyst was investigated. Characterization was performed by infrared absorption spectroscopy, elemental analysis and 29Si NMR. Infrared data show Si-C and -CH2- vibrational modes at 1250 to 1280 and 2920 to 2940 cm-1, respectively. The elemental analysis confirmed the presence of organic groups in the inorganic silica network. 29Si NMR results show different hydrolisys depending on the acid or base catalysis.

In this work the formation of multilayers composed by carboxymethylcellulose (CMC), chitosane and bovine serum albumin (BSA) was studied by ellipsometry. First, the adsorption behavior of carboxymethylcellulose onto amino-terminated surfaces was investigated as a function of molecular weight and average degree of substitution of CMC. The influence of these parameters on the adsorbed amount of CMC onto amino-terminated substrates was absent. However, the interaction of CMC covered surfaces with chitosane and BSA was favored when the average degree of substitution of CMC was increased. The adsorption of BSA onto the polysaccharide systems was studied as a function of pH. At the isoelectric point of BSA a maximum in the adsorbed amount was found.

A simple device for glass plate drilling, which is an important step in microfabrication procedures, is described. A reservoir of concentrated hydrofluoric acid with a hole in its bottom is affixed against the glass plate. Leakage is prevented by using a neoprene O-ring. A plastic pipet tip inserted in the reservoir, close to the corrosion region, provides forced convection by pressure variations inside it. A device to make 5 simultaneous holes in a plate is also presented. For a 140 µm thickness alkaline glass lamina and 1/8" O-ring, 5 holes are drilled in 20 min.

Acidic hydrolysis of samples is frequent in urinary 2,5-hexanodione determination. This hydrolysis should be performed under proper conditions, in order to avoid interference, such as the presence of 2,5-dimethylfurane and 2-acetylfurane. The results of the present work, as well as data from the literature, show that the use of non-polar or slightly polar chromatographic columns of 30 m length is an essential condition for 2,5-HD determination in an acid hydrolysed urine sample. In the same way the pH should be kept between 0.3 and 0.5. The mean concentrations of 2,5-HD in samples submitted to hydrolysis (0.50 ± 0.28 mg/g of creatinine) was about 3 times higher than those found in the samples without acid hydrolysis (0.19 ± 0.3 mg/g of creatinine).

Plant extracts are usually complex mixtures which contain several molecules of different sizes with varied functional groups. Such extracts are a challenge to the chemist of natural products. Ion exchange chromatography in non-aqueous medium, used for separation of basic or acidic fractions from plant extracts, is an important unit operation in preparative scale separations. Anionic macroporous resin in non-aqueous medium was used with success in this study for separation of the acid fraction of Copaifera multijuga (Copaiba oil), rich in labdanic diterpenes and for the methanolic extract of Croton cajucara (acetyl aleuritoric acid).

A series of bovine serum albumin-immobilized supports have been prepared and used as restricted access media (RAM) columns. Restricted-access supports combine size-exclusion of proteins and other high-molar-mass matrix components with the simultaneous enrichment of low-molar mass analytes. These characteristics were chromatographically evaluated for the columns. The RAM-BSA (Bovine Serum Albumin) columns showed excellent performance for exclusion of human plasma protein with good retention capacity for a series of acidic, basic, and neutral drugs.
Remediação de drenagem ácida de mina usando zeólitas sintetizadas a partir de cinzas leves de carvão

Zeolitic material was synthesized from coal fly ashes (baghouse filter fly ash and cyclone filter fly ash) by hydrothermal alkaline activation. The potential application of the zeolitic product for decontamination of waters from acid mine drainage was evaluated. The results showed that a dose of 30 g L-1 of zeolitic material allowed the water to reach acceptable quality levels after treatment. Both precipitation and cation-exchange processes accounted for the reduction in the pollutant concentration in the treated waters.

A comparison between silica by acid leaching of rice husk (RH) and silica obtained from thermal treatment of rice husk ash (RHA) is presented. The best leaching results were obtained using 10% hydrochloric acid followed by washing with water. The alternative method, calcination of RHA at 700 ºC for 6 h followed by grinding for 80 min, was more effective. Silica obtained from RH was about 97% amorphous, had a 17.37 µm mean particle size, and a specific surface area of 296 m²/g. On the other hand, for silica obtained from RHA the values were about 95% amorphous material 0.68 µm, and 81 m²/g.

NMR is a valuable screening tool for the binding of ligands to proteins providing structural information on both protein and ligands and is thus largely applied to drug-discovery. Among the recent NMR techniques to probe weak binding protein-ligand complexes we have critically evaluated the advantages and disadvantages of STD (Saturation Transfer Difference), WaterLOGSY (Water Ligand Observation with Gradient Spectroscopy), NOE pumping and DOSY-NOESY (Diffusion-Ordered NOESY) using a mixture of BSA (bovine serum albumin) plus salicylic acid, caffeine, citric acid, adipic acid and D-glucose.
Purificação e caracterização de uma gentioexaose obtida de botriosferana por hidrólise ácida parcial

A hexa-oligosaccharide was obtained by partial acid hydrolysis from botryosphaeran, an exopolysaccharide (EPS) β(1→3; 1→6)-D-glucan type, produced by the ascomyceteous fungus Botryosphaeria rhodina. The oligosaccharide was purified by gel filtration and charcoal-Celite column chromatography and the analysis was followed by HPAEC/PAD. The structure was determined by NMR spectroscopy and mass spectrometry, which showed that the oligosaccharide consists of six β-D-glucopiranosyl units O-6 substituted (gentiohexaose).

Acid mine drainage generated from coal mine showed a pH of 3.2, high concentrations of SO4(2-), Al, Fe, Mn, Zn and minor As, Cd, Co, Cr, Cu, Ni and Pb. The major reduction in the concentration occurred for Al, As, Cr, Fe and Pb after the treatment with CaO. The evolution of these acid waters within the tributary stream showed decreasing concentration for all soluble constituents, except Al. This natural attenuation was controlled by pH (6.4 to 10.8) as a result of concurrent mixing with tributary stream and reaction with local bedrock that contains limestone. Aluminum increasing concentration during this evolution seems to be related to an input of Al-enriched waters due to the leaching of silicate minerals in alkaline conditions.
