The authors complement the description of Triatoma matsunoi Fernandez-Loayza, 1989 and present the morphology of the male external genitalia, characterized by the presence of conjuntiva process (PrCj) denticulate, basal plate (Eplb) too long and median process pygophore (PrP) showing the apex streched and the surface rugose and scaled. The specimens examined (holotype [male symbol] and allotype [female symbol]) were deposited in the Collection Herman Lent of Triatomines of the Instituto Oswaldo Cruz, IOC, Rio de Janeiro, Brazil.

The essential oil (EO) of Ocimum gratissimum inhibited Staphylococcus aureus at a concentration of 0.75 mg/ml. The minimal inhibitory concentrations (MICs) for Shigella flexineri, Salmonella enteritidis, Escherichia coli, Klebsiella sp., and Proteus mirabilis were at concentrations ranging from 3 to 12 mg/ml. The endpoint was not reached for Pseudomonas aeruginosa (>=24 mg/ml). The MICs of the reference drugs used in this study were similar to those presented in other reports. The minimum bactericidal concentration of EO was within a twofold dilution of the MIC for this organism. The compound that showed antibacterial activity in the EO of O. gratissimum was identified as eugenol and structural findings were further supported by gas chromatography/mass spectra retention time data. The structure was supported by spectroscopic methods.

Cry11Bb is an insecticidal crystal protein produced by Bacillus thuringiensis subsp. medellin during its stationary phase; this -endotoxin is active against dipteran insects and has great potential for mosquito borne disease control. Here, we report the first theoretical model of the tridimensional structure of a Cry11 toxin. The tridimensional structure of the Cry11Bb toxin was obtained by homology modelling on the structures of the Cry1Aa and Cry3Aa toxins. In this work we give a brief description of our model and hypothesize the residues of the Cry11Bb toxin that could be important in receptor recognition and pore formation. This model will serve as a starting point for the design of mutagenesis experiments aimed to the improvement of toxicity, and to provide a new tool for the elucidation of the mechanism of action of these mosquitocidal proteins.

The seroprevalence of Chlamydia pneumoniae and Mycoplasma pneumoniae in hantavirus seronegative patients, who had symptoms and signs compatible with pneumonia was established. For this purpose we used the indirect fluorescent antibody test. Titers Symbol>³ 1:16 for C. pneumoniae and M. pneumoniae were found in 8.6% and 17.1% of the serum, respectively, showing evidence of recent or current infection.

Biopsies from human localized cutaneous lesions (LCL n = 7) or disseminated lesions (DL n = 8) cases were characterized according to cellular infiltration,frequency of cytokine (IFN-Symbol>g, TNF-alpha) or iNOS enzyme producing cells. LCL, the most usual form of the disease with usually one or two lesions, exhibits extensive tissue damage. DL is a rare form with widespread lesions throughout the body; exhibiting poor parasite containment but less tissue damage. We demonstrated that LCL lesions exhibit higher frequency of B lymphocytes and a higher intensity of IFN-gamma expression. In both forms of the disease CD8+ were found in higher frequency than CD4+ T cells. Frequency of TNF-alpha and iNOS producing cells, as well as the frequency of CD68+ macrophages, did not differ between LCL and DL. Our findings reinforce the link between an efficient control of parasite and tissue damage, implicating higher frequency of IFN-gamma producing cells, as well as its possible counteraction by infiltrated B cells and hence possible humoral immune response in situ.

We have compared the efficacy of two Leishmania (Leishmania) major vaccines, one genetically attenuated (DHFR-TS deficient organisms), the other inactivated [autoclaved promastigotes (ALM) with bacillus Calmete-Guérin (BCG)], in protecting rhesus macaques (Macaca mulatta) against infection with virulent L. (L.) major. Positive antigen-specific recall proliferative response was observed in vaccinees (79% in attenuated parasite-vaccinated monkeys, versus 75% in ALM-plus-BCG-vaccinated animals), although none of these animals exhibited either augmented in vitro gamma interferon (IFN-Symbol>g) production or positive delayed-type hypersensitivity (DTH) response to the leishmanin skin test prior to the challenge. Following challenge, there were significant differences in blastogenic responses (p < 0.05) between attenuated-vaccinated monkeys and naïve controls. In both vaccinated groups very low levels of antibody were found before challenge, which increased after infective challenge. Protective immunity did not follow vaccination, in that monkeys exhibited skin lesion at the site of challenge in all the groups. The most striking result was the lack of pathogenicity of the attenuated parasite, which persisted in infected animals for up to three months, but were incapable of causing disease under the conditions employed. We concluded that both vaccine protocols used in this study are safe in primates, but require further improvement for vaccine application.

Tuberculosis (TB) is a disease caused by Mycobacterium tuberculosis whose interaction with the host may lead to a cell-mediated protective immune response. The presence of interferonSymbol>-g (IFN-gamma) is related to this response. With the purpose of understanding the immunological mechanisms involved in this protection, the lymphoproliferative response, IFNSymbol>-g and other cytokines like interleukin (IL-5, IL-10), and tumor necrosis factor alpha (TNFSymbol>-a) were evaluated before and after the use of anti-TB drugs on 30 patients with active TB disease, 24 healthy household contacts of active TB patients, with positive purified protein derivative (PPD) skin tests (induration > 10 mm), and 34 asymptomatic individuals with negative PPD skin test results (induration < 5 mm). The positive lymphoproliferative response among peripheral blood mononuclear cells of patients showed high levels of IFNSymbol>-g, TNFSymbol>-a, and IL-10. No significant levels of IL-5 were detected. After treatment with rifampicina, isoniazida, and pirazinamida, only the levels of IFNSymbol>-g increased significantly (p < 0.01). These results highlight the need for further evaluation of IFNSymbol>-g production as a healing prognostic of patients treated.

Resistance to infection by Leishmania major has been associated with the development of a Th1 type response that is dependent on the presence of interleukin 12 (IL-12). In this work the involvement of this cytokine in the response to infection by L. braziliensis, a less virulent species in the mouse model, was evaluated. Our results show that while interferon (IFNSymbol>-g) deficient (-/-) mice inoculated L. braziliensis develop severe uncontrolled lesions, chronic lesions that remained under control up to 12 weeks of infection were observed in IL-12p40 -/- mice. IL 12p40 -/- mice had fewer parasites in their lesions than IFNSymbol>-g-/- mice. Lymph node cells from IL-12p40 -/- were capable of producing low but consistent levels of IFNSymbol>-g suggestive of its involvement in parasite control. Furthermore, as opposed to previous reports on L. major-infected animals, no switch to a Th2 response was observed in IL-12p40 -/- infected with L. braziliensis.

Type I diabetes mellitus (insulin-dependent DM = IDDM) is a chronic disease characterized by specific destruction of pancreatic beta cells, resulting in an absolute lack of insulin. Immune mechanisms, genetic susceptibility, and environmental factors are all implicated in the pathogenesis of Type 1 diabetes. This study was aimed at determining the efficiency of cytokines, natural killer (NK) cells in the pathophysiology of IDDM. Therefore, we evaluated the plasma levels of cytokines by specific enzyme-linked immunosorbent assay (ELISA) and the cytotoxicity activity of NK cells by anti-candididal index in rats with type I diabetes. We found that the cytotoxicity activity of NK cells in IDDM groups significantly decreased compared to the control groups. The levels of interferonSymbol>-g (IFNSymbol>-g) in IDDM groups were slightly higher than in healthy controls. These results indicate that the changes of T H1 type cytokines such as IFNSymbol>-g and NK cell activity can play a role in the etiology of IDDM. The data may provide new strategies for the treatment of IDDM.

This work aimed to study the T helper type 1/2 (Th1/Th2) cytokine profile in a co-infection murine model of Plasmodium chabaudi chabaudi and Leishmania infantum. Expression of interferon-gamma (IFNSymbol>-g) and interleukin-4 (IL-4) was analyzed, in spleen and liver of C57BL/6 mice, by reverse transcriptase-polymerase chain reaction. High levels of IFNSymbol>-g expression did not prevent the progression of Leishmania in co-infected mice and Leishmania infection did not interfere with the Th1/Th2 switch necessary for Plasmodium control. The presence of IL-4 at day 28 in co-infected mice, essential for Plasmodium elimination, was probably a key factor on the exacerbation of the Leishmania infection.

Adenosine triphosphate (ATP) is now established as a principle vaso-active mediator in the vasculature. Its actions on arteries are complex, and are mediated by the P2X and P2Y receptor families. It is generally accepted that ATP induces a bi-phasic response in arteries, inducing contraction via the P2X and P2Y receptors on the smooth muscle cells, and vasodilation via the actions of P2Y receptors located on the endothelium. However, a number of recent studies have placed P2X1 receptors on the endothelium of some arteries. The use of a specific P2X1 receptor ligand, Symbol>a, b methylene ATP has demonstrated that P2X1 receptors also have a bi-functional role. The actions of ATP on P2X1 receptors is therefore dependant on its location, inducing contraction when located on the smooth muscle cells, and dilation when expressed on the endothelium, comparable to that of P2Y receptors.

A sand fly fossil was found in amber, a vegetal resin, which allows all the external phlebotomine structures to be seen. The piece that contains the new species is 14 mm long Symbol>´ 8 mm wide Symbol>´ 3 mm high. All the structures from the head, thorax, and abdomen were examined under the microscope and measured with a calibrated micrometric eye-piece. The morphological aspects of the new species suggest its inclusion in the Pintomyia genus, Pifanomyia subgenus though it is not possible to include it in any of the series known for this subgenus. The presence of two atrophied spines on the gonostyles and gonocoxites without tufts of setae permit the exclusion of the new species from the other species of the subgenus Pifanomyia. The new species is named Pintomyia (Pifanomyia) brazilorum sp. nov.

The word tradition has a very specific meaning in linguistics: the passing down of a text, which may have been completed or corrected by different copyists at different times, when the concept of authorship was not the same as it is today. When reading an ancient text the word tradition must be in the reader's mind. To discuss one of the problems an ancient text poses to its modern readers, this work deals with one of the first printed medical texts in Portuguese, the Regimento proueytoso contra ha pestenença, and draws a parallel between it and two related texts, A moche profitable treatise against the pestilence, and the Recopilaçam das cousas que conuem guardar se no modo de preseruar à Cidade de Lixboa E os sãos, & curar os que esteuerem enfermos de Peste. The problems which arise out of the textual structure of those books show how difficult is to establish a tradition of another type, the medical tradition. The linguistic study of the innumerable medieval plague treatises may throw light on the continuities and on the disruptions of the so-called hippocratic-galenical medical tradition.

Os efeitos do tráfego de máquinas nos atributos do solo de acordo com o tempo de adoção do sistema plantio direto são ainda pouco pesquisados em ambientes tropicais, e muitas dúvidas ainda persistem sobre a variação dinâmica da estrutura do solo e a sua interação com máquinas e equipamentos. Objetivou-se com este estudo avaliar o efeito do tempo de adoção do sistema plantio direto, comparativamente com área de mata nativa e de preparo convencional, usando os modelos de compressibilidade do solo. O estudo foi realizado em um Nitossolo Vermelho distroférrico, sob mata nativa (MN), preparo convencional (PC), plantio direto com um ano (PD1), plantio direto com quatro anos (PD4), plantio direto com cinco anos (PD5) e plantio direto com 12 anos (PD12). Amostras indeformadas e deformadas foram coletadas em duas profundidades (0-5 e 10-15 cm). O tempo de adoção do sistema plantio direto alterou o comportamento compressivo dos solos em ambas as profundidades, por meio das mudanças na pressão de preconsolidação. A profundidade de 0-5 cm apresentou menor capacidade de suporte de carga do que a profundidade de 10-15 cm. A profundidade de 0-5 cm, em todos os sistemas de manejo, mostrou-se mais susceptível à compactação em relação à profundidade de 10-15 cm. Os sistemas de plantio direto e convencional apresentaram a capacidade de suporte de carga crescente na seguinte ordem: PD5 < PD12 < PD1 < PD4 Symbol>@ PC, para a profundidade de 0-5 cm e para a profundidade de 10-15 cm: MN Symbol>@ PD12 < PC Symbol>@ PD4 < PD5, enquanto o sistema PD1 apresentou comportamento diferenciado.

No contexto das Ciências Agronômicas e Ambientais, a função condutividade hidráulica K(teta), isto é, a condutividade hidráulica (K) em função da umidade (teta), tem especial importância para os estudos e solução de problemas práticos relacionados, dentre outros, com a irrigação, drenagem e lixiviação de nutrientes e poluentes. Quantificar e caracterizar a variabilidade deste parâmetro hídrico é fundamental para a obtenção de valores médios representativos que possam ser utilizados com segurança nas suas diversas aplicações. Objetivando caracterizar o comportamento estatístico e quantificar a variabilidade dos parâmetros da equação representativa da função K(teta), realizou-se um experimento de campo para determinação desta função pelo método do perfil instantâneo em um Latossolo Vermelho-Amarelo (Typic Hapludox), numa transeção de 50 pontos distanciados entre si de 1 m. As equações obtidas foram do tipo K = Kteta = 0 exp(Symbol>bq) e os resultados indicaram que os padrões de variabilidade dos parâmetros beta e lnKteta=0, bem como de teta, aumentaram em profundidade e não apresentaram relação entre si. Verificou-se também que a presença de valores extremos alterava as medidas estatísticas relativas aos parâmetros da função K(teta), com reflexos significativos quanto ao número de amostras necessárias para estimar o valor médio destes parâmetros, sendo importante sua identificação antes de aplicar determinado método para avaliação da condutividade hidráulica do solo.
